NaCl胁迫对嫁接黄瓜膜脂过氧化、渗透调节物质含量及光合特性的影响 总被引:28,自引:7,他引:21
以日本引进的耐盐品种‘帝王新土佐’南瓜为砧木,以‘津研4号’黄瓜品种为接穗,研究了NaCl胁迫对黄瓜嫁接植株和自根植株的膜脂过氧化、渗透调节物质含量及光合特性的影响。结果表明:在NaCl胁迫下,黄瓜嫁接植株叶片超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)和抗坏血酸过氧化物酶(APX)活性均显著高于自根植株,O·2-产生速率及丙二醛(MDA)含量显著低于自根植株,嫁接植株膜脂过氧化轻于自根植株;嫁接植株叶片游离脯氨酸、可溶性糖、可溶性蛋白质等渗透物质含量均显著高于自根植株;嫁接植株的净光合速率(Pn)、气孔导度(Gs)、胞间CO2浓度(Ci)、叶绿素含量均显著高于自根植株;虽然NaCl胁迫抑制光合作用,但嫁接植株仍表现出优势。以上结果证明,嫁接植株耐盐性优于自根植株。 相似文献

采用营养液栽培系统,以黄瓜品种中农8号为材料,研究了Ca2 对根际低氧胁迫下黄瓜幼苗体内超氧阴离子(O2?-)、过氧化氢(H2O2)、丙二醛(MDA)含量和超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)、谷胱苷肽还原酶(GR)活性的影响.结果表明:低氧胁迫下黄瓜体内活性氧含量和保护酶活性均高于对照;低氧缺钙处理的活性氧含量最高,而保护酶活性却较低;营养液Ca2 浓度提高到8mmol/L后,显著降低了低氧胁迫下黄瓜幼苗体内MDA、H2O2含量和O?2-产生速率,提高了SOD、POD、CAT、APX、GR活性,说明Ca2 可减少低氧胁迫下黄瓜幼苗体内活性氧的产生,提高抗氧化酶的活性、降低膜脂过氧化水平,减缓低氧胁迫对植株的伤害,增强黄瓜幼苗对低氧逆境的适应性. 相似文献

目的:提高纤维素的酶水解效率和开发高效的纤维素酶水解过程。方法:采用RT-PCR方法从黄瓜胚轴细胞中分离了膨胀素S1的cDNA,并使之与毕赤酵母表达质粒pPICZ(A连接,形成重组质粒pPICZ(A-exs1。通过电转化方法,用质粒pPICZ(A-exs1转化巴氏毕赤酵母GS115,得到重组菌株P.pastoris-exs1。在该重组菌株中,膨胀素的基因通过同源重组整合在毕赤酵母的染色体上,并处于毕赤酵母甲醇氧化酶启动子的下游。重组菌株P.pastoris-exs1在甲醇诱导下可合成并分泌膨胀素。结果:培养上清液没有纤维素酶活性,但具有破坏滤纸纤维素结晶结构的能力。培养上清液与里氏木霉纤维素酶等量混合后,可使纤维素酶的滤纸酶活力提高50%。结论:采用巴氏毕赤酵母GS115重组成功表达了黄瓜膨胀素,其表达产物可以促进纤维素酶对滤纸的水解。 相似文献

A study was conducted in 2007 and 2008 to investigate the effectiveness of grain amaranth (Amaranthus cruentus L.) planted at different times in an amaranth/cucumber intercrop to reduce the density of cucumber insect pests. Cucumber varieties, Marketmore and hybrid F1, were established from seed as a monocrop or intercropped with amaranth, and with amaranth established at two weeks before cucumber (2 WBC) on the same day as cucumber (SDC) or two weeks after cucumber (2 WAC). The population of cucumber beetles and fruit flies was lower with amaranth established 2 WBC and SDC than with 2 WAC. Populations of the insects were influenced by cropping system and time of introducing amaranth. Fruit damage by cucumber beetles and fruit flies was lower with 2 WBC and SDC than with 2 WAC. However, the damage was reduced in the intercrop compared with the monocropped cucumber. With SDC, the reduction was about 75 and 50% in 2007 and 2008, respectively. In intercrops and monocrops, cucumber fruit yield was influenced by the time of inter-cropping; in the mono- and inter-crop, the highest yield was with SDC. The highest land equivalent ratios for cucumber were 1.0 and 0.7 in 2007 and 2008, respectively, with SDC. A field survey was conducted in eight local government areas (LGA) of Ogun state, Nigeria to assess the incidence of viral diseases of yams in the areas. Leaf samples were collected from 90 yam plants which were either symptomatic or asymptomatic. These were bulked into 45 during serological tests and the viruses indexed include yam mosaic virus (YMV); Dioscorea alata bacilliform virus (DaBV) and cucumber mosaic virus (CMV). DaBV was the most prevalent virus on the field with incidence of 48.9% (22/45) followed by YMV which occurred in 42.2% (19/45). CMV had the lowest percentage of incidence; 2.2% (1/45). Of all the LGAs visited, Abeokuta north and Abeokuta south had the highest incidence of YMV and DaBV, respectively. Mixed virus infections were also detected. 相似文献

Summary Viviparity, where the embryos develop in the female reproductive system, is a rare form of reproduction in marine invertebrates, being described in only 14 species of echinoderm. In the intraovarian brooding sea cucumber, Leptosynapta clarki Heding 1928 (cf., Sewell et al. 1995), we used direct evidence (changes in energetic content) to show that significant additional nutrients are provided to the embryos during viviparous development (matrotrophy). In the transition from a structure used to produce gametes to a long-term brooding structure there are visual, histological and transmission electron microscopy (TEM) changes in the structure of the ovarian wall. Changes occur primarily in the cells of the visceral peritoneum and involve an increase in size of the connective tissue/genital haemal sinus (CT/GHS). In the latter part of the brooding period the visceral peritoneum returns to a flattened form, and new oocytes develop along the tubule wall. Similar changes in the intraovarian brooding sea cucumber Oneirophanta mutabilis affinis lead us to suggest that there is a role for the genital haemal sinus in providing nutrition during the brooding period in viviparous echinoderms. Future research is suggested to focus on changes in the ovarian wall structure during the different phases of reproduction (gamete production/brooding) in these species. 相似文献

QTL Mapping of Downy Mildew Resistance in an Introgression Line Derived from Interspecific Hybridization Between Cucumber and Cucumis hystrix 总被引:1,自引:0,他引:1
Downy mildew (DM), caused by Pseudoperonospora cubensis (Berk. & M.A. Curtis) Rostovzev, is a worldwide major disease of cucumbers (Cucumis sativus L.). By screening 10 introgression lines (ILs) derived from interspecific hybridization between cucumber and the wild Cucumis, C. hystrix, through a whole plant assay, one introgression line (IL52) was identified with high DM‐resistance. IL52 was further used as a resistant parent to make an F2 population with ‘changchunmici’ (susceptible parent). The F2 population (300 plants) was investigated for DM‐yellowing, DM‐necrosis and DM‐resistance in the adult stage. A genetic map spanning 642.5 cM with 104 markers was constructed and used for QTL analysis from the population. Three QTL regions were identified on chromosome 5 and chromosome 6. By interval mapping analysis, two QTLs for DM‐resistance were determined on chromosome 5 (DM_5.1 and DM_5.2), which explained 17.9% and 14.2% of the variation, respectively. QTLs for DM‐yellowing were in the same regions as DM‐resistance. For DM‐necrosis, by interval mapping analysis, one QTL was determined on chromosome 5 (Necr_5.1) that explained 18.3% of the variation and one on chromosome 6 (Necr_6.1) that explained 13.9% of the variation. Our results indicated that the identification of molecular markers linked to the QTLs could be further applied for marker‐assisted selection (MAS) of downy mildew resistance in cucumber. 相似文献

植物对昆虫取食产生的防御反应,在昆虫与植物相互作用关系中起着重要的作用。为明确南美斑潜蝇Liriomyza huidobrensis(Blanchard)取食与植物防御之间的作用关系,本文分别测定了南美斑潜蝇幼虫持续为害1、3、5、7及9d后黄瓜叶片中苯丙氨酸解氨酶(PAL)、多酚氧化酶(PPO)、过氧化物酶(POD)及超氧化物歧化酶(SOD)活性的变化。结果表明:在南美斑潜蝇幼虫持续取食下,PAL、PPO、POD和SOD等4种酶活性显著上升;PAL、PPO和POD等3种酶活性随受害程度的加重而上升幅度加大,即重度受害>轻度受害>系统对照>健康对照,而SOD活性变化没有规律。在系统对照黄瓜叶片中,PAL、PPO、POD和SOD等4种酶活性最大值分别在第5、1、5和5d;在轻度受害黄瓜叶片中,PAL、PPO、POD和SOD等4种酶活性最大值分别在第5、9、5和9d;在重度受害黄瓜叶片中,PAL、PPO、POD和SOD等4种酶活性最大值分别在第7、7、5和5d。黄瓜叶片受害后,POD和PPO活性上升幅度较大,PAL和SOD活性上升幅度较小,说明POD和PPO对取食胁迫响应比PAL和SOD更灵敏。 相似文献

In this study, a highly active foliar aminopeptidase preferentially releasing N-terminal alanine from artificial substrates was purified and characterized from cucumber (Cucumis sativus L. suyo). The enzyme had a molecular mass of 200 kDa consisting of two subunits of 95 kDa. It was a metalloprotease the pH optimum of which was 8 to 9. It cleaved Ala-, Gly-, Met-, Ser-, Leu-, Lys-, and Arg artificial substrates. An internal amino acid sequence was similar to those of aminopeptidase N (clan MA, family M1) of microorganisms, and was very similar to that of a putative aminopeptidase N of Arabidopsis thaliana. From these results, the highly active aminopeptidase in cucumber leaves was identified to be a plant aminopepitdase N. 相似文献
