用改进的细胞遗传学方法制备染色体标本,鉴定了小鼠晚期桑椹胚(晚桑)、囊胚和扩展囊胚(扩囊)的性别。在实验中,利用小鼠早期胚胎染色体标本制备的理想实验参数,鉴定了242枚小鼠晚桑、囊胚和扩囊的性别,性别鉴定成功率分别为80.4%、99.0%和94.5%。以细胞遗传学方法鉴定小鼠早期胚胎性别的结果为标准,得出用间接免疫荧光法和PCR(聚合酶链反应)扩增SRY(y染色体的性别决定期)部分序列法分别鉴定100枚和26枚小鼠胚胎性别的准确率相应地为74.0%和92.3%。  相似文献

根据C57BL6小鼠Y染色体重复序列145C5的碱基顺序,设计并合成一对引物,运用PCR扩增昆明白小鼠入前胚胎卵裂球DNA,以确定其性别,共对108枚活检胚胎的相应卵裂球进行了性别诊断,获雄性胚46枚,雌性胚62枚,移植后分别获雄性仔鼠4只,准确率100%(4/4),雌性仔鼠9只,准确率70%(9/13),本研究结果表明小鼠Y染色体重复序列145C5的碱基顺序在C57BL6小鼠和昆明白小鼠中基本一致,为农牧业动物进行性别选择和运用PCR进行单基因病植入前遗传学诊断提供了方法学基础。  相似文献

利用双引物PCR技术对优质良种牛体外受精的冷冻胚胎进行性别鉴定。并对分割后的半胚分别进行PCR扩增和染色体分析,两种方法的判定结果相吻合,在本实验条件下,利用减少PCR体系中细胞的数量来确定最低的模板DNA的检出量,2-细胞不能检测,4-细胞检出率为62.5%,8-细胞为90%,16-细胞以上为100%。对23牧经过性别鉴定后的胚胎进行移植,获得5头纯种试管牛,产犊率为21.7%,与正常的胚胎移植 结果27.3%(3/11)无显著差异,预测性比为100%符合。  相似文献

为了不断完善山羊早期胚胎性别鉴定技术,实现奶山羊雌性胚胎的体外生产与商业化,本研究对胚胎性别鉴定的两种采样方法进行比较。结果表明,两种取样方法性别鉴定结果的准确性没有差异,桑椹胚经打孔法采样后胚胎妊娠率(36.84%)明显高于切割法(31.25%),囊胚经切割法采样后胚胎妊娠率(31.58%)明显高于打孔法(23.53%),综合比较,胚胎性别鉴定应以打孔法采集早期桑椹胚为宜。  相似文献

研究了金不换鲜三七液特殊毒理学效应的致突变性。以小鼠骨髓细胞染色体畸变试验,小鼠睾丸减数分裂染色体畸变及小鼠致畸试验为指标,研究金不换鲜三七液的安全性。结果:(1)小鼠骨髓细胞染色体畸变试验:低,中,高3个剂量组小鼠肌髓细胞染色体畸变率分别为0.7%,0.2%和0.9%,与对照组相比无显著差异。阳性对照组染色体畸变率大大增高。(2)小鼠睾丸减数分别细胞染色体畸变;在本实验条例上,小鼠睾丸细胞染色体  相似文献

通过普通小麦与滨麦、簇毛麦及山羊草属7个种杂交幼胚培养,获得8个体细胞胚性无性系(8个组合10个胚)及大量试管苗。愈伤组织及胚性愈伤组织诱导率分别为65.79%和26.32%。形态学及细胞学鉴定结果,均为真杂种。不同染色体组及同一染色体组不同基因型的幼胚,在组织培养中有明显差异。非部体细胞具有遗传的全能性,但当染色体数目严重偏离双单倍体数目的,其全能性即丧失。胚状体的发育具有与合子胚极相似的典型结  相似文献

本实验用小鼠血液淋巴细胞为核供体进行了核移植研究。用淋巴细胞分离液(比重1.088)分离出小鼠血液中的淋巴细胞,直接用作核移植供体细胞,采用胞质内注射法成功构建的重构胚经常规培养2h后,SrCl2激活处理6h,然后添加mM16培养液和小鼠输卵管上皮细胞饲养层共培养。把发育至早期囊胚阶段的重构胚转移至小鼠胎儿成纤维细胞饲养层上,添加ES细胞培养液继续培养。对孵化出的内细胞团进行消化,然后接种培养。结果显示,小鼠血液淋巴细胞可以支持体细胞核移植重构胚的发育,核移植重构胚2-细胞率41.03%(128/312),桑葚胚和囊胚发育率分别为9.29%(29/312),1.92%(6/312)。重构囊胚在小鼠胎儿成纤维细胞饲养层上分离出2个内细胞团,分离率为0.64%(2/312)。实验证实利用小鼠血液淋巴细胞进行体细胞核移植是可行的,可用于深入研究。  相似文献

为了使牛体外受精卵能通过体外早期发育阻滞期,我们建立了卵丘细胞单层(A)和输卵管上皮单层(B)两种共同培养系统。A1共同培养实验是在本实验室进行的,A2和B共同培养实验均在日本岗山大学农学部动物繁殖学研究室进行,牛输卵管组织的分离使用0.76%EDTA—PBS溶液,共同培养系统均使用含10%小牛血清的TCM—199(Earle's salts)作为培养液。培养的卵泡卵母细胞体外成熟率为100%,体外受精率为99—100%。在A1共同培养实验中,越过阻滞期发育到16—细胞以上的胚胎占卵裂胚的35.7%,与A2共同培养实验中越过阻滞期的发育率(40.1%)无显著差异(P<0.05)。A1和A2共同培养实验,在卵裂基础上得到的桑椹胚和囊胚发育率分别为23.7%和27.9%。每百枚培养的卵母细胞,在A1共同培养实验中可获得桑椹胚和囊胚15.1枚,在A2共同培养实验中可获桑椹胚和囊胚的20.5枚。B共同培养实验中桑椹胚和囊胚发育率为54.1%,显著高于A1或A2共同培养实验的相应发育率(P<0.001),使用B共同培养系统每百枚培养的卵母细胞可以获得37枚桑椹胚和囊胚。  相似文献

本研究采用人工聚合技术,将小鼠不同品系和同品系之间的8细胞胚聚合为一体,分别用PBS FCS、BWW和BMOC-Ⅲ培养20—24小时后观察了发育率。其结果在昆明白←→C57中分别有88.1%(59/67)、95.0%(115/121)和94.8%(73/77),在昆明白←→昆明白中分别有93.3%(28/30)、87.7%(121/138)和86.7%(52/60)的嵌合胚发育为桑椹胚或胚泡,各处理组之间无显著差异。将141枚嵌合胚分别移植给12只受体,结果有7只小鼠妊娠共产仔25只。在昆明白←→C57的17只嵌合体小鼠中毛色为黑、白和杂色的各有3、4和10只。  相似文献

对波斯小麦、“中国春”小麦与珍珠粟远缘杂交的受精作用和胚胎发育进行了研究。珍珠粟花粉管能在小麦柱头上萌发,并且有多个花粉管进入花往及胚囊。波斯小麦已授粉的207个子房中,11.11%既形成胚又形成胚乳,7.25%只形成胚,而1.93%只形成胚乳,在这个杂交中受精率和成胚率分别为20.29%和18.36%。“中国春”小麦已授粉的167个子房中,相应的数字是299%、5.99%、0、8.98%和8.98%。杂交后形成的胚和胚乳核型极不稳定,来自珍珠粟的染色体很快被排除,最后产生的是单倍体的小麦胚。但因无胚乳或胚乳败育,幼胚缺乏营养供应而停止发育或夭亡。探讨了影响小麦×珍珠粟受精率的可能因素以及通过远缘杂交染色体排除法进行单倍体诱导和珍珠粟基因向小麦转移的可能性。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献
