荧光光谱法是目前我国药物分析的主要方法,在我国各科研机构的应用广泛,具有一定的优势。荧光光谱法能够从分子之间的相互作用中分析药物的分子的相互作用,在药物分析中具有非常广阔的发展前景。荧光光谱法的主要原理,是根据不同物质在光照下的不同反射颜色和强度为主,根据反射光分析物质的原理。本文主要对荧光光谱的传统分析方法、新型分析方法以及与其他方法之间的联用三个部分进行分析,总结荧光光谱法在药物分析中的应用,并做发展展望。  相似文献

三维荧光光谱法检测多次高温加热植物油,该方法能快速、准确的获得待测样品的三维荧光光谱特征图,但只是定性的分析。当荧光强度变化较小的时候,容易出现误检、漏检的情况。先用三维荧光光谱技术分析植物油样品的光谱信息,然后再通过荧光寿命法,计算多次高温加热植物油中荧光光子的寿命,通过与未加热油荧光寿命的对比,检测多次高温加热植物油。二者联合,进一步提高了检测的准确度,有利于解决误检、漏检的问题,从而为高温加热油的检测提供有力的参考。  相似文献

本文应用荧光光谱法,观测了药物分子头孢菌素Ⅳ、异烟肼、维生素B_6和氟哌酸对白蛋白荧光的猝灭。由Lineweaver-Burk双倒数作图法,确定了药物与白蛋白作用的离解常数。并通过Forster偶极-偶极无辐射能量转移机理确定了药物分子氟哌酸在人血清白蛋白中与色氨酸残基之间的距离R为2.55nm,由这一距离确定了药物分子可能进入的区域和位置。  相似文献

有机小分子与DNA相互作用机理研究已经成为药物作用机理研究与新药筛选的重要手段之一。槲皮素 (Quercetin) 是一种多羟基黄酮类化合物,具有抗癌、抗炎、抗菌、抗病毒、降糖降压、免疫调节及保护心血管的作用。实验研究的目的是发现与确认槲皮素与DNA之间是否具有相互作用,以及确定其相互作用的类型。根据荧光光谱法和共振散射荧光光谱法的分析结果,发现槲皮素与鲱鱼精DNA之间存在相互作用;使用紫外-可见分光光度法和荧光偏振分析,发现槲皮素与鲱鱼精DNA之间的相互作用模式不属于嵌插作用,而是沟槽嵌合或者静电相互作用;最后通过分子对接实验,成功佐证槲皮素与鲱鱼精DNA之间的相互作用模式是沟槽结合。该工作有利于理解槲皮素与DNA之间的体外作用方式,助力于相应疾病的药物开发。  相似文献

本研究旨在对罗丹明类荧光探针ZM-6与人血清白蛋白(HSA)的相互作用进行研究。采用了荧光光谱法、三维荧光光谱法、同步荧光光谱法以及CD光谱法在模拟生理条件下对二者的相互作用以及HSA的构象进行了研究。研究结果表明,探针与ZM-6之间的猝灭机理主要是静态猝灭方式。根据热力学数据确定了二者之间的作用力,类型为范德华力和氢键。二者之间的结合距离为4.45 nm。同时得出,ZM-6对HSA的构象产生了影响。此研究对于探针分子的设计以及修饰提供有效的数据以及理论支持。  相似文献

[目的]建立新的荧光染料DAPI与FITC标记Annexin V联合的细胞凋亡流式细胞术检测方法,以用于具有橙红色荧光的药物处理细胞的流式细胞术凋亡检测。[方法]以倒置荧光显微镜成像和流式细胞仪分别检测不同浓度DAPI对活和死细胞的标记作用。以荧光酶标仪检测不同浓度DAPI处理细胞的荧光信号,并进行相关性分析。以流式细胞术比较Annexin V-FITC/DAPI双染法与Annexin V-FITC/PI双染法对无色和含红/橙色荧光药物导致的细胞凋亡。[结果]DAPI标记可用于区分死细胞和活细胞,DAPI标记死细胞的荧光信号和DAPI的浓度呈线性正相关。Annexin V-FITC/DAPI双染法与Annexin V-FITC/PI双染法相比,二者对不含红橙色荧光药物诱导的多种肿瘤细胞的凋亡检测结果无显著差异。与Annexin V-FITC/PI双染法相比,Annexin V-FITC/DAPI双染法可有效避免药物自身荧光与PI通道重合导致的流式检测干扰。[结论]成功建立了新的Annexin V-FITC/DAPI双染法用于细胞凋亡的流式细胞术检测,该方法能够避免具有红、橙色荧光基团的药物对的干扰检测凋亡。  相似文献

荧光光谱分析法在地沟油鉴别中的应用研究   总被引:2,自引:0,他引:2  
由于地沟油的成分含量复杂性和不定量性,导致了现有的单一检测方法不能同时满足快速和准确的辨认。荧光光谱具有高灵敏度和分辨率的特性,由此提出了一种利用荧光光谱快速检测食用油中是否掺有地沟油的新方法。将花生油分成7组,每组油所含的地沟油的比例不同,用220 nm到800 nm的激发和发射光检测各组样品油,收集其荧光数据后做归一化处理进行分析。在荧光实验中,特别是在365 nm和720 nm激发波长波段和434 nm发射波长波段,样品油的荧光强度与所含地沟油的体积分数大小明显成反比,当地沟油的体积分数大于5%时,荧光强度的衰减更为明显。结果证明了荧光光谱法检测地沟油的可行性,而且步骤更为简单。利用荧光光谱的非接触和高灵敏度的优势,能够更为简便地检测到加入了5%以上地沟油的花生油。  相似文献

【目的】利用季也蒙毕赤酵母ZJC-1合成银纳米团簇并用于痕量Cr(Ⅵ)的检测。【方法】使用经耐银驯化的季也蒙毕赤酵母ZJC-1生物合成荧光银纳米团簇,并对其结构和荧光性能进行了表征,探究Cr(Ⅵ)对银纳米团簇荧光的选择性猝灭作用,建立了银纳米团簇荧光强度与Cr(Ⅵ)浓度的线性关系。同时还考察了体系p H和其他金属离子对Cr(Ⅵ)检测的影响。【结果】Cr(Ⅵ)浓度在一定的范围内(1–80μmol/L)与银纳米团簇荧光强度(F_0–F)/F_0有着良好的线性关系(R~2=0.9821),线性方程为(F_0–F)/F_0=0.0054×Ccr(Ⅵ)+0.1876,检测限为184 nmol/L(信噪比为3)。利用该方法检测实际水样(松花江、马家沟河)中的Cr(Ⅵ),回收率介于97.73%–102.88%之间。【结论】以季也蒙毕赤酵母ZJC-1为还原剂和稳定剂,制备了具有较好荧光性能的水溶性银纳米团簇,基于Cr(Ⅵ)对银纳米团簇荧光的选择性猝灭作用,建立了一种快速且灵敏检测痕量Cr(Ⅵ)的新方法,并成功地应用于松花江、马家沟河水样中Cr(Ⅵ)的测定,在分析检测领域中具有良好的应用前景。  相似文献

以斜生栅藻(Scenedesmus obliquus JNU20)为实验材料,采用尼罗红(NR)荧光光谱法和傅里叶变换红外光谱法(FTIR)测定该藻细胞中的油脂含量。研究结果表明NR的最佳染色条件为:染色前微波处理40 s,二甲基亚砜体积分数1%, NR最终质量浓度1.5 μg/ml,染色时间5 min,染色温度40℃。比较了NR荧光光谱法、FTIR与传统重量法测定的该藻在不同时相的油脂积累情况,利用激光共聚焦显微镜观察了细胞中油体形成的动态过程。实验结果表明NR荧光光谱法、FTIR与传统重量法测定的结果显著相关(R2=0.9258, R2=0.9844),但NR荧光光谱法和FTIR更简便快速,研究结果为规模化筛选高含油量藻株及跟踪产油微藻油脂积累过程奠定了基础。  相似文献

The algologically pure cultures of the green–brown cyanobacterium Chroococcidiopsissp. and three cyanobacteria of the genus Gloeocapsa, the blue–green Gloeocapsa sp.1, the brown Gloeocapsa sp.2, and the red–orange Gloeocapsa sp.3, were isolated from sandstones and rock fissures in the high-polar regions of Antarctica. These cyanobacteria are the most widespread phycobionts of cryptoendolithic lichens in these regions. The comparative analysis of the absorption and the second-derivative absorption spectra of the cyanobacteria revealed considerable differences in the content of chlorophyll a and in the content and composition of carotenoids and phycobiliproteins. In addition to phycocyanin, allophycocyanin, and allophycocyanin B, which were present in all of the cyanobacteria studied, Gloeocapsa sp.2 also contained phycoerythrocyanin and Gloeocapsa sp.3 phycoerythrocyanin and C-phycoerythrin (the latter pigment is typical of nitrogen-fixing cyanobacteria). The fluorescence spectra of Gloeocapsa sp.2 and Gloeocapsa sp.3 considerably differed from the fluorescence spectra of the other cyanobacteria as well. The data obtained suggest that various zones of the lichens may be dominated either by photoheterotrophic or photoautotrophic cyanobacterial phycobionts, which differ in the content and composition of photosynthetic pigments.  相似文献

A phycoerythrin-allophycocyanin (PE-AP) complex was obtained from intact phycobilisomes of the marine red alga Polysiphonia urceolata. Exposure of intact cells of Spirulina to high temperature (HT) stress (40–60 °C) caused decrease in absorption spectrum and fluorescence emission spectrum. Low temperature emission spectra were altered at phycocyanin (PC) level. The activities of photosystem 2 (PS2) and whole chain electron transport declined in high temperature treated cells at the room temperature beyond 35 °C, while photosystem 1 (PS1) showed increased activity. Thylakoid membrane studies did not exhibit increase in PS1 activity indicating that the enhancement of PS1 activity is due to permeability change of cell membranes. However, the electron transport activity measured from reduced duroquinone to methylviologen which involves intersystem electron transport was extremely sensitive to high temperature. The activity of PS2 at different irradiance, which was accompanied by alterations in absorption and fluorescence emission properties, indicated changes in the energy transfer processes within phycobilisomes. Thus high temperature has multiple target sites in photosynthetic electron transport system of Spirulina platensis.  相似文献

The molecular structure-function relationship of the Fenna-Matthews-Olson light-harvesting complex of the photosynthetic green bacterium Prosthecochloris aestuarii has been investigated. It has been assumed that the electronic excited states responsible for the function (transfer of electronic excitation energy) result from the dipole-dipole interactions between the bacteriochlorophyll molecules bound to the polypeptide chain of the complex at a specific three-dimensional geometry. The molecular structure-electronic excited states relationship has been addressed on the basis of simultaneous simulations of several spectroscopic observations. Current electronic excited state models for the Fenna-Matthews-Olson complex have generally been based on obtaining an optimal match between the information contents of the optical steady-state spectra and the bacteriochlorophyll organization. Recent kinetic and spectral information gathered from ultrafast time-resolved measurements have not yet been used effectively for further refinement of the excited state models and for quantification of the relation between the excited states and the energy transfer processes. In this study, we have searched for a model that not only can explain the key features of several steady-state spectra but also the temporal and spectral evolution observed in a recent absorption difference experiment and we have discussed the implications of this model for equilibration of the electronic excitation energy in systems at low temperatures.
