从皖南尖吻蝮蛇毒中经阴离子交换层析和凝胶过滤层析分离纯化得到抗血小板凝集蛋白agkisacutacin ,纯化的agkisacutacin由分子量为 1 4kD和 1 5kD的 2条肽链通过二硫键连接 ,能有效抑制ristocetin诱导的血小板凝集 (IC5 0 为 1 8.5mg/L) ,能轻微抑制凝血酶诱导的血小板聚集 (IC5 0 为 1 .2 2 g/L) ,但对ADP、胶原诱导的血小板聚集无影响。agkisacutacin不具有纤溶活性、抗凝活性、磷脂酶A2 活性和出血活性 ,是一种潜在的安全、有效的抗血小板性栓塞的药物  相似文献

从马槟榔(Capparis masaikai)种子中曾分离出二种甜味蛋白,称为Mabinlin Ⅰ和Ⅱ,分子量分别为11.6kD和10.4kD。用Edmail降解和反相HPLC鉴定PTH-aa方法测定证明:二者的N末端均因焦谷氨酸环化而封闭。采用1mol/l HCl-甲醇溶液35℃处理24小时的方法可使之开环。二种蛋白N端肽8个氨基酸残基顺序均为:Pyroglutamic acid-Pro-Arg-Gly-Pro-Ala-Leu-Arg-。羧肽酶酶解的动态分析证明MaI羧端的氨基酸顺序为-Phe-Gln-Leu-Ala-Ser,MaⅡ为-Phe-Gln-Leu-Ala。其差别仅在于MaⅡ缺一个Ser,这一结果表明MaⅠ和MaⅡ的主要差异不是在端肽。  相似文献

采用STI-Sepharose 4B亲和层析的方法,从鼠新鲜胰脏中分离得到纯的胰蛋白酶。大鼠胰蛋白酶的比活为24 615BAEEU/mg蛋白,总活性回收率47%,小鼠胰蛋白酶的比活为32 768BAEEU/mg蛋白,总活性回收率55%。经SDS-聚丙烯酰胺凝胶电泳鉴定,大鼠、小鼠胰蛋白酶均呈现单一蛋白带,两者的分子量都是24kD。用等电聚焦电泳测定,二者的等电点均为p19.5以上。对它们的动力学性质作了研究,大鼠胰蛋白酶的Km值为2.33×10~(-4)mol/L,K,值为0.92×10~(-5)mol/L,小鼠胰蛋白酶的Km值为5.60×10~(-4)mol/L,K:值为1.27×10~(-5)mol/L。  相似文献

以未诱导组作为空白对照研究比较真菌诱导、超声诱导和热诱导家蝇Musca domestica 幼虫血淋巴初提液的抗真菌肽效果,比较各种诱导方法诱导后的幼虫存活率;用凝胶层析法和高效液相分离纯化热诱导家蝇3龄幼虫抗真菌肽,检测其抗白假丝酵母菌Candida albicans和新生隐球菌Cryptococcus neoformans活性;SDS-PAGE分析抗真菌肽的蛋白分子量范围。结果表明:3种诱导方法诱导后家蝇幼虫均产生具有明显抗真菌作用的抗真菌肽,其初提液抑菌圈大小没有明显差别;真菌诱导组和热诱导组幼虫存活率低于对照组,而超声诱导组与对照组相比则无明显差别。经分离纯化后,抗真菌肽仍具有较好的抗真菌活性;SDS-PAGE分析表明该抗真菌肽有效成分的蛋白分子量在14.4 kD以下。结果提示热诱导家蝇幼虫产生抗真菌肽是一种方便、有效的诱导方式。  相似文献

商陆种子抗病毒蛋白的制备及毒性分析   总被引:1,自引:0,他引:1  
采用一种改进的方法,从商陆属两种植物——商陆(Phytolacca acinosa)和美州商陆(P. americana)种子中分别制备抗病毒蛋白(Pokeweed antiviral protein from seeds,简称PAP-s)。研究证明,两种植物中的PAP-s性质完全相同。经SDS-PAGE和凝胶过滤层析测得该蛋白质为单一肽链,分子量约30kD;聚丙烯酰胺等电聚焦电泳测得等电点为8.4;经Edman降解,测得N-末端氨基酸为Ile.PAP-s在兔网织红细胞裂解液中抑制蛋白质合成ID_(50)为2.6ng/ml(8.6×10~(-11)mol/1);将该蛋白质与脊髓灰质炎(?)型病毒混合接种于猴肾细胞培养液,显示出较强的抗病毒活性,浓度为4.17×10~(-8)mol/1时能抑制98%的病毒增殖。将PAP-s通过异型双功能连接剂SPDP与抗人T细胞单克隆抗体Wu71偶联制备免疫毒素,对T淋巴细胞白血病CEM细胞显示特异杀伤作用,在10~(-9)mol/1时可杀灭76.4%的靶细胞,~(14)C-亮氨酸掺入蛋白质合成抑制试验显示ID_(50)为10~(-10)—10~(-9)mol/l。  相似文献

 从慢性淋巴性白血病人的周血白细胞中纯化了DNA拓扑异构酶Ⅰ,经SDS-聚丙烯酰胺凝胶电泳分析,以蛋白质染色只有一条100kD的肽链,而用此酶的单克隆抗体探测同一纯化的酶则出现100kD,90kD,83kD,80kD和74kD五条肽链,并从部分纯化的酶制剂中检测到一条34kD的小分子具有相当高的酶活性。用此抗体进一步探测了不同类型白血病人周血白细胞的DNA拓扑异构酶,发现明显的差异,不同分子量仍具有此酶的活性,说明不同细胞固有DNA拓扑异构酶Ⅰ的不均一性。  相似文献

本文报道唐菖蒲球茎中抗真菌活性物质的成分及其分子量测定方法和结果。用沸水提取,经过超滤、透析、柱层析,纯化到一种有抗真菌活性的物质。排除了它是蛋白质、核酸、脂类及小分子物质的可能性,确定这是一种多糖,定名为唐菖蒲抗真菌多糖(Gladiolus hntifungal Polysaccharide,简称GAFS)。真空旋转蒸发干燥后,用苯酚-硫酸法测得干燥后的样品糖含量为44.08%,用FPLC(快速蛋白质液相层析仪)测其分子量为15750。  相似文献

从苦参 (SophoraflavescensAit.)根浸出液经硫酸铵分级 ,得苦参凝集素 (SFL)粗品 ,再经DEAE_Sepharose、SephadexG_1 5 0和HPLC层析 ,获得具有强凝集活性的SFL样品 ,用PAGE和SDS_PAGE检测均为单一蛋白染色带。SDS_PAGE显示SFL分子仅有一条肽链 ,SephadexG_1 0 0和SDS_PAGE测得其分子量均为 32kD。当SFL浓度为 0 .97μg/mL时能凝集兔红细胞 ,无血型专一性 ,其凝血活性可被甘露糖和果糖抑制 ,麦芽糖和葡萄糖有弱的抑制作用 ,凝集素分子含有 2 .89%的中性糖 ;当SFL量为 6 2 μg时 ,对棉花枯萎病菌 (FusariumvasinfectumAtk .)、小麦赤霉病菌(Gibberllasaubinetii (Mont.)Sacc .)和水稻稻瘟病菌 (PiriculariaoryzaeCav)菌丝体的生长发育有明显地抑制作用。用Edman法在蛋白测序仪上测出SFL的N端肽链 30个氨基酸的排列顺序为 :T/A/VDXLXFTFSDFDP NGEDLLFQGDAHVTSNN。  相似文献

白天麻(Gastrodia elata)顶生球茎中分离并纯化了一种抗真菌蛋白(Gastrodia Antifungal Protein),简称GAFP。在马铃薯葡糖琼脂培养基上,4μg GAFP滴加在直径0.6 cm圆纸片上可明显抑制木霉(Trichoderma reesei)菌丝生长。从每千克鲜球茎中可分离得GAFP约20 mg。用SDS-PAGE和凝胶过滤层析测得该蛋白为单多肽链,分子量14.0kD;用离子交换结合法测得电点为8.1;富含Asn,Ala,Gly和Leu,但无Met,Cys和Pro。未经热变性或SDS处理的GAFP与考马氏亮蓝试剂不发生显色反应。GAFP不具有几丁质酶和β-1,3-葡聚糖酶活性。球茎外层组织富含这种蛋白,内层薄壁组织无此蛋白。认为GAFP在阻止真菌侵染当年生顶生和侧生球茎的防卫机制中起重要作用。  相似文献

利用嗜硫色谱和POROS HQ20离子交换色谱从蕲蛇蛇毒中纯化得到一种新型P-Ⅲ型蛇毒金属蛋白酶(snake venom metalloproteases SVMP) AA-ACA-I.该酶经SDS-PAGE测得分子量为 47.6 kD,含有链内二硫键,加DTT处理后,分子量为50.8 kD,中性糖含量为3.5%,具有出血毒性和抗凝血活性.在70℃和pH 9.0条件下保温60 min,该酶会自降解成分子量30 kD左右的新蛋白,降解得到的新蛋白抗凝血活性高于原蛋白  相似文献

Bacillus sphaericus strain Ts-1 is highly insecticidal to larvae of the mosquito. It's insecticidal component is toxic proteins. The toxin was extracted from spore-crystal complexes by disruption in a Sonicator Cell Disruptor Model W-220F followed by treatment with 0.05 mol/L NaOH. Fraction recovered from chromatography of the spore-crystal complexes on column of Sephadex G-200 were assayed against mosquito larvae and the toxic fractions from gel chromatography were subjected to SDS-PAGE. The toxic proteins in B. sphaericus Ts-1 spore-crystal complex migrated in position corresponding to 42kD and 43kD. Bioassay of the two purified proteins prepared by PAGE indicated that they were all toxic to mosquito larvae. Toxic protein was further purified by DEAE-cellulose chromatography. The toxic protein with a molecular weight of 42kD was obtained.  相似文献

Two novel type I ribosome-inactivating proteins (RIPs) were found in the storage roots of Mirabilis expansa, an underutilized Andean root crop. The two RIPs, named ME1 and ME2, were purified to homogeneity by ammonium sulfate precipitation, cation-exchange perfusion chromatography, and C4 reverse-phase chromatography. The two proteins were found to be similar in size (27 and 27.5 kD) by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and their isoelectric points were determined to be greater than pH 10.0. Amino acid N-terminal sequencing revealed that both ME1 and ME2 had conserved residues characteristic of RIPs. Amino acid composition and western-blot analysis further suggested a structural similarity between ME1 and ME2. ME2 showed high similarity to the Mirabilis jalapa antiviral protein, a type I RIP. Depurination of yeast 26S rRNA by ME1 and ME2 demonstrated their ribosome-inactivating activity. Because these two proteins were isolated from roots, their antimicrobial activity was tested against root-rot microorganisms, among others. ME1 and ME2 were active against several fungi, including Pythium irregulare, Fusarium oxysporum solani, Alternaria solani, Trichoderma reesei, and Trichoderma harzianum, and an additive antifungal effect of ME1 and ME2 was observed. Antibacterial activity of both ME1 and ME2 was observed against Pseudomonas syringae, Agrobacterium tumefaciens, Agrobacterium radiobacter, and others.  相似文献

通过亲和层析和羧甲基一纤维素离子交换层析从萝卜的块根中分离到两个具溶菌酶活性的酶组份:CBP1和CBP2。两者经SDS-PAGE均显示单一蛋白染色条带,其对应的分子量分别为26.9kD和24.8kD。两种蛋白除有溶菌酶活性外,还有几丁质酶活性,但无壳聚糖酶活性。各种类型的几丁质对CBP1和CBP2都有较强的吸附作用,而在还原/非还原的单向SDS-PAGE中却观察不到两者分子中存在二硫键。  相似文献

试验以-1.2MPaPEG6000处理动小麦种子(TriticumaestlivumL.).SDS-PAGE图谱分析表明,水分胁迫诱导幼芽及整株均产生48.4kD、41.5kD二个蛋白质亚基。在幼根中未出现以上二个蛋白亚基。胁迫48h后,根干重/芽干重比呈上升趋势,幼芽细胞膜楔对透性增大和相对含水量降幅度均大于幼根。  相似文献

Three-day-old maize seedlings were subjected to 100 μM AlCl3 for 24 h. Cytoplasmic proteins were isolated from root tips, root base and from coleoptiles. After fractionation of cytoplasmic proteins on anion chromatography column Bio-Scale Q2 sodium dodecylsulphate polyacrylamide gel electrophoresis (SDS-PAGE) analysis was used to monitor Al-induced changes in polypeptide composition of particular fractions. Four (root) and 7 (coleoptile) fractions were eluted from the column with linear 0 - 1.0 M NaCl gradient. In fraction 1 of cytoplasmic proteins from root tips Al induced accumulation of polypeptide with molecular mass of 16 kD and simultaneous reduction of two polypeptides (67.5 and 60 kD). In fraction 1 isolated from mature zone of maize roots Al-induced accumulation of 22 kD polypeptide and reduction of 67.5, 60, and 14 kD polypeptides. Most pronounced changes were revealed in coleoptile. In three protein fractions increased accumulation of polypeptides with molecular mass of 14, 17.5, 20, 24.5, 28, 30, and 37.5 kD were observed. In the remaining three root or four coleoptile fractions of cytoplasmic proteins, no differences were found between Al-treated and control maize seedlings. 1. Dianthin 30 and dianthin 32, two proteins isolated from the leaves of Diathus caryophyllus (carnation), were purified to homogeneity by chromatography on CM-cellulose. 2. The mol.wt. of dianthin 30 is 29 500 and that of dianthin 32 is 31 700. Both dianthins are glycoproteins containing mannose. 3. Dianthins inhibit protein synthesis in a lysate of rabbit reticulocytes, with an ID50 (concentration giving 50% inhibition) of 9.15 ng/ml (dianthin 30) and 3.6 ng/ml (dianthin 32). They act by damaging ribosomes in a less-than-equimolar ratio. Protein synthesis by intact cells is partially inhibited by dianthins at a concentration of 100 microgram/ml. 4. Dianthins mixed with tobacco-mosaic virus strongly decrease the number of local lesions on leaves of Nicotiana glutinosa.  相似文献

利用硫酸铵沉淀、羟基磷灰石柱层析、Sephadex G-75凝胶过滤和DEAE-52离子交换柱层析的方法,将枯草芽孢杆菌SA-22 β-甘露聚糖酶纯化了30.75倍,同时,该酶比活达到3478056 u/mg,收率达到23.43%。利用SDS-PAGE凝胶电泳和Sephadex G-75凝胶过滤的方法测得枯草芽孢杆菌SA-22 β-甘露聚糖酶的分子量分别为38 kD和34 kD。实验发现该酶的最适pH为6.5,在pH 5~10的范围内稳定;该酶最适温度为70℃,在50℃保温4h后其活力不变,在60℃保温4 h后剩余酶活为74.2%,70℃的酶活半衰期为3h。实验还发现Hg2+对酶活力有明显抑制作用。该酶对槐豆胶和魔芋胶的KmVmax值分别为11.30mg/mL, 4.76mg/mL和188.68(μmol·mL-1·min-1), 114.94(μmol·mL-1·min-1)。  相似文献

Seeds of soybean (Glycine max L. Merr.) harvested at variousstages of development and allowed to dry in intact pods undergoa maturation process and are viable. Defatted powders of seedharvested 24–66 d after flowering were extracted to yieldbuffer-soluble and alkali-soluble proteins. Imposition of amaturation process increased the level of buffer-soluble proteinsbut had no effect on the disulflde content of these proteins.After undergoing maturation, seeds showed an accumulation ofbuffer-soluble polypeptides in the molecular weight range of43–94 kD. Maturation may be associated with the synthesisof specific polypeptides having a molecular weight of approximately85 kD. Alkali-soluble proteins, which represents the storageproteins, did not show any responses to maturation. Their quantityincreased substantially during seed development and the disulfidelevel was only half that of buffer-soluble proteins, attaininga maximum value of 10.9 mol S per 105 g protein. Matured seedat all harvest dates had a final starch content close to thatof normal seed, 10–20 mg g–1, and soluble sugarswere maintained at quite high levels, 51–83 mg g1.The metabolic program for synthesis and degradation of starchseems quite rigidly followed and is independent of harvest dateor of attachment to the parent plant. Soybean seeds retain considerablesoluble proteins and soluble sugars throughout maturation, andthese collectively may be important in maintaining a desiccationresistant structure.  相似文献

Plasma membrane (PM) of high purity was prepared through a two-phase aqueous system from roots of 3 ~ 4 d old com ( Zea mays L. ) seedling, and within which calcium-activated protein kinase was identified. The kinase was mainly located in the inner side of the PM with a semi-activation concentration to free calcium ion of 50 μmol/L. Exogenous CaM had no agonistic effect on the protein kinase but CaM antagonist TFP could strongly inhibit its activity with a semi-inhibition concentration of 75 μmol/L. The protein kinase showed high specificity for the exogenous substrate, Histone Ⅲ S. These results indicated that this protein kinase could be a calcium-dependent and calmodulin-independent protein kinase (CDPK). Moreover, two proteins (33 kD and 58 kD) in PM might be the endogenous substrates of the CDPK.  相似文献
