玉米隐性突变o2基因能通过减少醇溶蛋白的合成来显著提高赖氨酸含量,为培育高赖氨酸含量的优质蛋白玉米(quality protein maize, QPM)提供了良好的基因资源。对玉米o2基因的发现、研究现状及其修饰基因的研究进展,以及当前育种家利用这两种基因相互作用培育优质蛋白玉米的研究进展进行了综述,以期为高赖氨酸玉米育种提供参考。  相似文献

对含有opaque—2和flour—2基因型的高单系列玉米进行了赖氨酸含量和种子蛋白分析。高单3号的赖氨酸含量比当地品种高出105%,产量高6%。高单系列玉米种子内含赖氨酸高的盐溶和水溶蛋白含量提高了1.6~2.0倍,不含赖氨酸的醇溶蛋白在opaque—2基因型的品种中都减少了一半以上。  相似文献

扩展蛋白(Expansin)是一类能够使植物细胞壁松弛的活性蛋白,在植物生长发育过程中起着重要作用。利用PLAZA、NCBI、MaizeGDB、Uniprot、PLEXdb等基因组数据库,获得玉米Expansin家族的基因序列、染色体基因座位、蛋白质序列以及长度,构建玉米Expansin基因家族系统进化树,进行基因组织表达谱的分析。结果表明,玉米基因组中含有93个Expansin基因,分布于玉米的9条染色体上;多数Expansin具有250~300个氨基酸;玉米Expansin基因家族有40个Expansin A(EXPA)、47个Expansin B(EXPB)、6个Expansin-like A(EXLA),未发现Expansin-like B(EXLB);44个玉米Expansin基因在不同玉米组织中特异表达。该研究结果不仅为玉米扩展蛋白的深入研究奠定了基础,而且为其他研究人员对基因信息的获取提供了参考。  相似文献

用纤维素酶解离胚乳、滤膜法统计玉米胚乳细胞的数目,进一步借助Logistic方程模拟胚乳细胞增殖动态的结果表明,整个灌浆期间胚乳细胞增殖呈现“慢-快-慢”的变化趋势。授粉15d后,不同类型胚乳的细胞数目依序为普通玉米〉糯玉米〉甜玉米〉爆裂玉米;胚乳细胞数目主要取决于细胞的增殖速率,并与淀粉充实和粒重关系密切。胚乳发育前期以胚乳细胞增殖为主,后期以淀粉积累为主。  相似文献

不同类型玉米籽粒的营养品质及其与籽粒质地的关系   总被引:5,自引:0,他引:5  
测定普通玉米、爆裂玉米、糯玉米和甜玉米4种类型玉米籽粒不同发育时期的直链淀粉、支链淀粉、总淀粉、可溶性糖和蛋白的含量,分析这些营养物质与角质率的关系。结果表明:灌浆期间4种类型玉米的直链淀粉、支链淀粉和总淀粉含量呈上升趋势,总蛋白含量呈下降趋势,可溶性糖含量变化规律不明显。爆裂玉米的直链淀粉含量始终最高(4.7%-23.1%),甜玉米(1.4%-4.6%)和糯玉米(2.3%-4.9%)的始终较低;甜玉米的支链淀粉含量一直最低(15.7%-35.5%),除授粉10d以外,糯玉米的支链淀粉含量一直最高(65.5%-69.8%);甜玉米总淀粉含量始终最低(17.1%36.1%)、总蛋白含量(15.2%-26.9%)和授粉30d后的可溶性糖含量最高(14.2%-17.6%)。高蛋白含量可能是爆裂玉米和甜玉米角质率高的原因,糯玉米的角质率低可能与支链淀粉含量高和蛋白积累少有关。  相似文献

利用一套16个玉米B-A染色体易位的系列材料,和一套11个由wx基因标记易位断点的第9染色体相互易位材料,测定了一个新发现的玉米隐性突变基因os(opaque-small germ)所属的染色体。两个结果相互印证,证明该基因位于第2染色体短壁。os基因具有产生不透明胚乳和小胚面的双重遗传效应,是一个良好的遗传标记。这一特性有可能来检查异雄核受精或定向受精的存在和比数。os基因除缩小种胚体积之外,也  相似文献

对玉米Hz85(O2/O2)和 Hz85(o2/o2)以及S7913(O2/O2)和 S7913 (o2/o2)两种不同核背景下的近等基因系(NIL),采用cDNA芯片杂交技术研究玉米o2突变基因及赖氨酸形成和胚乳发生相关基因在RNA水平上的表达差异。在两套NILs中检测到共同的表达差异克隆87个,对其序列分析得到26个TUGs(tentative unique genes)、11个未命名蛋白和6个新序列。这些TUGs分别参与了生长发育、胁迫响应、物质运输、信号转导、电子传递链、细胞防御、代谢等细胞过程,以及作为细胞组分和贮存蛋白。基于O2/o2 NILs间胚乳发育中基因表达的差异,讨论了o2籽粒中的不透明粉质胚乳形成的分子机制。  相似文献

利用等电聚焦(IEF)电泳和不连续醋酸尿素聚丙烯酰胺凝胶电泳(NAU-PAGE)对玉米籽粒贮藏蛋白的等电点(pI)和在F1中的遗传表现以及贮藏蛋白在胚和胚乳中的分布进行研究,结果表明:(1)玉米籽粒贮藏蛋白的pI在3.5 ̄8.45范围内,可分离的有39条左右,60%左右的蛋白质属酸性,pI分布在3.50-6.85范围内,40%左右属中性偏碱,pI在6.85-8.45范围内。(2)籽粒贮藏蛋白在F1  相似文献

用10个遗传上和籽粒形态性状上具有差异的玉米自交系,依多种可能的交配方法获得亲本P1、P2、F1(P1× P2)、F2、B1(F1×P1)、B2(F1× P2)及其相应反交RF1、RF2、RB1、RB2共10个种子世代。种植2年。依广义遗传模型建立包括种子胚乳加性、胚乳显性、母体加性、母体显性和细胞质效应的遗传模型,运用种子数量性状的精细鉴别法[1]和混合模型分析法[2,3],对粒长、粒宽、粒长宽比、粒厚及百粒重作了性状表达遗传机制的鉴别与探讨。单个组合的遗传模型精细测验表明,5个籽粒性状的遗传主要受母体显性和胚乳基因型(包括加性和灵性)的控制,一个组合的粒宽、粒厚和百粒重上还检测到细胞质效应。对25对 F1正反交组合世代均值依MINQUE法分析的结果表明,5个籽粒性状的遗传方差中,母体遗传方差占60%以上,胚乳基因型方差低于40%,粒长和百粒重还有细胞质效应,约占10%~30%。可见,籽粒性状的遗传特点是受多套遗传系统控制,其中以母体基因型的作用最大。  相似文献

Maize, an important cereal crop, has a poor quality of endosperm protein due to the deficiency of essential amino acids, especially lysine and tryptophan. Discovery of mutants such as opaque-2 led to the development of nutritionally improved maize with a higher concentration of lysine and tryptophan. However, the pleiotropic effects associated with opaque-2 mutants necessitated the development of nutritionally improved hard kernel genotype, the present-day quality protein maize (QPM). The aim of present study was to analyze and compare the temporal profile of lysine and tryptophan in the developing maize kernel of normal, opaque-2 and QPM lines. A declining trend in protein along with tryptophan and lysine content was observed with increasing kernel maturity in the experimental genotypes. However, opaque-2 retained the maximum concentration of lysine (3.43) and tryptophan (1.09) at maturity as compared to QPM (lysine-3.05, tryptophan-0.99) and normal (lysine-1.99, tryptophan-0.45) lines. Opaque-2 mutation affects protein quality but has no effect on protein quantity. All maize types are nutritionally rich at early stages of kernel development indicating that early harvest for cattle feed would ensure a higher intake of lysine and tryptophan. Two promising lines (CML44 and HKI 1105) can be used for breeding high value corn for cattle feed or human food in order to fill the protein inadequacy gap. Variation in lysine and tryptophan content within QPM lines revealed that differential expression of endosperm modifiers with varying genetic background significantly affects nutritional quality, indicating that identification of alleles affecting amino acid composition can further facilitate QPM breeding program.  相似文献

The characteristics of storage protein accumulation of maize with qualified protein (MQP) and 02 maize were analysed basing on the genetical and biochemical point of views. The 22 kD and 20 kD zeins in the developing endosperms of maize accumulated 15 days after pollination. The structural genes encoding 22 kD and 20 kD zeins in the developing endosperms were simutaneously expressed. In the endosperms of MPQ and o2 maize the synthesis of 22 kD and 20 kD zeins was suppressed. That is to say, o2 gene negatively regulated the synthesis of 22 kD and 20 kD zeins. Two-dimentional electrophoretic analysis of zeins in the maize endosperms further revealed the effects of o2 gene and its modifiers on the synthesis of zeins. In Mol7 and Mo17/o2 endosperms the synthesis of 27 kD, 22 kD, 20 kD and 15 kD zeins was severely suppressed. In 041/oz and 040/o2 endosperms little difference existed SDS-PAGE analysis of the soluble proteins of Mol 7 and Mo17/o2 endosperms revealted that two bands with molecular weight (MW) of 38.7 kD and 26.7 kD were present in wild type but absent in o2 mutant, while two bands with MW 27.2 kD and 26.1 kD were present in o2 mutant but absent in wild type. These differences were resulted from the effect of o2 gene. In 040/02 and 041/o2 endosperms two bands with MW 18.6 kD and 17.6 kD were present in 041/o2 but absent in 040/02 while one band with MW 40. 2 kD was present in 040/02 and absent in 041/o2, which was closely related to the effects of the modifiers of o2 gene.  相似文献

Twenty-two selected quality protein maize (QPM) lines, including 13 lines developed in India (DMRQPM series) and nine lines released by CIMMYT, Mexico (CML series), were evaluated for their endosperm protein content and quality, besides kernel modification in terms of vitreousness. Endosperm protein contents in 13QPMlines were on par or better than that of the normal maize ‘checks’ (Trishulata and Parkash). The QPM endosperm proteins showed significantly higher % tryptophan as well as EF-1α (a multifunctional protein with a positive and highly significant correlation with lysine content in the endosperm) contents, in comparison with the normal maize genotypes. Evaluation of kernel modification revealed considerable scope for accumulation of endosperm modifiers in some of the QPM lines. Positive and highly significant correlation was revealed between tryptophan and EF-1α contents in the endosperm proteins, whereas the correlations between the quality parameters with kernel modification in the QPM genotypes were found to be non-significant. The study led to the identification of some promising QPM lines, such as DMRQPM-37, DMRQPM-44, CML176, CML142 and CML149, which could be effectively deployed in the QPM breeding programmes.  相似文献

A split pollination was used to produce normal (Su su su O2 o2 o2) and high lysine double mutant sugary opaque-2 (su su su o2 o2 o2) endosperms on the same ear of sugary opaque-2 maize plants. Amino acids were determined in the vascular sap of the ear peduncle. Lysine content in the sap was compared with lysine stored in both normal and sugary opaque-2 endosperm during kernel filling. Lysine content in the ear peduncle sap could account for all lysine found in both endosperms. Preformed lysine is highly catabolized in the normal endosperm, but not in the high lysine sugary opaque-2 endosperm. The rate of lysine breakdown appears to be an important mechanism by which the high lysine mutant controls lysine level in maize endosperm.  相似文献

The opaque-2 (o2) mutation of maize increases lysine content, but the low seed density and soft texture of this type of mutant are undesirable. Lines with modifiers of the soft kernel phenotype (mo2) called “Quality Protein Maize” (QPM) have high lysine and kernel phenotypes similar to normal maize. Prior research indicated that the formation of vitreous endosperm in QPM might involve changes in starch granule structure. In this study, we focused on analysis of two starch biosynthetic enzymes that may influence kernel vitreousness. Analysis of recombinant inbred lines derived from a cross of W64Ao2 and K0326Y revealed that pullulanase activity had significant positive correlation with kernel vitreousness. We also found that decreased Starch Synthase III abundance may decrease the pullulanase activity and average glucan chain length given the same Zpu1 genotype. Therefore, Starch Synthase III could indirectly influence the kernel vitreousness by affecting pullulanase activity and coordinating with pullulanase to alter the glucan chain length distribution of amylopectin, resulting in different starch structural properties. The glucan chain length distribution had strong positive correlation with the polydispersity index of glucan chains, which was positively associated with the kernel vitreousness based on nonlinear regression analysis. Therefore, we propose that pullulanase and Starch Synthase III are two important factors responsible for the formation of the vitreous phenotype of QPM endosperms.  相似文献

Polysome preparations obtained from opaque-2 and normal maize endosperms during development did not show any significant difference in sedimentation coefficient or nucleotide composition. The pattern of incorporation in vitro of lysine and leucine, however, differed quite distinctly in these two preparations. During early stages of maturity the polysomes from opaque-2 incorporated substantially more lysine and less leucine as compared with those from normal maize. Although the trend was reversed at 25 days post-pollination, this did not result in any significant zein accumulation since very little total protein was synthesized after that stage in opaque-2 maize endosperm. It is, therefore, suggested that the opaque-2 gene exerts a regulatory control on mRNA synthesis, required for zein formation at early stages of maturation.  相似文献

The effect of genotypes and texture on the content of proteins in maize grains was examined by assessing absolute amounts of six protein fractions in the whole endosperms of four wild‐type lines with high protein content and four quality protein maize (QPM) varieties and for hand‐dissected hard and soft endosperm regions from eight other lines. As previously reported for six wild‐type lines and their opaque‐2(o2) versions, zeins were predominant for all genetic backgrounds and all types of endosperms. From these data and others the amounts of zeins and true proteins (crude proteins free of non‐protein nitrogen) in developing and mature endosperms of wild‐type lines were correlated. The data points for zeins from hard endosperms lay between the regression line and the upper limit of confidence area. Those for zeins from soft endosperms were located at the lower part of confidence area and on a level with the points corresponding to the most immature endosperms. Furthermore, some data points for zeins from o2 and QPM samples lay near the lower limit while the others were outside the confidence area. This suggested an initial zein accumulation dependent on the genotype at a low relative rate, followed by an accumulation at higher rate. The conditions used for isolating and quantitating zeins are discussed.  相似文献

研究玉米种质的遗传关系及其杂种优势模式对玉米育种者有着极为重要的指导意义.本研究对从国际玉米小麦改良中心(CIMMYT)引入的及国内自育成的10个优质蛋白玉米(QPM)黄粒优良系进行了配合力分析及杂优模式的初步研究.这10个QPM优良系中有5个来自CIMMYT的热带、亚热带系;5个为国内自育成的骨干系.通过部分双列杂交获得45个杂交组合,将这些组合种植在云南省及广西自治区的三种不同生态条件下进行观察鉴定.产量的方差分析结果表明,品种之间、环境之间的差异达到极显著水平,而重复之间不显著;产量的一般配合力差异达极显著水平,而特殊配合力的差异不显著.杂交组合CML166×齐205具有最高产量(10880kg/hm2),杂交组合长631/02×中系096/02具有最低产量(5496kg/hm2).自交系CML161产量的一般配合力效应值最高(1010.53),自交系CML166产量的一般配合力效应值其次(947.11);而自交系中系096/02产量的一般配合力效应值最低(-1119.98).自交系CML194与忻9101/02具有最高的产量特殊配合力效应值(1813.50),自交系CML166与齐205产量的特殊配合力效应值也较高(1272.00);而自交系忻9101/02与齐205产量的特殊配合力效应值最低(-1670.96).根据杂交组合产量性状的配合力分析,可初步将这10个优质蛋白玉米自交系划分为4个杂种优势群和4种杂种优势模式.  相似文献
