大白菜霜霉菌诱导抑制性消减杂交cDNA文库的构建和分析   总被引:2,自引:0,他引:2  
以抗霜霉病大白菜双单倍体系(DH)‘T12-19’为材料,构建了霜霉病诱导表达的正向抑制性消减文库,并利用反向Northern斑点杂交技术对768个阳性克隆进行了筛选,共获得57个病原菌诱导上调表达的克隆。测序后得到55条通读表达序列标签(ESTs),对这些ESTs序列进行聚类和拼接分析,共获得50个unigenes。Blast分析表明,37个unigenes与已知基因高度同源,占全部非重复序列的67.3%。对已知功能基因按MIPS的分类方法进行功能分类,发现这些基因的功能主要涉及物质与能量代谢、转录调控、蛋白质合成与代谢、膜及转运、信号转导、抗病防御等。为了验证文库筛选结果的可靠性,采用实时荧光定量PCR技术分析了其中2个克隆BFCH10和BFIA7的表达谱。结果表明,这2个克隆在接种病菌6h后明显上调表达,与反向Northern斑点杂交结果基本一致。  相似文献

白粉病是我国小麦的主要病害之一,尝试用表达序列标签(expressed sequence tage,EST)技术,研究了经白粉病菌诱导后的小麦基因表达。从构建的普通cDNA库中随机挑取约1500个阳性克隆并进行测序,获不重复ESTs序列387条,不重复序列均获GenBank的存储号,其中49.4%的序列与已知基因同源,196条序列功能未知,84条序列为新ESTs。将不重复序列制备成高密度点阵膜,用差示杂交法筛选到几个抗病相关序列。  相似文献

口蹄疫是一种烈性传染病,其广泛流行给社会造成了巨大经济损失。为了研究口蹄疫灭活疫苗免疫的分子机制,同时也为抗口蹄疫病毒药物的研制奠定基础,本研究应用mRNA差异显示技术,以PK-15细胞为材料,系统比较了口蹄疫疫苗刺激组(A组)和正常的PK-15细胞(B组)的基因表达情况,回收差异片段,经二次扩增并纯化后,得到30条ESTs。将30条ESTs采用以地高辛标记的反向Northern点杂交鉴定,将阳性条带克隆测序,筛选出8条ESTs,编号E1~E8,应用BLASTn工具将8条ESTs对核酸数据库nr和dbEST中所有序列进行了同源性分析,其中E1,E2分别与猪的热休克蛋白基因、猪的MHCⅠ类基因同源,序列相似性都达到100%。E3,E4,E5,E7分别与已有核酸数据库中的基因克隆或EST具有较高同源性,为已知的EST,但功能未知;E6,E8在数据库中没有发现与其相似性较高的序列,为新的EST。应用数据库资源将E5、E7进行电子延伸后,将延伸序列进行开放阅读框分析,又经TBLASTx分析发现E7蛋白质序列与猪的精氨酸酶Ⅰ类蛋白序列有很高同源性。将E1、E2、E4、E5序列进行了基因表达谱分析,对E6、E8用BLASTx工具对非冗余蛋白质数据库nr进行了相似性搜索,在其他物种中找到了相似的基因序列。本研究筛选出的热休克蛋白基因、MHCⅠ类基因、精氨酸酶Ⅰ类基因和其他未知功能基因可以作为抗口蹄疫病毒研究中的侯选基因,其具体的功能有待今后进一步研究。  相似文献

小麦抗白粉病侵染初期的表达序列标签分析   总被引:19,自引:2,他引:17  
以抗白粉病品系“百农 32 17×Mardler” BC5F4为材料 ,构建了一个白粉病菌接种初期的抑制消减杂交cDNA文库 ,测序获得 76 0条ESTs。与GenBank序列进行BLASTx分析 ,获功能已知ESTs 2 71条。通过分析抗病相关基因 ,推测G蛋白介导的信号传导途径、SA信号传递系统、MAP相关信号传递系统等参与了小麦抗白粉病过程。SAR基因在抗病相关ESTs中的种类与数量最多。数据显示苯丙烷代谢途径、细胞壁结构修饰作用、细胞保卫机制参与了抗病过程。未知功能ESTs与GenBank序列进行BLASTn分析 ,其中许多与病原菌、非病原菌诱导cDNA文库来源的ESTs同源 ;新ESTs占全部ESTs的 16 6 %。  相似文献

以云南普通野生稻为材料,利用抑制差减杂交技术(SSH),构建了白叶枯病菌胁迫的云南普通野生稻特异表达基因的差减文库.通过对文库所有阳性单克隆进行测序,聚类分析后共获得494条高质量的表达序列标签(EST).经过BlastN分析,有417条与已知功能的序列有较高同源性;经BlastX分析,有104条EST与未知功能蛋白或假定蛋白有较高相似性,49条EST未能找到同源匹配,341条EST与已知功能蛋白有较高同源性.初步分析发现,这些基因主要涉及能量代谢、蛋白质代谢、核酸代谢、防御与抗逆应答反应、信号转导、光合作用及膜运输等代谢过程.使用半定量RT-PCR研究了7个可能与白叶枯病抗性相关的EST序列在云南普通野生稻对照和白叶枯病菌处理的叶片中的表达情况,并获得这些基因的表达谱.结果发现,克隆编号为OR7,OR68和OR826的EST受白叶枯病菌胁迫诱导上调表达,其中OR826 EST在蛋白数据库中无同源序列,可能是一类新的白叶枯病抗性基因,而组成型表达的OR143 EST在对照和接菌处理的叶片中均能检测到其mRNA的表达,但其表达量在白叶枯病菌胁迫48 h后逐渐增强,推测这些基因直接参与了云南普通野生稻抗病防御反应.本研究为从云南普通野生稻中发掘和克隆新的白叶枯病抗性基因提供了理论依据,为进一步研究云南普通野生稻抗白叶枯病的分子机制奠定了基础.  相似文献

以‘条中32’接种前后的小麦抗条锈病种质‘陕麦139’幼苗叶片为材料,利用抑制消减杂交技术,构建了条锈菌接种48 h小麦抗病品种叶片的抑制表达SSH-cDNA文库,通过测序以了解条锈菌侵染后被抑制表达的相关基因。从构建的文库中随机选取48个阳性克隆,进行测序,经过序列拼接等,获得高质量EST序列34条(Gen-Bank序列号为EL930132-EL930165),序列比对分析表明其中31条EST序列与已知功能的基因序列同源性较高,主要涉及植物的能量代谢途径、膜转运、信号传导及核酸加工等方面的基因,包括光合系统Ⅱ抗性蛋白D1、碳酸酵素、线粒体磷酸盐转运子、苹果酸脱氢酶、过敏性诱导反应蛋白和rRNA核酸内切酶等相关基因。此外,获得未知功能的EST序列3条。研究表明,小麦在条锈病菌侵染初期,许多基因的正常表达调控受到影响,如能量代谢和寄主细胞结构木质化,阻止小麦抗病过敏性反应等。  相似文献

为分析条锈菌诱导下的小麦抗病与感病近等基因系之间差异表达的基因,以接种小麦条锈菌CY26小种的抗病近等基因系Yr4/6×Taichung 29幼苗叶片cDNA作为实验方,接种CY26的感病亲本Taichung 29幼苗叶片cDNA为驱动方,利用抑制消减杂交(SSH)技术构建了一个包含1 300余克隆的消减文库。对文库中600个克隆进行了反向Northern点杂交筛选,对获得的阳性克隆进一步进行了Northern杂交验证,获得显著差异的克隆12个。经测序和BlastX分析,其中6个差异表达序列的推测产物分别为亮氨酸重复序列蛋白、过氧化氢酶、硫氧还蛋白、RNA结合蛋白、抗坏血酸过氧化物酶和热激蛋白。除亮氨酸重复序列为信号传导类蛋白外、其他几个均为抗病防御类蛋白。  相似文献

以小麦抗叶锈病近等基因系TcLr41和感病亲本Thatcher的叶片cDNA分别作为试验方和驱动方,利用抑制差减杂交技术,构建了一个包含2544个克隆的差减文库。随机提取阳性克隆质粒DNA后经PCR检测,插入片段大部分集中在200~1000bp之间,证明所构建的文库符合要求。在功能已知的基因中,推测过氧化氢酶(catalasc)基因、抗秆锈病基因(rust resistance gene)、铜蓝结合蛋白(blue copper—binding protein)基因、锌指蛋白(ring zinc finger protein)基因、胁迫反应蛋白(stress responsive protein)基因等可能是TcLr41中抗病相关差异表达基因。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
