2011和2012年山东栖霞、蓬莱、沂源等地套袋苹果果实出现了一种红褐色病斑。从病斑上分离获得两株枝顶孢属Acremonium真菌,其中一株产生头状排列的分生孢子,另一株产生链状排列的分生孢子。2个菌株都能从伤口侵染近成熟期的苹果果实,导致果肉细胞坏死,表面凹陷,形成红褐色坏死斑。2个菌株都不能侵染无伤果实。2个菌株的ITS、β-微管蛋白基因、核糖体大亚基(nucLSU)和小亚基(nucSSU)的DNA序列完全相同。依椐两个菌株的ITS和nucLSU的DNA序列和形态特征,将2个菌株鉴定为菌核生枝顶孢Acremonium sclerotigenumA. sclerotigenum是一种机会真菌,条件适宜时可侵染套袋的苹果果实,导致褐色坏死病斑,称之为枝顶孢褐点病。  相似文献

对富士苹果果实进行双层纸袋套袋处理,通过分析袋内微域环境变化,研究套袋微域环境对果皮扫描与透射结构的影响。结果表明:套袋内微域的黑暗环境导致果皮的光合能力缺失,表皮第一层细胞内容物减少,缺少脂质体,取而代之的是游离的脂体小球,说明表皮细胞形成角质层的物质来源缺乏,导致角质层变薄。与外界比较,套袋内昼夜具较高的温度与湿度,形成所谓的"小温室"环境,而且,由于受套袋的保护,使果实免受外界环境的直接刺激,导致套袋果实的果面光洁平滑,果点小且色淡,蜡质层龟裂均匀,且裂口深度远小于未套袋果;但摘袋后果面龟裂产生的裂纹频度增大,说明环境的改变影响蜡质层龟裂。研究结果从果皮结构的变化为套袋果实外观品质的变化提供了理论依据,同时为果实摘袋后的补钙实施提供了参考依据。  相似文献

构树Broussonetia papyrifera为桑科重要的资源植物。为调查逆境构树内生真菌的多样性,我们对铅锌矿区构树根、茎、叶和果实各部位进行内生真菌分离鉴定,从中分离得到187株内生真菌,根据形态学与分子生物学方法鉴定为20属,其中链格孢属Alternaria(50.27%)、镰孢属Fusarium(11.76%)为优势属。不同培养基的内生真菌分离率结果表明,马铃薯葡萄糖琼脂培养基(PDA)分离出的菌株数最多,占总数的73.96%;构树不同部位的菌株分离数目显示,叶中分离出的菌株较多,占38.02%。在属的分类水平上,根、茎、叶、果实不同部位的真菌数量也存在明显差异。Shannon-Wiener多样性指数和Simpson多样性指数分析结果提示不同样地的构树内生真菌的多样性也存在差异。Chao1指数和ACE指数也提示构树内生真菌丰富度与样地和组织有关。  相似文献

为揭示小麦种子所携带真菌的种类和多样性,丰富小麦相关微生物的可利用资源,我们对来自俄罗斯、澳大利亚、哈萨克斯坦、加拿大以及中国的57份小麦种子样品进行内携真菌分离并对其中的21份种子利用高通量测序进行多样性分析。结果显示,分离获得564株真菌,经ITS序列分析结合形态观察鉴定为31属69种,其中菌株数量相对多度最高的5个属为链格孢属Alternaria(51%)、核腔菌属Pyrenophora(6%)、附球菌属Epicoccum(6%)、镰刀菌属Fusarium(5%)和青霉菌属Penicillium(5%);同时有节菱孢属Arthrinium、烟管菌属Bjerkandera、毛壳属Chaetomium和小鬼伞属Coprinellus等17个属的真菌是首次从小麦种子内获得分离培养;此外还从小麦种子中分离得到多种潜在的重要植物病原菌,如互隔链格孢Alternaria alternata、麦根腐平脐蠕孢Bipolaris sorokiniana和禾谷镰刀菌Fusarium graminearum等。根据高通量测序结果进行的多样性分析表明来自中国的样品仅与来自俄罗斯样本存在显著差异。高通量测序获得的小麦种子内携真菌的优势属为链格孢属Alternaria、枝孢属Cladosporium和附球菌属Epicoccum,相对多度分别为23.95%、18.91%、13.32%;对小麦种子内携真菌OTU的营养类型进行注释后发现植物病原菌类型的相对多度占到71.36%。本研究结果初步揭示了小麦种子内携真菌的组成和多样性,结果显示小麦种子是多种种传病害病原菌的潜在载体,内携多种潜在的植物病原真菌,有可能在后续生产中条件适合的情况下发生病害,本文为小麦种传病害的防治和研究提供了重要的材料和科学依据。  相似文献

套袋黄冠梨果实花斑病发生与其组织结构变化的关系   总被引:1,自引:0,他引:1  
以黄冠梨果实为试验材料,在调查套袋处理果实花斑病发生的基础上,研究了套袋及未套袋黄冠梨果实发育过程中的组织结构变化,并分析了发病果实的病部与正常果实组织结构差异。结果显示:(1)套3层袋果实花斑病的发病率为22.5%,而不套袋果实的发生率仅为1.5%。(2)未套袋果实角质层厚,而且呈波纹状分布,并渗入到表皮细胞间隙中,果实发育过程中厚度变化幅度不明显;套袋果实表面的角质层均匀一致,随果实迅速膨大,角质层变薄,且显著薄于未套袋果实,尤其是套3层袋与未套袋果实差异更大;套袋果实表皮细胞呈长方形,且较未套袋果实排列致密;不同套袋处理果实单宁细胞层数随果实发育逐渐减少,在花后90 d后,套袋处理与未套袋处理的厚度无显著差异。(3)套袋果实果肉薄壁细胞排列紧密,胞间隙小,其石细胞大小和密度均较未套袋果实小。(4)套袋果实花斑病发病部位角质层模糊,发病果实正常部位的角质层、表皮层厚度和单宁层均低于正常果实。研究表明,套袋黄冠梨果实表面组织结构发生较明显变化,它对梨果实花斑病的发生有重要影响。  相似文献

为揭示燕麦孢囊线虫孢囊定殖菌物的多样性,从青岛市选择3个罹患燕麦孢囊线虫病的冬小麦地,在返青期至成熟期,定时采集小麦根际燕麦孢囊线虫病土样,采用孢囊刺破法、单孢分离法或单菌丝分离法对定殖菌物进行分离纯化,并对定殖菌物进行形态鉴定和分子鉴定以及多样性分析。结果表明,从3个地点分离得到1 072个菌株,这些菌株归属于19目(含3个未定目)28科(含10个未定科)35属(含3个未定属),其中轮枝孢属Verticillium的相对多度最高,为25.2%,是优势属;镰孢菌属Fusarium、黑团孢属Periconia及链格孢属Alternaria的相对多度分别为14.9%、13.5%及13.4%。示范园样品的Margalef’s丰富度指数,Shannon-Wiener多样性指数及Pielou均匀度指数均最高,分别为4.30、2.516及0.755;家属区样品的Margalef’s丰富度指数最低,为3.18;而葛家屯样品的Shannon-Wiener多样性指数和Pielou均匀度指数最低,分别为2.077和0.654。3个地点定殖菌物群落组成的Jaccard相似性指数在0.486-0.607之间。  相似文献

本研究使用6种不同培养基对深圳大鹏湾6种珊瑚上的共附生真菌进行平板涂布法分离培养,结合ITS-rDNA基因序列特征和形态学特征进行鉴定来研究深圳大鹏湾海域珊瑚共附生真菌的多样性。共分离培养出457株真菌菌株,分属于7个属,分别为红酵母属Rhodotorula(180株)、曲霉属Aspergillus(170株)、木霉属Trichoderma(50株)、青霉属Penicillium(34株)、枝孢属Cladosporium(21株)、透孢黑团壳属Massarina(1株)和弯颈霉属Tolypocladium(1株),其中优势属为红酵母属Rhodotorula和曲霉属Aspergillus,占菌株分离总数的76.59%。单独鹿角珊瑚上分离到的共附生真菌数量和种类最多,为160株9种;在该种珊瑚上还分离出弯颈霉属Tolypocladium sp.,该属真菌此前从未在珊瑚共附生真菌的研究中分离出来。本研究还发现SDA培养基分离出的共附生真菌数量最多,CDA培养基的真菌数量最少,表明不同培养基分离的共附生真菌多样性存在差异。  相似文献

从云南“世界烟草品种园”10种烟草的根、茎、叶中分离得到199株内生真菌,根据rDNA-ITS系统发育分析鉴定为17属25种,其中格孢腔菌目Pleosporales内生真菌的种类和数量最多;茎点霉属Phoma、链格孢属Alternaria和镰孢菌属Fusarium为主要优势属,相对频率分别为25.1%、24.6%和11.6%,优势度Y值分别为0.251、0.172和0.104。烟草不同组织内生真菌的种群结构存在显著差异,分离自根的内生真菌的主要优势属为FusariumPhoma,Y值分别为0.235和0.123;分离自茎的内生真菌的主要优势属为PhomaAlternaria,Y值分别为0.186和0.155;分离自叶片的内生真菌的主要优势属为Alternaria,Y值为0.286。Phoma从烟草根茎叶中均可分离得到,而Alternaria只分布在地上部茎叶中,Fusarium只分布在根茎中,表明这3个优势属真菌对根茎叶组织的专化性不同。  相似文献

以富士苹果'长富2号'品系为试材,研究了套袋后果实生长发育过程中果皮色素含量、果肉糖含量及取袋后果皮色素、糖含量和PAL活性等品质形成因子的变化规律.结果显示:套用双层纸袋能显著降低红富士苹果果皮叶绿素含量,并以叶绿素a降低幅度较大,且与总叶绿素含量呈基本一致的下降变化趋势,而果皮类胡萝卜素降低幅度较小;果皮的花青苷含量取袋前显著低于对照,取袋后迅速上升,并在取袋后6 d超过对照,采收时超过对照1倍以上;果实PAL酶活性取袋后呈先升后降的趋势,取袋后4 d活性超过对照,取袋后6 d活性达到最高值;套袋不同程度地降低了果实可溶性总糖和还原糖含量,取袋后可溶性总糖、果糖和葡萄糖含量上升,蔗糖含量在取袋后4 d内迅速上升而后下降,但套袋果始终低于对照果.研究表明,套袋可以显著降低果皮中叶绿素的含量,促进花青苷积累,从而减少叶绿素对果实着色的干扰,提高果实外观品质,但同时也部分减少了果实总糖的含量,对果实内在品质有一定负面影响.  相似文献

为了了解红豆杉Taxus chinensis内生真菌的种类及分布特点,通过采样,分离,培养与鉴定,对安徽黄山红豆杉内生真菌的群落结构及生态分布进行了研究.结果共分离出14属107株内生真菌,其中青霉属Penicillium,曲霉属Aspergillus,交链孢属Alternaria和拟青霉属Paecilomyces为优势种群.同时内生真菌的多样性具有组织专一性,以根和皮中分布较多,分别是分离菌株总数的37.38%和26.17%.  相似文献

研究了光对湖景蜜露桃果实成熟的影响。用1层、2层和3层橘黄色果袋(透光率分别为27.0%、13.9%和8.2%)为完全花后(DAFB)50d的果实套袋。分别在111、114、117、120DAFB(果实硬熟期)和124DAFB(果实完熟期)测定乙烯、呼吸速率和果实品质。套1层和3层袋的果实乙烯生成速率高于其他处理。111和114DAFB时,未套袋果实呼吸速率最高,其余测定期1层套袋果实最低。完熟期未套袋果实£值较高,而色角ho值较低。果实硬熟期前未套袋果实硬度高于套袋果实,完熟期则是套3层袋果实较高。完熟期套1层袋果实TSS高于其他果实。套1层袋果实分别在硬熟期和完熟期生成了最高量的内脂类物质和γ-癸内酯。根据以上结果可以认为完全花后50d左右,用一层橘黄色果袋为果实套袋,可以生产具有丰富桃香气的高品质桃果实。适当的光照强度明显增加了桃果实果香型香气物质,尤其是γ-癸内酯的合成。  相似文献

以幸水梨为试材分不同时期进行套袋处理,采集各个发育时期的果实,对其果实品质、石细胞团的密度、大小、含量及几种相关酶活性进行了分析.结果表明:(1)不同时期套袋后,幸水梨的硬度、可滴定酸含量均比对照显著增加,可溶性固形物、可溶性总糖含量以及单果重均有不同程度的降低.(2)不同时期套袋果的石细胞含量显著低于对照;与对照果相比套袋果果实表面光洁,果点小而稀,外观品质明显改善.(3)果实石细胞团的密度在幼果期较高,随着果实的发育膨大,密度逐渐减小,成熟前1个月左右基本稳定,石细胞团的纵横径随果实发育先逐渐增大而后减小,石细胞含量也表现出先增加后减少的趋势,在花后第49 d达到最大值.(4)果实内苯丙氨酸解氨酶在果实发育初期的活性较高,随着果实发育逐渐降低;多酚氧化酶活性变化与苯丙氨酸解氨酶相似;过氧化物酶活性随果实的生长呈现先上升后下降的趋势,其活性峰值在盛花后第28 d,其后缓慢下降.(5)不同时期套袋处理后,苯丙氨酸解氨酶、过氧化物酶以及多酚氧化酶活性整体上均比对照降低,与石细胞含量呈正相关关系.  相似文献

A study was performed to assess the preference of fourteen mango cultivars for fruit flies and their management by bagging. So the choice of Tephritid flies to mango cultivars during fruiting phase is crucial. Fourteen different cultivars of mango viz., ‘Dusehri’, ‘Malda’, ‘Langra’ early cultivars, ‘Chaunsa’, ‘Fajri Klan’, ‘Sensation’ medium whereas ‘Sanglakhi’, ‘Retaul-12’, ‘Mehmood Khan’, ‘Tukhmi’, ‘Kala Chaunsa’, ‘Chitta Chaunsa’, ‘Dai Wala’ and ‘Sobey De Ting’ late cultivars were assessed for their suitability for fruit flies. The results indicate that the population density of fruit flies was higher on late cultivars like ‘Sanglakhi’ (20.61 percent), ‘Mehmood Khan’ (20.22 percent) and ‘Reutal-12’ (19.92 percent) were proved to be highly susceptible to fruit flies. Among these the cultivar ‘Reutal-12’ was selected being commercial and future cultivar for the management of fruit flies through bagging. The results reported that the attack of tephritid fruit flies and other insect pests were zero in bagged fruits as compared with control. It was further recorded that the bagged fruits has maximum average fruit weight i.e. 203.50 and 197.83 g per fruit was noted in those treatments where butter paper bag and brown paper bag was wrapped with better coloration as compared with un-bagged fruit with 159.5 g per fruit. Similarly, on an average fruit length were more i.e. 90.17, 91.33 mm in bagged fruit and 85.33 in un-bagged fruits. Furthermore, bagged fruits have zero incidence of disease with reduced fruit crack, fruit sunburn, mechanical damage, bird damage, fruit blemished and agrochemical residues on the fruit. So, it is concluded that the special attention should be given on ‘Reutal-12’ for the management of fruit flies when devising an IPM program for the control of fruit flies. Further, bagging has proved to be the good agricultural practices for the production of quality mango.  相似文献

Covering apple fruits with double layer waterproof bags to enhance fruit quality and evenness of blush colour is typical on many cultivars in Korea and Japan. Aminoethoxyvinylglycine (AVG) applied to unbagged apple fruits at 3–4 weeks before commercial harvest reduces ethylene production in the fruit, delays fruit ripening and reduces pre-harvest fruit drop. Spray application of AVG to trees of bagged apples should have no effect on apple ripening as there is␣no direct contact with the fruit and the translocation of AVG in apple trees is regarded as negligible. However, preliminary experiments suggested that AVG applied to trees of bagged apples reduced pre-harvest fruit drop in “Kotgetsu” apples. This study investigated the effect of spray treatments of 125 ppm of AVG on fruit drop, fruit ripening (firmness, starch conversion and soluble solids) and ethylene production to whole trees with bagged or unbagged “Kogetsu” fruit, as well as sprays of only the bagged or unbagged fruit on trees on two orchards. AVG applied to whole trees with unbagged apples reduced fruit drop from an average of 58.9% to 10.4%, delayed starch conversion and decreased ethylene production. AVG applied to whole trees with bagged fruit was equally effective in reducing pre-harvest drop, delaying fruit ripening and reducing ethylene production. Application of AVG to unbagged fruit only was nearly as effective as application to whole trees with unbagged fruit but application to bagged fruit only had no effect on fruit ripening or ethylene production. Application of AVG to bagged fruit only did reduce fruit drop to an average of 42.5% but this was not as effective as spraying unbagged fruit only or whole trees with bagged fruit. Developmental studies were made on the dewaxed thin-sectionedapple fruit cuticles of 10 Malus sylvestris Mill, cultivarsfrom 4 weeks before anthesis through fruit maturation and harvest.Cuticular development appears to correlate well with the generalgrowth of the fruit. However, no correlation exists betweenfruit size and cuticle thickness. Cuticular pores were evidentas early as 1 week before anthesis and transcuticular canalsbecame evident by 1 week following anthesis. Dewaxed thin-sectioned,as well as isolated, mature fruit cuticles of 16 cultivars andfour crab-apples consistently revealed the distinct presenceof ubiquitous pores and canals. Evidence is provided by lightmicroscopy and scanning electron microscopy (SEM). Measurementswere taken of cuticle thicknesses during development and ofcuticular pore dimensions, and calculations were made of poreand canal numbers. Fruit size alone is not directly indicativeof total pore numbers per surface area. Canal lengths are directlyrelated to the developmental thickness of the cuticle. No correlationwas found between the thickness of the mature cuticle and eitherthe number of pores present or the pore diameters. 以翠冠和黄金梨为试材,测定套袋和未套袋(对照)梨果实发育时期果实中蔗糖、葡萄糖、果糖和山梨醇含量以及蔗糖代谢相关酶酸性转化酶(AI)、中性转化酶(NI)、蔗糖合成酶(SS)和蔗糖磷酸合成酶(SPS)的活性,并对果实中糖组分积累与酶活性的关系进行了分析.结果表明:(1)两梨品种套袋果实在发育过程中蔗糖、葡萄糖、果糖、山梨醇和糖代谢相关酶活性变化趋势与对照基本一致,套袋果实糖含量均低于对照但差异不显著,而各相关酶活性在两类果实间差异表现各异.(2)在梨果实发育早期,果实中以分解酶类为主,糖分积累低;发育后期以合成酶类为主,糖分积累多.(3)两品种套袋和对照果实AI活性与葡萄糖含量均呈显著或极显著正相关,SS合成方向活性与蔗糖含量均为极显著正相关,且翠冠对照果SPS活性与蔗糖含量呈极显著正相关.可见,套袋通过提高果实发育早期转化酶(Inv)活性,降低果实后期蔗糖磷酸合成酶(SPS)、蔗糖合成酶(SS)的活性来影响糖分积累,从而影响梨果品质.  相似文献

Pepper (Capsicum spp.) fruits are covered by a relatively thick coating of cuticle that limits fruit water loss, a trait previously associated with maintenance of postharvest fruit quality during commercial marketing. To shed light on the chemical‐compositional diversity of cuticles in pepper, the fruit cuticles from 50 diverse pepper genotypes from a world collection were screened for both wax and cutin monomer amount and composition. These same genotypes were also screened for fruit water loss rate and this was tested for associations with cuticle composition. Our results revealed an unexpectedly large amount of variation for the fruit cuticle lipids, with a more than 14‐fold range for total wax amounts and a more than 16‐fold range for cutin monomer amounts between the most extreme accessions. Within the major wax constituents fatty acids varied from 1 to 46%, primary alcohols from 2 to 19%, n‐alkanes from 13 to 74% and triterpenoids and sterols from 10 to 77%. Within the cutin monomers, total hexadecanoic acids ranged from 54 to 87%, total octadecanoic acids ranged from 10 to 38% and coumaric acids ranged from 0.2 to 8% of the total. We also observed considerable differences in water loss among the accessions, and unique correlations between water loss and cuticle constituents. The resources described here will be valuable for future studies of the physiological function of fruit cuticle, for the identification of genes and QTLs associated with fruit cuticle synthesis in pepper fruit, and as a starting point for breeding improved fruit quality in pepper.  相似文献

The synthesis of anthocyanin, the xanthophyll cycle, the antioxidant system and the production of active oxygen species (AOS) were compared between red and non‐red apple cultivars, in response to either long‐term sunlight exposure (high light intensity) during fruit development, or to exposure of bagged fruits to lower light intensity late in fruit development. During fruit development of red and non‐red apples, the xanthophyll cycle pool size decreased much more in red apple peel late in development. With accumulation of AOS induced by long‐term sunlight exposure, enhancement of the antioxidant system was found. However, this change became significantly lower in red apple than non‐red apple as fruit developed, which might serve to accelerate the anthocyanin synthesis in red apple peel. When, late in fruit development, bagged fruits were exposed to sunlight, the accumulation of AOS was lower in red apple peel than in non‐red peel. This could be due to the higher anthocyanin concentration in the red peels. Meanwhile, compared with that in non‐red cultivar, the xanthophyll cycle and the antioxidant system in red apple peel were protected first but then down‐regulated by its higher anthocyanin concentration during sunlight exposure. In conclusions, red and non‐red apples peel possess different photoprotective mechanisms under high light conditions. The relationship between anthocyanin synthesis and the xanthophyll cycle, and the antioxidant system, depends on the light conditions that fruit undergoes.  相似文献

A common modern cultivation practice is bagging the fruit bunch if date palm (Phoenix dactylifera L.) which may influence fruit maturity and nutraceutical quality. Exposure of fruits to photooxidative stress induces changes in the endogenous concentrations of plant hormones and other metabolites, which may cause accelerated fruit maturity. This study was conducted to examine the effect of exposure to direct and indirect sunlight on date palm fruit development. The indirect sunlight treatment was simulated by fruit bunch bagging, a common practice in modern date production. The exposure of date palm fruits to direct sunlight-induced photooxidative stress causing an increased concentration of ascorbic acid and decreased content of chlorophyll, anthocyanins, carotenoids, and phenols compared to the fruit bagging treatment. Direct sunlight also reduced the concentration of phytohormones, including indoleacetic acid, gibberellin, and zeatin, but increased abscisic acid accumulation. The directly-exposed fruits reached a partially-mature stage (Rutab) in August, whereas the bagged fruits remained at the immature stage (Khalal). This study is the first to describe the biochemical basis of the observed improvement of date palm fruit development in response to reduced light intensity. Besides, it provides insights into controlling date palm fruit maturity and subsequently prolonging the shelf life dates on the tree; thus, extending the marketing period for the benefit of the farmers.
