中国林蛙和中华蟾蜍皮肤抗菌肽的分离纯化及其抗菌活性   总被引:1,自引:0,他引:1  
分别以中国林蛙长白山亚种Rana chensinensis changbaishansis和中华蟾蜍Bufo gargarizans的鲜皮为原料,通过酸化乙醇法提取抗菌肽粗提液,再经葡聚糖凝胶层析进一步分离纯化获得抗菌肽纯品,采用滤纸片法进行抑菌活性研究.结果 表明,经Sephadex G-50和Sephadex G-100分离纯化后获得3种多肽,中国林蛙与中华蟾蜍皮肤中的活性多肽对革兰氏阴性和革兰氏阳性细菌都具有一定的抗菌作用,其中多肽Ⅲ具有最佳的抑菌效果.抗菌肽相对含量比较的结果表明,蟾蜍皮肤中抗菌活性肽的含量较高,是理想的抗菌肽提出和纯化的源材料.  相似文献

低能离子诱变筛选获得的枯草芽孢杆菌JA-206的发酵粗提液对红色毛癣菌和须毛癣菌有较强的抑菌或杀菌作用,粗提液经高温、蛋白酶处理后仍然有较高的稳定性,将JA-206粗提液经多步分离纯化,得到三种单一纯化产物抗菌肽AFP1、AFP2和AFP3,其中AFP1和AFP3对两种皮肤癣菌显示出明显的抑菌或杀菌作用。  相似文献

通过抑菌试验,确定不同培养基条件下多肽化合物的抑菌效果,并测定其最小抑菌浓度(Minimum inhibitory concentration,MIC)。加入Vc,比较抑菌效果,进一步优化筛选H37Ra抑制剂。先根据ELISA试验结果进行噬菌体肽库筛选,然后利用分子对接软件模拟多肽与ICL蛋白晶体(1F8I)的分子对接,将成功对接的多肽采用Fmoc固相合成法合成,并对其生物活性进行检测。用制备型反相高效液相色谱仪对合成的多肽进行纯化并进行质谱检测。共筛选得到4条多肽且均有抑菌效果,并与剂量相关。结果显示,浓度为800-1 500μg/mL时,各组多肽均抑菌。浓度为500μg/mL时,正常培养基中只有一种肽有抑菌作用,而限制碳源培养基中均不能抑菌。2号肽抑菌效果最好,正常培养基中MIC为200μg/mL,限制碳源中为500μg/mL。阳性对照组,两种培养基抑菌效果一致,利福平MIC为0.8μg/mL,异烟肼MIC为0.5μg/mL。根据MIC结果,在正常培养基中加入Vc,检测到多肽、利福平和异烟肼的抑菌效果呈剂量依赖性升高。  相似文献

目的 从变形链球菌临床株的液体培养基中分离纯化变链素,为进一步从分子水平研究变链素奠定基础.方法 通过抑菌活性检测,从变链临床株中选择出抑菌活性较强的菌株.用氯仿抽提法从该菌株的培养液中粗提变链素,经固相萃取和反相高效液相色谱(RP-HPLC)对粗提物进行纯化.结果 获得变链素活性较强的菌株"1G".从其200 ml液体培养基中粗提出变链素约15 μg,经固相萃取柱洗脱,再经过RP-HPLC 2次纯化,得到有抑菌活性的成分,此为纯化的变链素.结论 变链素分子量小,分离提纯步骤复杂,本实验得到纯化的变链素,为下一步研究变链素的氨基酸序列和基因序列奠定了基础.  相似文献

为了探究苦荞多肽粗提液抗氧化活性的主要来源,需要进一步对苦荞多肽进行分离纯化,通过结构鉴定来分析抗氧化活性与多肽结构之间的关系。该研究以苦荞功能提取物废渣为原料提取苦荞粗蛋白辅以植物乳杆菌发酵法制备苦荞多肽,采用大孔吸附树脂、葡聚糖凝胶、液相色谱对苦荞多肽粗提液进行分离纯化,并以抗氧化活性为指标,筛选抗氧化活性较强的组分,利用液相色谱-质谱联用技术鉴定抗氧化多肽结构。结果表明,纯度不同的苦荞多肽抗氧化能力也不同,经过Sephadex G-15葡聚糖凝胶分离纯化得到的T-3组分ABTS和DPPH自由基清除能力最优,清除率分别为96%、94%。纯化后苦荞多肽的抗氧化活性显著高于粗提液(P<0.05),但是抗氧化活性并没有完全与苦荞多肽纯度呈正相关,通过液相色谱进一步分离纯化T-3组分后得到的组分T-3-1,苦荞多肽纯度虽然达到了98%,但是对DPPH和ABTS抑制率不再增强,反而稍有下降,抑制率分别为89%和90%。由质谱鉴定结果得到,主要抗氧化肽的氨基酸序列为苯丙氨酸-脯氨酸-酪氨酸Phe-Pro-Tyr(FPY)和酪氨酸-亮氨酸-脯氨酸-苯丙氨酸Tyr-Leu-Pro-Phe(YLPF)。研究结果以期为苦荞多肽的进一步开发利用提供一定的理论基础。  相似文献

以金黄色葡萄球菌(Staphylococcus aureus)、克氏库克菌(Kocuria kristinae)、粪肠球菌(Streptococcus faecalis)、大肠杆菌(Escherichia coli)、鼠伤寒沙门氏菌(Salmonella typhimurium)、阪崎肠杆菌(Enterobacter sakazakii)和绿脓假单胞菌(Pesudomonas pyocyaneum)为受试菌,采用滤纸片法检测桦褐孔菌(Inonotus obliquus)发酵浸膏各极性部位的抑菌活性。利用色谱技术进行分离纯化,根据理化性质和谱学数据进行结构鉴定,确定了桦褐孔菌石油醚提取物的化学成分及乙酸乙酯层抑菌单体化合物的结构。结果表明:桦褐孔菌发酵浸膏的乙酸乙酯层抑菌活性较强,通过分离纯化,从该层分离得到了苯甲酸,并确定为主要抑菌活性成分。  相似文献

分别以总抗氧化活性(T-AOC)和多肽含量为评价指标,通过单因素实验分析和Box-Behnken响应面分析法对豆粕固态发酵工艺条件进行优化,得到最佳发酵工艺条件为:接种量9.6%,料水比1∶0.9,发酵温度35℃。豆粕在此条件下发酵48h,其总抗氧化活性与多肽含量分别由发酵前的207.2U/g和21.0mg/g提高到1091.1U/g和347.7mg/g,分别提高了4.3倍和15.6倍,与预测值相比,结果吻合性良好。总抗氧化活性与多肽含量的相关性良好(R2=0.8604),表明得到的多肽为抗氧化肽。  相似文献

采用凝胶层析对乳酸菌粗提液进行分离、纯化,得到乳酸菌肽纯化样品,对其含量、纯度及抑菌效果等指标进行了测定,得到纯化时间在14~18 h所收集的样品含量最高、纯度最高、抑菌效果最好。从而获得了重要的乳酸菌肽研究资料,为今后更进一步的研究打下基础。  相似文献

以脱脂豆粕粉为原料,接入产蛋白酶能力较强的菌株进行发酵,通过菌株所产蛋白酶作用于豆粕粉中的大豆蛋白将其水解为大豆多肽。以蛋白酶活力及水解度为指标逐步筛选降解豆粕的菌株,获得菌株CHD16;以水解度作为指标,对菌株CHD16发酵降解豆粕的条件进行了优化。通过液体发酵探讨菌株CHD16适宜的培养基组成、接种时间、接种量与发酵时间。结果表明:在豆粕含量11%,蔗糖含量2%的液体发酵培养基中接种3%的菌龄为21h的液体种子,于温度40℃、转速120r/min条件下发酵48h,豆粕蛋白的水解度可达58%,比条件优化前的水解度提高了88%。  相似文献

Two fermented milks containing angiotensin-I-converting-enzyme (ACE)-inhibitory peptides were produced by using selected Lactobacillus delbrueckii subsp. bulgaricus SS1 and L. lactis subsp. cremoris FT4. The pH 4.6-soluble nitrogen fraction of the two fermented milks was fractionated by reversed-phase fast-protein liquid chromatography. The fractions which showed the highest ACE-inhibitory indexes were further purified, and the related peptides were sequenced by tandem fast atom bombardment-mass spectrometry. The most inhibitory fractions of the milk fermented by L. delbrueckii subsp. bulgaricus SS1 contained the sequences of beta-casein (beta-CN) fragment 6-14 (f6-14), f7-14, f73-82, f74-82, and f75-82. Those from the milk fermented by L. lactis subsp. cremoris FT4 contained the sequences of beta-CN f7-14, f47-52, and f169-175 and kappa-CN f155-160 and f152-160. Most of these sequences had features in common with other ACE-inhibitory peptides reported in the literature. In particular, the beta-CN f47-52 sequence had high homology with that of angiotensin-II. Some of these peptides were chemically synthesized. The 50% inhibitory concentrations (IC(50)s) of the crude purified fractions containing the peptide mixture were very low (8.0 to 11.2 mg/liter). When the synthesized peptides were used individually, the ACE-inhibitory activity was confirmed but the IC(50)s increased considerably. A strengthened inhibitory effect of the peptide mixtures with respect to the activity of individual peptides was presumed. Once generated, the inhibitory peptides were resistant to further proteolysis either during dairy processing or by trypsin and chymotrypsin.  相似文献

Two distinct domains with antibacterial activity were isolated from a peptic hydrolysate of bovine alpha(s2)-casein. The digested alpha(s2)-casein was fractionated by cation-exchange chromatography, after which the peptides in the two active fractions obtained were separated by high-performance liquid chromatography and sequenced by electrospray-ionization tandem mass spectrometry. The major component in each active fraction, f(183-207) and f(164-179), was further purified and the antibacterial activity of these components was tested against several microorganisms. Depending on the target bacterial strain, these peptides exhibited minimum inhibitory concentrations between 8 and 99 microM. Peptide f(183-207) exhibited a consistently higher antibacterial activity than f(164-179), although both peptides showed a comparable hemolytic effect. A method of in situ enzymatic hydrolysis on a cation-exchange membrane to obtain a fraction enriched in the most active antibacterial domain is presented. The antibacterial and hemolytic activities are discussed in relation to the structure and hydrophobicity of the peptides.  相似文献

Soybean meal was fermented withAspergillus usamii to improve zinc availability through the degradation of phytic acid. Rats fed a diet containing fermented soybean meal showed greater femoral zinc than did animals fed a diet containing regular soybean meal. Zinc solubility in the small intestine was higher in the rats fed fermented soybean meal than in the rats fed regular soybean meal. These results suggested that fermentation withAspergillus usamii improved zinc availability in dietary soybean meal, which was induced by the increase of zinc solubility in the small intestine. Adding the same amount of phytate that was contained in the regular soybean mealbased diet did not affect the amount of zinc present in rats fed a fermented soybean meal-based diet with sodium phytate. Phytase activity was found in fermented soybean meal, and this activity may degrade added phytate in fermented soybean meal-based diet.  相似文献

实验研究黄颡鱼(Pelteobagrus fulvidraco)对分别含有豆粕、发酵豆粕、菜粕和发酵菜粕的4种饲料以及这4种原料的干物质、蛋白、能量及氨基酸的表观消化率, 为黄颡鱼饲料的配制提供科学依据, 同时测定不同原料对消化道胰蛋白酶、糜蛋白酶及淀粉酶活性以及血液指标谷丙转氨酶、谷草转氨酶、总蛋白及碱性磷酸酶的影响。结果显示: 黄颡鱼对4种原料的干物质的表观消化率有显著差异, 其利用率由高到低依次为豆粕、发酵豆粕、菜粕及发酵菜粕(P<0.05)。黄颡鱼对发酵菜粕的蛋白质表观消化率显著低于其他3种原料(P<0.05), 而4种原料能量的表观消化率没有显著性差异(P>0.05)。黄颡鱼对各原料蛋氨酸和组氨酸的表观消化率没有显著性差异且低于83%(P>0.05), 而对各原料中其他氨基酸的表观消化率均有显著性差异, 在85%—98%之间, 发酵菜粕各氨基酸的表观消化率显著低于其他3种原料(P<0.05)。胰蛋白酶活力在菜粕组显著高于其他原料组(P<0.05), 活力呈现出前肠>中后肠>胃的趋势, 糜蛋白酶活力在不同原料间无显著差异, 糜蛋白酶活力呈现胃>前肠>中后肠的趋势, 淀粉酶活性呈现前肠>胃>中后肠。血液指标谷草转氨酶在豆粕组显著高于其他原料组(P<0.05), 菜粕与发酵菜粕组最低, 而谷丙转氨酶、总蛋白、碱性磷酸酶在各原料组间没有显著性差异(P>0.05)。综上所述, 豆粕、发酵豆粕、菜粕和发酵菜粕均可以作为黄颡鱼饲料中优质的蛋白源。  相似文献

为探讨发酵豆粕替代鱼粉对大口黑鲈(Micropterus salmoides)幼鱼生长、脂质代谢、血清非特异性免疫及肠道菌群的影响, 试验在基础饲料中用发酵豆粕分别替代0、10%、20%、30%和40%鱼粉, 用晶体氨基酸平衡各组间蛋氨酸与赖氨酸的差异, 配制成共5种等氮等能(CP47%, GE19 MJ/kg)的试验饲料, 分别为FM、FSM10、FSM20、FSM30和FSM40。将初始体重为(22.05±0.09) g的大口黑鲈随机分成5组, 每组设3个重复, 养殖58d。生长试验结果表明: 各组之间增重率(WGR)、特定生长率(SGR)、肥满度(CF)、肝体比(HSI)和脏体比(VSI)均无显著差异(P>0.05), 但替代组饲料系数(FCR)均显著高于FM对照组(P<0.05), FSM40组的存活率(SR)和干物质表观消化率(ADDM)显著低于其余各组(P<0.05)。脂质代谢方面结果显示, FSM30和FSM40组的血清甘油三酯(TG)水平显著低于FM对照组(P<0.05), FSM40组的血清总胆固醇(T-CHO)水平显著低于FM对照组(P<0.05)。血清非特异性免疫方面结果表明, FSM40组的溶菌酶(LZM)和超氧化物歧化酶(SOD)活性有显著增加的变化(P<0.05)。对肠道菌群进行了Illumina Mi Seq高通量测序分析, 结果表明: 各组肠道菌群的Alpha多样性指数(Sobs、Shannon、Simpson、Ace和Chao)无显著差异(P>0.05), 但OTU数量发生了变化。在门水平上, 优势菌群为软壁菌门(Tenericutes)、变形菌门(Proteobacteria)和梭杆菌门(Fusobacteria); 而属水平上, 优势菌群为支原菌属(Mycoplama)、邻单胞菌属(Plesiomonas)和鲸杆菌属(Cetobacterium)。FSM20组与FM对照组相比, 软壁菌门和支原菌属的相对丰度显著提高(P<0.05), 变形菌门和邻单胞菌属的相对丰度显著降低(P<0.05)。综上所述, 在试验条件下, 大口黑鲈饲料中发酵豆粕替代20%鱼粉不会对大口黑鲈幼鱼生长、脂质代谢、血清非特异性免疫及肠道菌群多样性造成负面影响, 且能显著降低肠道中邻单胞菌属在内的有害菌的相对丰度, 因此, 发酵豆粕替代鱼粉的适宜比例为20%。  相似文献

本实验以灵芝、金耳以及冬虫夏草作为发酵菌种,选用豆浆为基本原料配制培养基,在发酵过程中定时取样并测定其中几种生物活性物质的含量,分析各生物活性物质在发酵过程中的变化。研究结果表明,三种食用真菌发酵豆浆的发酵液中总游离型大豆异黄酮含量在发酵过程中均缓慢上升并分别在发酵72 h、24 h和36 h达到最高,与发酵初期相比分别增加了31.81μg/m L、19.05μg/m L和15.71μg/m L;三种食用真菌发酵液中蛋白质含量随着发酵时间的推移而降低的同时氨基酸含量均有增加,且发酵液中的肽主要是分子量范围在300 Da~1 500 Da之间的具有较高生物活性的小分子肽,发酵液中的必需氨基酸含量以及占总氨基酸含量的比值均有增加。本实验探究了食用真菌发酵豆浆过程中生物活性物质的变化,为日后的富集游离型大豆异黄酮、多肽以及氨基酸等生物活性物质实验提供依据,并为工业化生产提供实践基础。  相似文献

Angiotensin I-converting enzyme (ACE) inhibitory activity was observed in a tofuyo (fermented soybean food) extract with an IC(50) value of 1.77 mg/ml. Two ACE inhibitors were isolated to homogeneity from the extract by adsorption and gel filtration column chromatography, and by reverse-phase high-performance liquid chromatography (HPLC). The purified substances reacted with 2,4,6-trinitrobenzensulfonic acid sodium salt. The amino acid sequences of these inhibitors determined by Edman degradation were Ile-Phe-Leu (IC(50), 44.8 microM) and Trp-Leu (IC(50), 29.9 microM). The Ile-Phe-Leu sequence is found in the alpha- and beta-subunits of beta-conglycinin, while the Trp-Leu sequence is in the B-, B1A- and BX-subunits of glycinin from soybean. Both of the peptides are non-competitive inhibitors. The inhibitory activity of Trp-Leu was completely preserved after a treatment with pepsin, chymotrypsin or trypsin. Even after successive digestion by these gastrointestinal proteases, the activity remained at 29% of the original value.  相似文献

When the usual assay method of proteinase using milk casein as substrate is applied to the crude extract of wheat bran culture of Aspergillus sojae KS, over 90% of the total activity at pH 7 to 8 is occupied by that of alkaline proteinase. However, lower hydrolytic activity of purified alkaline proteinase than that of crude extract was observed not only in the digestion of soybean meal but also in the digestion of soybean protein, in spite of the fact that each enzyme solution had the same proteolytic activity on milk casein. From the experiments to fractionate crude extract by chromatography on DEAE-Sephadex A-50, neutral proteinase I and II, whose contribution to the hydrolysis of milk casein was estimated to be under 10% of the total activity of crude extract, were suggested to have almost comparable effect to alkaline proteinase in the digestion, determined by the increase of 0.4 m TCA-soluble nitrogen, of soybean protein by crude extract. Based on the rapid increase of 0.4 m TCA-soluble nitrogen and slight increase of Formol-titration value, it seems that both neutral proteinase I and II act as endo-type enzyme similar to alkaline proteinase and are not effective in the liberation of low molecular peptides or amino acids.  相似文献
