不同产地西洋参皂甙成分的HPLC分析   总被引:4,自引:1,他引:3  
以西洋参的主要皂甙成分人参皂甙Re和Rb1为标准对照品 ,建立西洋参药材的HPLC定量分析技术 ,并参考人参皂甙Rc,Rd及Rg2 的相对峰面积进行主成分分析。色谱条件为 :C18柱 (5 μm ,3.9× 15 0mm) ,乙腈 :水流动相 ,二元梯度洗脱 ,检测波长 2 0 3nm。结果表明 ,就皂甙成分的组成与含量而言通过人参皂甙Re和Rb1的含量测定和皂甙的主成分分析 ,不同产地的西洋参药材皂甙成分存在一定的差别。吉林省靖宇县产的西洋参与进口品最为接近。  相似文献

本文旨在探究不同诱导子对西洋参愈伤组织生长、相关酶活性及其人参皂苷含量的影响,在西洋参愈伤组织中,应用HPLC检测了添加诱导子茉莉酸甲酯(methyl jasmonate,MeJA)和水杨酸(salicylic acid,SA)后西洋参愈伤组织皂苷生物合成的变化。结果显示,MeJA抑制生长,但SA对其生长影响较小;2种诱导子均可以显著激活西洋参愈伤组织中超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)、过氧化物酶(peroxidase,POD)活性和丙二醛(malondialdehyde,MDA)含量;培养基中MeJA浓度为100μmol/L时,愈伤组织中总皂苷含量和产量均达到最大值,人参皂苷Rg_(1)、Re、Rb_(1)、Rc的含量最高,在SA诱导子试验组中,当SA浓度为300μmol/L时,西洋参愈伤组织中总皂苷含量和产量均达到最大值,人参皂苷Rb_(1)的含量最高。结果表明,在西洋参愈伤组织中添加适当浓度MeJA和SA诱导子会提高人参皂苷化合物含量,其中以MeJA诱导人参总皂苷的效果最好,同时也会影响西洋参愈伤组织的生长量。说明在培养过程中添加外源诱导子,有利于提高西洋参愈伤组织中皂苷含量。  相似文献

三七.人参和西洋参细胞悬浮培养的比较研究   总被引:19,自引:1,他引:18  
用薄层层析对三七、人参和西洋参愈伤组织进行的初步鉴定表明,三种愈伤组织都含有皂甙和主要皂甙成分Rb_1、Rg_1,三七愈伤组织还含有一种抗癌皂甙Rh_1。对愈伤组织的生长,三七低于人参高于西洋参;对愈伤组织中总皂甙含量,三七均高于人参和西洋参。三种植物细胞悬浮培养结果类似于他们的愈伤组织培养,但生长又进一步提高。三七细胞悬浮培养中皂甙产生的时间进程几乎与生长平行,合适的收获期为培养30天。寡糖素不仅增强三七培养细胞的皂甙形成而且促进细胞生长,较合适的浓度为1.25 ppm。通过以上研究,使三七悬浮培养细胞的生长(干重增加178毫克)为最初培养愈伤组织的4倍以上,总皂甙产率高达20.6毫克,为最初培养愈伤组织的8.5倍。  相似文献

西洋参总皂苷经β-糖苷酶催化水解,采用HPLC检测分析确定西洋参总皂苷中的主要原人参二醇型皂苷Rb1、Rd、Rc和Rb2已经完全被水解。水解产物通过反复硅胶柱层析和反向硅胶柱层析分离纯化得到7个皂苷,通过NMR谱图分析分别鉴定为人参皂苷compound K(1)、人参皂苷Mc(2)、人参皂苷Rg1(3)、人参皂苷Rg2(4)、人参皂苷Re(5)、人参皂苷F1(6)和拟人参皂苷F11(7)。β-糖苷酶催化西洋参总皂苷水解实验表明,西洋参中原人参二醇型皂苷的水解产物是人参皂苷compound K和人参皂苷Mc。  相似文献

人参皂甙Rb1,Rg1,Re和Rh1对体外培养细胞增殖的影响   总被引:3,自引:0,他引:3  
本研究应用体外培养的人胚肺成纤维细胞(2BS)和人宫颈癌细胞(Hela)为实验模型。在培养液中分别加入人参根总皂甙(SRG)和人参皂甙Rb_1、Rg_1、Re、Rh_1孵育3~7d后,用MTT法和细胞计数法测定细胞的增殖。MTT测定结果表明,Rb_1、Rg_1、Re、Rh_1可以促进衰老细胞的增殖,但对未衰老的细胞增殖没有显著影响,对Hela细胞的增殖有抑制作用。细胞计数法测定结果表明,总皂甙和4种皂甙单体都可以降低Hela细胞的群体增殖率和克隆生长率,其中Re、Rh_1作用显著。同时,增加衰老细胞的群体增殖率和克隆生长率,其中Rb_1和Rg_1作用显著。  相似文献

采用TLC和HPLC方法分析比较西洋参(Panax quinquefolium L.)、人参(P.ginseng C.A.Mey.)及其加工品红参(red ginseng),以及不同规格的西洋参中人参皂甙的含量。结果表明,西洋参中人参皂甙总量及人参二醇型皂甙的含量明显高于人参及红参,且含有1种人参及红参中未发现的未知人参皂甙Rx,但不含人参及红参中含有的Rf;人参中人参二醇型皂甙的含量高于人参三醇  相似文献

本文采用Shibata提取方法,以人参皂甙Re为标准品,通过对样品提取液的薄层分离,用香草醛—高氯酸比色法对不同产地、不同年生、不同规格的西洋参及其不同部位中的总皂甙和分组皂甙作了含量测定。结果表明:各种西洋参样品总皂甙含量差异较大;三醇型与二醇型皂甙含量比为1:2.93~1:3;花蕾皂甙含量最高。  相似文献

我们从三七(Panax notoginseng(Burk.)F.H.Chen)根中分离得皂甙A和皂甙C。根据熔点、比旋光度及元素分析、红外、核磁证明皂甙A与S.Shibata等从同属植物人参(Panax ginseng)中分离得到的Ginsenoside-Rg_1(6,20—di—O—β—D—glucosyl—20S—Protopanaxatriol)是同一化合物;皂皂甙C与Ginsenoside—Rb_2(20S—Proto-panaxadiol—3—[O—β—D—glucopyranosyl(1→2)—β—D—glucopyranoside]—20—[O—α—L—arabino-pyranosyl(1→6)—β—D—glucopyranoside])是同一化合物。  相似文献

从三七叶乙醇提物中分离到16个化合物,分别鉴定为人参皂甙(ginsenoside)Rh2(1),F2(4),Rg3(5),Rg1(7),Rd(8),Re(11),Rb3(13),Rb1(14),Rc(15),七叶胆皂甙(gypenoside)XIII(2),IX(9),XVII(10),三七皂甙(noto-ginsenoside)R1(12),Fa(16),甘草素(liquiritigenin)(3),以及芹糖甘草甙(liquiritin apioside)(6)。其中化合物1,2,3和6首次分离自该植物中,化合物1~8及10~12首次从三七叶中分离到。研究结果进一步证实,三七叶以含原人参二醇型皂甙为主,同时含有微量的原人参三醇型皂甙和黄酮类化合物。  相似文献

本文采用MPG-ODS色谱柱,以醋酸铵作HPLC流动相的改性荆在15 min以内较好地分离出单体皂甙Rg_2、Rb_1、Rc、Rd、Rg_1、Re等,首次比较了化学上醇提取方法和食用时水提取方法对单体皂甙及总皂甙提取效果的不同,结果表明,食用水对Rg组类皂甙的提取量高于Rb组,这将为人参的药理学研究及临床食用提供科学依据。  相似文献

In order to determine the active ingredients in root extracts of Panax quinquefolius (American ginseng), a gradient HPLC method involving UV photodiode array detection was applied to separate and quantify simultaneously the ginsenosides Rb1, Rb2, Rc, Rd, Re, Rf and Rg1. All ginseng saponins were baseline-resolved under the selected conditions, and the detection limits were 1.0 microg/mL or less. The method has been applied to analyse ginsenosides extracted from American ginseng cultivated in both Wisconsin and Illinois. Ginsenosides Re and Rb1 were the two main ginseng saponins in the root. The amounts of Re in 5- and 7-year Illinois-cultivated samples were greater than those found in ginseng cultivated for 3 or 4 years in Wisconsin, whereas the levels of Rb1 were greater in the younger Wisconsin samples.  相似文献

目的:探讨人参皂苷Rb1、Rg1、Re对急性髓系白血病细胞株(KG1α)增殖的影响.方法:取对数生长期KG1α细胞,分设人参皂苷Rb1、Rg1、Re组和常规培养组,以MTT比色法检测作用24h、48h、72h时对KG1α细胞增殖抑制作用,并计算Rb1的IC_(50)值,以此浓度为工作浓度,设常规培养组和处理组,台盼蓝计数法观察对KG1α细胞增殖的影响;流式细胞术测定细胞周期分布的变化.结果:MTT、台盼蓝计数显示人参皂苷单体Rb1、Rg1可抑制KG1α细胞增殖,呈浓度依赖性,以Rb1抑制效应最佳,于作用48h抑制率最高.台盼蓝计数显示人参皂苷单体Rb1-120μmol/L作用48h时抑制率达50.22%;流式细胞术结果提示,与对照组比较,Rb1-120μmol/L组G_2/M期KG1α细胞比例增加(P<0.05).结论:Rb1可抑制KG1α细胞体外增殖,其增殖抑制作用与将KG1α细胞阻滞于G_2/M期有关.  相似文献

为探究人与大鼠肠道菌群对三七水煎液中三醇型人参皂苷Rg1、Re及二醇型人参皂苷Rb1、Rd体外代谢的差异性及发现其代谢产物原人参二醇PPD与原人参三醇PPT,实验利用UPLC方法测定三七水煎液分别与人、大鼠肠道菌群在厌氧条件下共培养24h后的孵育液中4种皂苷的含量及代谢产物PPD与PPT的含量。结果表明三七中含有三醇型人参皂苷Rg19.4500mg/g、Re1.8872mg/g,二醇型人参皂苷Rb18.5816mg/g、Rd1.9456mg/g。与人源肠道菌共培养后,三七中含有的二醇型、三醇型人参皂苷含量显著降低,重要的是,在培养液中检测到代谢产物PPD和PPT的存在,含量分别为0.2136mg/g及0.0344mg/g,与大鼠肠道菌共培养后,三七中含有的二醇型皂苷含量有轻微降低,而三醇型皂苷含量未见明显变化,但有少量PPT(0.0184mg/g)的生成。由此可见:在体外条件下,三七水煎液中人参皂苷会被人肠道菌群降解生成代谢产物PPD和PPT,而大鼠肠道菌群的降解产物却仅有PPT生成,二者存在种属差异。  相似文献

Utx is a candidate tumor suppressor gene that encodes histone H3 lysine 27 (H3K27) demethylase. In this study, we found that ectopic expression of Utx enhanced the expression of retinoblastoma tumor suppressor gene Rb and its related gene Rbl2. This activation was dependent on the demethylase activity of Utx, and was suggested to contribute to the decreased cell proliferation induced by Utx. A chromatin immunoprecipitation assay showed that over-expressed Utx was associated with the promoter regions of Rb and Rbl2 resulting in the removal of repressive H3K27 tri-methylation and the increase in active H3K4 tri-methylation. Furthermore, siRNA-mediated knockdown of Utx revealed the recruitment of endogenous Utx protein on the promoters of Rb and Rbl2 genes. These results indicate that Rb and Rbl2 are downstream target genes of Utx and may play important roles in Utx-mediated cell growth control.  相似文献

羽叶三七(Panax japonicus C. A. Meyer var. bipinnalifidus (Scem.) Wu et Feng)又称疙瘩七,产我国西北部至西南部山区,是人参届植物中分布海拔和纬度均较高的一个种类。在陕西省秦岭地区主要产于南北坡海拔2100—2900米的针叶林下阴湿处。民间以其根茎入药,具有清热解毒、顺气健胃、活血祛瘀、滋补强壮之效。作为国产人参属植物皂甙成分系统研究的一个部分,本文报告秦岭产羽叶三七根茎的皂甙成分,并讨论其化学分类学意义。  相似文献

Some plant extracts and products are known to affect mammalian cells, tissues and organisms as they contain a toxic substance or a metabolic stimulant. Our biochemical investigations revealed that some plant saponins can increase the cellular DNA repair activity and the general recombinase activity measured by in vitro assay (1). In the experiments described here, HeLa cells were cultured for several days with plant saponins or flavonoids and analyzed to measure i) recombination activity of the cell extract by induction of Tcr colonies from two mutant DNAs (mutants 1 and 2, which are both tetracycline sensitive) after transformation into E. coli recA-, and ii) repair synthesis of nuclear DNA followed by incorporation of 3H-thymidine. Saikosaponins a, b1, d, ginsenosides Rb1, Re, Rh and flavonoid baicalin caused a significant stimulation of intermolecular recombination. It is worth noting that none of the plant saponins and flavonoids had any inhibitory or toxic effect at concentrations less than 25 micrograms/ml in the culture media.  相似文献

Ginseng saponins (ginsenosides) were isolated from soil associated with the roots of commercially grown American ginseng (Panax quinquefolius L.), identified via LC-MS and quantified via analytical HPLC. The ginsenosides, including F(11), Rb(1), Rb(2), Rc, Rd, Re and Rg(1), represented between 0.02 and 0.098% (average 0.06%) of the mass of the soil collected from roots annually between 1999 and 2002. The same ginsenosides were also isolated from run-off of undisturbed plants grown in pots in a greenhouse using a root exudate trapping system. To investigate (1) whether these saponins could influence the growth of pythiaceous fungi pathogenic to ginseng, and (2) whether soil levels of ginsenosides were sufficient to account for any effects, bioassays were completed using a crude saponin extract and an ecologically relevant concentration of purified ginsenosides. Thus, when cultured on media containing crude saponins, the colony weight of both Phytophthora cactorum and Pythium irregulare was significantly greater than that of control, indicating a strong growth stimulation by ginsenosides. The growth of Pythium irregulare was also significantly stimulated after addition of an ecologically relevant, low concentration (i.e. 0.06%) of purified ginsenosides to culture medium. By contrast, growth of the saprotrophic fungus Trichoderma hamatum was slightly (but not significantly) inhibited under the same conditions. These results imply that ginsenosides can act as allelopathic stimulators of the growth of pythiaceous fungi in the rhizosphere, and this may contribute to the disease(s) of this crop.  相似文献

为了测定西洋参总皂苷及单体皂苷是否有降脂活性,通过体外实验分别测定脂肪分解活性、小肠刷状缘膜小囊吸收脂肪酸、三油酸甘油酯油酸的释放率。结果表明,西洋参茎叶总皂苷在0.5g/L浓度时,对胰脂肪酶活性的抑制率为90%;人参皂苷Rc,Rb1,Rb2对胰脂肪酶活性均显示很强的抑制作用,在0.5g/L浓度时抑制率分别为100%,96%,97%。西洋参总皂苷和人参皂苷Rc,Rh,Rb2可以通过抑制胰脂肪酶活性起到降脂作用。  相似文献
