低双乙酰啤酒酵母菌株BEZ112的选育   总被引:16,自引:1,他引:15  
以啤酒酿造生产菌株啤酒酵母(Saccharomyces cerevisiae)FB作为出发菌株,用甲基磺酸乙酯(EMS)诱变,经分离筛选得到一株优良的啤酒酵母菌株BEZ112。该菌株的絮凝性、发酵度、酒精度、发酵液的总酯和总高级醇的含量等特性保持了亲株的优良性状。但以12°Bx麦芽汁为培养基用500mL三角瓶在12℃下发酵,该菌株发酵至第4d,发酵液中的双乙酰含量达到峰值(0.291mg/L),比出发菌株FB发酵4d的峰值降低了30%,发酵至第8d,BEZ112发酵液中的双乙酰含量比出发菌株FB的降低了23%。以12°Bx麦芽汁为培养基用500L罐在12℃下发酵8d,BEZ112发酵液中的双乙酰含量(0.091mg/L)比出发菌株FB的(0.124mg/L)降低了27%。发酵得到的啤酒口感纯正清爽。  相似文献

高温高浓发酵技术作为一项新兴的啤酒生产技术,它为啤酒生产带来诸多利益的同时,也存在着发酵结束后酵母絮凝性下降、高级醇生成量过高等系列问题。为提高高温高浓发酵条件下酿酒酵母的絮凝性同时降低高级醇的合成能力,首先构建了以酿酒酵母BAT2基因为整合位点过表达FLO5基因的菌株,重组菌株S6-BF的絮凝性达到67.67%,比出发菌株S6提高了29%,而高级醇生成量仅降低5.9%;进一步构建以BAT2基因为整合位点再次过表达FLO5基因的菌株,与出发菌株S6相比,重组菌株S6-BF2的絮凝性提高了63%,达到85.44%,高级醇生成量下降至159.58 mg/L,降低了9.0%;通过弱化线粒体支链氨基酸转氨酶(BAT1)的表达,高级醇的生成量得到进一步的降低,达到142.13 mg/L,比原始菌株S6降低了18.4%,同时重组菌株S6-BF2B1的絮凝性没有受到影响;风味物质的测定结果表明啤酒中醇酯比例较为合理。研究结果对工业啤酒酵母发酵后的沉降分离和提高啤酒风味品质有着重要的意义。  相似文献

应用原生质体融合技术,得到乙醇产量较高的两株多倍体酵母菌融合子F_(28)和F_(38),融合频率为2×10~(-5)。测定融合子F_(28)、F_(38)每个细胞内DNA含量分别为2.14×10~(-8)、2.34×10~(-8)μg,而亲株DNA含量分别为0.78×10~(-8)和1.24×10~(-8)μg。并进行了融合子细胞增殖率、乙醇发酵能力及同功酶分析等试验。用固定化细胞发酵乙醇试验结果表明,在pH4.0,17%糖蜜为基质情况下,发酵3小时,融合子F_(28)、F_(38)的乙醇产量分别为77.21和77.09mg/ml;亲株乙醇产量仅为65.00mg/ml和68.40mg/ml。为固定化细胞发酵乙醇提供优良菌株。  相似文献

通过灭活原生质体融合选育啤酒酵母新菌株   总被引:33,自引:0,他引:33  
将生产用啤酒酵母(\%Saccharomyces cerevisiae)\% LQ16和QSB7分别进行单倍体化,并筛选LQ16(Ile\+-,Dat\+r)和QSB7(Ala\+-,H\-2S\+-)的遗传标记。经摇振培养后,酶解制备原生质体,对前者进行紫外灭活,而后者进行热灭活,再利用PEG进行融合,在MMR和SMMR培养基上再生,挑选融合子。连续传代稳定后鉴定融合子QSB\|XI\-6。通过细胞体积和生物量的测定,以及遗传型的分析和细胞DNA含量测定等均证实了获得的是融合子。经对啤酒感观评价,双乙酰含量测定与多种成分的气相色谱分析以及啤酒的发酵度,絮凝性、稳定性测定,并经连续多次生产试验证实QSB\|XI\-6是一株口味独特、发酵度高、絮凝性强、遗传性能稳定兼具有多种优良性状的啤酒酵母生产新菌株。  相似文献

为获得具有多种优良性状的高效乙醇生产菌株,以絮凝性酿酒酵母RHZ-1及耐酸耐温酿酒酵母SEB2为亲本,通过紫外诱变获得营养缺陷型标记菌株,并利用原生质体融合技术选育耐酸絮凝性酿酒酵母。通过对融合子的筛选获得一株优秀的耐酸絮凝性酿酒酵母菌株RS-1;该菌株在30℃,pH2.2,15%YPD条件下发酵24 h,乙醇产生量为47.87 g/L,糖消耗速率和基于糖消耗的乙醇收率分别比亲本菌株RHZ-1提高了19.5%和9.8%。在35℃,p H2.2,15%YPD条件下发酵48 h,乙醇产生量为38 g/L,糖消耗速率和乙醇收率分别比亲本菌株RHZ-1提高了46.8%和21.6%。结果表明,获得的菌株可为提高燃料乙醇的生产效率奠定基础。  相似文献

对402株不同种属的酵母菌株进行初筛、复筛,筛选到一株生物量较高的二倍体菌株ZY-46(Saccharomyces cerevisiae)和一株铁富集量较高的二倍体菌株ZY-173(Saccharomyces kluyveri)。然后以它们为出发菌,分别进行单倍体分离、硫酸二乙酯(DES)诱变,并通过原生质体融合,得到一株高生物量富铁酵母融合菌株ZYF-15。在优化的发酵条件下,该融合菌株生物量可达11.2g/L,细胞铁含量达24.5mg/g干细胞,细胞总铁含量分别比原始亲株ZY-46和ZY-173提高了2.6倍和1.9倍。  相似文献

啤酒酵母是啤酒酿造的灵魂,可以直接影响啤酒品质。在啤酒酿造过程中,由于啤酒酵母被多次传代和保藏,造成优良菌种发酵性能衰退等问题,导致发酵不彻底,影响最后啤酒的风味质量。为此以8株Lager型啤酒酵母为出发菌株,通过平板分离纯化获得80株分离菌株,再经过三角瓶发酵初筛和复筛、发酵罐中试发酵实验最终获得了8株发酵性能优良的啤酒酵母。其中,6株酵母可应用于酿造双乙酰含量低于0.1 mg/L的啤酒;3株酵母发酵度高于70%,适合酿造干啤酒;1株酵母发酵度低于50%,适合酿造低醇啤酒。在风味方面:1株酵母酿造的啤酒醇酯比为3.3,啤酒酯香味较突出;另1株酵母酿造的啤酒醇酯比为4.5,啤酒高级醇含量较高。8株经过选育的啤酒酵母发酵特征明显,便于精酿啤酒厂实际应用。  相似文献

采用10 Kev低能N~+注入啤酒酵母,经筛选获得一菌株Lz37,再用150 MPa超高压处理菌株Lz37,经双乙酰平板筛选获得一菌株Gy3,其凝聚性很强,适合于在小麦汁中发酵啤酒,其发酵度为66%~68%,双乙酰含量低于口味阈值,遗传稳定性良好。将Gy3酵母定为全小麦啤酒生产应用酵母,命名为商啤3号(Sp-03)。SP-03啤酒酵母菌株的各项生理及生产性能都较优良,特别是在全小麦芽啤酒的酿造中适用性较强,经过对发酵工艺等的调整,用其酿制的啤酒口感纯正、淡爽、柔和。  相似文献

为筛选猕猴桃酒专用酵母,通过酵母菌的分离、显微镜观察和杜氏管产气试验、耐受试验、发酵力测定试验以及酵母凝集性试验,筛选出一株性能优良的菌株命名为L36。实验表明,该菌能够耐受的酒精度为14%,可耐受的二氧化硫质量浓度为250 mg/kg,主发酵CO2失重量为15.04 g,凝集性强。利用L36菌株酿造的猕猴桃原酒酒精度可达10.57%vol,Vc含量可达29.1 mg/100 m L,酸度为11.52 g/L。通过分子生物学鉴定,确定该菌为毕次酵母属。本试验通过从猕猴桃表面筛选出优良的野生酵母,可作为果酒酵母的筛选的参考依据。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献
