本文分别采用环己烷、正丁醇、乙酸乙酯、丙酮、甲醇5种极性不同的溶剂分别提取青蒿内生真菌Alternaria sp.(PQH12)的菌丝体。采用MTT法分别测定不同极性溶剂提取物对3种肿瘤细胞株(HL-60、MCF-7和COLO205)的生长抑制率,采用DPPH自由基清除法和超氧阴离子O-·2清除法评价了青蒿内生真菌Alternaria sp.(PQH12)不同极性溶剂提取物的抗氧化活性。结果显示,正丁醇和乙酸乙酯提取物均表现出对肿瘤细胞生长的抑制作用;浓度为20μg/mL时,正丁醇提取物对HL-60细胞的生长抑制率为80.56%,乙酸乙酯提物对MCF-7和COLO205细胞的生长抑制率分别为54.47%和83.09%;在两种抗氧化活性评价体系中,甲醇提取物的抗氧化活性均明显优于其他溶剂的提取物,其IC50值分别为0.20 mg/mL(DPPH)和0.19 mg/mL(O-·2)。该结果表明,青蒿内生真菌Alternaria sp.(PQH12)具有较好的细胞毒和抗氧化潜力,具有一定的开发利用价值,可以作为进一步实验研究的对象。  相似文献

复方木鸡冲剂诱导人白血病细胞HL-60凋亡机制研究   总被引:1,自引:0,他引:1  
目的:探讨复方木鸡冲剂诱导人白血病细胞HL-60细胞凋亡的作用和机制,为相关中药开发提供实验资料.方法:用MTT法检测复方木鸡冲剂对HL-60细胞增殖活性的影响,光镜下观察细胞形态的变化;流式细胞仪(Annexin V/PI双染法)检测细胞凋亡,并分析细胞周期;免疫细胞化学法检测Bcl-2、caspase-3、p21WAF1的表达.结果:MTT法显示复方木鸡冲剂能抑制HL-60细胞的生长,细胞呈凋亡形态学变化.流式细胞仪检测结果为细胞凋亡率明显增高,出现GO/G1期阻滞.Bcl-2表达降低,caspase-3表达增高,p21WAF1表达强阳性.结论:复方木鸡冲剂明显抑制HL-60细胞的生长,其抗肿瘤的机制与诱导肿瘤细胞凋亡、促进细胞分化有关.  相似文献

用MTT法观察羊栖莱多糖(SFPS)在体外抗人白血病HL-60细胞增殖作用;扫描电镜、透射电镜、DNA电泳和流式细胞仪检测HL-60细胞凋亡。结果表明SFPS对HL-60细胞具有显著生长抑制作用,并呈量效和时效关系,药物作用24,36,48,72h的IC50分别为390,362,402,421mg/L;药物浓度为300mg/L和500mg/L作用HL-60细胞后,琼脂糖凝胶电泳显示有凋亡细胞特有的DNA梯状条带,细胞微绒毛减少、染色质固缩、边集,凋亡小体形成;DNA直方图出现亚G1峰。在一定浓度范围内,SFPS诱导细胞凋亡的作用呈现浓度和时间依赖性,同时G2/M期细胞比例增多。因此,SFPS抗肿瘤作用与诱导细胞凋亡和G2/M期细胞阻滞有关。  相似文献

目的:研究白花蛇舌草豆甾醇(stigmasterol from Hedyotis diffusa willd.,SHD)对人肝癌细胞SMMC-7721、BEL-7402的体外抑制作用,对肝癌H22的体内抑制作用及对其增殖周期、凋亡的影响。方法:MTT法评价SHD对人肝癌细胞SMMC-7721、BEL-7402的抑制率变化规律。昆明雄性小鼠60只,随机取10只为正常对照组,余接种H22瘤株,随机分为模型对照组、5-FU阳性对照组(30mg/kg)和高中低剂量SHD给药组(剂量分别为15、30、60mg/kg),腹腔给药10 d后,比较各组瘤重抑制率、H22细胞周期分布、凋亡率。结果:SHD对SMMC-7721、BEL-7402细胞具有体外抑制作用;SHD显著抑制H22肿瘤,增加G0-G1期细胞比例,降低G2/M期细胞比例,促进肿瘤细胞凋亡。结论:SHD在体外、体内均具有抑制肝癌细胞的作用,此作用与阻滞肿瘤细胞增殖周期,促进肿瘤细胞凋亡有关。  相似文献

目的研究藻蓝蛋白对人结肠癌SW480细胞的体外抑瘤作用,为进一步探讨藻蓝蛋白抑制肿瘤的机制提供依据。方法用不同浓度的藻蓝蛋白处理结肠癌SW480细胞后,应用MTT实验来检测藻蓝蛋白对细胞增殖的抑制作用并计算出半数抑制率,利用HE染色法和电镜技术观察凋亡细胞形态结构,采用流式细胞术分析其对SW480细胞周期的影响。结果 MTT试验证明藻蓝蛋白能够抑制SW480细胞的增殖,且呈时间和剂量依赖性;HE染色以及电镜的观察结果显示藻蓝蛋白能够诱导SW480细胞的凋亡,流式细胞术显示SW480细胞被阻滞在G2/M期,G0/G1期细胞比例降低。结论藻蓝蛋白在体外能够抑制结肠癌SW480细胞的增殖,具有可开发人结肠癌治疗光敏剂应用前景。  相似文献

炎症在肿瘤的发生发展过程中扮演重要角色,脂氧素是一类重要的内源性抗炎介质。但是迄今为止,脂氧素对肿瘤的影响报道极少。为此,本文研究了脂氧素对HL-60和K562白血病细胞增殖和凋亡的影响。体外培养白血病细胞株HL-60和K562,Western印迹和实时荧光定量PCR检测脂氧素受体的表达情况;CCK-8法(cell counting kit-8 assay)检测HL-60和K562的增殖能力;PI染色后利用流式细胞仪进行细胞周期分析;膜联蛋白V试剂盒检测脂氧素对细胞凋亡的影响。实验结果表明脂氧素抑制HL-60和K562白血病细胞增殖(P0.05);脂氧素处理组S期细胞比例明显减少而G_0/G_1期细胞比例增加;脂氧素还可以诱导HL-60和K562白血病细胞凋亡。由此可见,脂氧素抑制HL-60和K562白血病细胞增殖,其机制可能与诱导白血病细胞G_0/G_1期阻滞和细胞凋亡有关。  相似文献

为探讨连香树精油的体外抗肿瘤活性。以人肝癌SMMC-7721细胞为受试细胞株,利用MTT法、流式细胞术、JC-1法和Western blot法评价连香树精油的体外抗肿瘤活性。结果表明:连香树精油处理24 h后可降低SMMC-7721细胞存活率;细胞中G0/G1期的比例下降,G2/M期的比例升高;细胞线粒体膜电位降低;自噬相关蛋白LC3-Ⅱ、Beclin-1表达上升,加入自噬抑制剂氯喹后LC3-Ⅱ蛋白与Bcl-2抗凋亡蛋白表达下降。综上说明连香树精油能抑制SMMC-7721细胞的增殖活性,通过阻滞G2/M期抑制细胞分裂,通过降低线粒体膜电位诱导SMMC-7721细胞凋亡,而自噬在SMMC-7721细胞凋亡过程中作为一种保护机制而存在。  相似文献

目的:观察外源性RASSF1A基因对食管癌细胞的增殖作用并探讨机制.方法:将包含RASSF1A基因的质粒转染食管癌细胞EC9706,建立稳定转染细胞克隆,Western blot检测RASSF1A基因表达,通过细胞生长曲线检测细胞生长活性和增殖能力、流式细胞仪检测对细胞周期的影响、裸鼠移植瘤实验检测转染细胞的体内成瘤特性.结果:稳定表达RASSF1A基因的EC9706细胞中RASSF1A蛋白表达增加;细胞生长速度明显减慢;细胞周期中G1/G0期比例明显增加,S期比例减少;EC9706细胞的裸鼠致瘤能力被抑制.结论:RASSF1A基因能显著抑制食管癌细胞在体内外的生长,其机制可能与该基因诱导凋亡、抑制增殖作用有关.  相似文献

从不同品种的灵芝中筛选出了多糖含量最高的菌株GL2为材料,利用柱层析技术从子实体和菌丝体提取物中分离得到多个组分。实验发现子实体组分P3,P31及P32对人白血病细胞株K562的生长有明显地抑制作用,这三个组分中只有P32对另一白血病细胞株HL-60有抑制作用。免疫活性测试的结果显示子实体各组分在刺激小鼠脾淋巴细胞,T和B细胞的增殖,提高人外周血中NK细胞杀伤活性方面比菌丝体的作用强;进一步的实验发现子实体与菌丝体相应组分在刺激人外周血中的T和B淋巴细胞增殖方面的活性差异不大。子实体和菌丝体提取物各组份均可剂量依赖型的促进PBMC分泌TNF-α。菌丝体提取物各组分对TNF-释放量的影响在低浓度时与子实体各组份相当,在高浓度时要明显好于赤芝子实体提取物各组分。  相似文献

Five ethanolic extracts from the mycelia of Ganoderma lucidum,G.tsugae,G.oerstedii,G.subamboinense,and G.resinaceum were respectively studied on their anticancerous activities against leukemic HL-60 cell line in vitro.Results showed that all five extracts potently inhibited HL-60 proliferation.The extract from G.lucidum mycelia exerted the highest activity.Annexin V/PI bivariate flow cytometric analysis further revealed that the five extracts significantly induced early apoptosis in HL-60 cells.The results illustrate that not only G.lucidum but also other Ganoderma species can inhibit cancer cells,and their mechanisms are related to induction of apoptosis.  相似文献

先在基础培养基中添加苦参水煎汁,然后培养灵芝,得到灵芝培养液,再按乙醇氯仿-5%碳酸氢钠溶液-氯仿的顺序提取培养液中的有机酸成分,用制备高效液相色谱分离,得到6个新组分,用这些组分分别作用于转染了乙型肝炎病毒DNA的2.2.15细胞,用固相放射免疫法测定细胞培养上清液中的乙肝病毒表面抗原(HBsAg)和e抗原(HBeAg)的含量,用四甲基偶氮唑盐(MTT)法测定细胞的存活率。结果表明,其中的3个组分对2.2.15细胞分泌HBsAg和HBeAg有抑制作用,提示可能具有抗乙肝病毒的作用。  相似文献

对纳米级灵芝子实体粉末及破壁灵芝孢子粉石油醚提取物(PE)、氯仿提取物(CE)、丙酮(AE)、甲醇提取物(ME)、水提取物(WE)与灵芝子实体及灵芝孢子提取量进行对比,利用GC-MS联用仪对石油醚提取物进行了成分分析鉴定,对水提物中总糖进行了含量测定,并利用宫颈癌细胞Hela和晶体上皮细胞SRA01/04进行了体外增殖作用和剂量效应关系研究,为灵芝资源的保护及进一步开发利用提供理论基础。结果表明,纳米化灵芝子实体及破壁灵芝孢子不同溶剂提取量显著增加,纳米级灵芝子实体粉末水提取物具有抑制宫颈癌细胞Hela和晶体上皮细胞SRA01/04增殖的作用。破壁灵芝孢子各溶剂提取物对宫颈癌细胞Hela和晶体上皮细胞SRA01/04没有明显的增殖抑制作用。  相似文献

Griseofulvin (GF), an oral antifungal agent, has been shown to exert antitumorigenesis effect through G2/M cell cycle arrest in colon cancer cells. But the underlying mechanisms remained obscure. The purpose of this study is to test the cytotoxic effect of GF on HL-60 and HT-29 cells and elucidate its underlying molecular pathways. Dose-dependent and time-course studies by flow cytometry demonstrated that 30 to 60 microM GF significantly induced G2/M arrest and to a less extend, apoptosis, in HL-60 cells. In contrast, only G2/M arrest was observed in HT-29 cells under similar condition. Pretreatment of 30 microM TPCK, a serine protease inhibitor, completely reversed GF-induced G2/M cell cycle arrest and apoptosis in HL-60 cells but not in HT-29 cells. The GF-induced G2/M arrest in HL-60 cells is reversible. Using EMSA and super-shift analysis, we demonstrated that GF stimulated NF-kappaB binding activity in HL-60 cells, which was completely inhibited by pretreatment of TPCK. Treatment of HL-60 with 30 microM GF activated JNK but not ERK or p38 MAPK and subsequently resulted in phosphorylation of Bcl-2. Pretreatment of TPCK to HL-60 cells blocked the GF-induced Bcl-2 phosphorylation but not JNK activation. Time course study demonstrated that activation of cdc-2 kinase activity by GF correlated with Bcl-2 phosphorylation. Taken together, our results suggest that activation of NF-kappaB pathway with cdc-2 activation and phosphorylation of Bcl-2 might be involved in G2/M cell cycle arrest in HL-60 cells.  相似文献

The development of radioprotective agents has been the subject of intense research, especially in the field of radiotherapy. In this study, we examined the radioprotective activity of the total triterpenes isolated from Ganoderma lucidum (Fr.) P. Karst in mouse splenic lymphocytes in vitro. Using the MTT assay, Ganoderma triterpenes were found to have no effect on cell viability, indicating that they are non-toxic to splenic lymphocytes. The effect of the total triterpenes on DNA damage and apoptosis induced by radiation was analyzed using the comet assay, DNA ladder assay and flow cytometric analysis. Total triterpenes were found to be highly effective in preventing DNA laddering, even at low concentrations (25μg/ml). The comet assay demonstrated that the G. triterpenes effectively prevented DNA damage, and flow cytometry revealed a reduction in apoptotic cells. The effect of the total triterpenes on intracellular reactive oxygen species (ROS) level and endogenous antioxidant enzyme activity in splenic lymphocytes were determined to elucidate possible radioprotective mechanisms. Total triterpenes successfully reduced the formation of intracellular ROS and enhanced endogenous antioxidant enzyme activity in splenic lymphocytes following irradiation. Thus, these findings indicate that the total triterpenes isolated from G. lucidum have a remarkable ability to protect normal cells from radiation-induced damage, which suggests therapeutic potential.  相似文献

Ganoderma lucidum is a well-known traditional Chinese medicinal herb containing many bioactive compounds. Ganoderic acid T (GA-T), which is a lanostane triterpenoid purified from methanol extract of G. lucidum mycelia, was found to exert cytotoxicity on various human carcinoma cell lines in a dose-dependent manner, while it was less toxic to normal human cell lines. Animal experiments in vivo also showed that GA-T suppressed the growth of human solid tumor in athymic mice. It markedly inhibited the proliferation of a highly metastatic lung cancer cell line (95-D) by apoptosis induction and cell cycle arrest at G(1) phase. Moreover, reduction of mitochondria membrane potential (Delta psi(m)) and release of cytochrome c were observed during the induced apoptosis. Our data further indicate that the expression of proteins p53 and Bax in 95-D cells was increased in a time-dependent manner, whereas the expression of Bcl-2 was not significantly changed; thus the ratio of Bcl-2/Bax was decreased. The results show that the apoptosis induction of GA-T was mediated by mitochondrial dysfunctions. Furthermore, stimulation of the activity of caspase-3 but not caspase-8 was observed during apoptosis. The experiments using inhibitors of caspases (Z-VAD-FMK, Z-DEVD-FMK and Z-IETD-FMK) confirmed that caspase-3 was involved in the apoptosis. All our findings demonstrate that GA-T induced apoptosis of metastatic lung tumor cells through intrinsic pathway related to mitochondrial dysfunction and p53 expression, and it may be a potentially useful chemotherapeutic agent.  相似文献

In this study, we investigated the anti-tumor activity both in vitro and in vivo of a polysaccharide obtained from Ganoderma lucidum on HL-60 acute myeloid leukemia cells, and focused on its targeting effect on mitogen-activated protein kinase (MAPK) pathways. It was found by the methods such as western blot and flow cytometry (FCM), that G. lucidum polysaccharide (GLP) blocked the extracellular signal-regulated kinase/MAPK signaling pathway, simultaneously activated p38 and JNK MAPK pathways, and therefore regulated their downstream genes and proteins, including p53, c-myc, c-fos, c-jun, Bcl-2, Bax, cleaved caspase-3 and cyclin D1. As a result, cycle arrest and apoptosis of HL-60 cells were induced. Therefore, GLP exerted anti-tumor activity via MAPK pathways in HL-60 acute leukemia cells.  相似文献
