食品和饲料的生物催化转化和发酵技术成为保障餐桌安全及绿色发展的重要领域,其用酶量在工业酶制剂市场中占比最大,且增长最快。性能优良的酶制剂能显著缩短发酵周期,降低生产成本,提高目标产物的转化率,进而提高市场竞争力。食品和饲料加工产业除了应用分离提取的酶制剂外,很多发酵过程仍以微生物为细胞工厂进行多种酶催化转化。该发酵过程通常涉及复杂的代谢网络和调控机制,且很多体系不能采用基因工程技术,因此理性基因工程改造通常难以满足应用需求。诱变育种仍然是食品和饲料加工用酶及微生物菌株改造的主要手段,需要与时俱进不断发展新技术。近年发展起来的常压室温等离子体(ARTP)诱变育种技术因其具有操作简单、安全、环境友好、突变率高、突变库容大等特点,在食品和饲料加工微生物育种中得到了广泛应用,取得了良好的成效。重点介绍近年ARTP在食品与饲料加工酶制剂及发酵微生物改造中的相关应用进展。 相似文献

生物发酵是以微生物菌种为生物催化剂,以淀粉糖、生物质等可再生资源为原料发酵生产各种食品、化学品、燃料、材料等物质的生产过程,具有绿色、低碳和可持续等特征。我国拥有全球规模最大的生物发酵产业,尤其氨基酸、维生素等传统发酵产品占全球市场份额的60%–80%。发展生物发酵产业对于我国实现“碳中和、碳达峰”的战略目标和生物经济发展具有重要的意义。微生物工业菌种是生物发酵产业的核心,直接影响原料路线、产品种类和生产成本。创新发酵工业菌种,提升其原料转化利用效率,提高产物生产水平,拓展产品种类,是生物发酵产业高质量发展的关键。近年来,合成生物学、系统生物学等学科的发展,进一步加深了研究者对微生物底盘细胞生理代谢机制的理解,加速了基因编辑等菌种设计创制使能技术的发展,为发酵工业菌种改造提升提供了新动能。本文选取了具有代表性的大宗氨基酸、B族维生素、柠檬酸、燃料乙醇等发酵产业,从其工业微生物底盘的基础研究和技术开发角度,综述发酵工业菌种改造提升的最新进展,并展望人工智能、自动化与生命科学交叉融合将对工业菌种迭代产生的重要影响。 相似文献

氨基酸是蛋白质的基本组成单元,对人和动物的营养健康十分重要,广泛应用于饲料、食品、医药和日化等领域。目前,氨基酸主要通过微生物发酵可再生原料生产,氨基酸产业是我国生物制造的重要支柱产业之一。氨基酸菌株主要通过随机诱变和代谢工程改造结合筛选获得。菌株生产水平进一步提高的核心限制之一是缺乏高效、快速和准确的筛选方法,因此,发展氨基酸菌株的高通量筛选方法对关键功能元件挖掘及高产菌株的创制筛选至关重要。本文综述了氨基酸生物传感器的设计,及其在功能元件、高产菌株的高通量进化筛选和代谢途径动态调控中的应用研究进展,讨论了现有氨基酸生物传感器存在的问题和性能提升改造策略,并展望了开发氨基酸衍生物生物传感器的重要性。 相似文献

工业生物技术是以微生物细胞工厂利用可再生的生物原料来生产能源、材料与化学品等的生物技术,在解决资源、能源与环境等问题方面起着越来越重要的作用。系统生物学是全面解析微生物细胞工厂及其发酵过程从"黑箱"到"白箱"的重要研究方法。系统生物学借助基因组、转录组、蛋白质组、代谢组以及代谢流组等多组学数据,可解析微生物细胞工厂在RNA、蛋白与代谢物等不同水平上的变化规律与调控机制。目前,系统生物学在微生物细胞工厂的设计创建与发酵工艺优化中起着越来越重要的指导作用,许多成功应用实例不断涌现,推动着工业生物技术的快速发展。文中重点综述基因组、转录组、蛋白质组、代谢组与代谢流组以及基因组规模的网络模型等各组学技术的最新发展及其在工业生物技术尤其是菌株改造与发酵优化中的应用,并就工业生物技术中系统生物学的未来发展方向进行展望。 相似文献

生物燃气俗称沼气,是微生物群体在厌氧条件下协同发酵可降解有机废弃物的产物,传统能源供应的萎缩和增加可再生能源在能源消费中份额的需求使沼气的重要性越来越突出。经过不断研究与工程实践,已经开发出了不同的发酵工艺处理生活垃圾和工农业生产废弃物等有机质生产生物燃气,其中全混式中温发酵占主导地位,欧洲的技术处于领先水平,特别是德国的沼气发电、热电联产。结合作者多年沼气研究积累的经验,综述了沼气技术的最新进展,包括厌氧发酵菌群、消化反应器结构和发酵工艺,沼气生产和应用等,指出了今后发展的重点和方向。 相似文献

Adhesion properties of microorganisms are crucial for many essential biological processes such as sexual reproduction, tissue

in liquid environments is a process of asexual aggregation of cells which is also referred to as flocculation. This process
has been the subject of significant scientific and biotechnological interest because of its relevance for many industrial
fermentation processes. Specifically adjusted flocculation properties of industrial microorganisms could indeed lead to significant
improvements in the processing of biotechnological fermentation products such as foods, biofuels and industrially produced
peptides. This review briefly summarises our current scientific knowledge on the regulation of flocculation-related phenotypes,
their importance for different biotechnological industries, and possible future applications for microorganisms with improved

Microbial surfactants are amphipathic molecules that consist of hydrophilic and hydrophobic domains, which allow partition of two fluid phases of varying degree of polarity. They are classified into two main groups: bioemulsifier and biosurfactant, depending on their molecular weight. Microbial surfactants occur in various categories according to their chemical nature and producing organisms. These biomolecules are produced by diverse groups of microorganisms including fungi, bacteria, and yeasts. Their production is significantly influenced by substrate type, fermentation technology and microbial strains. Owing to inherent multifunctional properties and assorted synthetic aptitude of the microbes, microbial surfactants are mostly preferred than their chemical counterparts for various industrial and biomedical applications including bioremediation, oil recovery; as supplements in laundry formulations and as emulsion-stabilizers in food and cosmetic industries as well as therapeutic agents in medicine. The genus Gluconobacter comprises some of the most frequently used microorganisms when it comes to biotechnological applications. Not only has it been involved in "historical" production processes, such as vinegar production, but in the last decades many bioconversion routes for special and rare sugars involving Gluconobacter have been developed. Among the most recent are the biotransformations involved in the production of L-ribose and miglitol, both very promising pharmaceutical lead molecules. Most of these processes make use of Gluconobacter's membrane-bound polyol dehydrogenases. However, recently other enzymes have also caught the eye of industrial biotechnology. Among them are dextran dextrinase, capable of transglucosylating substrate molecules, and intracellular NAD-dependent polyol dehydrogenases, of interest for co-enzyme regeneration. As such, Gluconobacter is an important industrial microbial strain, but it also finds use in other fields of biotechnology, such as biosensor-technology.
