一例智力低下患者7q~ 标记染色体的来源鉴定

以人类染色体显微切割、PCR技术构建的现有人类染色体特异性和染色体区带特异性探针池作为绘画探针,采用正向染色体绘画技术,结合染色体筛查方法,查明了一例7q~ 标记染色体患者的染色体附加片段来源于3q26→3qter。确定该患者的核型为46,XX,-7, der(7)t(7;3)(7pter→7q32::3q26→3qter)。应用这个策略,能够快速有效地鉴定标记染色体的来源。     

一例带有两条9号染色体次缢痕丢失的患者及其家系的细胞遗传学研究

本文报道了一例两条9号染色体次缢痕缺失的女性病例,临床表现为严重智力低下和语言障碍。经外周血淋巴细胞染色体G、C显带分析,确定其核型为46,XX,del(9)del(9)(pter→q11∷q13→qter)。又对其父母及妹妹进行了染色体分析,患者之父与两个妹妹的核型均正常,其母是46,XX,del(9)(pter→q11∷q13→qter)核型的携带者。     

六例性腺发育不全症的实验室及临床研究

中国人体细胞G式显带染色体的鉴别及模式图

本文提出“中国人体细胞G式显带染色体的鉴别及模式图”。 (一)显带方法 1.湖南医学院医学遗传研究室所用的方法(1973):用外周血和骨髓标本按常规方法培养和滴制染色体玻片标本后,立即置70一80℃烤箱内烤2一3小时,关闭烤箱,让其自然冷却后,按下列程序进行显带:(1)在无菌条件下,用0.8,%NaCI溶液配制0.25多胰蛋白酶溶液,玻璃滤菌器过滤,冰箱保存。(2)将上述0.25铸胰蛋白酶溶液倒人染色缸中,用1 MNaOH调pH至7.0左右,用水浴加温至37℃。(3)将玻片标本投人胰蛋白酶溶液中,不断轻轻地摆动2一3分钟左右。(D取出玻片标本,直立于吸水纸上数秒钟,…

Effect of Human WEE1 and Stem Cell Factor on Human CD34^＋ Umbilical Cord Blood Cell Damage Induced by Chemotherapeutic Agents

Myelosuppression is one of the major side-effects of most anticancer drugs. To achieve myeloprotection, one bicistronic vector encoding anti-apoptotic protein human WEE l （WEElHu） and proliferation-stimulating stem cell factor （SCF） was generated. In this study, we selected human umbilical cord blood CD34^＋ cells as the in vitro model in an attempt to investigate whether WEEIHu, rather than conventional drug-resistant genes, can be introduced to rescue cells from the damage by chemotherapeutic agents such as cisplatin, adriamycin, mitomycin-c and 5-fluorouracil. Cell viability and cytotoxicity assay, colony-forming units in culture assay and externalization of phospholipid phosphatidylserine analysis showed that the expression of WEElHu and SCF in CD34^＋ cells provided the cells with some protection. These findings suggest that the expression of WEElHu and SCF might rescue CD34^＋ cells from chemotherapyinduced myelosuppression.     

数据综合分析鉴定人类Auxilin基因

Auxilin蛋白诱导Hsp70c蛋白与笼形蛋白的结合,在真核细胞衣被小泡脱衣被的过程中扮演了重要的角色.通过对已有EST,STS等数据库的综合分析,我们将人类auxilin基因定位到1p31,D1S515和D1S198标记之间.26个EST构成的5个重叠群,占该基因中共约2.3 kb的部分cDNA序列,其中编码区长501 bp,得到的序列与牛的auxilin基因显示有极高的同源性.各EST数据显示,auxilin在人胚胎的多种组织中表达,在成人脑、表皮组织中也有表达.     

人多肽链延伸因子1α四个反转录假基因的鉴别与定位

人多肽链延伸因子1α基因EEF1A是一个在蛋白质合成过程中起重要作用的看家基因。通过对已有GenBank、GDB等数据库的综合分析,鉴别了4个人类多肽链延伸因子1α基因的反转录假基因,并分别将其精细定位于4q2 5、7p15-21、9q34和19q13上。 Abstract:The gene for human polypeptide chain elongation factor-1α(EEF1A)is a house-keeping gene which plays an important role in the process of protein synthesis.By means of comprehensive analysis in the database of Genbank,GDB and ect,we identify 4 retropseudogenes of EEF1A and finely localized to 4q25,7p15-21,9q34 and 19q13,respectively.
