Biocarbazin (DTIC synonym) is an anticancer drug acting as a purine analogue, as an alkylating agent, as a SH-group blocker. Biocarbazin administration in doses 50 and 200 mg/kg of body weight to adult male BALB/c mice suppresses the process of spermatogenesis. Seminiferous tubule of the testis displayed dose-dependent histological alterations manifested with decrease of mitotically dividing cells and increase in the number of multinucleated spermatocytes and spermatids. RNA synthetic activity studied by means of autoradiography showed a tendency for a reduction in the spermatogonia and pachytene spermatocytes.  相似文献

Intracellular cysteine aspartate-specific proteases (caspases) play both signaling and effector roles in realizing the program of cell death. Caspases function as proteolytic cascades unique for each cell type and signal triggering apoptosis. All parts of the proteolytic cascades are duplicated and controlled by feedback signals. Amplification cycles between pairs of caspases (the third and the sixth, the ninth and the third, the twelfth and the sixth, and others) help multiply the initial apoptotic signal. The presence of physiological inhibitors of apoptosis that directly interact with caspases creates a multilevel regulatory network of apoptosis in cell. The caspase proteolytic cascades are also regulated by sphingolipid secondary messengers, among them ceramide, sphingosine, and their phosphates. Moreover, an association of the caspase signaling with ubiquitin-dependent proteolysis is shown in cells. In particular, the use of extracellular activators and inhibitors of caspases allows irreversible activation of apoptosis in tumor cells or the prevention of apoptosis in cortical neurons under neurodegenerative diseases.  相似文献

The mammalian zona pellucida (ZP) is an extracellular glycoprotein structure formed around growing oocytes, ovulated eggs and preimplantation embryos. The specific functions of ZP are highly determined by its morphological structure. Studies on cat oocytes during maturation and after fertilization were undertaken, using routine transmission (TEM) and scanning electron microscopy (SEM). Two basic ZP layers – outer with rough spongy appearance and inner with smaller fenestrations and smooth fibrous network – were visible. Deposits, secreted by oviductal cells formed new layer, the so called oviductal ZP. After fertilization outer ZP showed rougher meshed network due to fusion between filaments as a consequence from sperm penetration while the inner was smoother with melted appearance. The presented data on the SEM and TEM characteristics of cat oocytes, together with our previous studies on carbohydrate distribution suggest that during oocyte maturation and fertilization ZP undergoes structural and functional rearrangements related to sperm binding and penetration.  相似文献

Fumonisins have been reported to have diverse effects on animals, including immunosuppression in chickens and feeder calves; therefore, the effects of fumonisin B1 (FB1) on immune function in BALB/c mice was investigated. When administered i.p. with sheep red blood cells (SRBC), 5 to 100 µg of FB1 reduced the number of plaque-forming cells (PFC) produced against SRBC; however, when administered daily, 1 to 50 µg of FB1 caused a 4 to 12-fold increase in the number of PFC after SRBC injection. Therefore, FB1 is not only immunosuppressive; but also, immunostimulatory. To test the possibility that there may have been an immune response to FB1 as an antigen, FB1 was injected into mice and the number of splenic cells forming rosettes on FB1-treated SRBC was determined. There were dose-dependent increases in the antigen-binding cells, with up to 4.9- and 4.6-fold increases, respectively, upon primary and secondary immunization. FB1-binding immunoglobulins could be detected in sera from some treated mice, but this response was not obtained in every experiment. This paper is an overview of Russian literature dealing with the accumulation, the transformations and the release of phosphate and nitrogen compounds in a great number of Russian lakes and reservoirs. A considerable data bank has been analysed. Special attention is given to the relations of N- and P-accumulation with the input and transformation of organic carbon, as well as to the release mechanisms, often in relation to eutrophication of the lakes and reservoirs. It is shown that the major input of organic matter into the sediments comes from autochthonous material, and is usually > 70 %. The relative importance of phytoplankton and macrophytes as sources of organic matter is discussed; it appears that trophic state, depth and other factors may have a large influence on this ratio. In shallow eutrophic lakes macrophytes may be the source of organic matter, which source can amount to 1.5–2.5 times that of phytoplankton. It is also shown that the C/N ratio is not a good indicator of the source of the organic matter, because their C/N ratios often are not very different. The decomposition rate of organic matter was analysed; it depends on trophic state and other factors. Sediment N accumulation is mostly (> 90%) in organic form, and depends on nitrogen and organic matter inputs coming from phytoplankton or macrophytes. A correlation coefficient of 0.9–0.95 was found in 176 lakes. In 113 lakes the N accumulation was 0.11 x C accumulation, with C/N ratios between 7.4 and 12.9. Ammonification was rather constant in different groups of lakes; values were often about 20–25 mg m−2 d−1. The presence of the different forms of nitrogen in interstitial water and in adsorbed forms is discussed. The N in interstitial water is usually in the form of NH3. Sediment P-accumulation is usually in inorganic form and is related to primary production. Three different groups of sediments could be distinguished with C/P ratios of 31–100, of 101–350 and > 350. In hard water lakes P sedimentation was found to be 0.3–0.5 times that in soft water lakes with comparable primary production. The relative occurrence of apatite, non-apatite and residual P in sediments was calculated. In the interstitial water the P concentration appeared to be controlled by the input and decomposition of organic matter. The concentration of phosphate dissolved in the interstitial water of the top 2 cm layer is often 10–100 times lower than that of the dissolved N. The concentrations of interstitial phosphate are from a few μgl−1 up to 15 mgl−1, but the higher concentrations occur only rarely. Different types of vertical profiles of P compounds in the sediments were shown to be related with the presence of an oxidised zone, the presence of clay etc. Autochthonous apatite and non-apatite phosphates are more mobile than the allochthonous ones and are in equilibrium with interstitial phosphate. Accumulation of autochthonous apatite in sediments is controlled by decomposition of organic matter and accumulation of carbonates.  相似文献

The mouse sperm nucleus, after the removal of protamines and DNA, consisted of a skeletal structure that conformed to the original nuclear shape. Sperm were extracted with 1% SDS, and the isolated nuclei, along with the enveloping perinuclear theca, were incubated in 25 mM dithiothreitol, and exposed to different reagents in an effort to displace the protamines, P1 and P2. Protamines, labeled with [3H]arginine, were displaced from the nucleus by CaCl2.MgCl2, but only partially by anionic detergents, monovalent cations, and polyvalent anions. Displacement of P1 and P2 was achieved by digesting the nuclei with DNase I and simultaneously extracting with CaCl2.MgCl2 (3:2; mol:mol) in stepwise increments of 125, 150, 175, 200, and 250 mM. Protamine displacement was concentration-dependent, occurring with an EC50 of approximately 205 mM and with maximal displacement at approximately 250 mM CaCl2.MgCl2. The nucleus was reduced to a skeletal structure consisting of the perinuclear theca and an internal network of transverse fibers. The evidence was consistent with the former being derived from the perforatorium and postacrosomal nuclear sheath (both cytoplasmic structures), whereas the fibers were most likely of nuclear origin. By SDS-PAGE and isoelectric focusing (IEF), perinuclear matrices consisted of greater than or equal to 230 protein spots, with M(r)s in the range of 70,000 to 8000 and pIs of greater than or equal to 7.5 to approximately 4.7, respectively. Monoclonal antibodies prepared against perinuclear matrices bound to specific proteins on IEF immunoblots and, based on light and electron microscopic observations, to discrete domains of the sperm perinuclear theca and nucleus.  相似文献

The role of gangliosides in the reception of low density lipoproteins (LDL) was studied using as targets mouse ascites hepatoma 22a (MAH) cells which bind LDL through a specific high affinity receptor. Low density lipoprotein binding and uptake by MAH cells decreased after brief treatment of the cells with neuraminidase to partially remove surface sialic acid residues. The LDL uptake capability of the neuraminidasetreated MAH cells was fully restored after incorporation of exogeneous GM1- and GD1a-gangliosides into the cell surface. In contrast, free (extracellular) gangliosides inhibited LDL uptake by native MAH cells. This inhibitory effect was seen at ganglioside concentrations corresponding to the ganglioside content of serum and was most pronounced with gangliosides whose sialic acids were linked to a terminal galactose residue (GM3, GD1a, GT1b) but was smaller or absent with gangliosides whose sialic acids were attached to an internal galactose (GM1, GM2). The binding of gangliosides to LDL was structure and concentration dependent, saturable and trypsin sensitive. The LDL-ganglioside interaction was further investigated by steady state fluorescence spectroscopy. Changes in the LDL fluorescence polarization were observed with as little as 0.01 M concentrations of the gangliosides. The magnitude and nature of the effect depended on the type of ganglioside. We conclude that the LDL surface possesses sites recognizing specific carbohydrate sequences of glycoconjugates and that changes in the cell surface concentrations of sialic acids significantly modulate the LDL uptake.
