蚕豆根端细胞核中微核仁的研究

以蚕豆(Vicia faba)根端分生组织细胞为材料研究了微核仁的超微结构和细胞化学特点。结果表明;微核仁是直径0.3—0.5μm 的卵圆形或球形结构。常规染色时,微核仁与集缩染色质的电子密度相仿,但两者之间在结构上没有任何联系。细胞化学研究指出,微核仁含有 RNA 和蛋白质,其结构成分主要是与核仁颗粒组分十分相似的 RNP 颗粒。报道了植物细胞核中微核仁发生于核仁的过程并对微核仁的本质和功能进行了讨论。     

3-Hydroxypropyl amide formation from 1-aminocyclopropane-1-carboxylic acid by a cell-free ethylene-forming system from olive leaves

The low ethylene yield in a cell-free ethylene-forming system from olive tree leaves ( Olea europaea L. cv. Picual) was investigated. During the incubation, 1-aminocyclopropane-1-carboxylic acid (ACC) was extensively transformed into 3-hydroxypropyl amide (HPA). Enzyme extract, Mn²⁺ and oxygen are responsible for this reaction. Horseradish peroxidase (EC 1.11.1.7) can substitute for the enzyme extract in this reaction. HPA formation could be one reason for the poor in vitro conversion efficiency of ACC to ethylene.     

Nucleotide regulation of vasoactive intestinal peptide binding to bovine thyroid plasma membranes

The specific binding of vasoactive intestinal peptide (VIP) to bovine thyroid plasma membranes is inhibited by guanine nucleotides. Guanosine 5-triphosphate (GTP) and the non-hydrolyzable GTP analogs guanosine 5-,-imidotriphosphate (Gpp(NH)p) and guanosine 5-O-(3-thiotriphosphate) (GTP--S) inhibited markedly the binding of VIP to its receptors. This inhibition was higher with GTP than with Gpp(NH)p and GTP--S and was due to an increase of the rate of dissociation of peptide bound to membranes. Other nucleotides did not show any effect.     

Somatostatin binding to dissociated cells from rat cerebral cortex

A method has been developed for the study of somatostatin (SS) binding to dissociated cells from rat cerebral cortex. Binding of [¹²⁵I][Tyr¹¹]SS to cells obtained by mechanical dissociation of rat cerebral cortex was dependent on time and temperature, saturable, reversible and highly specific. Under conditions of equilibrium, i.e., 60 min at 25°C, native SS inhibited tracer binding in a dose-dependent manner. The Scatchard analysis of binding data was linear and yielded a dissociation constant of 0.60±0.08 nM with a maximal binding capacity of 160±16 fmol/mg protein. The binding of [¹²⁵I][Tyr¹¹]SS was specific as shown in experiments on tracer displacement by the native peptide, SS analogues, and unrelated peptides.     

红松越冬针叶的电子自旋共振（ESR）波谱

用ESR波谱仪在越冬红松针叶及叶绿体冻干粉中检出了g值分别为2.0044和2.0037的明确的单峰信号。针叶伤害愈重,信号愈强。此信号可被Vc或富含SOD等多种自由基清除剂的沙棘果汁所抑制。针叶受害愈重,沙棘果汁对信号的抑制作用愈强,Vc的抑制作用愈弱。据此认为红松苗越冬伤害和有机自由基的形成有关。在伤害发展的不同阶段,有机自由基的种类组成也不同。     

长白山树舌水溶性色素多糖CF_1的分离纯化与结构研究

 从长白山树舌子实体中分离色素多糖,其均一性检查用Sepharose CL-4B柱层析、高压玻璃纤维纸电泳、醋酸纤维素薄膜电泳、超离心分析等方法,用凝胶层析测定的分子量为18.5万。 均一的脱色多糖(CF_1a)分子量为14万。用红外光谱,G.C,~1H-N.M.R,~(13)CN.M.R,高碘酸氧化与Smith降解,甲基化分析等确定其结构为葡聚糖,其基本结构可能如下式: 总色素用次氯酸氧化法测定为24％。在色素多糖中的色素可能是聚合形式。色 素经薄层层析、紫外吸收性质检查等表明可能是新黄酮类化合物。     

家兔早期胚胎细胞发育能力的研究

本文利用胚泡注射法制作嵌合体对家兔交配后96,120和144小时的ICM细胞的发育能力进行了研究。供体胚胎取自青紫兰灰免,受体胚胎取自新西兰白兔,结果表明96和120小时供胚的ICM细胞与96小时受胚胚泡组合后均能参与发育,形成嵌合兔,144小时者未获得嵌合体。由于120小时的ICM细胞发育的2只表型为雄性的嵌合兔,其中1只不育,其性腺和外周血核型表明不育兔为xx/xy性嵌合,性腺中有处于不同发育程度的卵巢和精细管,外周血含xx和xy两种核型。本实验结果首次证明家兔交配后120小时胚泡的ICM细胞仍具有参与嵌合体发育的能力。它不仅能参与体细胞的分化,并具有形成生殖细胞的能力。交配后144小时胚泡的ICM细胞其发育能力似乎已发生了局限。
