缙云山特有植物缙云黄芩的遗传多样性研究   总被引:6,自引:3,他引:3  
采用聚丙烯酰胺凝胶垂直平板电泳检测了重庆缙云山特有植物缙云黄芩7个居群70个个体的过氧化物酶、细胞色素氧化酶、超氧化物歧化酶、淀粉酶和酯酶5种等位酶,获得20个基因位点的资料,由此分析了其遗传分化水平。结果表明:缙云黄芩遗传多样性水平较高,多态位点比率P为45.7%,平均每个多态位点的等位基因数目A为1.46,平均预期杂合度He为0.205,平均观察杂合度Ho为0.352。缙云黄芩居群间遗传分化水平较高,Gst=0.401,居群间遗传一致度和遗传距离的平均值分别为0.741和0.300。缙云黄芩遗传多样性的40.1%来源于居群之间的基因差异,59.9%属于居群内的遗传分化,因而对缙云黄芩遗传多样性的保护,应保护其较多的居群。  相似文献

采用RAPD和PCR—RFLP技术,分析了长江中游两个中国胭脂鱼群体的遗传结构。50个随机引物进行RAPD分析,有3个引物显示了多态,宜昌、金口群体内个体之间的遗传相似度分别为0.9274、0.9313,群体之间遗传相似度为0.9000。12个限制性内切酶分析了两群体线粒体DNAND-5/6基因的限制性片段长度多态性,仅内切酶Ncil的酶切图谱显示了多态,基因型间的核苷酸序列歧化距离为0.235%,核苷酸多样性为0.004。分析表明,长江中游两个中国胭脂鱼群体遗传结构较为单一,群体之间表现了较为明显的遗传分化。  相似文献

目的:比较两个封闭群SPF级昆明( KM)小鼠的遗传差异,调查引进的SPF级KM小鼠封闭繁殖6年后,其遗传构成是否发生变化。方法应用微卫星DNA标记方法对18个位点在两个群体中的遗传差异进行分析,主要包括观察等位基因数( Na)、有效等位基因数( Ne)、观察杂合度( Ho)、期望杂合度( He)、多态信息含量(PIC )、Shan-non信息指数、遗传分化系数( F st)、遗传距离等遗传参数。结果两个封闭群SPF 级KM小鼠在18个微卫星位点共发现67个等位基因,Na为2~8个,平均3.7222个;Ne为1.9459~6.5442,平均2.7966个;Ho为0.4225~1.0000,平均0.8823;He为0.4892~0.8527,平均0.6162;Shannon信息指数0.6792~1.9526,平均1.0598;PIC为0.3680~0.8301,平均0.5317;Fst平均值为0.0159,表明群体间的遗传差异仅1.59%,二者间的遗传距离(DA)为0.0499。结论两个封闭群SPF级KM小鼠遗传结构相似度极高,它们与原引进群体的分化差异极小。  相似文献

采用垂直板型聚丙烯酰胺凝胶电泳测定了海南粗榧5个种群遗传多样性和遗分化程度。结果表明:海南粗榧种群遗传多样性水平较低,多态位点比率P=0.33,等位基因平均数A=1.33,平均期望杂合度为He=0.135,观察杂合度为Ho=0139。种群间遗传分化程度较低,基因分化系数Gst=0.123,种群间遗传一致度和遗传距离的均值分别为0.9719、9.0288。黎母山种群与其它种群分化最大,其原因可能与传粉时盛行的风向有关。  相似文献

利用超薄平板聚丙烯酰胺等电聚焦技术对陕西板栗4个实生居群,8个酶系统的20个位点进行了遗传多样性及遗传结构分析,结果表明:4个居群中以宝鸡居群遗传多样性最高,其P为80.0%,Ho,He分别为0.478和0.343,4个居群的居群遗传分化度Gst为6.2%,平均总遗传多样性为0.3372,总遗传多样性的93.8%属于居群内的遗传变异,聚类分析结果表明,以秦岭为界可以分为秦岭以南与秦岭以北2个略有差异的板栗自然亚分布区。  相似文献

中国主要家鹅品种的遗传分化研究   总被引:14,自引:0,他引:14  
利用PCR和DNA测序技术扩增了15个中国家鹅品种线粒体DNA控制区部分序列(1042bp)。研究结果表明:伊犁鹅与14个品种间的核苷酸分歧度最高,为3.805%~4.067%;不同品种内核苷酸多样度表现出较大的差异,为0~0.116%。除伊犁鹅外的14个家鹅品种中,豁眼鹅与其他品种间的核苷酸分歧度为0.211%~0.272%,明显高于其他品种间的0~0.094%。中国家鹅品种的遗传分化格局与地理分布有关,豁眼鹅的分歧时间较早,遗传漂变是导致豁眼鹅遗传分化的主要因素(Nm=0.02~0.54),基因流则是另外13个家鹅品种间遗传分化不明显的主要因素(Nm=12.0~65.33).  相似文献

目的检测国内Et本大耳白兔、青紫蓝兔、新西兰白兔的遗传背景及遗传结构,为封闭群兔遗传检测方法建立和标准化提供基础资料。方法应用18个微卫星标记及荧光标记一半自动基因分型技术对三个群体95个个体进行Hardy.Weinberg检测,统计每个位点等位基因频率、杂合度、F值、遗传距离等信息。结果三个种群平均等位基因观测数为3.167、4.556、3.444,平均观测杂合度为0.444、0.5230、0.4976。18个位点平均多态信息含量(PIC)为0.410、0.549、0.470,日本大耳白兔6个位点HWE检验(P〈0.05),显著偏离Hardy—Weinberg平衡,并在Satl3,INRCCDDV0088位点基因型完全纯和,新西兰白兔和青紫蓝兔分别有2个位点显著偏离Hardy—Weinberg平衡。三个种群遗传距离:青紫蓝兔与新西兰白兔遗传距离最近,为0.124,与日本大耳白兔遗传关系最远,为0.320;新西兰白兔与日本大耳白兔较远,为0:10。结论三个种群有各自不同遗传特征,遗传多样性较高,种群间分化明显。个别位点偏离遗传平衡,推测人工繁育过程中存在一定问题。  相似文献

选用20个微卫星标记分析了2001年和2002年引进的H高产蛋鸡配套系祖代各品系之间的遗传关系,计算杂合度、多态信息含量(PIC)、有效等位基因数、Nei氏遗传距离等统计量,分析了品系内和品系间的遗传变异,并采用非加权类平均法(UPGMA)对各品系进行聚类。结果显示:每个微卫星位点的平均等位基因数为3.250,平均有效等位基因数为2.395,20个微卫星标记共有等位基因65个,各位点的PIC在0.102—0.729之间变动,平均为0.454;各位点杂合度的变动范围是0.108—0.765,各品系杂合度的分布范围是从A2001的0.390到D2001的0.452之间,各品系的杂合度较低,群体内变异较小。配套系各品系是经过高度选育的,所以群体内变异较小,本研究结果与育种实际相符合,也从另一方面说明了品系的遗传特征能够由各微卫星位点的多态性得到真实的反映。A、B两系之间的遗传距离为0.005—0.016,遗传相似系数大于0.984;C、D两系之间的遗传距离为0.094~0.119,遗传相似系数为0.900左右。这些结果表明,A、B应为同一个品系或遗传关系非常近的两个品系,C、D是两个遗传关系较远的品系。  相似文献

采用AFLP技术对我国具有代表性的7个省市的10个野桑蚕(Bombyx mandarina)种群和2个家蚕(Bombyx mori)品种的遗传多样性进行了研究。结果表明:杭州、陕西和重庆3个地区的野桑蚕种群间及种群内个体间的遗传距离都比家蚕品种大。野桑蚕存在广泛的变异,7个省市的10个野桑蚕种群之间的遗传距离为0.164-0.444(平均值为0.3826),平均杂合度为0.7061,表明野桑蚕自然群体的遗传多样性十分丰富。  相似文献

利用RAPD标记技术检测了厦门和汕头沿海2个短沟对虾群体基因组DNA的多态性,并对其遗传多样性进行了分析。从40条随机引物中筛选出13个10bp引物。共扩增出65条清晰可重复的DNA片段,片断长度为100—2200bp,在2个群体间没有检测到特异的片段。厦门和汕头群体的多态片段比例分别为87.69%和89.23%,杂合度分别为0.212和0.218,遗传多样性指数分别为0.2847和0.2913,两群体间的遗传距离为0.018,FST值为0.004。可见两野生群体种质资源仍然维持在良好水平,遗传分化程度很低,可能是同一种群,具有进一步开发的潜力。  相似文献

采用水平切片淀粉凝胶电泳测定了东北地区分布的不同生态环境条件下7个三裂叶豚草种群的遗传结构。统计分析了10个酶系统的10个位点,结果表明:三裂叶豚草种群内存在着丰富的遗传变异。多态位点百分率为80.00%,等位基因平均数为1.80,期望杂合度为0.375,遗传一致度和遗传距离为0.959和0.042。  相似文献

调查陕西渭南地区汉族群体17个Y-STR基因座的多态性,探讨其群体遗传学及法医学应用价值。应用Y-fi ler荧光标记复合扩增系统,对413名陕西渭南地区汉族无关男性个体17个Y-STR基因座进行复合扩增,用ABI3130遗传分析仪进行基因分型,计算各基因座的群体遗传学参数,并结合已经发表的其他10个群体相应基因座的单倍型资料,分析各群体间的遗传距离。413名陕西渭南汉族个体共检出405种单倍型,其中397种单倍型仅出现1次,单倍型多样性达0.9999,基因多样性(GD)为0.4130(DYS391)~0.9734(DYS385a/b),累计GD值为0.9999。遗传距离分析提示,陕西渭南汉族与辽宁满族的遗传距离最小(0.00110),与青海藏族的遗传距离最大(0.22333)。结果表明,17个Y-STR基因座在陕西渭南汉族群体中具有丰富的遗传多态性和较高的非父排除能力,在法医学和人类群体遗传学研究中具有重要价值。  相似文献

光叶珙桐的等位酶分析及其生物地理学意义   总被引:12,自引:0,他引:12  
采用水平切片淀粉凝胶电泳实验方法,对光叶珙桐(Davidia involucrata var.vilmoriniana)4个居群、78个个体、11种酶系统,21个位点进行了等位酶分析。各居群的多态位点百分比P=28.6%-47.6%,实际杂合度Ho=0.177—0.308,平均每个位点的等位基因数A=1.3-1.6。在变种水平上,多态位点百分比P=52.4%,平均每个位点的等位基因数A=1.8,实际杂合度Ho=0.272,期望杂合度Ho=0.216。基因分化系数FST=0.1928,说明光叶珙桐居群问分化小。各项指标表明光叶珙桐的遗传多样性水平较高,比原变种珙桐高,外部环境可能是影响珙桐分布格局的重要因素。昭通与宝兴的遗传多样性明显高于其他两个居群,遗传多样性保存较为完整,而处于分布区东、西两侧边缘(云南西北部和湖北五峰)遗传多样性则相对较低,推测四川盆地边缘山地是该种的遗传多样化中心,可能是在地质灾难中(如第四纪冰期),光叶珙桐真正的避难所。分布区东西两侧的居群可能是从四川盆地边缘山地扩散而来。  相似文献

研究了AFLP标记在研究小香羊遗传多态性方面应用的可行性和该山羊个体基因组DNA的AFLP扩增结果。实验应用10条AFLP引物,用PstI酶切,对15只小香羊基因组DNA进行AFLP反应,共获得113个AFLP标记,单引物获得的标记数在2~19之间,小香羊群体相似系数AFLP研究结果为0.913(0.814~0.980)。该研究为评价小香羊的遗传稳定性提供了相关的参数,准确评价尚待和其它品种对比研究后确定。  相似文献

采用等位酶电泳技术了云南省中北部昆明、富民、宾川3县(市)及四川省西南布拖县的硬枝野荞麦(Fagopyrum urophyllum)6个天然居群的遗传多样性和分化。硬枝野荞麦居群内维持有较高的遗传多样性,多态位点比率为50.0%,预期杂合度和观察杂合度分别为0.251和0.471。并对硬枝野荞麦(F.urophyllum)与栽培荞麦之间遗传变异作了比较。  相似文献

Summary Allozyme variation in the tetraploid wild emmer wheat, Triticum dicoccoides, the progenitor of all cultivated wheats, was studied for the proteins encoded by 42 gene loci in 1815 plants representing 37 populations - 33 from Israel and 4 from Turkey - sampled in 33 localities from 1979 to 1987. The results showed that: (a) 6 loci (14%) were monomorphic in all populations, 15 loci (36%) were locally polymorphic, and 21 loci (50%) were regionally polymorphic. These results are similar to those obtained earlier on 12 Israeli populations. All polymorphic loci (except 4) displayed high local levels of polymorphism (>/ 10%). (b) The mean number of alleles per locus, A, was 1.252 (range: 1.050–1.634); the proportion of polymorphic loci per population averaged 0.220 (range: 0.050–0.415); genic diversity, He, averaged 0.059 (range: 0.002–0.119). (c) Altogether there were 119 alleles at the 42 putative loci tested, 114 of these in Israel, (d) Genetic differentiation was primarily regional and local, not clinal; 70% of the variant alleles were common (>/ 10%) and not widespread, but rather localized or sporadic, displaying an archipelago population genetics and ecology structure. The coefficients of genetic distance between populations were high and averaged D = 0.134; range: 0.018–0.297, an indication of sharp genetic differentiation over short distances, (e) Discriminant analyses differentiated Israeli from Turkish populations, and within Israel, between central and 3 marginal regions, as well as between different soil-type populations, (f) Allozymic variation comprised 40% within and 60% between populations, (g) Gametic phase disequilibria were abundant, their number being positively correlated (rs = 0.60, P<0.01) with the humidity, (h) Multilocus organization was substantive, also positively correlated with humidity, (i) Allozyme diversity, overall and at single loci, was significantly correlated with, and partly predictable by, climatic and edaphic factors, (j) The distrubition of the significant positive and negative values and the absence of autocorrelations in the correlogram revealed no similar geographic patterns across loci, eliminating migration as a prime factor of population genetic differentiation. These results suggest: (I) during the evolutionary history of wild emmer, diversifying natural selection, through climatic and edaphic factors, was a major agent of genetic structure and differentiation at both the single and multilocus levels; (II) wild emmer harbors large amounts of genetic diversity exploitable as genetic markers in sampling and abundant genetic resources utilizable for wheat improvement.  相似文献

采用聚丙烯酰胺凝胶电泳技术,对福清黑松群体进行醇溶蛋白的多样性分析,共获得40种图谱,9个等位基因位点。对这些位点的统计分析表明,福清黑松的多肽位点百分率P=55.56%, 等位基因平均数A=3.00,平均等位基因有效数目Ae=2.28,预期杂合度He=0.533,平均实际杂合度Ho=0.402,固定指数F仅为0.246>0。该群体表现出遗传多样性水平较高,但该群体偏离了Hardy-Weinberg平衡定律,原因可能是取样偏差、群体中个体密度分布不均和个体生长状况不好,不能产生充分的随机交配,出现遗传漂移导致的。  相似文献

为了探讨蒙新干旱区景观和环境因素对子午沙鼠种群遗传结构的影响,我们利用8 个多态性的微卫星位点,对子午沙鼠14 个局域种群的160 个个体进行了种群遗传多样性和遗传结构的分析。微卫星结果显示种群具有高的遗传多样性,微卫星位点的等位基因数(A)为22. 50 ±3.02 (19 ~ 28),多态信息含量(PIC)为0.912 ±0. 02 (0.872 ~ 0. 929);种群观测杂合度(HO )为0.68 ± 0. 19 (0. 52 ~0.85),期望杂合度(HE )为0.79 ± 0. 08(0. 71 ~ 0.85)。分子变异分析(AMOVA)结果显示,所得到的3 个组之间已发生显著的遗传分化(P < 0. 001)。
Mantel 检测结果表明,子午沙鼠种群遗传结构与地理距离无显著的相关性(P = 0.270 0),而在多元线性模型中,蒙新干旱区的海拔高度是影响种群遗传结构的关键因素。  相似文献

Microsatellite markers were used to assess the genetic diversity and population structure in four populations of Elymus trachycaulus from British Columbia and one population of Elymus alaskanus from Northwest Territories. Fourteen microsatellite loci were used in this study. Our results indicated that E. trachycaulus is highly polymorphic, with an average percentage of polymorphic loci of 96.5% over the four populations. Average expected heterozygosity values (HE or gene diversity) varied from 0.418 to 0.585 with a mean of 0.497. Most of the genetic variation was found within populations (85%) and the differentiation among populations was found to be 15% (Fst = 0.15). Interpopulation genetic distances corresponded well with the geographic distance between the population sites of origin, as well as morphological characteristics. Tests for Hardy–Weinberg equilibrium (HWE) for all loci and all populations revealed that all loci significantly differ from HWE. Subsequent analysis indicated that departure from HWE at some loci was due to an excess of heterozygotes. Possible explanations for heterozygote excess are discussed. The most likely reason for observed heterozygote excess could be due to the polyploidy nature of the species.  相似文献

A population genetic study in the Ochamchir region, Abkhazia, SSR   总被引:1,自引:0,他引:1  
The reported longevity of residents of the Soviet Socialist Republic of the Caucasus has focused considerable attention on this population. However, little is known of the genetic composition of this population. With this in mind, several village populations of the Ochamchir Region, Abkhazia, SSR, were typed for 37 discrete genetic blood groups, erythrocyte and plasma protein loci. Gene and haplotype frequencies calculated for the polymorphic markers were determined and the results used in an analysis of intervillage heterogeneity and genetic distance analysis comparing the Abkhazians to European and Asian reference populations. The Abkhazians are approximately equal distance from European and West Asian populations in a genetic sense, and this is consistent with their geographical location. In addition to the usual genetic polymorphisms, rare electrophoretic variants were encountered at the lactate dehydrogenase A and phosphohexose isomerase loci. These results suggest that the population of the Ochamchir Region is relatively homogeneous and not distinctly different from its geographical neighbors.  相似文献
