【目的】探究不同腐殖酸浓度下参与含砷水铁矿转化的微生物类群组成和丰度变化及对砷释放的影响,预测原位高砷含水层中功能微生物群参与有机质—含砷铁矿物转化过程对砷转化释放的作用。【方法】对河套平原高砷地下水和同深度高砷沉积物中的铁还原功能群落进行富集培养,构建室内厌氧微宇宙体系,将富集菌群分别加入到实验室条件下合成的不同浓度腐殖酸(0、1.5、7、14 mg C/L)-含砷水铁矿体系中,通过体系中砷、铁形态及浓度的变化分析,结合高通量测序技术、X-射线衍射(X-ray diffractometer, XRD),探究不同条件下砷的释放固定和群落的演替。【结果】高砷地下水组(G组)和沉积物组(S组)富集得到的铁还原功能群落具有明显差异,G组中以Aeromonadaceae为特殊优势菌群,而S组中以Shewanellaceae为特殊优势菌群。微宇宙实验结果显示,S组的铁还原量相对较高且速率较快;G组与S组中液相砷形态存在明显差异,整个培养期内G组均以As(Ⅴ)为主,而S组中前期以As(Ⅴ)为主,当反应到达20 d时液相As(Ⅲ)高达3.4μmol/L,推测此时具有砷还原功能的群落占优势地位。当反应...  相似文献

【目的】冷泉系统广泛存在于大陆边缘地区,其典型特征是在海底渗漏出大量富含以甲烷为主的碳氢化合物和硫化氢等成分的低温流体。冷泉也因其独特的地球化学条件孕育着独特的原核微生物群落结构,然而,原核微生物组成与冷泉环境之间的响应关系却并不清楚。【方法】本文以莫克兰大陆边缘活跃冷泉区沉积物柱状样为研究对象,沿深度剖面分析了沉积物中的CH4以及孔隙水SO42–、H2S浓度等关键地球化学参数,并基于16S rRNA基因高通量测序对冷泉沉积物原核微生物的群落结构及其空间变化进行了系统分析。【结果】根据其硫酸盐-甲烷浓度剖面特征,从上向下,将沉积物垂向剖面划分为硫酸盐还原区(SZ)、硫酸盐-甲烷转换区(SMTZ)和产甲烷区(MZ)。通过原核微生物α多样性与基因定量研究发现,随着深度增加微生物多样性与丰度呈逐渐降低的趋势。16SrRNA基因高通量测序结果表明,SZ中以硫氧化细菌γ-变形菌纲、α-变形菌纲和埃普西隆杆菌门为主,且以硫酸盐为电子受体的与有机质降解相关的原核微生物JS1、绿弯菌门、洛基古菌纲、深古菌纲及底栖古菌纲的相对含量也较高;SMTZ存在较高含量的ANME-1a、ANME-1b与SEEP-S...  相似文献

【目的】从海洋沉积物中富集获得硫酸盐还原菌群,改变pH值进行培养,分析pH值对硫酸盐还原性质的影响,明确菌群组成和进行硫酸盐还原功能基因预测,探究硫酸盐还原机制。【方法】分析硫酸盐还原菌群在不同pH值条件下的硫酸盐还原率,在此基础上,利用高通量测序技术和PICRUSt软件分析硫酸盐还原菌群优势菌组成及硫酸盐还原相关基因相对丰度。【结果】硫酸盐还原菌群在不同pH值培养条件下的生长和硫酸盐还原率出现显著变化(P<0.01),在pH 5.0时达到峰值,分别为0.34±0.01和96.52%±0.44%。高通量测序数据显示,pH 5.0时菌群丰富度和多样性最高,优势菌属为假单胞菌(Pseudomonas)和芽孢杆菌(Bacillus),相对丰度较高的基因为同化性硫酸盐还原相关基因。【结论】硫酸盐还原菌富集生长的最适pH 5.0,在此条件下的高硫酸盐还原率由同化性硫酸盐还原途径主导,为揭示硫酸盐还原机制提供了实验支持,并拓宽了硫酸盐还原菌实践应用方面的种质资源。  相似文献

【背景】海草床是重要的"蓝碳"生态系统,对全球碳汇有重要贡献。海草床沉积物剖面的垂直梯度特征显著,表层呈现氧化态,富含活性有机质,而深层呈还原态,以惰性有机质为主。【目的】探究这种垂直特征如何影响微生物的丰度和群落分布。【方法】利用荧光定量PCR和16SrRNA基因高通量测序等手段,测定了山东荣成天鹅湖大叶藻海草床不同深度(5、10、15、20、25和30 cm)沉积物中细菌和古菌丰度、多样性和群落结构的变化。【结果】细菌和古菌16S rRNA基因拷贝数随深度的增加而降低,在沉积物5cm深处,细菌的16SrRNA基因拷贝数显著高于20cm和30cm层(ANOVA,P0.05)。深度对细菌和古菌α多样性指数没有显著影响(P0.05)。细菌中相对丰度最高的是变形菌门,其次是绿弯菌门,拟杆菌门,浮霉菌门等,其中δ-变形菌和浮霉菌的相对丰度随深度显著增加(P0.05)。古菌群落中深古菌门比例最高,在25cm深处达到70%以上;其次是乌斯菌门、洛基古菌门、广古菌门和奇古菌门等。奇古菌门比例随深度增加而显著降低(P0.05),其他古菌类群在不同深度间差异不显著(P0.05)。【结论】海草床沉积物细菌和古菌的丰度、多样性和群落分布具有明显的垂直特征,这种特征可能受沉积物有机质组成和氧化还原状况影响。  相似文献

【背景】从活性污泥中分离出一类具有硫酸盐还原能力的菌株,探讨了其用于铅锌冶炼渣重金属污染修复的可行性。【目的】研究硫酸盐还原菌(Sulfate reducing bacteria)对铅锌冶炼渣中重金属的固化作用。【方法】将从活性污泥中分离出的硫酸盐还原菌接种到铅锌冶炼渣中进行修复,采用X射线衍射、Tessier、电感耦合等离子体发射光谱仪检测、高通量测序等方法进行实验。检测铅锌冶炼渣中矿物组成,以及修复过程中重金属化学形态、各金属离子浓度和微生物群落结构的变化。【结果】修复实验表明,体系中电位降低、pH值提高、各重金属稳定态增加、离子浓度降低且微生物群落结构变化显著,硫酸盐还原菌成为优势菌群。【结论】接种硫酸盐还原菌后铅锌冶炼渣中的重金属原位固化效果显著,从而降低生物可利用性,将恶性循环变为良性循环,所以硫酸盐还原菌可用作重金属污染修复的固化药剂。  相似文献

【背景】土壤微生物对其生存的微环境变化极为敏感,鸟岛作为湖滨湿地,对气候变化具有敏感性,但目前关于青海湖鸟岛的土壤微生物鲜有研究。【目的】探究气候变暖后青海湖鸟岛土壤微生物群落特征的变化。【方法】利用开顶箱模拟增温,通过高通量测序方法了解温度升高后土壤细菌及真菌的群落结构以及多样性的变化情况。【结果】温度的升高并未改变青海湖鸟岛土壤微生物的优势菌群,细菌优势菌群为变形菌门和酸杆菌门;真菌优势菌门为子囊菌门,优势菌纲为座囊菌纲。但增温改变了土壤微生物的群落结构,显著升高了拟杆菌门、蓝细菌门、Patescibacteria及球囊菌纲的相对丰度,显著降低了锤舌菌纲的相对丰度。土壤微生物群落的多样性指数也发生了变化,温度上升后微生物的ACE指数及Chao1指数均降低,细菌的Simpson指数及真菌的Shannon指数降低。【结论】青海湖鸟岛土壤微生物对温度升高的响应明显,增温改变了土壤细菌拟杆菌门、蓝细菌门、Patescibacteria的相对丰度及真菌的球囊菌纲、锤舌菌纲的相对丰度,降低了土壤微生物的多样性。  相似文献

【背景】甲烷厌氧氧化(anaerobic oxidation of methane, AOM)包含反硝化型甲烷厌氧氧化和硫酸盐还原型甲烷厌氧氧化。目前,人们向水体中排放过量的含氮及含硫污染物,引起了严重的环境污染和生态破坏。【目的】利用甲烷厌氧氧化微生物燃料电池(microbial fuel cell, MFC)研究同步脱氮除硫耦合反应机理及反应过程中微生物的多样性信息。【方法】构建了3个微生物燃料电池(N-S-MFC、N-MFC、S-MFC),以甲烷作为唯一碳源,探究其同步脱氮除硫性能,并采用16S rRNA基因高通量测序技术对微生物群落结构进行分析。【结果】N-S-MFC中硝酸盐和硫酸盐的去除率分别为90.91%和18.46%。阳极室中微生物的相对丰度提高,与反硝化及硫酸盐还原菌相关的微生物大量富集,如门水平上拟杆菌门(Bacteroidota)、厚壁菌门(Firmicutes)和脱硫杆菌门(Desulfobacterota),同时属水平上Methylobacterium_Methylorubrum、Methylocaldum、Methylomonas等常见的甲烷氧化菌增多。【结论...  相似文献

【目的】通过研究林地转型耕地对土壤编码碱性磷酸酶基因的细菌群落丰度、多样性和结构的影响,为丘陵区耕地长期施肥下农田土壤微生物多样性丧失的影响机制以及未来的退耕还林过程中土壤微生物多样性的提升和土地可持续利用研究提供一些基础数据和技术支撑。【方法】采用实时荧光定量PCR (real-time quantitative PCR,qPCR)和高通量测序技术解析土壤编码碱性磷酸酶基因的细菌群落的丰度、多样性和结构变化,并耦合土壤化学性质分析,明确土壤编码碱性磷酸酶基因的细菌群落丰度和多样性与土壤化学性质的关系以及关键的驱动因子。【结果】林地垦殖为农田后,长期施肥导致土壤酸化,pH从5.58降至4.72,而土壤速效磷则从2.49 mg/kg增至49.3 mg/kg。相应地,耕地土壤编码碱性磷酸酶基因的细菌群落的丰度和Shannon指数均显著低于林地。基于编码碱性磷酸酶的phoD基因(alkaline phosphatase-encoding gene)序列的物种分类表明,丘陵区土壤编码碱性磷酸酶基因的细菌群落的优势门为变形菌门(Proteobacteria)、蓝藻门(Cyanobacteria)、浮霉菌门(Planctomycetes)、放线菌门(Actinobacteria)、厚壁菌门(Firmicutes)和疣微菌门(Verrucomicrobia),其中林地土壤的蓝藻门的相对丰度显著高于耕地。耕地土壤的慢生根瘤菌属(Bradyrhizobium)和芽孢杆菌属(Bacillus)的相对丰度显著高于林地,而中慢生根瘤菌属(Mesorhizobium)、假单胞菌属(Pseudomonas)、Chlorogloea属、Gemmata属、Phormidesmis属和Pseudolabrys属的相对丰度显著低于林地。土壤编码碱性磷酸酶基因的细菌群落结构因林地转型耕地而发生显著改变。phoD基因丰度和Shannon指数与pH显著正相关,而与总磷、速效磷、硝态氮和铵态氮均显著负相关,其中土壤速效磷是这些影响因素中影响最强烈的,长期施用无机磷肥导致含碱性磷酸酶的土壤细菌群落对有机磷分解的能力退化。【结论】林地转型耕地加之长期施肥改变了土壤pH和速效磷,并在其他理化因子的协同驱动下,导致土壤编码碱性磷酸酶基因的细菌群落丰度、多样性和结构的显著变化。  相似文献

【目的】通过考察Mn2+对A/O-BAF系统处理效能及微生物群落多样性的影响,研究了15℃下不同浓度Mn2+对A/O-BAF系统处理效能的影响,并通过高通量测序考察了细菌群落多样性的变化情况。【方法】在温度15℃、水力负荷0.50 m3/(m2·h)、气水比10:1的条件下,在进水中投加Mn2+,考察反应器处理效能的变化情况,并通过高通量测序对BAF生物膜样品进行分析。【结果】2.0 mg/L Mn2+作用下A/O-BAF系统的COD、NH4+-N、TN、TP去除率分别提高3.51%、2.21%、6.26%和12.13%;4.0 mg/L Mn2+作用下COD、NH4+-N、TN、TP去除率分别提高了4.24%、1.92%、7.75%和10.73%;Mn2+作用下A/O-BAF系统内反硝化细菌和亚硝酸菌的数量明显增加,硝酸菌数量有所减少,生物膜量和生物膜活性提高。高通量测序结果可知:2.0 mg/L和4.0 mg/L Mn2+作用下,微生物群落多样性大幅增加,样本OTU数量从4430分别提高到5659和5556;BAF柱内变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)、黄色单胞菌科(Xanthomonas)、红环菌科(Rhodocyclaceae)、丛毛单胞菌科(Comamonadacea)、丛毛单胞菌属(Comamonas)、陶厄式菌属(Thauera)、Denitratisoma、Sulfuritalea和热单胞菌属(Thermomonas)的相对丰度增加。部分微生物丰度[例如Denitratisoma、Sulfuritalea、红环菌科(Rhodocyclaceae)和丛毛单胞菌科(Comamonadacea)]的增加幅度随着Mn2+浓度的提高有所减少;Mn2+作用下硝化螺旋菌门(Nitrospirae)和硝化螺菌属(Nitrosospira)的相对丰度减少。【结论】一定浓度下Mn2+主要是通过促进A/O-BAF系统的亚硝化和反硝化作用和除磷微生物的生长提高A/O-BAF系统污染物去除效果,但对硝化的促进效果并不明显,且硝酸菌更易被Mn2+的抑制和毒害作用所影响。  相似文献

深部热水硫酸盐还原菌微滴数字PCR检测技术的建立与应用   总被引:1,自引:1,他引:0  
【背景】地下深部存在一个生物圈,深部沉积岩、玄武岩、花岗岩和变质岩等岩性环境的微生物群落已被调查,而地下深部碳酸盐岩岩溶-裂隙热储层微生物群落特征仍然不清。硫酸盐还原菌(sulfate-reducing bacteria,SRB)是地下深部频繁检出的微生物。【目的】建立快速准确定量深部热水硫酸盐还原菌的微滴数字PCR (droplet digital PCR,ddPCR)技术。【方法】以SRB的功能基因dsrB为检测目标,优化SRB ddPCR技术的退火温度,考察其线性范围、敏感性、重复性和特异性,并将该技术用于实际样品的检测。【结果】SRB ddPCR技术的最佳退火温度为54 °C,检测的线性范围为1.1×100?1.1×105 copies/μL-DNA,相关系数R2为0.996,检出限为1 copy/μL-DNA,重复性的相对标准差优于9%,对3种非SRB人工构建的质粒均没有扩增,显示该技术具有很好的线性关系、敏感性、重复性和特异性。利用该技术对冀中地热区深部热水、浅层水和土壤样品进行了检测,平均含量分别为(4.0±8.4)×103 copies/mL、(1.6±3.5)×102 copies/mL和(1.5±1.2)×103 copies/g-dw。与浅层水和土壤相比,深部热水富含SRB菌。【结论】为了提高地下深部生物圈认识和合理开发利用深部热水,建立了一种快速、灵敏、准确的SRB ddPCR检测技术,同时为其他指示菌检测技术的建立提供了参考。  相似文献

The mature embryo of seven species belonging to five genera of Indian bamboos is described. In all these the basic pattern of embryo organisation is same: the scutellar and coleoptilar bundles are not separated by an internode, the epiblast is absent, the lower portion of the scutellum and the coleorhiza are separated by a cleft and the margins of embryonic leaves overlap. The features unique to fleshy fruited bamboos are: presence of a massive scutellum, the juxtaposition of plumule and radicle and the occurrence of a bud in the axil of the coleoptile. The fleshy fruit bearing bamboos should be classified into one group, the tribeMelocanneae. Evidence is provided to recognise additional groups in the subfamilyBambusoideae.  相似文献

Komárek has recently reviewed the various species assigned to the green algal genusNeochloris Starr (Chlorococcales, Chlorococcaceae) and removed those with uninucleate vegetative cells to a new genus,Ettlia. Watanabe & Floyd, unaware ofKomárek's work, also reviewed the species ofNeochloris and distributed them among three genera—Neochloris, Chlorococcopsis gen. nov., andParietochloris gen. nov.—on the basis of details of the covering of the zoospore and the arrangement of the basal bodies of the flagellar apparatus. This paper reconciles these two treatments and makes additional recommendations at the ranks of genus, family, order, and class.  相似文献

The cardinalfishes (Apogonidae) are a diverse clade of small, mostly reef-dwelling fishes, for which a variety of morphological data have not yielded a consistent phylogeny. We use DNA sequence to hypothesize phylogenetic relationships within Apogonidae and among apogonids and other acanthomorph families, to examine patterns of evolution including the distribution of a visceral bioluminescence system. In conformance with previous studies, Apogonidae is placed in a clade with Pempheridae, Kurtidae, Leiognathidae, and Gobioidei. The apogonid genus Pseudamia is recovered outside the remainder of the family, not as sister to the superficially similar genus Gymnapogon. Species sampled from the Caribbean and Western Atlantic (Phaeoptyx, Astrapogon, and some Apogon species) form a clade, as do the larger-bodied Glossamia and Cheilodipterus. Incidence of visceral bioluminescence is found scattered throughout the phylogeny, independently for each group in which it is present. Examination of the fine structure of the visceral bioluminescence system through histology shows that light organs exhibit a range of morphologies, with some composed of complex masses of tubules (Siphamia, Pempheris, Parapriacanthus) and others lacking tubules but containing chambers formed by folds of the visceral epithelium (Acropoma, Archamia, Jaydia, and Rhabdamia). Light organs in Siphamia, Acropoma, Pempheris and Parapriacanthus are distinct from but connected to the gut; those in Archamia, Jaydia, and Rhabdamia are simply portions of the intestinal tract, and are little differentiated from the surrounding tissues. The presence or absence of symbiotic luminescent bacteria does not correlate with light organ structure; the tubular light organs of Siphamia and chambered tubes of Acropoma house bacteria, those in Pempheridae and the other Apogonidae do not.  相似文献

The Drosophila Pax6 homolog twin of eyeless (toy) is so far the first zygotically expressed gene involved in eye morphogenesis in Drosophila. The study of its expression during embryogenesis is therefore informative of the initial events of eye development in Drosophila. We have analyzed how the initial expression domain of toy at cellular blastoderm is regulated. We show that the three maternal patterning systems active in the cephalic region (the anterior, terminal and dorsal-ventral systems) cooperate with zygotically activated gap genes to shape the initial expression domain of toy. Whereas Bicoid, Dorsal and Torso signaling synergistically act as activators, Hunchback, Knirps and Decapentaplegic act as repressors.  相似文献

Twenty three species in 11 genera were examined in the field to determine hosts. OnlyStriga asiatica andSeymeria cassioides have a narrow host range being restricted to grasses and pines, respectively. Cyanobacteria and/or azolla were inoculated, with urea at 0, 72 or 144 kg N/ha, in plots in which azolla-free Indica rice var. IR 28 was grown. Productive tillers, yield and nitrogen contents of grain and straw positively responded to inoculation with cyanobacteria or azolla, even with fertilizer-N up to 144 kg N/ha. Inoculation improved colonization by cyanobacteria. Azolla were superior to the asymbiotic cyanobacteria in enhancing rice performance. Urea at a rate of 72 kg N/ha was found to support the best colonizations when applied with cyanobacteria or azolla or, to give maximum rice yields, both inoculants.  相似文献

Intracellular cysteine aspartate-specific proteases (caspases) play both signaling and effector roles in realizing the program of cell death. Caspases function as proteolytic cascades unique for each cell type and signal triggering apoptosis. All parts of the proteolytic cascades are duplicated and controlled by feedback signals. Amplification cycles between pairs of caspases (the third and the sixth, the ninth and the third, the twelfth and the sixth, and others) help multiply the initial apoptotic signal. The presence of physiological inhibitors of apoptosis that directly interact with caspases creates a multilevel regulatory network of apoptosis in cell. The caspase proteolytic cascades are also regulated by sphingolipid secondary messengers, among them ceramide, sphingosine, and their phosphates. Moreover, an association of the caspase signaling with ubiquitin-dependent proteolysis is shown in cells. In particular, the use of extracellular activators and inhibitors of caspases allows irreversible activation of apoptosis in tumor cells or the prevention of apoptosis in cortical neurons under neurodegenerative diseases.  相似文献

通过对采自广西24个县(市)的1 147份青藓科植物标本的逐一鉴定及相关文献的查阅,确认有广西青藓科植物11属、44种,其中包括广西青藓科植物新记录属1属,即拟异叶藓属(Pseudokindbergia),新记录种7种,分别为匐枝青藓(Brachythecium procumbens)、阔叶尖喙藓(Oxyrrhynchium latifolium)、泛生尖喙藓(O. vagans)、拟异叶藓(Pseudokindbergia dumosa)、华东细喙藓(Rhynchostegiella sinensis)、长肋拟青藓(Sciurohypnum populeum)和弯叶拟青藓(S. reflexum)。该文提供了修订后的广西青藓科植物名录,并对其中的新记录属、种的主要形态学识别特征、生境和地理分布等进行了详细描述。  相似文献

Four phenolic amides, dihydro-N-caffeoyltyramine (1), trans-N-feruloyloctopamine (2), trans-N -caffeoyltyramine (3), and cis-N-caffeoyltyramine (4), were isolated from an ethyl acetate extract of the root bark of Lycium chinense Miller. All had an anti-fungal effect; compounds 1-3 were potent at 5-10 microg ml(-1) and were without hemolytic activity against human erythrocyte cells. Compound 4 was active at 40 microg ml(-1). All four compounds impeded the dimorphic transition of pathogen, Candida albicans.  相似文献
