【背景】啶虫脒等新烟碱类杀虫剂的残留易对非靶标生物造成伤害,投加高效降解细菌进行生物强化,可促进其快速降解。【目的】从蚯触圈中分离筛选啶虫脒降解菌并优化其降解条件,提高降解效率。【方法】制备蚯触圈基质富集筛选降解菌;通过生理生化特征和16S rRNA基因序列分析对其进行鉴定;利用单因素筛选、Plackett-Burman试验、最陡爬坡试验及Box-Behnken design试验优化菌株降解条件。【结果】分离得到1株啶虫脒降解菌D35,可在72 h内降解55.46%初始浓度为50 mg/L的啶虫脒,将其鉴定为一株假单胞菌(Pseudomonas sp.)。优化得到菌株降解啶虫脒的最佳环境条件为:胰蛋白胨10.19 g/L、温度为30℃、接种量为5.24%,pH 7.0、初始农药浓度50 mg/L,在此条件下72 h内菌株降解率为80.21%,较未优化前提高了24.75%。【结论】本研究对分离筛选新烟碱类杀虫剂降解菌的方法进行了探索,获得的菌株D35可高效降解啶虫脒,为快速消除环境中啶虫脒污染提供了新的微生物资源。  相似文献

【背景】近年来,苯胺类化合物加重了生态环境的污染,而生物法处理苯胺类废水具有较大发展潜力与广阔的应用前景。【目的】从长期受苯胺类化合物污染的活性污泥中分离获得一株能高效降解苯胺的菌株,优化其培养基及降解条件,为苯胺生物修复提供菌株与基因资源。【方法】采用平板法从富集驯化的菌株中筛选出以苯胺为唯一碳氮源和能源的高效降解菌,通过16S rRNA基因测序鉴定菌种,利用单因素筛选实验对降解条件进行优化,通过正交试验优化培养条件。【结果】筛选到一株苯胺降解菌BA-6,经鉴定为微杆菌属(Microbacterium)。菌株BA-6对初始浓度为600mg/L苯胺的日降解率可达98%以上。其高效降解的温度范围是30–37℃,pH范围是6.5–7.5。底物利用实验表明,菌株BA-6具有降解多种苯胺类化合物的能力。发酵培养基优化实验获得一种发酵培养基,活菌量高达3.06×1010CFU/mL。【结论】苯胺降解菌BA-6对苯胺有较强的降解能力和环境调节能力,在修复苯胺类化合物的生态污染方面有一定的应用前景。  相似文献

【目的】以发酵液纤溶酶活力为指标,优化海洋来源的链霉菌菌株MY0504的发酵条件。【方法】在菌株生长曲线及单因素试验基础上,采用Plackett-Burman设计筛选影响纤溶酶活性的主要因素,进一步用最陡爬坡试验及Box-Behnken中心组合设计法优化发酵条件。【结果】纤溶酶活性最高的发酵条件为:葡萄糖21.68 g/L,酵母粉25.31 g/L,NaCl5.0 g/L,K_2HPO_4·3H_2O3.0 g/L,MgSO_4·7H_2O 0.5 g/L,FeSO_4·7H_2O 0.02 g/L,装液量50 mL(250 mL摇瓶),接种量10%(体积比),初始pH 7.5,温度24°C,转速200 r/min,培养时间4.5 d。发酵液纤溶酶活性可达2 190.6 U/mL。【结论】确定了MY0504菌株产纤溶酶的最优发酵条件,为该酶的进一步分离纯化及性质研究奠定基础。  相似文献

【背景】拟除虫菊酯类农药的降解已成为食品安全和环境卫生领域的研究热点,而生物降解被认为是一种绿色高效的解决方法。【目的】从长期受拟除虫菊酯类农药污染的草莓根系土壤分离一株溴氰菊酯(deltamethrin,DM)降解菌,并优化其培养基及降解条件,从而提高DM降解菌的降解效率。【方法】采用富集驯化、分离纯化法筛选DM降解菌,通过形态学和生理生化特征,以及16S rRNA基因序列分析进行鉴定。通过Plackett-Burman因素筛选试验、最陡爬坡试验和Box-Behnken试验优化菌株降解条件。【结果】筛选获得一株DM降解菌LH-1-1,96h对DM(100mg/L)的降解率为53.43%,经鉴定为琼氏不动杆菌(Acinetobacter junii);通过优化后,在DM浓度75mg/L、胰蛋白胨3 g/L、pH值6.8、硫酸铵1.5 g/L、氯化铁0.01 g/L、接种量为5%(体积比)、菌龄12 h、培养温度30℃条件下,菌株LH-1-1对DM降解率达82.36%,较未优化前提高了28.93%。【结论】A. junii LH-1-1具有较高的DM降解能力,该菌可为生物修复受DM或拟除虫菊酯类农药污染的环境提供优良的微生物资源。  相似文献

【背景】木聚糖广泛存在于木质纤维类生物质中,是世界上含量最丰富的半纤维素,利用产酶微生物对木质纤维类生物质进行发酵处理是木质纤维类生物质资源化和能源化的有效手段。【目的】通过产木聚糖酶菌株的筛选鉴定、酶学特性分析和发酵条件优化,获得开发多纤维农林废弃物生产新型多元化饲料添加剂的材料。【方法】利用青藏高原土壤筛选产木聚糖酶菌株,通过形态学观察和rDNAITS区域序列分析鉴定菌株XC70种属,对其所产酶的酶学特性及该菌的生长规律和产酶规律进行分析,并利用单因素法和正交试验法优化其发酵条件。【结果】菌株XC70经形态学和分子生物学方法鉴定为草酸青霉(Penicillium oxalicum)。菌株XC70所产木聚糖酶的最适反应条件为:pH 5.0,70°C,温度低于50°C时稳定性较好,具备一定的耐酸性,Na+和K+对木聚糖酶活力具有促进作用(P0.05),在发酵54 h后菌体量和上清液酶活力大小均达到高峰。经过单因素法和正交试验法优化后确定了该菌的最优发酵条件为:蛋白胨7 g/L,玉米秸秆50 g/L,KCl 4 g/L,培养基初始pH 4.0,28°C,摇床转速200r/min,接种量2%。在此发酵条件下,木聚糖酶活力可达到1 489.33U/mL,与优化前相比提高了3倍多。【结论】从青藏高原土壤中筛选获得的菌株XC70具有一定的产木聚糖酶能力,其所产生的酸性木聚糖酶可用于降解多纤维物质开发新型饲料添加剂,具有一定的应用潜力和开发价值。  相似文献

褐藻胶降解菌的筛选、鉴定及产酶条件优化   总被引:1,自引:0,他引:1  
【目的】筛选一株能降解褐藻胶的菌株,并优化产酶条件以提高褐藻胶裂解酶活力。【方法】从漳州海域采集到海水和海泥,以海藻酸钠为唯一碳源,通过富集培养、初筛、复筛筛选到一株能够降解褐藻胶的菌株。依据16S rRNA序列分析、生理生化特征、菌体形态及菌落特征对该菌进行鉴定。通过单因素和正交试验对该菌的产酶条件进行优化。【结果】该菌属于海科贝特氏菌,命名为Cobetiamarina HQZ08。该菌株最佳的产酶培养基组成为:海藻酸钠7.00g/L、蛋白胨3.00g/L、NaCl30.00g/L,K2HPO4·3H2O 1.25 g/L。最佳发酵条件为:接种量2%,接种龄12 h,培养基起始pH为7.0,培养温度25°C,培养时间24 h。优化后褐藻胶裂解酶活力达到68.5 U/mL,TLC法分析酶解产物为褐藻胶寡糖。【结论】HQZ08菌株可以用于降解褐藻胶,产生聚合度为2–6的褐藻胶寡糖。  相似文献

木质素降解菌BYL-7的筛选及降解条件优化   总被引:3,自引:3,他引:0  
【背景】微生物降解木质素因其具有降解效率高和环保等特点而备受关注。【目的】筛选高效木质素降解真菌,并对其降解条件进行优化。【方法】通过愈创木酚-马铃薯葡萄糖琼脂(potato dextrose agar,PDA)和苯胺蓝平板法筛选高效木质素降解菌株,利用单因素筛选及响应面试验对培养条件进行优化。【结果】筛选到一株高效木质素降解菌BYL-7,经形态和多序列分析初步确定为Trametes versicolor。单因素试验证明初始pH、温度和接种量为降解木质素显著影响因子,响应面试验确定降解木质素最优条件为:初始pH 6.7,温度25 °C,接种量8%。在此条件下,碱性木质素降解率为36.5%,比未优化前提高54.0%;水稻秸秆木质素、半纤维素和纤维素降解率分别为32.8%、21.5%、13.2%,其中木质素降解率比未优化前提高36.1%;漆酶活性在第6天达到峰值120.0 U/L,比未优化前提高25.0%;木质素过氧化物酶活性在第6天达到峰值1 343.8 U/L,比未优化前提高36.0%;锰过氧化物酶活性在第5天达到峰值463.8 U/L,比未优化前提高31.7%。【结论】研究结果为木质素的降解提供了良好的菌种资源,同时也为后续木质素的研究积累了相关数据。  相似文献

【背景】洛伐他汀(lovastatin)是红曲霉的次生代谢产物,是重要的临床用降血脂药物。在液态发酵条件下,红曲霉的洛伐他汀产量较低,难以满足工业化生产的要求。【目的】筛选获得一株高产洛伐他汀的红曲霉株,并通过优化液态发酵条件提高洛伐他汀的产量。【方法】从红曲米中筛选获得一株高产洛伐他汀的红曲霉株,依据形态学特征、生理生化特性及18S rRNA基因序列分析对分离菌株进行鉴定;通过响应面法对其产洛伐他汀的液态发酵条件进行优化。【结果】获得一株产洛伐他汀的紫红曲霉(Monascus purpureus M4),该菌在甘油57.80g/L、酵母浸粉5.52 g/L、接种量为6.90%条件下,洛伐他汀产量(173.60 mg/L)较优化前提高了4.8倍。【结论】菌株M4产洛伐他汀最优液态发酵条件的建立,为洛伐他汀的大规模生产及该菌株的工业化应用提供了技术支撑。  相似文献

本研究从造纸厂的污泥及污水筛选获得了一株产木聚糖酶能力较强的菌株,经鉴定该菌株为异硫链霉菌(Streptomyces althioticus),专利保藏号为CCTCC M 2014110。通过单因素试验初步优化了碳源、氮源、温度、初始p H、发酵周期、摇床转速、接种量及装液量。试验结果显示该菌株最佳产酶条件为:以为玉米芯为碳源,用量30 g/L;以大豆粉与硝酸钾复合物为氮源,用量分别为20g/L和10 g/L;发酵周期120 h,发酵温度40℃,接种量6%,初始p H 5.0,装液量50 m L/250 m L三角瓶,摇床转速为160 r/min。  相似文献

【目的】从葡萄园土壤中分离L-天冬氨酸α-脱羧酶的产生菌株,对其进行分类鉴定,优化其产生L-天冬氨酸α-脱羧酶的发酵条件,为β-丙氨酸的生物合成提供基础。【方法】采用变色圈法和液体复筛培养基分离筛选具有L-天冬氨酸α-脱羧酶活力的菌株,对菌株进行形态、生理生化特征试验及16S r RNA序列同源性分析鉴定菌株的系统发育学地位,采用单因素及正交设计试验优化培养基及发酵条件。【结果】筛选到一株L-天冬氨酸α-脱羧酶高产菌株Pan D37,其亲缘关系和特基拉芽孢杆菌(Bacillus tequilensis)较近,且形态与培养特征、生理生化特性与特基拉芽孢杆菌基本相符。研究表明其最佳发酵配方和培养条件为:蔗糖22.5 g/L、富马酸7.5 g/L、蛋白胨20 g/L、L-天冬氨酸6 g/L、Triton X-100 2g/L,起始p H为7.0,装液量50 m L/500 m L,摇床转速220 r/min,种子液接种量为5%(V/V),35°C培养28h。在最优条件下L-天冬氨酸α-脱羧酶活力可达44.57 U/m L,比初筛时提高2.57倍。【结论】分离并获得一株特基拉芽孢杆菌(Bacillus tequilensis)Pan D37,经条件优化后具有较高的L-天冬氨酸α-脱羧酶产生能力,有望应用于β-丙氨酸的工业生产。  相似文献

The mature embryo of seven species belonging to five genera of Indian bamboos is described. In all these the basic pattern of embryo organisation is same: the scutellar and coleoptilar bundles are not separated by an internode, the epiblast is absent, the lower portion of the scutellum and the coleorhiza are separated by a cleft and the margins of embryonic leaves overlap. The features unique to fleshy fruited bamboos are: presence of a massive scutellum, the juxtaposition of plumule and radicle and the occurrence of a bud in the axil of the coleoptile. The fleshy fruit bearing bamboos should be classified into one group, the tribeMelocanneae. Evidence is provided to recognise additional groups in the subfamilyBambusoideae.  相似文献

Komárek has recently reviewed the various species assigned to the green algal genusNeochloris Starr (Chlorococcales, Chlorococcaceae) and removed those with uninucleate vegetative cells to a new genus,Ettlia. Watanabe & Floyd, unaware ofKomárek's work, also reviewed the species ofNeochloris and distributed them among three genera—Neochloris, Chlorococcopsis gen. nov., andParietochloris gen. nov.—on the basis of details of the covering of the zoospore and the arrangement of the basal bodies of the flagellar apparatus. The Drosophila Pax6 homolog twin of eyeless (toy) is so far the first zygotically expressed gene involved in eye morphogenesis in Drosophila. The study of its expression during embryogenesis is therefore informative of the initial events of eye development in Drosophila. We have analyzed how the initial expression domain of toy at cellular blastoderm is regulated. We show that the three maternal patterning systems active in the cephalic region (the anterior, terminal and dorsal-ventral systems) cooperate with zygotically activated gap genes to shape the initial expression domain of toy. Whereas Bicoid, Dorsal and Torso signaling synergistically act as activators, Hunchback, Knirps and Decapentaplegic act as repressors.  相似文献

Cyanobacteria and/or azolla were inoculated, with urea at 0, 72 or 144 kg N/ha, in plots in which azolla-free Indica rice var. IR 28 was grown. Productive tillers, yield and nitrogen contents of grain and straw positively responded to inoculation with cyanobacteria or azolla, even with fertilizer-N up to 144 kg N/ha. Inoculation improved colonization by cyanobacteria. Azolla were superior to the asymbiotic cyanobacteria in enhancing rice performance. Urea at a rate of 72 kg N/ha was found to support the best colonizations when applied with cyanobacteria or azolla or, to give maximum rice yields, both inoculants.  相似文献

Intracellular cysteine aspartate-specific proteases (caspases) play both signaling and effector roles in realizing the program of cell death. Caspases function as proteolytic cascades unique for each cell type and signal triggering apoptosis. All parts of the proteolytic cascades are duplicated and controlled by feedback signals. Amplification cycles between pairs of caspases (the third and the sixth, the ninth and the third, the twelfth and the sixth, and others) help multiply the initial apoptotic signal. The presence of physiological inhibitors of apoptosis that directly interact with caspases creates a multilevel regulatory network of apoptosis in cell. The caspase proteolytic cascades are also regulated by sphingolipid secondary messengers, among them ceramide, sphingosine, and their phosphates. Moreover, an association of the caspase signaling with ubiquitin-dependent proteolysis is shown in cells. In particular, the use of extracellular activators and inhibitors of caspases allows irreversible activation of apoptosis in tumor cells or the prevention of apoptosis in cortical neurons under neurodegenerative diseases.  相似文献

通过对采自广西24个县(市)的1 147份青藓科植物标本的逐一鉴定及相关文献的查阅,确认有广西青藓科植物11属、44种,其中包括广西青藓科植物新记录属1属,即拟异叶藓属(Pseudokindbergia),新记录种7种,分别为匐枝青藓(Brachythecium procumbens)、阔叶尖喙藓(Oxyrrhynchium latifolium)、泛生尖喙藓(O. vagans)、拟异叶藓(Pseudokindbergia dumosa)、华东细喙藓(Rhynchostegiella sinensis)、长肋拟青藓(Sciurohypnum populeum)和弯叶拟青藓(S. reflexum)。该文提供了修订后的广西青藓科植物名录,并对其中的新记录属、种的主要形态学识别特征、生境和地理分布等进行了详细描述。  相似文献

The cardinalfishes (Apogonidae) are a diverse clade of small, mostly reef-dwelling fishes, for which a variety of morphological data have not yielded a consistent phylogeny. We use DNA sequence to hypothesize phylogenetic relationships within Apogonidae and among apogonids and other acanthomorph families, to examine patterns of evolution including the distribution of a visceral bioluminescence system. In conformance with previous studies, Apogonidae is placed in a clade with Pempheridae, Kurtidae, Leiognathidae, and Gobioidei. The apogonid genus Pseudamia is recovered outside the remainder of the family, not as sister to the superficially similar genus Gymnapogon. Species sampled from the Caribbean and Western Atlantic (Phaeoptyx, Astrapogon, and some Apogon species) form a clade, as do the larger-bodied Glossamia and Cheilodipterus. Incidence of visceral bioluminescence is found scattered throughout the phylogeny, independently for each group in which it is present. Examination of the fine structure of the visceral bioluminescence system through histology shows that light organs exhibit a range of morphologies, with some composed of complex masses of tubules (Siphamia, Pempheris, Parapriacanthus) and others lacking tubules but containing chambers formed by folds of the visceral epithelium (Acropoma, Archamia, Jaydia, and Rhabdamia). Light organs in Siphamia, Acropoma, Pempheris and Parapriacanthus are distinct from but connected to the gut; those in Archamia, Jaydia, and Rhabdamia are simply portions of the intestinal tract, and are little differentiated from the surrounding tissues. The presence or absence of symbiotic luminescent bacteria does not correlate with light organ structure; the tubular light organs of Siphamia and chambered tubes of Acropoma house bacteria, those in Pempheridae and the other Apogonidae do not.  相似文献

Four phenolic amides, dihydro-N-caffeoyltyramine (1), trans-N-feruloyloctopamine (2), trans-N -caffeoyltyramine (3), and cis-N-caffeoyltyramine (4), were isolated from an ethyl acetate extract of the root bark of Lycium chinense Miller. All had an anti-fungal effect; compounds 1-3 were potent at 5-10 microg ml(-1) and were without hemolytic activity against human erythrocyte cells. Compound 4 was active at 40 microg ml(-1). All four compounds impeded the dimorphic transition of pathogen, Candida albicans.  相似文献

Resin glycosides are secondary metabolites exclusive to the convolvulaceous plants. In this study, crypthophilic acids A–C (13), the first resin glycosides occurring in another family (Scrophulariaceae), and the other constituents of Scrophularia cryptophila were examined for in vitro antiprotozoal and antimycobacterial potentials. Except for crypthophilic acid B (2), all tested compounds exhibited growth-inhibitory effect against Trypanosoma brucei rhodesiense, with l-tryptophan (6) and buddlejasaponin III (7) being the most potent ones (IC50's 4.1 and 9.7 μg/ml). In contrast, the activity towards Trypanosoma cruzi was poor, and only crypthophilic acid C (3), 6 and 7 were trypanocidal at concentrations above 40 μg/ml. With the exception of 2 and 6, all compounds were active against Leishmania donovani. Harpagide (4) and 3 emerged as the best leishmanicidal agents (IC50's 2.0 and 5.8 μg/ml). Only compounds 3, 6 and 7 showed antimalarial activity against Plasmodium falciparum with IC50 values of 4.2, 16.6 and 22.4 μg/ml. Overall the best and broadest spectrum activity was presented by compounds 3 and 7, as they inhibited all four parasitic protozoa. None of the isolates had significant activity against Mycobacterium tuberculosis (MICs >100 μg/ml) or were toxic towards mammalian (L6) cells. This is the first report of antiprotozoal activity for natural resin glycosides, as well as for harpagide (4), acetylharpagide (5), tryptophan (6) and buddlejasaponin III (7).  相似文献
