普通大麦和球茎大麦抗病种间杂种的产生及其同工酶标记   总被引:2,自引:0,他引:2  
以二棱大麦(Hordeum vulgare L.)“苏啤1 号”为母本与四倍体球茎大麦(H. bulbosumL.)“GBC141”杂交, 并通过幼胚培养获得11 株三倍体F1 植株. 三倍体F1 植株与二倍体母本二棱大麦“苏啤1 号”回交, 获得7 株二倍体回交后代BC1. 7 个回交后代株系通过大麦黄花叶病病圃鉴定, 其中BC1-2株系抗大麦黄花叶病. 利用聚丙烯酰胺凝胶电泳对父本、母本和BC1-2株系进行了同工酶分析,结果表明二倍体二棱大麦与四倍体球茎大麦的过氧化物酶同工酶差异显著,并且在BC1-2株系幼根的过氧化物酶同工酶谱中,发现一条来自父本球茎大麦“GBC141”的过氧化物酶谱带,该酶带的相对迁移率(Rf)为0.47, 重复性好并且易于检测.由于回交后代BC1-2抗大麦黄花叶病, 又带有来自球茎大麦特异的幼根过氧化物酶谱带作为易于检测的同工酶标记,因此该回交后代株系可以作为大麦抗病育种工作重要的抗源.  相似文献

本试验对7个品种变叶木的形态特征进行比较,并利用聚丙烯酰胺凝胶电泳,对各品种过氧化物酶同工酶进行分析。结果表明,各品种的过氧化物酶同工酶谱不一;7个品种变叶木共有14条谱带,2号谱带最多为11条,5号谱带最少为6条;其谱带分为3个区(A=0~0.18,B=0.18~0.35,C=0.35~0.5);并对7个品种的亲缘关系进行聚类分析。  相似文献

以芦荟属植物元江芦荟和皂质芦荟为亲本进行远缘有性杂交试验。正交对3株103朵小花进行授粉,获得7枚果实,平均结实率为6.80%;反交对2株88朵小花授粉,获得5枚果实,平均结实率为5.88%。获得了19株F1代实生苗,并对亲本和F1代实生苗以及库拉索芦荟、华芦荟等5种芦荟属植物进行了过氧化物酶同工酶(POD)的比较研究,结果显示,5种芦荟种间的POD同工酶酶谱具有较高的相似程度,同时各种又具有各自的特征酶带,很容易区分。特别是F1代实生苗的酶谱与父母本的酶谱有显著不同的酶带,证明产生了新的中间类型。  相似文献

采用电泳技术结合同工酶染色,分析了罗汉果雌雄株叶片的过氧化物酶同工酶、酯酶同工酶、超氧化物歧化酶同工酶、多酚氧化酶同工酶和过氧化氢酶同工酶。结果表明:罗汉果雌雄叶片在同工酶谱上,存在着与性别性状相关的酶带;雌雄间的差异酶带在每一种同工酶中均有一条以上,可作为罗汉果雌雄株间的性别鉴定。此外,还比较了高产、低产、不结果雌株之间同工酶的酶带和活性差别。  相似文献

用淀粉凝胶电泳,分析苹果实生苗新梢皮层的过氧化物酶同工酶,其酶9带(A_9)与苹果的矮生习性相关,且不受树龄、苹果类型以及栽培条件等影响。在矮生型苹果育种中,A_9可以作为预选标志,其准确率为86.14%。  相似文献

对栽培大麦(HordeumvulgareL.品种“Arupo”)与纤毛鹅观草(Roegneriaciliaris(Trin.)Nevski)属间杂种后代F5和回交后代BC1F4(以“Arupo”作回交亲本)的不同类型株系与其双亲的幼根、幼芽、幼穗、幼嫩籽粒分别进行了酯酶、过氧化物酶同工酶分析。结果F5和BC1F4各株系的酯酶同工酶、过氧化物酶同工酶酶谱具有母本“Arupo”全部或绝大多数酶带,父本R.ciliaris的1~3条酶带和数量不等的双亲所没有的新酶带,也出现酶带的减少。结果表明,纤毛鹅观草的遗传物质已稳定地遗传给了其自交和回交后代。酶谱变异与植株习性出现一定程度的关联。由于在不同时期内结构基因的表达起不同作用,因此,在用同工酶分析法鉴定易位系时,最好不同的器官采用不同的酶类或一种同工酶多时期连续分析。  相似文献

获得了粳型水稻77-170品系原生质体再生植株(R_2)6个株系的206棵后代植株。对其中96株进行性状观察和细胞学染色体鉴定,发现再生植株子代在株高、剑叶和主穗长、单株有效穗数、每穗粒数、育性、生育期等性状上都产生了变异,除株高外其他性状在第2代遗传上不稳定,染色体倍性稳定(2_n=24)。对56株再生植株子代作酯酶、过氧化物酶同工酶测定,其酶谱谱带与对照实生株相似。  相似文献

本文对根瘤菌属12株不同根瘤菌的过氧化物酶,谷氨酸草酰乙酸转氨酶及酯酶同工酶进行了测定,结果发现:所有菌株的过氧化物酶和谷氨酸草酰乙酸转氨酶均呈阴性。酯酶同工酶酶带数在快生型和慢生型根瘤菌之间存在明显的区别,在YMA培养基上,快生型根瘤菌仅有1—2条,迁移率在0.71—0.98之间变化;而慢生型根瘤菌却有4—6条酶带,迁移率在0.15—0.98之间变化,同一菌株在不同碳源上酯酶同工酶酶带数也发生显著变化。  相似文献

小麦叶片的过氧化物酶活性随年龄增大而上升,同工酶谱也随不同叶层叶龄的增大而加强,并出现新的酶带。 SO_2熏气促进小麦叶片的过氧化物酶活性,不同层次叶片比较,幼嫩叶片促进程度比老叶大些。SO_2熏气对小麦叶片过氧化物酶同工酶的某些酶带有加强作用,并能诱导产生新的酶带。同一叶片的前半叶伤害比后半叶重,过氧化物酶活性也以前半叶为大,说明过氧化物酶活性的提高与伤害有一定关系。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献
