3个山羊群体中4个微卫星DNA多态性及其与杂种优势的关系   总被引:23,自引:0,他引:23  
利用4个微卫星标记(OarFCB11,OarAE101,McM218,McM38)对波尔山羊、太行山羊和河北奶山羊的等位基因频率、群体多态信息含量、有效等位基因数、杂合度和遗传距离进行了遗传检测,并测定了波尔山羊与河北奶山羊及太行山羊的杂交效果。结果表明:4个微卫星标记在波尔山羊、太行山羊和河北奶山羊3个品种中存在多态性,可以用于山羊遗传多样性的评估;从不同品种来看,太行山羊的遗传变异程度最大,而波尔山羊的遗传变异程度相对较小;波尔山羊与河北奶山羊的遗传距离大于与太行山羊,波尔山羊与河北奶山羊的杂种优势高于与太行山羊,与实际杂种优势测定结果相符。 Abstract: Gene frequency, polymorphism information contents, number of effective alleles, heterozygosity and genetic distances were studied in Boer goat, Taihang goat and Hebei dairy goat using four microsatellite markers(OarFCB11,OarAE101,McM218,McM38). The crossing effects on Hebei dairy goat and Taihang goat with Boer goat were tested. The results indicated that there are genetic polymorphisms at four microsatellite markers in three goat breeds. Four microsatellite markers can be used for genetic diversity evaluation in goat breeds. The genetic variability of Taihang goat is the highest, and Boer goat is the lowest in three goat breeds. Genetic distances between Boer goat and Hebei dairy goat is bigger than that between Boer goat and Taihang goat. The heterosis between Boer goat and Hebei dairy goat is higher than that between Boer goat and Taihang goat. It accords with testing results on actual heterosis.  相似文献

本研究以柴达木山羊、柴达木绒山羊和辽宁绒山羊三个群体共147只山羊为材料,利用聚丙烯酰胺凝胶电泳(PAGE)技术检测了5种血液蛋白质(酶)基因座的遗传多态性,并进行了结构基因遗传共适应的研究,结果发现:45个基因座组合中有10个基因座组合处于遗传不平衡状态,并且这些遗传不平衡皆单纯由遗传共适应差异造成;除辽宁绒山羊Tf-PA-3组合的遗传不平衡包含非等位基因间的遗传共适应差异外,其他基因座组合的遗传不平衡皆由等位基因间的共适应差异,即单基因座的遗传不平衡造成;LAP-EsD组合的共适应差异在群体间有遗传传递现象。 Abstract:With the technology of PAGE,the genetic polymorphism of blood protein and enzyme was investigated,and genetic co-adaptability among structural genes was studied in three goat populations(147 goats) including Chaidamu goat(CS),Chaidamu Cashmere goat(CRS) and Liaoning Cashmere goat(LRS) in Qinghai Province,China.The results were showed that the genetic disequilibrium of 10 locus combinations was found among 45 locus combinations in the three goat populations,and these genetic disequilibria were caused only by the difference of genetic co-adaptability among genes,because there didn′t exist the linkage disequilibrium among non-allelic genes.The genetic disequilibrium including the difference of genetic co-adaptability between non-allelic genes was only found at Tf-PA-3 locus combinations in LRS population,the other ones were all caused by the genetic disequilibrium at a single locus.The difference of genetic co-adaptability of LAP-EsD locus combinations could be messaged among different populations.  相似文献

利用RAPD标记和POPGENE、PHYLIP及SPSS等统计分析软件,研究了波尔山羊及其杂交改良后代群体的遗传结构及遗传亲缘关系。结果表明,波尔山羊与其杂交改良后代间特别是高代杂种间具有相似的多态基因座百分率、等位基因数和基因多样性。随着级进杂交改良代数的递增,高代杂种之间以及与改良亲本之间的群体遗传结构的差异逐渐缩小,群体间的基因分化降低,基因流动和遗传相似度增加,遗传距离缩小,高代杂种与波尔山羊间具有更近的遗传亲缘关系。 Abstract:The genetic structure and relationship of Boer goat and its upgrading offspring to Tangshan Diary goat were studied using the RAPD marker and some statistical program,such as POPGENE,PHYLIP and SPSS.The results indicated that there were the similar percentage of polymorphic loci,observed and effective number of alleles,gene diversity between Boer goat and its upgrading offspring,especially higher upgrading offspring.With the increasing of upgrading,the difference of population structure decreased as well as the genetic distance and differentiation among higher upgrading offspring and their improving parental,but gene flow and genetic identity increased.There was a close genetic relationship between higher upgrading offspring and Boer goat.  相似文献

利用随机扩增多态DNA技术研究了波尔山羊、唐山奶山羊、青龙本地山羊以及波尔山羊与这两个山羊群体杂交后代共计128个山羊个体的随机扩增多态DNA。结果表明:(1)总群体平均遗传多样性指数(Hsp)为0.6974,群体遗传分化指数为0.9706,山羊群体间平均遗传距离指数(0.1314~0.2052)明显大于群体内的相应值(0.0582~0.1440),上述结果说明,所研究山羊群体不仅具有较为丰富的遗传多样性,而且其核基因组遗传变异主要存在于群体间。(2)山羊群体间的分子聚类关系与各群体间的亲缘关系基本一致。 Abstract:The random amplified polymorphic DNA(RAPD)of 128 individuals was studied,which were from Boer goat, Tangshan dairy goat,Qinglong native goat and their hybrids crossbred with Boer goat.The average index of genetic polymorphism for whole population(Hsp)and the index of genetic differentiation were 0.6974 and 0.9706,respectively.The average index of genetic distance between populations(0.1314~0.2052)was significantly higher than that within populations(0.0582~0.1440).All of these indicated that the genetic polymorphism was not only abundant,but also the genetic variation was mainly existed between goat populations.The molecular dendrogram among goat populations was in accord with their genetic relationship.  相似文献

我国主要地方绵羊品种随机扩增多态DNA研究   总被引:1,自引:0,他引:1  
对蒙古羊、湖羊、滩羊、小尾寒羊、乌珠穆沁羊、藏绵羊、阿勒泰羊7个地方绵羊品种和无角陶赛特羊、德国美利奴羊、萨福克羊3个引入品种基因组DNA进行了RAPD分析。结果表明:(1)RAPD可作为一种有效的标记用于绵羊品种之间遗传亲缘关系的分析。(2)在所使用的43种随机引物中,有35种引物扩增出多态谱带,多态频率为66.24%,说明RAPD技术用于研究绵羊核DNA的遗传变异具有较高的检出率和灵敏度。(3)总群体平均遗传多样性指数(HSP)为0.9139,说明绵羊群体具有较为丰富的遗传多样性。(4)我国地方绵羊品种间的分子聚类关系与其所处的地理位置、考古学结果,以及细胞遗传学研究结果基本一致,引入品种间的分子聚类关系也与其育成史基本一致。 Abstract:The genetic polymorphism and relationship of 7 indigenous sheep breeds of China and 3 imported sheep breeds were studied using random amplified polymorphic DNA (RAPD).The results indicated that the RAPD was an effective marker for the analysis of genetic relationship among sheep breeds.Among 43 arbitrary primers,35 were polymorphic.The percentage of polymorphic markers was 66.24%,which indicated that the RAPD had higher efficiency of polymorphism detection and sensitivity in studying the genetic variation among sheep breeds.The average index of genetic polymorphism for whole population (Hsp) was 0.9139,which showed that the genetic polymorphism was abundant between sheep populations.The genetic relationship between different indigenous sheep breeds in China was in accord with their localities,the results from archeology and cytogenetics and the genetic relationship between imported sheep breeds was in accord with their breeding history.  相似文献

Genetic diversity and phylogenetic relationships among 568 individuals of two red jungle fowl subspe- cies (Gallus gallus spadiceus in China and Gallus gallus gallus in Thailand) and 14 Chinese domestic chicken breeds were evaluated with 29 microstaellite loci, the genetic variability within population and genetic differentiation among population were estimated, and then genetic diversity and phylogenetic relationships were analyzed among red jungle fowls and Chinese domestic fowls. A total of 286 alleles were detected in 16 population with 29 microsatellite markers and the average number of the alleles observed in 29 microsatellite loci was 9.86±6.36. The overall expected heterozygosity of all population was 0.6708±0.0251, and the number of population deviated from Hardy-Weinberg equilibrium per locus ranged from 0 to 7. In the whole population, the average of genetic differentiation among population, measured as FST value, was 16.7% (P<0.001), and all loci contributed significantly (P<0.001) to this differentiation. It can also be seen that the deficit of heterozygotes was very high (0.015) (P<0.01). Reynolds' distance values varied between 0.036 (Xiaoshan chicken-Luyuan chicken pair) and 0.330 (G. gallus gallus-Gushi chicken pair). The Nm value ranged from 0.533 (between G. gallus gallus and Gushi chicken) to 5.833 (between Xiaoshan chicken and Luyuan chicken). An unrooted consensus tree was constructed using the neighbour-joining method and the Reynolds' genetic distance. The heavy-body sized chicken breeds, Luyuan chicken, Xiaoshan chicken, Beijing Fatty chicken, Henan Game chicken, Huainan Partridge and Langshan chicken formed one branch, and it had a close genetic relationship between Xiaoshan chicken-Luyuan chicken pair and Chahua chicken-Tibetan chicken pair. Chahua chicken and Tibetan chicken had closer genetic relationship with these two subspecies of red jungle fowl than other domestic chicken breeds. G. gallus spadiceus showed closer phylogenetic relationship with Chinese domestic chicken breeds than G. gallus gallus. All 29 microstaellite loci in this study showed high levels of polymorphism and significant genetic differentiation was observed among two subspecies of red jungle fowl and 14 Chinese domestic chicken breeds. The evolutional dendrogram is as follows: evolutional breeds→primitive breeds (Chahua chicken and Tibetan)→red jungle fowl in China (G. gallus spadiceus)→red jungle fowl in Thailand (G. gallus gallus). The results supported the theory that the domestic fowls might originate from different subspecies of red jungle fowl and Chinese domestic fowls had independent origin.  相似文献

采用简单随机抽样法对柴达木山羊群体进行了遗传检测,从毛色、外型特征、体尺、血液蛋白与DNA多态性等5个方面进行了群体遗传结构分析,并探索了其系统地位。结果表明:(1)毛色和形态特征的平均表型异质度分别为0.3419、0.5207;(2)血液蛋白在6个基因座上具有多态性,平均基因座杂合度为0.2584;(3)DNA-RAPD标记表现丰富的多样性,基因多样度为0.4085~0.5318。通过对柴达木山羊与其他国内山羊的系统聚类,初步探索了该山羊群体的形成及所属,这些都表明柴达木山羊是一个比较古老的未经较高强度选育的地方山羊品种,其基因资源是我国山羊遗传资源中宝贵的一部分。Abstract: Genetic structure and character of Chaidamu Goats were studied through simple random sampling. Genetic structure was analysed from five aspects, and phylogeny status was also investigated. The results indicated that: (1) the average phenotypic heterogeneity degree of coat color and morphological character were 0.3419 and 0.5207,respectively; (2) polymorphous blood albumen existed in 6 loci and the average loci heterozygosity was 0.2584; and (3) polymorphism existed in marked genes by DND-RAPD with diversity of 0.4085~0.5318. Phylogeny status was investigated through clustering by Ward’s method on Chaidamu Goats and other domestic goats. All these indicated that Chaidamu Goats was a domestic goat with less intensively selected breed.  相似文献

In order to verify the effect of social behavior and geographical isolation on the genetic structure of the Himalayan marmot (Marmota himalayana) population, we examined the genetic diversity of Himalayan marmots alongside the Qinghai–Tibet Railway using microsatellite markers. Eight microsatellite loci were used to examine 120 animals of 4 populations: Ulan (U), Delhi (D), Tuotuohe (T) and Ando (A). The results show that: (1) Himalayan marmots alongside the Qinghai–Tibet Railway are highly genetically diversified. The allele number (Na), effective allele number (Ne), observed heterozygosity (Ho), Nei’s expected heterozygosity (He) and polymorphism information content (PIC) of the total Himalayan marmot population were 4.75, 3.0332, 0.6990, 0.6672, 0.6102, respectively. (2) Himalayan marmots may be able to avoid inbreeding by a mechanism that will prevent the genetic diversity reduction caused by their social lifestyle. Heterozygote excess was observed at most loci. The inbreeding coefficients within the subpopulation (FIS), in the total population (FIT), the differentiation index of population (FST), and the gene flow (Nm) were ?0.2265, ?0.0477, 0.1458, and 1.4646, respectively. (3) The genetic differentiation of the Himalayan marmot population was in accordance with Wright’s “isolation by distance” theory. The Mantel test indicates that the correlation between genetic distance and geographic distance was significant (P < 0.05, r = 0.698). (4) Each of the four geographical populations had moderate differentiation. Both geographic distance and isolation could affect the population genetic structure of the Himalayan marmot. The maximum gene flow (3.5915), the smallest genetic differentiation index (0.0651), the lowest genetic distance (0.0700) and the highest genetic identity (0.9526) were all between the Ulan population and Delhi populations. (5) The cluster analysis, based on Nei’s standard genetic distance, showed that the populations of Delhi and Ulan were first merged in a cluster, and then Tuotuohe population was merged in the clustering. 分析了4个微卫星基因座BM143、OarHH35、OarAE101、BMS2508在7个绵羊群体(小尾寒羊、湖羊、乌珠穆沁羊、萨福克羊、多赛特羊、夏洛来羊、多赛特公羊×小尾寒羊母羊F1代杂种羊)286只绵羊中的遗传多态性。结果表明,这4个微卫星标记在7个绵羊群体中的等位基因数分别为9、11、14和9,其多态信息含量/有效等位基因数/杂合度分别为0.7073/3.7231/0.7314、0.8267/6.4399/0.8447、0.5743/2.5178/0.6028、0.6172/3.0712/0.6744,其中OarHH35的遗传变异最大,OarAE101最小。7个绵羊群体中小尾寒羊的遗传变异最大,湖羊的最小。基于Nei氏DA距离和DS标准遗传距离,采用UPGMA方法构建了系统发生树。该发生树将中国地方品种(小尾寒羊、乌珠穆沁羊、湖羊)和法国的夏洛来羊归为一类,将F1杂种羊、英国品种(萨福克羊和多赛特羊)归为另一类。绵羊微卫星基因分型技术为检查品种(群体)之间的遗传关系提供了一个有用的工具。 Abstract:The genetic polymorphisms of four microsatellite loci BM143,OarHH35,OarAE101,and BMS2508 were analyzed in 286 sheep of seven sheep populations (Small Tail Han sheep, Hu sheep, Ujumqin sheep, Suffolk sheep, Dorset sheep, Charolais sheep, F1 of Dorset♂ × Small Tail Han sheep♀). The numbers of alleles for BM143,OarHH35,OarAE101,and BMS2508 are 9, 11, 14 and 9 in seven sheep populations, respectively. The polymorphism information content/number of effective alleles/ heterozygosity of BM143,OarHH35,OarAE101 and BMS2508 were 0.7073/3.7231/0.7314, 0.8267/6.4399/0.8447,0.5743/2.5178/0.6028,0.6172/3.0712/0.6744 in 286 sheep, respectively. The results revealed the greatest genetic variation at OarHH35 locus and the lowest at OarAE101, the greatest genetic variation in Small Tail Han sheep and the lowest in Hu sheep among seven sheep populations. In the unweighted pair group method with arithmetic mean (UPGMA) dendrograms based on Nei's DA distance and Nei's DS standard genetic distance, the Chinese native breeds (Small Tail Han sheep, Ujumqin sheep, Hu sheep) were grouped together, then with Charolais sheep. The F1 crossbred sheep, and the two British native sheep (Suffolk sheep, Dorset sheep) also clustered together. Microsatellite genotyping in sheep provided a useful tool for examining the genetic relationships among breeds(populations).  相似文献

用40条多态引物对乌羊、小香羊、南江黄羊、黑山羊、川东白山羊、波尔山羊和马头山羊7个品种(或群体)进行RAPD分析,其中28条引物扩增出多态性普带,并用于进一步对12只乌羊个体和12只小香羊个体基因组进行扩增。扩增产物以1.5%琼脂糖凝胶(含0.5μg/ml溴化乙锭)电泳分离。Nei氏计算品种间的遗传距离指数和品种内的遗传相似指数。NJ法构建系统聚类图。结果表明:乌羊和川东白山羊间的遗传距离最小,亲缘关系较近,而小香羊与品种间的遗传距离都较大,亲缘关系较远。乌羊群体及小香羊品种都具有一定的遗传稳定性。  相似文献

A progressive paresis was encountered in herds of Swedish goats. The symptoms developed during a period of weeks or months, and were initially often seen as a weakness of the hind limbs before the animals became paralytic. The development and the histopathological lesions of the disease in the GNS and the lungs were similar to those of visna in sheep. In vitro grown choroid plexus cells, prepared from affected goats, showed foci of polykaryocytes. Electron microscopy revealed the presence of particles morphologically similar to those of sheep visna virus (SVV). Goats experimentally infected with the goat visna virus (GVV) developed GNS lesions similar to those of visna in sheep and became seropositive to SVV. The results of complement fixation tests, carried out on sera from 11 goat herds, showed a coincidence between seropositiveness and the occurrence of disease in one and the same herd. Using the ELISA method, an average of 80 % of the goats in 5 herds were found to be seropositive to GVV.  相似文献

Abstract: I investigated seasonal altitudinal movements of 42 mountain goats (Oreamnos americanus) in the Cascade Range of Washington, USA. Because mountain goats typically move to lower elevations during the winter, I partitioned locations from Global Positioning System collars into summer and winter seasons based on elevation. Using an iterative narrowing search, I identified summer and winter start dates for each individual and year and derived several measures of altitudinal movements from these, and examined differences in these measures on the basis of sex and year and their interrelationship. Generally, female mountain goats started summer about 2 weeks earlier than nondispersing males; winter start dates varied among years. Horizontal distance moved between seasons was unrelated to measures of altitudinal movement. Based on elevation, winters were generally longer than summers for mountain goats I studied, suggesting that the common perception of mountain goats as inhabitants of alpine and subalpine terrain is biased, because they spent the greater part of the year at lower elevations. Mountain goats showed a wide range of responses to seasonal environmental changes and individuals cannot be easily classified as migratory or nonmigratory. Because ecological conditions in mountain environments are closely related to elevation and horizontal and altitudinal movements were unrelated, studies of seasonal movements of mountain animals based on horizontal movement may be misleading. Because seasonal altitudinal movements of mountain goats are highly variable, the management needs of each population must be considered separately.  相似文献

ABSTRACT Unbiased estimates of mountain goat (Oreamnos americanus) populations are key to meeting diverse harvest management and conservation objectives. We developed logistic regression models of factors influencing sightability of mountain goat groups during helicopter surveys throughout the Cascades and Olympic Ranges in western Washington during summers, 2004–2007. We conducted 205 trials of the ability of aerial survey crews to detect groups of mountain goats whose presence was known based on simultaneous direct observation from the ground (n = 84), Global Positioning System (GPS) telemetry (n = 115), or both (n = 6). Aerial survey crews detected 77% and 79% of all groups known to be present based on ground observers and GPS collars, respectively. The best models indicated that sightability of mountain goat groups was a function of the number of mountain goats in a group, presence of terrain obstruction, and extent of overstory vegetation. Aerial counts of mountain goats within groups did not differ greatly from known group sizes, indicating that under-counting bias within detected groups of mountain goats was small. We applied Horvitz-Thompson-like sightability adjustments to 1,139 groups of mountain goats observed in the Cascade and Olympic ranges, Washington, USA, from 2004 to 2007. Estimated mean sightability of individual animals was 85% but ranged 0.75–0.91 in areas with low and high sightability, respectively. Simulations of mountain goat surveys indicated that precision of population estimates adjusted for sightability biases increased with population size and number of replicate surveys, providing general guidance for the design of future surveys. Because survey conditions, group sizes, and habitat occupied by goats vary among surveys, we recommend using sightability correction methods to decrease bias in population estimates from aerial surveys of mountain goats.  相似文献

In isolated bundles of external intercostal muscle from normal goats and goats with hereditary myotonia the following were determined: concentrations and unidirectional fluxes of Na+, K+, and Cl-, extracellular volume, water content, fiber geometry, and core-conductor constants. No significant difference between the two groups of preparations was found with respect to distribution of fiber size, intracellular concentrations of Na+ or Cl-, fiber water, resting membrane potential, or overshoot of action potential. The intracellular Cl- concentration in both groups of preparations was 4 to 7 times that expected if Cl- were distributed passively between intracellular and extracellular water. The membrane permeability to K (PK) calculated from efflux data was (a) at 38°C, 0.365 x 10-6 cm sec-1 for normal and 0.492 x 10-6 for myotonic muscle, and (b) at 25°C, 0.219 x 10-6 for normal and 0.199 x 10-6 for myotonic muscle. From Cl- washout curves of normal muscle usually only three exponential functions could be extracted, but in every experiment with myotonic muscle there was an additional, intermediate component. From these data PPcl could be calculated; it was 0.413 x 10-6 cm sec-1 for myotonic fibers and was 0.815 x 10-6 cm sec-1 for normal fibers. The resting membrane resistance of myotonic fibers was 4 to 6 times greater than that of normal fibers.  相似文献

Viable Transgenic Goats Derived from Skin Cells   总被引:3,自引:0,他引:3  
The current study was undertaken to evaluate the possibility of expanding transgenic goat herds by means of somatic cell nuclear transfer (NT) using transgenic goat cells as nucleus donors. Skin cells from adult, transgenic goats were first synchronized at quiescent stage (G0) by serum starvation and then induced to exit G0 and proceed into G1. Oocytes collected from superovulated donors were enucleated, karyoplast-cytoplast couplets were constructed, and then fused and activated simultaneously by a single electrical pulse. Fused couplets were either co-cultured with oviductal cells in TCM-199 medium (in vitro culture) or transferred to intermediate recipient goat oviducts (in vivo culture) until final transfer. The resulting morulae and blastocysts were transferred to the final recipients. Pregnancies were confirmed by ultrasonography 25-30 days after embryo transfer. In vitro cultured NT embryos developed to morulae and blastocyst stages but did not produce any pregnancies while 30% (6/20) of the in vivo derived morulae and blastocysts produced pregnancies. Two of these pregnancies were resorbed early in gestation. Of the four recipients that maintained pregnancies to term, two delivered dead fetuses 2-3 days after their due dates, and two recipients gave birth to healthy kids at term. Fluorescence in situ hybridization (FISH) analysis confirmed that both kids were transgenic and had integration sites consistent with those observed in the adult cell line.  相似文献

Adult animals from 2 herds were examined clinically and serologically, 5 (Herd A) and 4 (Herd B) times during the same period of 3% years. Serum samples were examined for antibodies against Gorynebac-terium pseudotuberculosis using the bacterial agglutination test (BAT) and the hemolysis inhibition test (HIT). The results of the first examination showed that no animal in Herd A was seropositive, while in Herd B 1 animal showed a high positive titre in BAT. The prevalence of animals with superficial swellings was then 2 % in Herd A and 4 % in Herd B. In both herds, the prevalence of animals with superficial swellings and seropositive reactions increased during the following 1–2 yeairs. About 30 % of animals had superficial lesions and close to 100 % were seropositive. The proportion, of animals with superficial swellings and seropositive reactions was almost constant on subsequent examinations. In some of the animals, superficial swellings were found during 2 or more of the examinations, a few animals having such lesions at the same site on both or all occasions. Animals in Herd A became infected through grazing together with goats from infected herds. Caseous lymphadenitis was introduced into Herd B by animals obtained from infected herds.  相似文献

Six castrated male kids from 2 herds free from caseous lymphadenitis, were inoculated subcutaneously with about 1 million colony forming units of a Corynebacterium pseudotuberculosis strain isolated from goat. The animals exhibited a febrile response and marked inflammatory reaction at the inoculation site during the first days after infection. Post mortem examination carried out 2 months after inoculation revealed abscesses in the regional lymph node (Inn. subiliaci) of all animals, in 3 of which lesions also appeared in other lymph nodes. The antibody titre (mean value) in the bacterial agglutination test peaked during the first 2 weeks following inoculation, while the titre in the hemolysis inhibition test began to rise from 2–3 weeks after the infection and continued to increase during the rest of the investigation period (9 weeks after inoculation). Hemoglobin and hematocrit levels decreased significantly after inoculation, and remained below the preinfection levels for 2 weeks. A significant leucocytosis was also seen during the same period. The present investigation indicates that subcutaneous inoculation with C. pseudotuberculosis might be a suitable challenge system to study the efficacy of vaccines against caseous lymphadenitis in goats.  相似文献

用10对山羊微卫星DNA多态性引物对2只体细胞克隆济宁青山羊、青山羊供体细胞、受体奶山羊母羊以及具有亲缘关系的3只对照济宁青山羊进行微卫星DNA分析.结果表明有5对山羊微卫星DNA多态性引物,即SR-CRSP1, SR-CRSP5, SR-CRSP6, SR-CRSP7和SR-CRSP24,扩增产物有明显的多态性.扩增产物经过6%变性聚丙烯酰胺凝胶电泳后银染,结果2只体细胞克隆山羊的微卫星DNA指纹与供体细胞完全相同,而且不同于其受体母亲也不同于其他所有同品种不同个体的对照青山羊.证明体细胞克隆山羊基因组来源于供体细胞.  相似文献

In 2 goat herds, one infected with Gorynebacterium pseudo-tuberculosis and one free from the infection,, goats were examined for superficial swellings on the shoulder and chest. All animals in this study had been vaccinated against paratuberculosis before the age of 4 weeks. The vaccine had been applied subcutaneously behind the shoulder. Twenty-two of 40 (55 %) and 31 of 45 (69 %) goats had such lesions in the infected and non-infected herds, respectively. The difference between the herds was not significant, P > 0.05. Swellings found behind the shoulder in 19 goat carcasses derived from 4 herds in which G. pseudotuberculosis infection occurred were examined bacteriologically. No bacteria could be isolated from such lesions in 15 animals, while G. pseudotuberculosis in pure culture was isolated from 3 carcasses, and a mixed bacterial flora from the re-maining carcass. Bacteria could not be isolated from lesions situated behind the shoulder in 7 carcasses from 3 herds free from G. pseudo-tuberculosis infection. It is concluded that most swellings on the shoulder and chest in goats were granulomas resulting from vaccination against paratuber-culosis.  相似文献
