登革热病毒反义寡核苷酸的合成及抗病毒活性   总被引:3,自引:0,他引:3  
根据碱基互补原理, 反义寡核苷酸可与特定病毒基因结合从而选择性地抑制该病毒的复制, 这是病毒性疾病治疗的新途径. 基于上述原理设计并合成了D2-04 RNA特异的6个5′末端脂肪链修饰的硫代反义寡核苷酸. 体外抗病毒活性评价显示互补于5′端起始密码、3′端重复序列和末端序列的3个反义寡核苷酸呈较强的抗病毒活性.  相似文献

目的探索MDCK细胞在微载体上的培养条件,并研究H1N1型流感病毒在MDCK细胞上的增殖条件。方法在微载体上培养好MDCK细胞上用H1N1型流感病毒在不同的病毒感染复数(MOI)、胰酶浓度两个关键的病毒增殖条件进行流感病毒在细胞上的增殖研究。结果微载体质量浓度为6 g/L时,MDCK细胞培养密度可以达到4.5×106cells/mL。在MOI为0.05接种流感病毒,胰酶质量浓度4μg/mL,流感病毒在MDCK细胞上可获得较高的滴度。结论 MDCK细胞用微载体培养可以达到较高的细胞密度,可以作为规模化生产新型流感病毒疫苗的主要细胞基质进行进一步的研究。  相似文献

合成 2 0mer随机寡核苷酸文库 ,与体外转录出的全长survivincRNA杂交 ,RNaseH酶切割后 ,经引物延伸、放射自显影 ,共筛选出 13个针对survivin基因的反义结合位点 (antisenseaccessiblesites ,AAS) .运用RNADraw软件分析、选定具有显著茎环结构的 4个位点 ,合成互补性反义寡核苷酸AS ODN1、AS ODN2 、AS ODN3 、AS ODN4并转染高表达survivin基因的胃癌细胞株MKN 4 5 .逆转录聚合酶链反应和Western印迹检测发现MKN 4 5细胞的survivinmRNA和蛋白水平均有显著的下降 ;MTT比色法证实 6 0 0nmol LAS ODN1~AS ODN4转染 2 4h后细胞生长受到明显抑制 ,透射电镜、annexinⅤ FITC和PI双染色流式细胞术均检测到细胞凋亡 .说明运用随机寡核苷酸文库 RNaseH酶切割与计算机分析相结合的方法 ,在体外有效筛选出survivin的反义核酸结合位点 ,其相应的反义寡核苷酸能阻断survivin基因的生物学功能 .  相似文献

目的:研究硫代反义寡核苷酸prop5在细胞水平的抗流感活性及其作用机制。方法:cy3标记prop5用于考查人肺腺癌细胞A549对硫代反义寡核酸的摄取;利用实时荧光定量PCR检测流感病毒RNA拷贝数,Western印迹检测prop5对PDCD5蛋白表达和caspase-3蛋白剪切的抑制;利用间接免疫荧光和Western印迹检测prop5对病毒核糖核蛋白复合体(RNP)出核的影响;利用TUNEL检测prop5对流感病毒引起细胞凋亡的抑制作用。结果:流感病毒感染促进A549细胞摄取prop5;prop5下调感染病毒的A549细胞中PDCD5蛋白的表达,并能抑制流感病毒的复制;prop5抑制流感病毒引起的A549细胞的凋亡;prop5抑制病毒RNP出核。结论:prop5在细胞水平具有抗流感病毒活性,其作用机制可能同抑制RNP出核有关;本研究为进一步探讨宿主-病毒相互作用和抗流感药物开发奠定了基础。  相似文献

Lipofectin介导反义磷脂酰肌醇3(IP3)-激酶寡核苷酸(ODN)转染HepG2细胞.用逆转录PCR法检测IP3-激酶mRNA表达水平,以Sandwich ELISA法检测AP-1的活化.结果表明a)反义IP3激酶ODN抑制IP3激酶mRNA表达;b)白介素-18(IL-18)诱导AP-1活化,AP-1的光密度值从基础水平的O.134±O.009上升至1. 704±0.019;c)反义IP3-激酶ODN呈时间 (5~24h)和剂量(1~8μg)依赖性地抑制IL-18诱导的AP-1活化,反义IP3-激酶ODN 2μg与细胞孵育8h的抑制作用最强,AP-1的光密度值从对照组的1.704±O.019下降到O.722±0.026,抑制丰达57.6%.上述结果表明,IP3-激酶调控白介素-18诱导的AP-1活化.  相似文献

反义RNA是反义技术的一个重要领域,为探索丙型肝炎病毒治疗新途径及验证转基因细胞模型HepG2.9706的有效性,设计了一条互补于HCV 5′NCR及翻译起始区(39713核苷酸)的反义RNA序列,将其插入pGL3载体SV40启动子下游,构建了HCV特异性的反义RNA真核表达载体(pHCV-asR)。通过PCR扩增、酶切反应及序列分析进行了初步鉴定,并将其转染HepG2细胞,通过RTPCR方法检测其在细胞中的表达并将pHCV-asR转染转基因细胞模型HepG2.9706,评价其对HCV 5′NCR的抑制活性。结果表明,构建的反义RNA表达载体插入序列正确,并能在HepG2细胞中表达;pHCV-asR在HepG2.9706中对HCV 5′NCR调控荧光素酶基因表达具有特异性的剂量依赖性抑制活性,最高抑制率可达57%。  相似文献

摘要:【目的】为研究流感病毒突破种间屏障分子机制,筛选流感基因工程疫苗株。【方法】本实验以猪流感病毒A/Swine/Henan/S4/01(H3N2)为亲本株,利用反向遗传学操作技术,采用RT-PCR技术对该病毒的8个基因片段分段进行扩增,通过与双向转录载体pHW2000连接, 重组质粒转染293T和MDCK共培养细胞,拯救出全部基因均来自于亲本株的猪流感病毒rgH3N2,并分别以人流感病毒A/ PR/8/34(H1N1)、禽流感病毒A/Duck/Nanchang /4-165/2000 (H4N6 )、马流感病毒A/Equine/Fuyun/2008/(H3N8)的HA和NA基因替换A/Swine/Henan/S4/01的相应基因,【结果】生物学实验结果表明rgH3N2在鸡胚半数感染量、组织培养半数感染量、稳定性试验等方面都与亲本株保持一致。rgH3N2经鸡胚多次传代后血凝价最高可达到1:256,接种MDCK细胞60 h后,血凝价可以达到1:64。基因替换后成功拯救出的重组病毒rgH1N1、rgH4N6和rgH3N8在鸡胚和细胞上均具有较高的增殖能力。【结论】病毒的成功拯救为流感病毒突破种间屏障分子机制,HA、NA基因在流感病毒跨种属传播中所扮演角色的研究和流感基因工程疫苗株的筛选奠定了基础。  相似文献

应用反向遗传学技术在哺乳动物细胞中产生甲型流感病毒   总被引:19,自引:1,他引:18  
在国内首次应用反向遗传学技术将多个质粒共转染哺乳动物细胞,得到了具有活性的甲型流感病毒.采用自行构建的含有polⅠ和polⅡ启动子和终止子序列的pIVV2质粒,将A/PR/8/34(H1N1)流感病毒基因组全部8个RNA节段的cDNA分别克隆到该质粒的两个启动子之间,得到了8个可在真核细胞中复制和表达的质粒.将这8个质粒共转染293T细胞,培养48h后吸取上清液,转接入MDCK细胞中,再经过72h培养,发现在MDCK细胞中有明显的流感样细胞病变产生,测上清病毒血凝滴度为1:10.将MDCK细胞冻融液继续接种鸡胚和MDCK细胞,培养后将鸡胚尿囊液和MDCK细胞冻融上清液进行血凝和血凝抑制实验,证明所得病毒为A/PR/8/34(H1N1).此结果将有助于国内今后对流感疫苗的研究.  相似文献

为了探讨 A S O D N 化学修饰形式与 A S O D N 稳定性,体外细胞毒性以及抗流感病毒活性之间的关系,合成了 7 种不同化学修饰形式的 A S O D N:硫代 A S O D N 及其 3′端分别磷酸化和胆固醇修饰;3′与 5′端硫代,中间为天然结构的混合骨架 A S O D N;天然结构 A S O D N 及其 3′端分别磷酸化和胆固醇修饰等.测定了 7 种修饰体在小鼠血清, M D C K 细胞裂解液,含 2% 胎牛血清的 D M E M培养液以及水中的稳定性,体外细胞毒性和在细胞水平抗流感病毒活性.结果表明,混合骨架 A S O D N,硫代 A S O D N 及其 3′端接磷酸和胆固醇的修饰形式在小鼠血清, M D C K 细胞裂解液与含2% 胎牛血清的 D M E M 培养液中稳定性相对较高,作用 24~48 h 仅混合骨架 A S O D N 与硫代 A S O D N 发生部分降解;天然结构 A S O D N 及其 3′端接磷酸和胆固醇修饰体在 24 h 内大部分降解.所有 A S O D N 修饰体在水中具有很高稳定性,48 h 内未见降解作用.7 种 A S O D N 修饰形式在 M D C K 细胞中未表现明显的细胞毒性.硫代 A S O D N 及其 3′端接磷酸和胆  相似文献

Abstract

The antiviral activity of PS-ODNs, complementary to different regions of the CVB3 genome, was investigated under in vitro conditions. Inhibition of CVB3 replication was detected only after prolonged pretreatment of HeLa cells with antiviral active PS-ODNs, but not when virus and PS-ODN were applicated simultaneously. Results from flow cytometric analysis indicate that a low cellular uptake of anti-CVB3 oligonucleotides into HeLa cells might be a reason for their moderate antiviral activity.  相似文献

本实验探讨了寡聚核苷酸对CSFV复制的影响以及作为抗CSFV新型药物的可行性。实验结果表明针对CSFV5'um端非编码区NS3蛋白丝氨酸蛋白酶功能区的寡聚核酸对CSFV复制均有显著的抑制作用,而针对CSFV3' 端非编码区寡聚核苷酸仅有轻微抑制作用,5'端寡聚核苷酸具有最佳抑制作用,同时发现相应序列中正义聚核苷酸的作用要优于反义寡聚核苷酸;脂质体介导转染能显著提高寡聚核苷酸对CSF复制的抑制作用。这些初步结果也提示,CSFV5'端和3'端非编码区对CSFV复制的重要性和作用有所不同。  相似文献

Lipofectin, which is a mixture of neutral lipid with a cationic lipid, has been widely used to enhance cellular delivery of phosphorothioate, 2'-sugar-modified, and chimeric antisense oligonucleotides. Phosphodiester oligonucleotides delivered with Lipofectin usually do not elicit antisense activity probably because cationic lipid formulations do not sufficiently protect unmodified oligonucleotides from nuclease degradation. We show that a cationic polymer, polyethylenimine (PEI), improves the uptake and antisense activity of 3'-capped 20-mer and 12-mer antisense phosphodiester oligonucleotides (PO-ODN) targeted to different regions of Ha-ras mRNA and to the 3'-untranslated region (3'-UTR) of C-raf kinase. In contrast, PEI, which forms a very stable complex with the 20-mer phosphorothioate oligonucleotide (PS-ODN), does not enhance its antisense activity. Using fluorescently labeled carriers and ODN, we show that PEI-PS-ODN particles are very efficiently taken up by cells but PS-ODN is not dissociated from the carrier. Our results indicate that carrier-ODN particle size and stability and ODN release kinetics vary with the chemical nature of the ODN and the carrier being transfected into the cells. The very low cost of PEI compared with cytofectins and the increased affinity for target mRNA and decreased affinity for proteins of PO-ODN compared with PS-ODN make the use of PEI-PO-ODN very attractive.  相似文献

Phosphorothioate cytosine-guanine oligodeoxynucleotides (CpG PS-ODNs) has been reported to induce Th1 immune responses against coadministered Ags more efficiently than phosphodiester CpG ODNs (CpG PO-ODNs). Here, we demonstrated that PS-ODNs, but not PO-ODNs, have a chemotactic effect on primary macrophages, which is independent of the CpG motif. In addition, the conjugation of a hexameric dG run (dG(6) run) at the 3' terminus reduced the concentration required for the optimal chemotactic activity of PS-ODNs by approximately 10-fold. Endosomal maturation blockers, such as monensin and chloroquine, inhibited the chemotactic effect of PS-ODNs. The inhibition of the activities of p38 mitogen-activated protein (MAP) kinase, and extracellular signal-related kinases (ERKs) as well as phosphoinositide 3-kinase with their specific inhibitors also resulted in suppressing the chemotaxis of primary macrophages induced by PS-ODNs. These results indicate that the PS-ODN-mediated chemotaxis requires the activation of ERKs, p38 MAP kinase, and phosphoinositide 3-kinase as well as endosomal maturation. In addition, the phosphorylations of the p38 MAP kinase, ERKs, and protein kinase B, Akt, were induced by PS-ODN, which were further enhanced by the presence of both a dG(6) run and CpG motifs. Our findings suggest that the chemotactic activity of PS-ODNs may be one of the mechanisms by which PS-ODNs exhibit stronger immunomodulatory activities than PO-ODNs in vivo.  相似文献

为了研究具有临床应用前景的 A S O D N 脂质体转运系统,以临床药用大豆磷脂为主要原料制备了p H 敏脂质体,并测定了脂质体体外转染活性、p H 敏特性、细胞毒性和对 A S O D N 抗流感病毒活性的影响 结果发现,批号为 98051903,98051102 和 98051202 的脂质体具有较高转染活性,但只有lipofectin 转染活性的 1/50~1/100当质粒/脂质体( W / W )为 1∶4~1∶8,转染时间为 3~5 h,质粒量为 05 μg,转染后 24~48 h 内检测时转染活性最高 脂质体 98051202 表现明显 p H值依赖溶解红细胞膜特性,而脂质体 98051102 和 98051903 的 p H 敏特性不明显 脂质体细胞毒性明显降低,如 98051903、98051102 和 98051202 的毒性分别是 lipofectin 毒性的 1/16、1/8 和 1/4p H 敏脂质体 98051202 具有促进 A S O D N 抗流感病毒作用,当 A S O D N 浓度为 02 μm ol/ L 时,p H 敏脂质体 98051202 使其抗病毒活性提高 5 倍,但 A S O D N 浓度较高时p H 敏脂质体对 A S O D N抗  相似文献

Novel antisense peptide nucleic acid (PNA) constructs targeting p75NTR as a potential therapeutic strategy for amyotrophic lateral sclerosis (ALS) were designed, synthesised and evaluated against phosphorothioate oligonucleotide sequences (PS-ODN). An 11-mer antisense PNA directed at the initiation codon dose-dependently inhibited p75NTR expression and death signalling by nerve growth factor in Schwann cell cultures. Inhibition of p75NTR production was not detected in cultures treated with the nonsense PNA or antisense PNA directed at the 3'-terminus sequence. The 19-mer PS-ODN sequences also failed to confer any activity against p75NTR but, unlike the PNA sequences, were toxic in vitro at comparable doses.  相似文献

Oligodeoxyribonucleotides (ODN) linked at their 5'-end with dimethoxytrityl (DmTr) residue were examined for antiviral activities against human immunodeficiency virus type 1 (HIV-1). We found that guanine-rich oligonucleotides exhibit anti-HIV activity upon 5'-end modification with DmTr. One oligonucleotide, DmTr-TGGGAGGTGGGTCTG (SA-1042), showed potent anti-HIV activity in vitro. A greater than 95% reduction of infectivity was observed if the cells were treated with 10 micrograms/ml of SA-1042 at the time of viral infection, PCR analysis confirmed that there was a significant reduction of provirus in the cells exposed to virus in the presence of SA-1042. By contrast, no inhibition was observed if the cells were treated with the oligomer 1 h after virus adsorption. SA-1042 prevented syncytium formation between chronically infected cells and CD4 positive uninfected cells. Furthermore, the oligomer interfered the interaction of purified gp120 to the CD4 receptor. By contrast, SA-1042 had no inhibitory effect on chronically HIV-infected cells. These results strongly suggest that the DMTr-ODNs with appropriate base sequences antagonize HIV-1 infection during the stage of virus-cell interaction.  相似文献

The use of antisense oligodeoxynucleotides (ODNs) to inhibit the expression of specific mRNA targets represents a powerful technology for control of gene expression. Cationic lipids and polymers are frequently used to improve the delivery of ODNs to cells, but the resulting complexes often aggregate, bind to serum components, and are trafficked poorly within cells. We show that the addition of a synthetic, pH-sensitive, membrane-disrupting polyanion, poly(propylacrylic acid) (PPAA), improves the in vitro efficiency of the cationic lipid, DOTAP, with regard to oligonucleotide delivery and antisense activity. In characterization studies, ODN complexation with DOTAP/ODN was maintained even when substantial amounts of PPAA were added. The formulation also exhibited partial protection of phosphodiester oligonucleotides against enzymatic digestion. In Chinese hamster ovary (CHO) cells, incorporation of PPAA in DOTAP/ODN complexes improved 2- to 3-fold the cellular uptake of fluorescently tagged oligonucleotides. DOTAP/ODN complexes containing PPAA also maintained high levels of uptake into cells upon exposure to serum. Addition of PPAA to DOTAP/ODN complexes enhanced the antisense activity (using GFP as the target) over a range of PPAA concentrations in both serum-free, and to a lesser extent, serum-containing media. Thus, PPAA is a useful adjunct that improves the lipid-mediated delivery of oligonucleotides.  相似文献

A marine Pseudomonas species WAK-1 strain simultaneously produces extracellular glycosaminoglycan and sulfated polysaccharide. Among the antiviral activities tested for these polysaccharides, the latter showed anti-HSV-1 activity in RPMI 8226 cells (50% effective concentration is 1.4 μg/ml). Oversulfated derivatives of these polysaccharides prepared by dicyclohexylcarbodiimide-mediated reaction for both polysaccharides showed antiviral activities against influenza virus type A (for glycosaminoglycan, 50% effective concentration is 11.0 μg/ml; for another, 2.9 μg/ml). Glycosaminoglycan, sulfated polysaccharide, and their chemically synthesized oversulfated derivatives did not show antiviral activities against influenza virus type B and human immunodeficiency virus type 1. No cytotoxicity of these products was noted against host cells at the 50% cytotoxic concentration of 100 μg/ml, except that naturally occurring sulfated polysaccharide had 50% cytotoxicity against MT-4 cells at 8–21 μg/ml.
