目的研究紫穗槐种子提取物的抑菌活性。方法将紫穗槐种子乙醇提取物分别通过石油醚、乙酸乙酯和正丁醇萃取,选择金黄色葡萄球菌和肺炎克雷伯杆菌为供试菌,采用试管二倍稀释法测定紫穗槐种子提取物的最小抑菌浓度(MIC)和最小杀菌浓度(MBC),涂布平板法绘制杀菌曲线,电镜下观察药物对细菌超微结构的影响。结果紫穗槐种子提取物经乙酸乙酯萃取后对供试菌抑制作用较强,其中对金黄色葡萄球菌的MIC和MBC分别为2.5、5.0mg/mL;对肺炎克雷伯杆菌的MIC和MBC分别为5.0、10.0mg/mL;杀菌曲线结果表明,药物对供试菌的抑制作用存在浓度和时间依赖性;电镜结果说明,药物的作用可能与破坏菌体细胞壁、改变细胞膜通透性有关。结论紫穗槐种子提取物具有显著的抗菌活性。  相似文献

褐藻硫酸多糖对老年人肠道歧杆菌促增殖作用的研究   总被引:3,自引:0,他引:3  
目的观察褐藻硫酸多糖对老年人肠道中双歧杆菌促增殖作用的影响。方法选择青岛地区健康老年人56例,随机分为两组,试验组和对照组各28例,试验组口服褐藻硫酸多糖每人每天2.5g(冷提取物SPC和热提取物SPH各1.25g),共口服5d,对照组不服用。试验组于口服前及口服后第1、2、3、5、8、14天无菌操作留取粪便,定量进行10倍系列稀释,采用滴注法接种于BS和EMB平板培养基上,分别于35℃厌氧和需氧培养48h,取出后分别计数双歧杆菌和大肠杆菌菌落数,对照组同步进行。结果在试验剂量下,褐藻硫酸多糖(SP)能有效地扶持、促进健康老年人肠道内双歧杆菌的生长繁殖,试验组口服SP后第3天,粪便中的双歧杆菌数明显增多,竞争地抑制大肠杆菌,第5天双歧杆菌增至11.88±0.98,大肠杆菌降至6.61±1.01,B/E值增加到1.67±0.88,与口服前比,有显著性和非常显著性差异(P<0.05,P<0.05,P<0.01);结果还显示,试验组口服SP后第3、5天粪便中培养计数的双歧杆菌、大肠杆菌数及B/E值与口服后第8、14天(即停服SP的第3、9天)比,均无显著性差异(P>0.05)。结论褐藻硫酸多糖是一种有效的双歧因子。在试验剂量下,对受试者肠道中双歧杆菌有显著的增值作用,且有较好的定植性。  相似文献

对药用植物墨旱莲Eclipta prostrata提取物的抑菌活性物质进行研究。结果表明:(1)通过不同部位提取物的抑菌试验,发现只有叶和根茎含有抑菌活性物质;(2)墨旱莲提取物对金黄色葡萄球菌、大肠杆菌、枯草杆菌、藤黄球菌的最低抑菌浓度分别为0.6、2.0、0.8、1.0 g/mL,最小杀菌浓度均为1.0 g/mL,对白色念珠菌有很强的抑菌作用,但对黑曲霉无效;(3)通过不同极性溶剂的抑菌试验表明,抑细菌活性物质易溶于石油醚、乙醚等非极性溶剂,具有最佳抑细菌效果的提取溶剂为偏碱性的65%乙醇溶液;(4)墨旱莲的抑菌活性物质可能为黄酮类物质。  相似文献

【目的】药用植物内生真菌是一种新型抗生素的微生物资源,研究绞股蓝内生真菌JY25的抗菌机制对内生真菌的研发具有重要意义。【方法】以二倍稀释法测定发酵液的最小抑菌浓度(MIC)和最小杀菌浓度(MBC);以MIC测定发酵液对大肠杆菌生长的抑制作用;扫描电镜观察发酵液作用下的大肠杆菌形态变化;同时,以β-半乳糖酶、碱性磷酸酶和电导率测定发酵液对细胞膜和细胞壁的损伤效果;采用考马斯亮蓝法检测发酵液对蛋白质合成的影响。【结果】JY25发酵液对大肠杆菌MIC为7 g/L、MBC为14 g/L;MIC浓度的发酵液使细菌对数生长期延迟12 h,菌体形态发生严重的畸形和破损;随着抑菌作用时间的延长,β-半乳糖酶含量增加、电导率增加,同时,实验发现大肠杆菌蛋白质合成异常,未检测到碱性磷酸酶。【结论】绞股蓝内生真菌JY25主要以破坏细菌的细胞膜及影响细菌蛋白质合成而抑制细菌生长。  相似文献

本文研究了绿茶和桑叶提取物在不同浓度时对细菌的抑制作用,采用D值计算法快速评价了它们的抑菌效果。结果表明,绿茶提取物对金黄色葡萄球菌和大肠杆菌的最低抑制浓度分别为0.034%,0.42%;而桑叶提取物分别为0.86%,1.54%。绿茶的抑菌效果更好。  相似文献

目的观察柠檬提取物对细菌生物膜的消除作用,对柠檬提取物抑制多重耐药金黄色葡萄球菌的机制进行初步研究,以及对环境中富集的病毒消除作用的研究。方法采用高分子滤膜法制备金黄色葡萄球菌生物膜后,加入柠檬提取物分别作用1、2和3h,通过细菌菌落计数判定不同生物膜的细菌存活数;采用SDS—PAGE电泳法,观察在柠檬提取物作用下菌体蛋白质的变化;EcoRⅠ酶切金黄色葡萄球菌DNA,观察酶切图谱的改变;在低渗环境下动态观察柠檬提取物在不同作用时间对细菌细胞壁的影响;通过柠檬提取物对病毒作用后RNA量的变化,说明柠檬提取物具有杀灭病毒的作用。结果柠檬提取物对生物膜中的细菌具有杀菌作用,且随作用时间的延长而逐渐增强;柠檬提取物对细菌蛋白质的组成和表达量均有一定影响,同时细菌DNA的EcoRⅠ酶切图谱发生改变;柠檬提取物作用15min时细菌呈现明显的胞壁缺损现象,即细菌个体胀大,呈现大球形;经柠檬提取物作用后,RNA病毒被杀灭。结论结果显示柠檬提取物对耐药金黄色葡萄球菌所形成的生物膜具有明显抑制作用;在柠檬提取物作用下,菌体蛋白合成量发生改变,同时对细菌的DNA和细胞壁有明显的影响。柠檬提取物处理病毒悬液后,病毒RNA量显著减少,说明病毒RNA结构被破坏。  相似文献

以常见6种细菌和5种真菌为供试菌。采用滤纸片水平扩散法。对黑刺菝葜根的4种溶剂提取物和剩余水相大孔吸附树脂5种乙醇梯度洗脱物共9种提取物进行了室内抗菌生物活性测试,并对其主要的化学成分-常规的抗菌活性物质黄酮和甾体皂苷类化合物进行了定量测定。结果表明:黑刺菝葜根剩余水相大孔吸附树脂100%乙醇的洗脱物表现出极强的广谱性抗菌活性,80%乙醇的洗脱物对绿木霉、大肠杆菌显示出极强抑菌力;黑刺菝葜根的石油醚提取物、乙酸乙酯提取物和正丁醇提取物均对枯草芽孢杆菌、大肠杆菌显示较强的抗性;根的石油醚提取物和乙酸乙酯提取物对金黄色葡萄球菌、蜡状芽孢杆菌和巨大芽孢杆菌等有较强的抑制作用;根中所含的常规抑菌活性成分黄酮的含量为2.83%;甾体皂苷元的含量为4.55%。  相似文献

用肉汤二倍稀释法和琼脂平板培养计数法,研究四川宜宾七种中药的提取物对大肠杆菌、沙门氏菌和巴氏杆菌的体外抑制作用.结果表明:黄柏提取物的抗菌活性最强,最小抑菌浓度(Minimum inhibitory concentration,MIC)为15.6~125 mg·mL-1,最小杀菌浓度(Minimum bactericidal concentration,MBC)为31.25~250 mg·mL-1;栀子提取物抗菌活性次之,MIC 为62.5~125 mg·mL-1,MBC 为125~>250 mg·mL-1;佛手提取物有较稳定的抗菌活性,MIC均为125 mg·mL-1,MBC为125 mg·mL-1和250 mg·mL-1;姜黄、杜仲、何首乌、细毡毛忍冬的提取物抗菌活性相对较差,大部分 MIC≥250 mg·mL-1.说明四川宜宾的黄柏和栀子的提取物抗菌活性较强,佛手提取物抗菌活性较稳定,有进一步研究的价值.  相似文献

研究海带水提液和酵母提取物促大肠杆菌生长的机理。采用二维电泳 (2 DE)和生物质谱等蛋白质组技术对细菌的全蛋白质表达进行研究。发现 8种蛋白出现有规律的变化 ,其中 ,2种蛋白在对数期各组表达量均增高 ,4种蛋白在各组表达量下降且实验组下降更明显 ,1种蛋白在实验组表达量明显降低而对照组无明显变化。  相似文献

A low concentration of yeast extract (0·1%) in liquid media favoured rapid growth and high percentage of viable cells in cultures of Rhizobium japonicum (CB 1809), R. lupini (WU 425), R. meliloti (SU 47), R. trifolii (TA1) and a cowpea strain (CB 756). Concentrations of yeast extract > 0·35% depressed viability and produced distorted cells in all strains except SU 47: TA1 was especially sensitive. When used at 0·5–1% (w/v), each yeast extract (Difco, Oxoid, Vegemite) or casein hydrolysate produced greatly enlarged abnormal cells of TA1, each containing several granules of poly-β-hydroxybutyrate and whorls of intracytoplasmic membranes, and showing greater internal disorganisation than that seen in root nodule bacteroids. Lysogenic and non-lysogenic cultures of R. trifolii were all sensitive to yeast extract, and such sensitivity, for strains of several species, was unrelated to effectiveness in nodulating host plants. Glycine inhibited growth of all strains tested. Several other amino acids occurring in casein hydrolysate inhibited TA1 strongly and induced formation of distorted cells and spheroplasts; this distortion was partly counteracted by adding salts of calcium or magnesium. In media with 0·1% yeast extract the use of mannitol, sucrose, lactose or galactose as alternative carbon sources, each at a concentration of 0·02–1%, did not affect numbers of viable rhizobia or cell shape in all strains tested.  相似文献

The active component(s) in yeast extract required by Thermoplasma acidophilum for growth is polypeptide in nature. A fraction from yeast extract was isolated and partially characterized as one or more peptides of molecular weight about 1,000 containing 8 to 10 amino acids. Although it was composed largely of basic and dicarboxylic amino acids, only one amino group per molecule was free. The polypeptide(s) appeared to bind avidly to cations. No other organic compounds except glucose were needed by Thermoplasma. Among several hundred known compounds tested, only glutathione plus Fe2+ or Fe3+, clostridial ferredoxin, and spinach ferredoxin elicited any growth response.  相似文献

Bacillus thuringiensis subsp. galleriae 69/6 was cultivated in a synthetic medium containing 5 amino acids and nicotinic acid. The dynamics of the culture growth and amino acid assimilation were studied in this medium and in a medium containing yeast extract. The phase of spore germination increased, the yield decreased and the maximal growth rate became higher when the culture grew in the synthetic medium. The percentage of thermoresistant spores was slightly lower in the synthetic medium comparing to the medium with yeast extract.  相似文献

The nutritional requirements of the rice stem borer,Chilo suppressalis Walker were studied by rearing on synthetic food media in relation to the constituents of the rice plant. The larvae required amino acids, vitamins, carbohydrates, cholesterol, minerals and water extract of the rice plant for their growth. Among amino acids, arginine, histidine, isoleucine, leucine, lysine, methionine, phenylalanine, threonine, tryptophane and valine were indispensable for the growth, and these amino acids could not be synthesized in the larval body. Among vitamines, thiamine, riboflavin, nicotinic acid, pyridoxine, Ca-pantothenate, folic acid, biotin, cholin chloride and ascorbic acid were essential for the growth. Cholesterol is the only essential nutrient among fat-soluble factors. Fructose, glucose and sucrose showed high nutritive values among carbohydrates. Addition of the water extract of the rice plant to the media improved the development of the insects, but the excess amounts of it resulted inhibition of the growth. Two aromatic acids, benzoic and salicylic acids, were isolated as the growth inhibitory substance. p-methyl acetophenone isolated from the rice plant seemed to play an important role for the attraction of the insect. Mass rearing of the rice stem borer in a laboratory was carried out on two kinds of foods: living rice plant seedlings and synthetic food media. The rearing the insects on rice seedlings is easy if rice seeds are available. The rearing on synthetic food media is modified to simplify from these for the nutritional study. The rearing practices, merit and demerit of both methods are discussed.  相似文献

Although wheat mashes contain only growth-limiting amounts of free amino nitrogen, fermentations by active dry yeast (Saccharomyces cerevisiae) were completed (all fermentable sugars consumed) in 8 days at 20 degrees C even when the mash contained 35 g of dissolved solids per 100 ml. Supplementing wheat mashes with yeast extract, Casamino Acids, or a single amino acid such as glutamic acid stimulated growth of the yeast and reduced the fermentation time. With 0.9% yeast extract as the supplement, the fermentation time was reduced from 8 to 3 days, and a final ethanol yield of 17.1% (vol/vol) was achieved. Free amino nitrogen derived in situ through the hydrolysis of wheat proteins by a protease could substitute for the exogenous nitrogen source. Studies indicated, however, that exogenously added glycine (although readily taken up by the yeast) reduced the cell yield and prolonged the fermentation time. The results suggested that there are qualitative differences among amino acids with regard to their suitability to serve as nitrogen sources for the growth of yeast. The complete utilization of carbohydrates in wheat mashes containing very little free amino nitrogen presumably resulted because they had the "right" kind of amino acids.  相似文献

An optimized medium containing Trypticasein, phytone, yeast extract and glucose is proposed to improve growth of bifidobacteria with high yeast extract concentration and decreased amounts of Trypticasein and phytone. 在重组枯草芽孢杆菌24/pMX45核黄素发酵中,酵母粉促进核黄素合成,酵母抽提物抑制核黄素合成。分析显示,酵母抽提物的无机离子和游离氨基酸含量均高于酵母粉。在酵母粉基础发酵培养基中,添加各种无机离子和游离氨基酸,使其含量与酵母抽提物相同。摇瓶发酵结果表明:过量的无机离子和谷氨酸对核黄素合成有显著的抑制作用。酵母抽提物含有较高浓度的谷氨酸,是其抑制核黄素合成的主要原因。  相似文献
