根据植物偏爱密码子人工合成的纳豆激酶基因sNK和其中插入番茄果实特异表达基因E8第一内含子的纳豆激酶基因sNK-E8i,通过农杆菌侵染的方法转化到烟草NC89中。经PCR检测,得到12株转sNK基因烟草植株和10株转sNK-E8i基因烟草植株,初步证明目的基因已整合到烟草基因组中;RT-PCR检测有9株转sNK基因烟草植株和10株转sNK-E8i基因烟草植株为阳性;通过RT-qPCR将两种基因在烟草中的表达量进行比较,结果表明转sNK-E8i基因的植株中纳豆激酶的表达量比转sNK基因的植株中高;通过纤维蛋白平板法检测其活性,共有5株转sNK基因烟草植株和3株转sNK-E8i基因烟草植株检测到溶圈,说明目的基因在部分转基因烟草中可正常转录和翻译并表现出溶栓活性。经加代培养已得到T1代转基因植株。  相似文献

为了揭示铁皮石斛(Dendrobium officinale)甾醇C-24甲基转移酶2基因(DoSMT2)在甾醇代谢过程的功能,该研究通过根癌农杆菌介导法将来源于铁皮石斛的DoSMT2基因转化烟草(Nicotiana tabacum),并采用qRT-PCR技术检测DoSMT2基因在转基因烟草叶片中的表达,采用气相色谱质谱法分析菜油甾醇和谷甾醇的含量。结果显示:(1)成功获得DoSMT2基因的开放阅读框(1 119 bp),并成功构建正义植物表达载体质粒pCXSN-DoSMT2,经农杆菌介导的烟草叶盘转化法转化烟草并鉴定,获得4株阳性转基因烟草植株。(2)Southern blot结果表明,4株转基因烟草植株都有1条杂交信号带,而非转基因烟草植株没有,说明外源DoSMT2基因都以单拷贝整合到4株转基因烟草基因组中。(3)qRT-PCR检测显示,非转基因烟草未检测到外源DoSMT2基因的表达,4株转基因烟草都能检测到DoSMT2基因的表达,且表达水平差异极显著,各株系表达量高低依次为P3P1P2(P4)。(4)气相色谱质谱分析显示,转DoSMT2基因烟草叶片的菜油甾醇含量均极显著低于非转基因烟草叶片,而谷甾醇含量均极显著高于非转基因烟草叶片。研究表明,DoSMT2具有催化24-亚甲基胆甾烯醇转化形成24-亚乙基胆甾烯醇活性。  相似文献

目的:构建可高效生产有活性的纳豆激酶的大肠杆菌工程菌。方法:将纳豆激酶酶原(pro-nattokinase,pro-NK)基因和纳豆激酶(natokinase,NK)基因,并分别克隆到表达融合蛋白的高效表达载体pJN上,构建出表达质粒pJNK1和pJNK2,并转化大肠杆菌BL21(DE3)。结果:IPTG诱导下,两个融合蛋白的表达量均达到30%,活性检测显示表达纳豆激酶酶原融合蛋白的菌株pJNK-1(BL)诱导后菌体破碎上清的溶栓活性比表达纳豆激酶融合蛋白的菌株pJNK-2(BL)高2-3倍,结论:纳豆激酶酶原融合蛋白部分自减切产生纳豆激酶成熟肽。  相似文献

利用PCR方法从分泌纳豆激酶的枯草杆菌基因组DNA中扩增得到纳豆激酶基因(NK),利用基因重组技术构建了纳豆激酶基因的表达载体pETNK。在诱导下,实现了在大肠杆菌中高效表达,经SDS-PAGE电泳分析和薄层扫描结果显示,表达的目的蛋白占菌体蛋白的21.5%。将表达产物经过DEAE-Cellulos-DE52和Sephedax-G100两个柱分离纯化,得到纯的纳豆激酶蛋白干粉,经琼脂糖-纤维蛋白平板法测出纳豆激酶干粉的溶栓活性相当于200u尿激酶。从基因工程角度研究纳豆激酶基因的克隆、表达及纯化,为用基因工程菌生产纳豆激酶奠定了基础。  相似文献

AtNYE1是拟南芥叶片衰老过程中叶绿素降解的重要调控基因,本文用AtNYE1为诱饵基因,通过NCBI tblastn在毛竹的cDNA文库中找到3个与其相似性较高的cDNA全长序列,分别命名为PeNYE1、PeNYE2和PeNYE3。为了验证其是否具有AtNYE1相似的功能,分别将它们的编码区构建到带有花椰菜花叶病毒35S强启动子的植物表达载体上,并通过冻融法将这3个表达载体导入GV3101农杆菌。通过农杆菌介导法,将这3个基因分别在烟草叶片中瞬时表达及在拟南芥植株中稳定表达,结果显示,瞬时过表达和组成型过表达PeNYE1均导致了叶片的黄化,而瞬时或组成型过表达PeNYE2或PeNYE3均未观察到黄化表型。这些结果表明PeNYE1是毛竹中叶绿素降解的重要调控基因。  相似文献

分离并克隆了水稻5-烯醇丙酮莽草酸-3-磷酸合酶基因的第一内含子(EPI). 序列分析表明, EPI长704 bp, GC含量为36.2%. 为进一步研究EPI序列在转基因植物中对外源基因表达水平的影响, 将EPI序列插入CaMV35S启动子和报导基因β-葡糖醛酸酶(β-glucuronidase, GUS)基因(gus)之间. 采用基因枪法转化烟草叶片, gus基因瞬时表达表明, EPI序列的存在可以使GUS的表达水平提高. 利用农杆菌法转化烟草, 获得了gus基因稳定表达的植株. GUS活性检测表明EPI内含子的存在可以显著提高GUS基因的表达(P<0.01). Northern blot 分析表明, 在转录水平上gus基因在含EPI内含子的转基因植株中的表达高于不含EPI gus基因的表达, 并且成熟的gus mRNA中EPI被正确剪取掉了, 表明是一种非转译的内含子. GUS定量检测表明, EPI存在可以使GUS的平均表达水平提高3倍, 最高单株可提高6倍.  相似文献

目的:构建纳豆激酶基因的表达载体,鉴定其在大肠杆菌中的表达及表达产物的生物活性鉴定.方法:以纳豆芽胞杆菌基因组为模板,PCR技术克隆出纳豆激酶基因的成熟肽序列,分别克隆进具有信号肽的pMAL-p2x及无信号肽pMAL-c2x质粒中,经酶切和测序鉴定其正确性,分别将重组质粒转化至大肠杆菌中表达.结果:成功构建的两组重组质粒在IPTG诱导下,均能分别在37℃及16℃条件下表达出可溶性的融合蛋白,SDS-PAGE胶检测证实重组质粒在大肠杆菌中可表达出相对分子量约76kDa的纳豆激酶蛋白.纤维蛋白平板实验证明两种融合蛋白均有活性,且有信号肽的融合蛋白的酶活较无信号肽的融合蛋白高.结果:成功构建了两组重组纳豆激酶基因的表达质粒,且该两组重组基因在大肠杆菌中可溶性表达并具有生物活性,因此为下一步研究表达产物纳豆激酶的功能、应用和生产奠定了基础.  相似文献

翻译和非翻译马铃薯Y病毒外壳蛋白基因介导的抗病性比较   总被引:16,自引:0,他引:16  
利用RT-PCR方法克隆获得马铃薯Y病毒烟草叶脉坏死株系(PVY^N)的可翻译和不可翻译外壳蛋白(CP)基因,并分别插入pROKⅡ质粒中获得重组双元表达载体。通过根癌农杆菌(Agrobacterium tumefaciens)介导的基因转化方法,将可翻译和不可翻译的PVY^N-CP基因分别导入烟草栽培品种NC89叶片组织中,得到抗卡那霉素的再生植株,对所得到的抗卡那霉素的再生苗进行PCR检测表明,被导入可翻译和不可翻译CP基因的植株分别占卡那霉素抗性植株的95%和98%。攻毒实验表明,两种类型的转基因烟草对PVY^N的抗病性具有相似性,其表现型为:免疫、抗病和感病。免疫型转基因植株的抗病性不受接种物类型及其剂量的影响。Southern印迹杂交结果显示,目的基因已经整合到烟草基因组中。Northern印迹杂交证明,两种类型的CP基因都已在RNA水平上得到了表达,但细胞内RNA的积累量与转基因植株的抗病强度成负相关。Western印迹杂交表明,在表达不可翻译PVY^N-CP基因的转基因植株内 以CP蛋白,而在导入可翻译的PVY^N-CP基因的植株内检测到了CP蛋白,且CP蛋白含量与抗病性不存在正相关。本研究结果证明,表达可翻译与不可翻译PVY^N-CP基因的转基因烟草对PVY^N的抗病性均为RNA介导的抗病性。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
