研究壳聚糖吸附和戊二醛交联对木聚糖酶固定化条件 .将酶液加入到经醋酸溶液处理过的脱乙酰壳聚糖的pH 4 8的悬液中 ,加入浓度为 0 3%~ 0 4 %的戊二醛溶液 ,室温下 ,8h后得到固定化酶 .固定化酶的半失活温度比游离酶高 ,由 5 1℃升至 71℃ ,Km 值由游离酶的 1 2mg ml增加到1 5mg ml ,最适反应温度也由 5 5℃增加到 71℃ ,而最适反应pH由 4 6下降到 3 8.该固定化木聚糖酶可用于制造低聚木糖 .经过 10次连续应用实验后 ,该固定化酶的活力保持 81%  相似文献

对谷胱甘肽硫转移酶的固定化、游离酶和固定化酶的酶学特性进行了研究,通过试验,确定谷胱甘肽硫转移酶的最佳固定化条件为先用2%壳聚糖吸附酶,然后再加戊二醛交联,交联用戊二醛浓度为1.2%,交联时间6h;游离酶的最适温度为45—55℃,最适pH值为6.5-7.0:固定化酶的最适温度为45-50℃,最适pH值为7.0;游离酶和固定化酶的最适酶促反应时间为30min。  相似文献

从青霉菌m8提取出木聚糖酶,将其固定在用戊二醛交联的壳聚糖载体上。1.0g壳聚糖与4%的二醛结合固定3.5mg蛋白,酶活回收率为46.6%。在酶的最适pH为4.6,固定化酶为pH3.8。原酶的最适温度为55℃,固定化酶在60-75℃都具有较高活性。固定化酶的耐热性优于原酶,固定化酶的表现Km值略低于原酶,前者为5.0×10-2g/L,后者为3.58×10-2g/L。  相似文献

聚乙二醇二缩水甘油醚(PEGDGE)作为双功能环氧试剂,在实验中被用于交联氨基载体LX-1000EA共价固定化海洋脂肪酶,经过处理后的载体共价固定化脂肪酶具有良好的效果。实验经过单因素初筛和正交试验,得到最佳的交联及固定化条件为0. 75%交联剂浓度、交联温度35℃、交联时间3h、载体量1. 25g、pH9. 0、固定化温度55℃、固定化时间1h。对LX-1000EAPEGDGE固定化酶与游离酶、戊二醛(GA)交联LX-1000HA-GA的固定化酶进行酶学性质的比较,发现LX-1000EA-PEGDGE固定化酶较游离酶最适反应温度未改变,与LX-1000HA-GA相同的是最适反应pH都由7. 0提高为8. 0。在最适条件中所测LX-1000EA-PEGDGE酶活达到78. 84U/g,固定化改变了游离酶的酸碱耐受性,热稳定性和操作稳定性较游离酶和LX-1000HA-GA固定化酶均有提高。LX-1000EA-PEGDGE的热稳定表现优异,在60℃孵育3h后保留90%酶活;使用5次后仍能残余50%酶活;保存30天酶活仍保留60%。首次使用新型双环氧交联剂PEGDGE交联有机氨基载体共价结合固定化脂肪酶,为更有效的固定化方法提供了技术支持,同时也发现交联剂对固定化酶的性质存在较大影响。  相似文献

分别采用海藻酸钠、明胶和壳聚糖为载体,并以戊二醛为交联剂,通过包埋-交联和吸附-交联两种耦合固定化方法制备固定化锰过氧化物酶。探讨了酶的不同固定化条件和固定化酶的部分性能。与游离酶相比,制备的3种固定化酶最适反应pH分别由7·0降低到5·0、5·0和3·0,最适反应温度分别由35℃升高到75℃、55℃和75℃。3种固定化酶的耐热性都显著提高,其中用壳聚糖制成的固定化酶在pH2·2~11的宽范围内表现出很好的酸碱耐受性。30℃连续测定6~9次酶活力,重复使用的3种固定化酶显示出良好的稳定性。将固定化酶应用在偶氮染料的脱色中,用明胶制成的固定化酶在静置和摇床条件下,以及用海藻酸钠制成的固定化酶在摇床条件下,均表现出与游离酶相近的脱色能力,并且在重复进行的摇床实验中,脱色能力未降低,反应前后的酶活力均没有损失。  相似文献

将胶原纤维用三价铁改性后作为载体,通过戊二醛的交联作用将过氧化氢酶固定在该载体上。制备的固定化过氧化氢酶蛋白固载量为16.7 mg/g,酶活收率为35%。研究了固定化酶与自由酶的最适pH、最适温度、热稳定性、贮存稳定性及操作稳定性。结果表明:过氧化氢酶经此法固定化后,最适pH及最适温度与自由酶相同,分别为pH 7.0和25 ℃;但固定化酶的热稳定性显著提高,在75 ℃保存5 h后,仍能保留30%的活力,而自由酶则完全失活;固定化酶在室温下保存12 d后,酶活力仍保持在88%以上,而自由酶在此条件下则完全失  相似文献

目的:利用壳聚糖固定日本血吸虫谷胱甘肽硫转移酶,并对固定化酶性质及体外催化活性进行研究。方法:利用大肠杆菌BL21(DE3)表达日本血吸虫谷胱甘肽硫转移酶,并从中粗提谷胱甘肽硫转移酶,将其固定在用戊二醛交联的壳聚糖载体上,对游离酶和固定酶的最适pH、温度,游离酶和固定化酶对底物1-氯-2,4-二硝基苯(CDNB)和谷胱甘肽(GSH)的亲和力,温度的稳定性进行了研究。结果:固定化酶活回收率可达41.6%,最适pH6.5~7.0,最适温度37℃,对底物(CDNB和GSH)的亲和力略有下降,对温度稳定性大大提高。在体外固定化酶有很好的催化解毒作用。  相似文献

在浸润条件下,以0.5%(v/v)戊二醛交联的高分子膜尼龙载体固定化木瓜蛋白酶。对固定化条件进行了优化,比较了固定化酶与游离酶的酶学参数。结果表明,4℃、pH6.0条件下,将膜载体浸润于2mg/mL酶液中5h,固定化酶活为303.4U/g。固定化酶最适反应pH为6.0~7.0,最适反应温度为65℃。其pH稳定性、热稳定性均比游离酶高。  相似文献

采用壳聚糖微球对琼脂酶进行固定化,在单因素实验的基础上用正交试验法确定最佳固定化工艺。结果表明:在戊二醛体积分数为2.5%,交联时间为6 h,加酶量为15 mL,固定时间为3 h时固定酶的活力最高;固定化酶的最适反应温度及最适pH分别为50℃和8.5,高于游离酶;同时其热稳定性及操作稳定性均高于游离酶。  相似文献

共价结合法是重要的工业酶固定化方法,利用稳定的共价键固定化工业酶,在载体和酶间形成多点共价连接,可以制备稳定性较好的固定化酶,更具有实际应用价值。利用氨基载体共价结合固定化海洋假丝酵母脂肪酶,采用较为廉价的戊二醛进行辅助交联,通过单因素和正交试验,确定最佳固定化条件为:25℃、pH5. 0、0. 1%戊二醛、0. 25g载体、交联0. 5h、固定化1h、加酶量为800U,最终得到的固定化酶酶活达到83. 01U/g。固定化脂肪酶的最适pH较游离酶向碱性方向偏移,最适反应温度提高10℃,固定化酶的热稳定性和酸碱稳定性比游离酶好且重复使用性和储存稳定性明显优于游离酶。同时发现交联剂是制备固定化脂肪酶的重要因素,因此探索新型交联剂对于固定化效果的提高具有重要意义,为海洋假丝酵母脂肪酶的固定化工艺技术和工业应用奠定了良好基础。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.
