一种多基因串联共表达载体的构建   总被引:1,自引:0,他引:1  
为了构建一种可用于串联共表达多个基因的原核表达载体,通过PCR引入点突变,对商业载体pET-22b的酶切位点进行定向改造,设计独特的XbaⅠ/SpeⅠ模块。获得改造成功的载体pET-m22b,测序结果显示启动子、核糖体结合位点、多克隆位点及终止子均位于XbaⅠ和SpeⅠ两酶切位点间。选取不同来源的基因进行串联组合及表达鉴定,四个基因成功组合于同一载体中,表达效果良好,各基因表达效率不受明显影响。研究构建的载体pET-m22b,使多基因的共表达更加便捷,为蛋白复合物的表达与纯化、代谢通路的异源重建及其优化等研究奠定了良好的基础。  相似文献

将我国登革 2、4型病毒分离株PrM E基因通过RT PCR以病毒RNA为模板扩增我国登革 2型 4 3株和 4型B5株的PrM E基因 .并分别克隆至pGEM TEasy载体 ,然后亚克隆至双顺反子表达质粒的两个多克隆位点 ,获得同时含有登革 2型 4 3株和 4型B5株PrM E基因的双顺反子重组表达质粒pIDME2 4 .在用该重组质粒转染的BHK2 1细胞中 ,不但可检测到PrM E基因的转录产物 ,而且采用间接免疫荧光法还可观察到针对登革 2型 4型病毒的特异荧光 .研究结果表明 ,重组的双顺反子表达质粒在真核细胞中可共表达登革两个血清型PrM E基因 ,为双价登革核酸疫苗的研究奠定基础  相似文献

从口蹄疫病毒Asia I/Jiangsu毒株的细胞毒中提取总RNA,通过RT-PCR方法分别获得FMDV的P12A及3C基因;同时以pMD18-T-α-IFN质粒为模板,PCR扩增得到α-IFN基因.将α-IFN基因及FMDV P12A及3C基因连接至双启动子表达载体pBudCE4.1上,构建成双效表达质粒pBudCE4.1-α-IFN-P12A3C,经电泳、PCR、双酶切和DNA测序鉴定表明双效质粒载体构建成功.用此重组质粒转染BHK-21细胞后并对其表达情况进行检测,表明该双表达质粒在BHK-21细胞中能够成功表达.以此重组质粒免疫乳鼠后12 h,按100 TCID50/O.1 mL的量进行攻毒,结果发现该质粒能够抑制病毒的增殖,对乳鼠有一定的保护作用.结果表明成功构建了牛α-IFN及FMDV P12A3C组合基因双表达载体,为进一步研究口蹄疫基因疫苗提供前期基础.  相似文献

多基因共表达在多领域有重要的应用价值,大肠杆菌共表达系统包括多顺反子系统和双质粒系统,两者各有优缺点。多顺反子系统不需外界2种抗生素的同时存在,但操作较为复杂。通常认为,具有相同复制子的质粒是不相容的,但近来的实验表明,在双抗生素的选择压力下,不相容的双质粒系统也能稳定传代,且操作简单,周期较短。双质粒系统已经应用于生产、医学等各个领域。  相似文献

Gene coexpression networks inferred by correlation from high-throughput profiling such as microarray data represent simple but effective structures for discovering and interpreting linear gene relationships. In recent years, several approaches have been proposed to tackle the problem of deciding when the resulting correlation values are statistically significant. This is most crucial when the number of samples is small, yielding a non-negligible chance that even high correlation values are due to random effects. Here we introduce a novel hard thresholding solution based on the assumption that a coexpression network inferred by randomly generated data is expected to be empty. The threshold is theoretically derived by means of an analytic approach and, as a deterministic independent null model, it depends only on the dimensions of the starting data matrix, with assumptions on the skewness of the data distribution compatible with the structure of gene expression levels data. We show, on synthetic and array datasets, that the proposed threshold is effective in eliminating all false positive links, with an offsetting cost in terms of false negative detected edges.  相似文献

 rnc,era基因是大肠杆菌两个相邻的基因。用S_1核酸酶图谱法测得转录从rnc基因伸延到era基因。当rnc基因突变使所产生的RNaseⅢ丧失活性时,大肠杆菌中RNaseⅢ和Era蛋白的合成速度同对上升,且上升幅度相同,合成量相等。将此二基因分别或一起克隆入质粒并置于P_L起动子下游时,rnc能表达产生RNaseⅢ;rnc-era能表达产生RNaseⅢ和Era两种蛋白质,单独era则虽能转录却不产生Era蛋白。实验证明:era基因的表达依赖于rnc基因的表达,在体内两者共表达体现在转录和翻译水平上,两个基因同属于一个操纵子,共同受RNaseⅢ活性的反馈调节。单独转录的era-mRNA因缺乏有效的翻译起始序列而不能被翻译。  相似文献

One of the most fundamental tenets in the field of olfaction is that each olfactory receptor neuron (ORN) expresses a single odorant receptor. However, the one receptor-one neuron principle is difficult to establish rigorously. Here we construct a receptor-to-neuron map for an entire olfactory organ in Drosophila and find that two receptor genes are coexpressed in one class of ORN. Both receptors are functional in an in vivo expression system, they are only 16% identical in amino acid sequence, and the genes that encode them are unlinked. Most importantly, their coexpression has been conserved for >45 million years. Expression of multiple odor receptors in a cell provides an additional degree of freedom for odor coding.  相似文献

The gene coexpression study has emerged as a novel holistic approach for microarray data analysis. Different indices have been used in exploring coexpression relationship, but each is associated with certain pitfalls. The Pearson's correlation coefficient, for example, is not capable of uncovering nonlinear pattern and directionality of coexpression. Mutual information can detect nonlinearity but fails to show directionality. The coefficient of determination (CoD) is unique in exploring different patterns of gene coexpression, but so far only applied to discrete data and the conversion of continuous microarray data to the discrete format could lead to information loss. Here, we proposed an effective algorithm, CoexPro, for gene coexpression analysis. The new algorithm is based on B-spline approximation of coexpression between a pair of genes, followed by CoD estimation. The algorithm was justified by simulation studies and by functional semantic similarity analysis. The proposed algorithm is capable of uncovering both linear and a specific class of nonlinear relationships from continuous microarray data. It can also provide suggestions for possible directionality of coexpression to the researchers. The new algorithm presents a novel model for gene coexpression and will be a valuable tool for a variety of gene expression and network studies. The application of the algorithm was demonstrated by an analysis on ligand-receptor coexpression in cancerous and noncancerous cells. [目的]改造大肠杆菌缬氨酸合成途径,使其能够代谢合成异丁醇.[方法]将乳酸乳球菌(Lactococcus lactis) 1.2829的2-酮异戊酸脱羧酶基因(kivD)和醇脱氢酶基因(adhA)串联克隆到大肠杆菌DH5α宿主中表达.[结果]经过改造的宿主菌发酵24 h后异丁醇产量为0.12 g/L.酶活测定实验发现,kivD和adhA基因在宿主菌中均得到表达,但由于KivD的低表达量导致宿主菌最终的异丁醇合成能力偏低.通过研究温度和pH对KivD和AdhA酶活的影响,最终选定二者的最适温度为30℃,最适pH为6.5. [结论]通过向宿主菌导入外源异丁醇合成基因能够改造其自身代谢途径,从而合成异丁醇.  相似文献
