从210株海洋细菌中筛选出30株产蛋白酶活力较高的菌株。以N1为出发菌,用UV诱变处理,获得突变株。其中的N1-3。菌株最适生长温度为20℃;在21℃下培养44小时,产酶最高。酶反应最适温度为50℃,最适pH8.0。30℃以下酶活稳定。  相似文献

目的:从青藏高原冰川雪中筛选出一菌多酶的菌株.方法:对恢复出的4个细菌,通过平板透明法研究其产淀粉酶、脂肪酶和蛋白酶的特性.结果:LHG-C-9为惟一可以产淀粉酶的菌株,所产脂肪酶活性最高.4个菌株均不产蛋白酶.结论:LHG-C-9最适生长温度为15℃,属于耐冷菌.对该菌所产淀粉酶和脂肪酶的性质进行了初步研究,其随产淀粉酶的最适作用温度为50℃;最佳产酶pH值为7.0,该pH值所产酶活为83.9U/mL;在60℃的高温下温浴10min后酶活为0%.该菌株所产脂肪酶的最适作用温度为20℃;最佳产酶pH值为7.0,该pH值所产酶活为9.2U/mL;50℃温浴1h后酶活力不足34%.  相似文献

通过对大量极区低温海洋菌株的分离、筛选及进一步的生理生化特性研究,获得1株最适生长温度为15℃、生长温度上限为35℃、产蛋白酶及多种多糖水解酶的耐冷细菌。该菌过氧化氢酶为阳性,具有弱嗜盐性;蔗糖、可溶性淀粉是有利于菌株生长的碳源物质,而酵母膏则是效果最佳的氮源物质。该菌株所产蛋白酶的最适作用温度为55℃,而淀粉酶、琼脂酶及纤维素酶的最适作用温度皆为35℃。  相似文献

从深海的 3 8份水样和泥样中筛选到一株产蛋白酶的菌株DY A ,其最适生长温度为1 0℃ ,能适应较大范围的pH值和盐度。此菌株只在酵母膏存在的情况下产酶 ,不利用单一氮源 ,最适产酶条件为 :pH1 0 0 ,1 0℃ ,接种量 0 5 % ,2 0 0r/min摇床培养 48~ 72h。粗酶液的最适作用温度为 40℃ ,最适pH值为 1 0 ,在pH9~ 1 2内稳定 ,是一碱性蛋白酶。  相似文献

从深海的38份水样和泥样中筛选到一株产蛋白酶的菌株DY-A,其最适生长温度为10℃,能适应较大范围的pH值和盐度,此菌株只在酵母膏存在的情况下产酶,不利用单一氮源,最适产酶条件为,pH10.0,10℃,接种量0.5%,200r/min摇床培养40-72h,粗酶液的最适作用温度为40℃,最适pH值为10,在pH9-12内稳定,是一碱性蛋白酶。  相似文献

中国冰川1号产适冷蛋白酶耐冷菌的分离鉴定及产酶条件   总被引:6,自引:0,他引:6  
从中国冰川 1号样品分离获得一株产适冷蛋白酶耐冷菌株SYP- A2 - 3,鉴定为蜡状芽孢杆菌 (Bacilluscereus)。该菌生长温度范围为 0~ 38℃ ,最适生长温度 2 5℃ ,而最适产酶温度为 15℃。所产蛋白酶为中性金属蛋白酶 ,最适催化温度为 4 2℃ ,低温催化活力较高 ,适宜作用pH为 7. 0~ 8 .5 ,SDS PAGE测定的分子量为 34 2kD。SYP A2 3产酶条件的研究结果显示酪蛋白是较好的氮源 ,葡萄糖、淀粉是较好的碳源 ,产酶最佳pH为 6. 5~ 7. 0 ,在优化的条件下 ,15℃摇瓶产酶达到 380 0U mL ,5L发酵罐通气培养产酶达 4 80 0U mL。  相似文献

【目的】筛选鉴定产右旋糖苷酶的海洋细菌,并对其所产右旋糖苷酶的酶学性质及在变异链球菌牙菌斑生物膜中的应用进行初步研究。【方法】利用平板透明圈法从海洋环境中筛选产右旋糖苷酶的细菌,根据菌株形态特征、生理特征及16S rDNA序列确定其分类学地位,采用体外生物膜模型研究该酶对变异链球菌牙菌斑生物膜形成的抑制作用。【结果】从海泥中筛选出一株产右旋糖苷酶的细菌KQ11,初步鉴定为节杆菌(Arthrobacter sp.)。该菌株的最适生长温度为30°C,最适生长pH 7.5,最适生长NaCl浓度为0.4%。右旋糖苷酶的最适作用温度为45°C,最适作用pH为5.5。该酶能有效地抑制变异链球菌牙菌斑生物膜的形成。【结论】菌株KQ11右旋糖苷酶能够抑制变异链球菌牙菌斑生物膜的形成,可望用于漱口液等口腔护理产品中。  相似文献

【目的】筛选产蛋白酶嗜碱菌并对其进行鉴定和特性分析。【方法】利用碱性脱脂牛奶培养基分离纯化产蛋白酶嗜碱菌,通过形态特征、生理生化、16S rRNA基因序列分析以及DNA-DNA杂交实验确定菌株的分类地位,利用酪蛋白水解法分析所产蛋白酶的pH和温度作用范围、稳定性和耐氧化剂能力。【结果】从我国西藏盐碱湖样品中分离到一株产碱性蛋白酶的菌株ZL223,该菌株为革兰氏阳性菌,最适生长温度为37℃,最适生长pH9.0,16S rRNA基因序列分析显示,菌株ZL223与假强芽孢杆菌Bacillus pseudo firmus OF4亲缘关系最近,16S rRNA基因序列相似性为98.6%,DNA-DNA杂交结果显示与B.pseudofirmus OF4同源性为86%。菌株ZL223产生的蛋白酶作用的最适pH为12.0,最适温度为40℃。【结论】结合生理生化指标测定的结果,鉴定菌株为假强芽孢杆菌ZL223(B.pseudofirmus ZL223)。该菌株产生的碱性蛋白酶具有较高的pH适应性,值得进一步研究。  相似文献

【目的】明确柑桔全爪螨Panonychus citri(Mc Gregor)高致病力菌株芽枝状枝孢霉Cladosporium cladosporioides(CQBBW3)的生物学特性。【方法】通过单因素试验,测试了不同培养基、碳源、氮源、温度及pH对CQBBW3菌株生长和产孢的影响。【结果】CQBBW3菌株的LC50为4.66×108个孢子/m L,LT50为6.2353 d。最佳生长培养基是淀粉琼脂培养基,最适产孢培养基为察氏培养基;菌丝生长最适碳源和氮源为甘露醇和甘氨酸,产孢最适碳源和氮源为乳糖和赖氨酸;最适生长温度为25℃,最适产孢温度为15℃和30℃;最适生长pH值为6.0~8.0,最适产孢pH值7.0。【结论】CQBBW3菌株对柑桔全爪螨有较好的杀虫活性,喜好25℃、中性、富含有机营养的环境。本研究为菌株快速培养及其真菌杀虫制剂的开发与田间应用奠定基础。  相似文献

从新疆昌吉市油脂化工厂的含油冻土中筛选到一株产低温脂肪酶的菌株ch-3,形态鉴定及18SrDNA序列分析表明该菌株为白地霉,命名为Geotrichum candidum ch-3。我们对其生长及产酶情况和酶性质做了初步研究。该菌株的最适生长温度是20℃。玉米浆、豆油可显著促进脂肪酶的产生。粗酶液经双水相萃取和Sephadex G-75凝胶过滤分离纯化后进行酶学性质的研究。该脂肪酶最适作用温度为35℃,在0℃可保持66%的相对酶活性,最适pH值为7.0,对热较敏感,50℃处理20min,剩余酶活为55%,Fe2+、Cu2+、Mn2+以及Ca2+对酶有较强的激活作用。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
