筛选产类细菌素乳酸菌及类细菌素特性研究   总被引:1,自引:0,他引:1  
从健康鸡肠道内容物及新鲜粪便中分离到21株乳酸菌,通过单层琼脂平板扩散实验,筛选出1株对藤黄微球菌(Micrococcus luteus)、金黄色葡萄球菌(Staphylococcus aureus)和枯草杆菌(Bacillus subtilis)等革兰氏阳性菌和大肠杆菌(Escherichia coli)等革兰氏阴性菌有明显抑菌活性代谢产物的乳酸菌,经细菌鉴定为乳杆菌属。排除酸和过氧化氢的干扰后,发酵液上清对指示菌仍有抑菌活性;用胰蛋白酶和蛋白酶K处理后抑菌活性明显降低,而胃蛋白酶对其活性无影响;用过氧化氢酶作用上清液,抑菌效果不变,说明过氧化氢未起作用;pH在2.0~7.0时,发酵上清液有抑菌活性;培养液粗提物经120℃处理20 min仍有部分活性,表明培养上清中有蛋白质类细菌素。  相似文献

青海湖裸鲤肠道乳酸菌多样性与抑菌活性   总被引:1,自引:0,他引:1  
【目的】通过生理生化特性,结合16S r RNA基因序列分析研究青海湖裸鲤肠道乳酸菌分离株的多样性,并对这些代表株的抑菌活性进行初步探讨,以期筛选具有高效抑菌活性的鱼源益生菌。【方法】对分离的47株乳酸菌代表株进行p H、温度生长范围、耐盐性等生理生化特征检测,结合16S r RNA基因序列对已分离到的乳酸菌进行基因分型和菌种鉴定,采用牛津杯双层平板法检测乳酸菌代表株的抑菌活性。【结果】鉴定结果显示:23株为Lactobacillus fuchuensis(48.94%),12株为Lactobacillus curvatus(25.53%),3株为Leuconostoc fallax(6.38%),2株为Lactobacillus sakei(4.26%),2株为Weissella ceti(4.26%);2株为Lactococcus cremoris(4.26%),1株为Leuconostoc lactis(2.13%),1株为Weissella minor(2.13%),1株为Enterococcus devriesei(2.13%)。qz1217、qz1196、qz1220所在的A、B、C三组乳酸菌在5-50°C的温度范围内生长良好,qz1196、qz1220所在的B、C组在pH 3.0-10.0的范围内生长良好,几乎所有乳酸菌都具有耐6.5%盐浓度特性。13株乳酸菌菌株对6种病原菌都具有抑制作用。通过排除酸、过氧化氢实验,发现上清液仍然具有抑菌活性。对qz1251发酵液进行蛋白酶处理,抑菌活性消失,确定其抑菌物质属于蛋白类物质,是一种细菌素。【结论】青海湖裸鲤肠道附着乳酸菌的多样性为益生性乳酸菌的筛选提供优质资源及数据参考。  相似文献

目的 从阴道炎患者的棉拭子样本中分离筛选具有抗菌活性乳酸菌,为研究对细菌性阴道炎具有确实疗效的微生态制剂提供理论与菌种保证.方法 应用纯培养技术及双层平板法从阴道样品中分离筛选具有较好抑菌活性的乳酸菌.通过有机酸排除试验、过氧化氢排除试验和蛋白类抑菌物质排除试验对具有较好抑菌活性菌株的主要抑菌物质分析,并通过PCR反应对是否具有细菌素相关基因进行鉴定.结果 从7个棉拭子样本中共分离得到26株乳酸菌,其中有6株呈现抑菌活性.K4M2、K4M5、K4M6和K4M8菌株的主要抗菌物质为有机酸.而K4M9、K4M10两株植物乳杆菌的主要抗菌物质除有机酸外,尚可以产生细菌素.病原菌拮抗试验结果表明,这两株菌株的混合培养上清对具有较好的抑菌效果.结论 K4M9、K4M10菌株有望作为微生态制剂用于细菌性阴道炎的临床防治.  相似文献

动物乳杆菌的分离鉴定及其抑菌蛋白的特性分析   总被引:2,自引:0,他引:2  
通过滤纸片法从健康肉猪猪大肠、小肠中分离得到212株抗生物质产生菌, 以杯碟法复筛, 得到1株对溶壁微球菌(Micrococcus lysodeikticus)、金黄色葡萄球菌(Staphylococcus aureus)、枯草芽孢杆菌(Bacillus subtilis)、蜡状芽孢杆菌(Bacillus cereus)等革兰氏阳性菌和大肠杆菌(Escherichia coli)、鼠伤寒沙门氏菌(Salmonella typhimurium)等革兰氏阴性菌以及部分真菌如禾谷镰刀霉(Fusarium graminearum)均有强烈抑制作用的乳酸菌。经形态学、生理生化特征及16S rDNA序列同源性分析等手段鉴定该菌株为动物乳杆菌(Lactobacillus animalis)。排除酸和过氧化氢的干扰后, 该菌株的发酵上清液对指示菌仍有明显抑菌活性; 用蛋白酶处理该菌株的发酵上清液后, 抑菌活性丧失; 发酵液粗提物具有较好的热稳定性(经121°C处理20 min仍有较强抑菌活性)以及较宽的抑菌活性pH值范围(3.5~5.5), 因此初步认为该菌株产生一类具有广谱抑菌活性的类细菌素物质。  相似文献

目的 从植物性材料中筛选产细菌素的乳酸菌。方法 琼脂扩散法。结果 所筛选的产细菌素R260菌株经鉴定为植物乳杆菌。排除有机酸、过氧化氢等干扰因素后,发酵液仍有很强的抑菌作用;用胰蛋白酶和胃蛋白酶处理后,发酵液抑菌活性急剧下降,因而确定产生的抑菌物质具有蛋白质性质,是一种细菌素。抑菌谱试验测定表明,此菌株的发酵液不仅抑制革兰阳性菌,而且对部分革兰阴性菌也有抑制作用,因此产生的是一类广谱细菌素。结论筛选到了1株产广谱细菌素的乳酸菌。  相似文献

目的通过对一株植物乳杆菌代谢产物的抑菌作用以及该产物的某些物理特性的研究,为进一步发现可替代现有抗生素的抗菌物质奠定基础。方法首先利用低温离心和超滤法对植物乳杆菌培养后的上清液进行初步提取,用打孔法行抑菌试验,明确其对铜绿假单胞菌、大肠埃希菌、肺炎克雷伯菌和金黄色葡萄球菌等临床分离菌株的抑菌活性;并进一步经加热、调整pH及过氧化氢酶等处理上清液后,再验证其活性变化。结果植物乳杆菌的培养上清液对铜绿假单胞菌等多株临床分离菌株均具有一定的抑菌活性,且以pH在6以内时该物质抑菌活性较好,过氧化氢酶处理后或加热至100℃、30min,上清液的抑菌活性依然存在。结论在植物乳杆菌的培养上清液中存在着具有抑菌活性的物质,对从临床标本所分离的部分革兰阴性菌及革兰阳性菌有抑菌作用,该物质对热耐受,其活性受pH变化的影响。  相似文献

一株植物病原拮抗细菌的分离筛选及拮抗物质   总被引:1,自引:0,他引:1  
从黄豆酱中分离筛选对植物病害有防治效果的拮抗菌,提取发酵液中的拮抗物质,研究其抑菌机理及基本性质。采用稀释平板法分离细菌、平板对峙法筛选拮抗菌,通过形态特征、生理生化特征及16S rRNA鉴定菌种,硫酸铵沉淀法提取发酵液中的拮抗物质,考马斯亮蓝法测得蛋白含量,扫描电镜观察拮抗物质抑菌作用,琼脂孔扩散法测定拮抗物质抑菌谱及其活性的稳定性。从黄豆酱中分离出18株细菌,通过筛选,得到一株拮抗效果最好的菌株,菌体抑菌率62.78%,发酵液抑菌率53%,命名为4-z-3,经鉴定为Bacillus siamensis。4-z-3发酵液在硫酸铵饱和度为20%~30%时提取的粗提物抗菌活性最大,为60.8%。最适饱和度下提取的粗提物的蛋白质含量为3.45%。拮抗物质对多种病原菌有良好的拮抗效果。经过4-z-3粗提物处理的病原真菌,菌丝受到破坏,细胞形状改变,细胞完整性缺失。粗提物的稳定性方面,对热不敏感,仅在90和100℃时抑菌活性下降明显;对酸碱不敏感,pH值为4时仍有81.6%的抑菌活性;对蛋白酶不敏感,抑菌活性最低时降低了19.6%;对紫外线不敏感,照射12 h之后抑菌活性仍有92.5%。本研究表明,菌株4-z-3是一株具有发展潜力的农业生防菌,分泌的胞外拮抗物质能够有效防治多种植物病害,且抑菌活性稳定。  相似文献

从健康成年尼罗罗非鱼(Oreochromis niloticus)肠道中分离纯化得到乳酸菌,采用牛津杯双层平板法筛选具有抑制嗜水气单胞菌(Aeromonas hydrophila)活性的乳酸菌菌株,借助16S rRNA分子鉴定乳酸菌种类。通过排除有机酸和过氧化氢干扰及蛋白酶敏感性等试验分析抑菌活性最佳乳酸菌的抑菌活性物质成分,并进行热、酸碱稳定性和扫描电镜观察等确定抑菌活性物质对嗜水气单胞菌的抑菌作用。结果显示,尼罗罗非鱼肠道中有3株乳酸菌对嗜水气单胞菌表现出良好的抑菌效果,经鉴定后分别为植物乳酸菌(Lactobacillus plantarum)和唾液乳杆菌(Lactobacillus salivarius)。其中,菌株LX3的抑菌活性最佳,抑菌圈直径可达到(25.25±0.23) mm且对多种蛋白酶敏感,经不同温度和酸碱性处理后抑菌活性物质仍保持较高的抑菌活性,特别是在pH 12和100 ℃处理后,抑菌圈直径仍达到(13.87±0.12)和(16.65±0.26) mm。此外,扫描电镜观察发现菌株LX3抑菌活性物质主要破坏了嗜水气单胞菌的细胞结构,使细胞内容物流出,致其死亡。上述结果表明,尼罗罗非鱼肠道中拥有较为丰富的抑制嗜水气单胞菌乳酸菌资源,尤其是菌株LX3显示出良好的抑菌活性与抑菌作用,这对于替代抗生素防治嗜水气单胞菌引发的相关鱼类疾病具有重要价值。  相似文献

目的:对番茄早疫病菌拮抗菌ZJNU27发酵液稳定性进行分析,从而为抑菌活性物质的分离纯化提供一定理论依据.方法:以番茄早疫病菌为指示菌,采用杯碟法测定发酵液的热稳定性、酸碱稳定性及光稳定性.结果:拮抗菌发酵液抑菌活性随着温度升高而降低,90℃时抑菌圈直径减至2.70cm,121℃时则无抑菌活性.随着酸碱性增强,抑菌活性均呈逐步下降趋势,但在pH 2.63和pH 10.52下抑菌圈直径仍有3.57cm和3.28cm.紫外光照射4h后,拮抗菌发酵液抑菌圈直径仍有3.13cm.自然光照射9h后,拮抗菌发酵液抑菌活性与对照无显著差异.结论:番茄早疫病菌拮抗菌ZJNU27发酵液具有较强的热稳定性、酸碱稳定性和光稳定性,具有一定开发价值和应用前景.  相似文献

目的筛选具有抑菌能力的乳杆菌菌株并对其抑菌物质进行初步探究。方法采用琼脂扩散法进行抑菌试验,筛选出具有抑菌能力的乳杆菌菌株;通过热稳定性试验检测抑菌物质耐高温的能力;通过有机酸排除与过氧化氢排除试验检测这两种物质对抑菌作用是否有影响;用胃蛋白酶、胰蛋白酶和蛋白酶K消化处理各株乳杆菌无细胞发酵液后进行抑菌试验,判断抑菌物质是否为蛋白多肽类物质。结果 2株副干酪乳杆菌与1株保加利亚乳杆菌对大肠埃希菌、铜绿假单胞菌、伤寒沙门菌和痢疾志贺菌有抑菌效果。这3株乳杆菌无细胞发酵液中的抑菌物质经过高温处理后仍具有抑菌能力,但抑菌能力与处理前相比显著降低(P0.05);有机酸对照组未产生明显的抑菌圈;过氧化氢排除后的无细胞发酵液的抑菌能力未受影响;经过蛋白酶作用3株乳杆菌无细胞发酵液的抑菌能力显著降低(P0.05)或消失。生物被膜态副干酪乳杆菌2的抑菌能力与浮游态相近,其无细胞发酵液中的抑菌物质可以完全耐受100℃处理与胃蛋白酶的消化作用,可部分耐受胰蛋白酶的消化作用。结论副干酪乳杆菌1、副干酪乳杆菌2和保加利亚乳杆菌具有良好的抑菌能力,它们产生的主要抑菌物质为蛋白多肽类,此物质具有较好的耐高温能力与耐蛋白酶能力;被膜态副干酪乳杆菌产生的抑菌物质表现出了更强的抗胁迫能力与稳定性。  相似文献

The evaluation of the potentiality of lactic acid bacteria (LAB) strains isolated from different origins to inhibit mould growth and to identify and characterize the antifungal metabolites were the aims of this study. From a total of ninety-one LAB strains tested, ten were selected due to their high inhibitory effect (>80%). The antifungal activity of the majority of the selected LAB strains was lost after the neutralization treatment determining the acidic nature of the antifungal metabolites. Lactic, acetic and phenyllactic (PLA) acids were identified as being responsible for antifungal effect in the 10 cell-free supernatants (CFS) evaluated. Amongst the strains evaluated, only Lactobacillus fermentum CRL 251 produced fungus inhibitory peptide/s, smaller than 10 kDa, thermostable, active in the pH range of 4–7 and sensitive to trypsin. This is the first report on antifungal peptide/s produced by a L. fermentum strain.  相似文献

A strain of Pediococcus acidilactici LAB 5 was isolated from vacuum-packed fermented meat product, in order to obtain a novel bacteriocin from food-grade organisms. Optimized culture conditions for bacteriocin production in different media (viz., MRS, TGE, TGE + buffer, TGE + Tween 80, and TGE + Tween 80 + buffer) and at different temperatures and pH conditions were reported. TGE + Tween 80 + buffer medium was found to be most effective for bacteriocin production (about 2400 AU/ml) by this strain, when incubated at 37 degrees C for 24 h. Bacteriocin, partially purified by adsorption-desorption method showed molecular mass of 10.3 kDa and produced prominent inhibition zone in activity gel. It showed significant storage stability both at high as well as in low temperatures for up to 6 months and retained its activity in a number of organic solvents, except in 2-mercaptoethanol. The treatment with amylase and lysozyme did not change its activity, but it lost its activity on proteinase K treatment. Antibacterial efficacy of bacteriocin was proved against some food spoilage and human pathogenic bacteria like Enterococcus, Leuconostoc, Listeria, Staphylococcus and Streptococcus.  相似文献

【目的】为认识乳酸菌在中国白酒酿造过程中的作用与影响, 分析、比较了酱香型与清香型白酒发酵过程中乳酸菌的菌群结构及其差异。【方法】运用PCR-DGGE技术分析酱香型与清香型白酒发酵过程中乳酸菌群的演变规律。并利用传统微生物分离筛选方法进一步确定酱香型白酒发酵中的主要乳酸菌种。【结果】DGGE图谱表明, 白酒发酵过程中的主要乳酸菌种是乳杆菌。但两种香型白酒发酵过程中乳酸菌群组成及动态变化均呈现出明显的差异。清香型白酒酒醅中Lactobacillus fuchuensis是优势菌种, 而酱香型白酒发酵中检测到多种含量较高的乳酸菌种。利用MRS培养基从酱香型白酒酒醅中共筛选获得5种乳酸菌种。通过两种方法, 确定Lactobacillus homohiochii是酱香型白酒发酵过程中含量最高的乳酸菌。【结论】深入研究白酒发酵过程中乳酸菌的组成及分布规律, 对于更好地认识中国白酒酿造中主要的细菌类群——乳酸菌的作用, 具有重要的理论意义和实践价值。  相似文献

AIMS: The propionibacteria are commercially important due to their use in the cheese industry, and there is a growing interest for their probiotic effects. Stimulatory effects of lactic acid bacteria (LAB) on propionic acid bacteria have been observed. This study was designed to examine the possibility of using spent media previously used to grow LAB for the production of biomass and metabolites of Propionibacterium freudenreichii subsp. shermanii. METHODS AND RESULTS: Seventeen MRS and vegetable juice media were prefermented by various LAB and evaluated for their ability to subsequently support the growth of Propionibacterium, using automated spectrophotometry (AS). Growth of Propionibacterium in spent media was strongly affected by the LAB strain used to produce the spent medium. The native MRS medium (not prefermented) yielded the highest optical density values followed by prefermented media by Lactobacillus acidophilus, Bifidobacterium longum and Lactococcus lactis. Prefermented cabbage juice enabled good growth of Propionibacterium. For the production of organic acids and vitamin B12, cells of Propionibacterium were concentrated and immobilized in alginate beads in the aim of accelerating the bioconversions. More propionic acid was obtained in spent media than in native MRS. The concentration of vitamin B12 was higher in media fermented with free cells than those with immobilized cultures; with the free cells, its concentration varied from 900 to 1800 ng ml(-1) of media. CONCLUSIONS: It was demonstrated that spent media could be recycled for the production of Propionibacterium and metabolites, depending on the LAB strain that was previously grown. Media remediation is needed to improve the production of vitamin B12, especially with immobilized cells. SIGNIFICANCE AND IMPACT OF THE STUDY: This study presents an option for recycling of spent media generated by producers of LAB or producers of fermented vegetables. The propionic fermentation may result in three commercial products: biomass, vitamin B12 or organic acids, which may be used as starters, supplements or food preservatives. AIMS: Food-borne pathogen inhibition was tested in the presence of a mixture of Lactobacillus acidophilus and Lactobacillus casei during fermentation under controlled pH conditions. METHODS AND RESULTS: The growth of Escherichia coli O157:H7, Salmonella serotype Typhimurium, Staphylococcus aureus, Listeria innocua, Enterococcus faecium and Enterococcus faecalis was evaluated for 48 h at 37 degrees C. In the presence of the lactic acid bacteria (LAB), an increase of the generation time was observed for all the gram-positive bacteria evaluated. Staphylococcus aureus was the most sensitive strain showing an increase of the generation time by 210%. However, for all the gram-negative bacteria evaluated, no inhibition occurred after 8 h of fermentation. The soluble portion of Lact. acidophilus- and Lact. casei-fermented milk was recuperated and tested for its antimicrobial activity. Listeria innocua and Staph. aureus were the most sensitive to the presence of fermented milk supernatant showing an inhibition of 85.9% and 84.7%, respectively. This soluble fraction was neutralized to eliminate the antimicrobial effect of the organic acids produced; the most sensitive strains were L. innocua and E. coli O157:H7 showing an inhibition of 65.9% and 61.9%, respectively. Finally, the soluble fraction was neutralized and irradiated at 45 kGy using a (60)Co source to eliminate the possible antimicrobial effect of both organic acids and bacteriocin-like substances. Enterococcus faecalis, E. coli O157:H7 and Staph. aureus were the most affected bacteria by this fraction, showing 39.1, 32 and 31.2% inhibition, respectively. CONCLUSIONS: The results obtained in this study suggest the implication of both organic acids and bacteriocin-like inhibitory substances in the antimicrobial activity observed in the soluble fraction of the probiotic preparation. SIGNIFICANCE AND IMPACT OF THE STUDY: This study revealed the antimicrobial mechanisms of action of Lact. acidophilus- and Lact. casei-fermented milk used to prevent antibiotic-associated diarrhoea.  相似文献

乳酸菌发酵可赋予茶饮料独特的香气与滋味,且可改变其物质组成,产生益生因子等。目前,针对乳酸菌在不同发酵阶段对茶汤中风味物质形成影响的研究较少。本研究以从中国传统泡菜中筛选获得的棒状乳杆菌FZU63为发酵菌株,对不同发酵阶段红茶汤中的挥发性香气成分、还原糖、游离氨基酸、有机酸等含量的变化过程进行分析,并对发酵红茶汤的感官品质进行评价。结果表明,棒状乳杆菌FZU63以红茶汤中的葡萄糖、果糖、甘露糖和木糖作为发酵过程中的主要碳源物质。红茶汤经棒状乳杆菌FZU63发酵作用后,香气成分丰度显著增加,且主要香气组分结构发生改变,发酵红茶汤在花香、坚果香的基础上增添了水果香;此外,部分苦味氨基酸含量下降,甜味和鲜味氨基酸含量增加;并且,乳酸、苹果酸、柠檬酸等有机酸含量在发酵过程中呈现积累。同时,感官评定结果表明棒状乳杆菌FZU63发酵可改善红茶汤的感官品质,且在发酵48h后达到较优。本文系统分析了经棒状乳杆菌发酵不同阶段对红茶汤风味的影响,可为乳酸菌发酵茶饮料的品质控制与产业化应用提供理论参考。  相似文献

【背景】细菌素是一类由细菌产生的抗菌肽活性物质,对多种致病菌有抑制和杀灭作用。昆虫是世界上种类最多、肠道菌群资源最为丰富且多样的动物类群之一。昆虫肠道很可能蕴含着丰富的产细菌素菌种资源。【目的】以马尾松毛虫(Dendrolimus punctatu)幼虫为研究材料,对其肠道细菌进行分离、培养和鉴定,获得对典型致病菌有明显抑制作用的产细菌素细菌,推测其抑菌物质,并对产高活性细菌素细菌的抑菌特性进行研究,进一步丰富产细菌素细菌的资源。【方法】采用传统细菌纯培养方法分离细菌,利用牛津杯法检测抑菌物质活性,结合菌落形态特征及16SrRNA基因序列确定产细菌素细菌种类;进一步排除有机酸和过氧化氢的干扰,初步证实抑菌物质有类蛋白性质。【结果】从马尾松毛虫肠道中筛选得到13株产细菌素细菌,隶属于芽孢杆菌属(Bacillus)、葡萄球菌属(Staphylococcus)、嗜冷杆菌属(Psychrobacte)和肠杆菌属(Enterobacter)。抑菌实验结果显示:13株产细菌素细菌对金黄色葡萄球菌(Staphylococcus aureus)和大肠杆菌(Escherichia coli)两种重要病原指示菌有一定程度的抑制效果,尤其是解淀粉芽孢杆菌(Bacillusamyloliquefaciens)MW-1产生的细菌素经不同温度和酸碱性处理后仍保持较好的抑菌活性,而且经单因素方差分析发现在pH 7.0和37℃时抑菌活性最佳。【结论】马尾松毛虫肠道内存在多种产细菌素细菌且对重要病原菌有抑制作用。菌株MW-1细菌素具有高效的病原菌抑菌能力,所产细菌素对热和酸碱处理稳定性较好,显示了替代抗生素的应用潜力。  相似文献

【目的】解析江苏洋河酒厂浓香型白酒窖内发酵过程酒醅微生物群落结构,建立酒醅微生物与主要有机酸合成的关联性。【方法】通过宏基因组测序获得白酒发酵过程中微生物群落结构变化规律,利用主成分分析和偏最小二乘回归分析寻找酒醅中影响主要有机酸合成的关键微生物。【结果】根据微生物组成结构变化和有机酸合成变化规律,可将白酒窖内发酵分为两个时期(0–14 d和15–60 d)。其中窖内发酵0–15 d与主要有机酸合成相关的微生物数量显著高于15–60 d的。窖内发酵过程与主要有机酸合成相关的微生物包括7个菌属,分别为乳杆菌属(Lactobacillus)、葡萄球菌属(Staphylococcus)、酵母属(Saccharomyces)、Naumovozyma、伊萨酵母属(Issatchenkia)、嗜冷芽孢杆菌属(Psychrobacillus)和根霉属(Rhizopus)。【结论】本研究识别了白酒窖内发酵过程中与主要有机酸合成相关的核心和关键微生物,可为阐明白酒窖内发酵产酸机理和保障白酒品质的稳定性奠定研究基础和理论依据。  相似文献

AIMS: Isolation of bacteriocinogenic lactic acid bacteria (LAB) from the Malaysian mould-fermented product tempeh and characterization of the produced bacteriocin(s). METHODS AND RESULTS: LAB were present in high numbers in final products as well as during processing. Isolates, Enterococcus faecium B1 and E. faecium B2 (E. faecium LMG 19827 and E. faecium LMG 19828, respectively) inhibited Gram-positive indicators, including Listeria monocytogenes. Partially purified bacteriocins showed a proteinaceous nature. Activity was stable after heat-treatment except at alkaline pH values. Both strains displayed a bacteriostatic mode of action. Bacteriocin production was associated with late exponential/early stationary growth. Molecular mass, calculated by SDS-PAGE, was 3.4 kDa for B1 bacteriocin, and 3.4 kDa and 5.8 kDa for B2 bacteriocins. PCR screening of enterocin-coding genes revealed three amplified fragments in total genomic DNA that may correspond with PCR signals for enterocin P, enterocin L50A and enterocin L50B. Both B1 and B2 contained a 42-kb plasmid. No differences in bacteriocinogenic capacity were found between wild type strains and plasmid-cured strains. CONCLUSIONS: It was possible to isolate bacteriocinogenic E. faecium active against various Gram-positive bacteria from final products of tempeh. SIGNIFICANCE AND IMPACT OF THE STUDY: A first step in applying biopreservation to fermented South-east Asian foods is to obtain bacteriocinogenic LAB from this source. Such isolates may also be used for biopreservation of mould-fermented foods in general, including various types of mould-ripened cheese.  相似文献

Purpose

Ogi is an indigenous edible fermented cereal slurry but the steep liquor is usually wasted or administered as therapeutic to suppress certain illnesses. The combination of lemon juice and ogi steep liquor (OSL) is known to possess bioactive metabolites.

Method

This study evaluated potential probiotic lactic acid bacteria (LAB) in different OSL (Zea mays, Sorghum bicolor, and Pennisetum glaucum L.) and lemon juice-ogi steep liquor (LJOSL) based on low pH, bile and lysozyme tolerances, hydrophobicity and auto-aggregation, antibiotic, cholesterol removal, exopolysaccharide production, β-galactosidase, and antimicrobial and hemolytic activities using standard methods. Presumptive LAB were sequenced and assayed for radical scavenging using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and lipid peroxidation inhibitory (LPI) tests.

Results

Presumptive LAB counts were higher in maize OSL (0 h:5.09 log CFU/ml) and combined cereal OSL (24–48 h:7.65 and 7.72 log CFU/ml) but decreased in all steep liquors at 72 h, except in millet OSL (7.72 log CFU/ml). A total of 120 LAB isolates were randomly selected. Based on pH and bile tolerances, 14 isolates were comparable to reference strains. All these isolates demonstrated probiotics properties except for three that did not show γ-hemolysis. Sequenced LAB isolates were identified as Lactobacillus plantarum, Lactobacillus fermentum, Pediococcus pentosaceus, and Weissella cibara. DPPH activities of LAB gradually increased during fermentation with the highest activity of DPPH (58.77%) and LPI (57.94%) activity in L. plantarum. Strong correlations were found between DPPH and LPI in all the selected isolates.

Conclusion

The antioxidant property of probiotic LAB in OSL and LJOSL could contribute to its therapeutic nature.
