粘红酵母发酵生产类胡萝卜素培养条件的优化   总被引:5,自引:0,他引:5  
目的是通过测定不同条件下类胡萝卜素的产量找出粘红酵母发酵生产类胡萝卜素的最优条件。探讨了不同碳源、氮源对粘红酵母菌体生长和色素形成的影响,并通过正交实验确定了最佳条件组合。实验结果表明,最适发酵培养条件为:蔗糖40g/L、酵母粉20g/L、转速150r/min、装液量30mL/500mL、发酵时间84h。在此条件下,粘红酵母摇瓶发酵的生物量、类胡萝卜素含量及产量分别达15.17g/L、718.6μg/g、10.9mg/L,依次比初始发酵提高了1倍、7.4倍和15.8倍。发酵过程动态分析表明,84h色素产量达最高峰。  相似文献

目的 对海洋红酵母Y2高产类胡萝卜素的发酵条件进行优化.方法 在摇瓶条件下,研究培养基成分和培养条件对海洋红酵母Y2生长和类胡萝卜素合成的影响,同时进行海洋红酵母Y2发酵过程的动态分析.结果 海洋红酵母Y2优化培养基组合为葡萄糖45 g/L,蔗糖15 g/L,酵母粉5 g/L,蛋白胨2.5 g/L,磷酸二氢钾1 g/L,磷酸二氢钠3 g/L,硫酸镁7.5 g/L,氯化钾3 g/L,氯化钠5 g/L.最适培养参数为:温度20℃,培养基初始pH为5,接种量为10%,250 mL摇瓶装液量为10~50 mL.类胡萝卜素的合成主要集中在对数生长期和稳定期.海洋红酵母Y2最适收获时间为72 h.种龄以36 h为宜.结论 利用优化培养基,在最适条件下培养海洋红酵母Y2,类胡萝卜素产量达到4.97 mg/L,比基础培养基提高了60.32%.  相似文献

目的:对黑曲霉WP124发酵产橙皮苷酶的工艺条件进行优化,旨在为利用酶法改造橙皮苷打下基础.方法:采用摇瓶培养,对培养基的成分和培养条件进行了优化.结果:黑曲霉WP124发酵产橙皮苷酶的最佳培养基组成是:蔗糖30g/L,酵母膏20g/L,磷酸二氢钾3g/L,桔皮粉50g/L;最佳培养条件是:起始pH为5.5,培养温度为30℃,摇瓶转速是180r/min.在上述条件下,经过72h的培养,橙皮苷酶酶活达到1398U/mL.结论:该菌株发酵过程具有较好的工业化应用的前景.  相似文献

以代谢控制发酵理论为指导,重点对C.glutamicum 366菌株进行摇瓶发酵条件的优化。应用响应面法优化发酵培养基的配比,优化后的发酵培养基:葡萄糖63.33 g/L、精氨酸196.96 mg/L、(NH4)2SO445.79 g/L、生物素35.72μg/L、K2HPO4·3H2O 1.0 g/L、KH2PO41.0 g/L、Mg SO4·7H2O、0.25 g/L、Mn SO4·H2O 0.02 g/L、Fe SO4·7H2O 0.02g/L、Zn Cl21 mg/L、Cu SO40.2 mg/L、VB1200μg/L、Ca CO330 g/L。摇瓶发酵培养条件:温度30℃、摇床转速200r/min、初始p H 7.0。在此发酵条件下,菌株进行摇瓶发酵72 h,产L-瓜氨酸14.96 g/L,相比优化之前提高了75.8%。  相似文献

目的:提高凝结芽孢杆菌芽孢形成率及芽孢数量,为凝结芽孢杆菌芽孢制剂的产业化生产提供理论依据.方法:在摇瓶、15L自动发酵罐中考察了碳源、氮源、无机盐、微量元素Mn2+、pH、温度、接种量、溶氧水平对凝结芽孢杆菌液体培养形成芽孢的影响.结果:芽孢形成的最适培养基组成为:麸皮20g/L,酵母膏5g/L,豆粕粉10g/L,NaCl 5g/L,K2HPO4 3g/L,MnSO4.3g/L;最适培养条件为:初始pH7.0,接种后最适起始芽孢浓度为106CFU/mL,培养温度为40℃,180r/min摇瓶培养,250mL三角瓶中最适装液体积为15mL.在15L自动发酵罐中扩大培养,控制溶氧在30%以上,培养20h,芽孢数量可达5.8×109CFU/mL,芽孢率达96.7%.结论:试验获得的最佳培养条件可进一步应用于生产.  相似文献

从废弃食用菌培养基周围土壤中分离得到一株产壳聚糖酶的菌株,结合形态学特征与26SrDNA序列进行了分类学鉴定,结果表明,该菌株与高山被孢霉(Mortierella alpina)的同源性较高,达99%,初步鉴定为被孢霉属的一种,命名为KB-1001。并对该菌株的产酶特性进行了研究,结果表明,该菌株液体发酵培养产酶高峰出现在第84h,最适碳源为1%的水溶性壳聚糖,最适氮源为1.87%的(NH4)2SO4,摇瓶培养的最适初始pH值为6.0,最适温度为28℃~30℃,接种量为4%,最佳装瓶量为70 mL/250 mL,150 r/min摇瓶培养,经优化培养后,该菌株发酵液中壳聚糖酶活力最高达到8.130 U/mL。比原始的未经发酵条件优化的产酶活性提高了12.78%。  相似文献

以红酵母菌株NZ-01为试验菌株,研究其发酵工艺与中试生产。采用摇瓶发酵优化的方式,研究培养基组分与发酵工艺条件对该菌发酵的影响,并进行中试放大生产。结果显示,该菌最适生长培养基组分为葡萄糖10g/L,蔗糖10g/L,酵母膏10g/L,牛肉膏2.5g/L;色素合成最适培养基组分为葡萄糖15g/L,蔗糖10g/L,酵母膏2.5g/L,牛肉膏5g/L。最适生长起始pH值为6.0,最适接种量为8%,生长周期为44h;最适色素合成起始pH值为7.0,最适色素合成接种量为8%,色素合成周期为48h。发酵优化后的色素产量3.88μg/mL较优化前1.71μg/mL提高了127%。中试产量达3.05μg/mL。红酵母菌NZ-01优化后的发酵条件可以应用于中试生产虾青素,有规模化生产应用潜力。  相似文献

一株高产脯氨酸的嗜醋酸棒杆菌的选育及发酵条件优化   总被引:1,自引:0,他引:1  
以嗜醋酸棒杆菌为出发菌株,经过片段化全基因组体外诱变、重组和连续的磺胺胍抗性筛选,获得一株L-脯氨酸的高产菌株。摇瓶发酵优化结果表明,葡萄糖、生物素和硫胺素的最适用量分别为16%、300μg/L、400μg/L,最适pH为6.8~7.0,装液量为25ml/500ml摇瓶,发酵培养72h后L-脯氨酸产率高达到75.6g/L,与对照相比提高了5%。考察了50L发酵罐中细胞生长对L-脯氨酸产量的影响,补料分批发酵结果表明(比生长速率分别为0.06/h、0.08/h和0.1/h),比生长速率在0.08/h左右时L-脯氨酸的产率最高,L-脯氨酸的比生产速率QP达到0.091 g/(g.h),产率高达82.1 g/L,比优化前提高了14%。  相似文献

对一株从腐烂海带中筛选得到的产褐藻胶裂解酶的菌株进行鉴定,并对其产酶条件进行发酵优化。经形态学、生理生化特征和分子生物学鉴定,将其鉴定为盐单胞菌属,并命名为Halomonas sp. WF6。通过在摇瓶培养水平上进行单因素和多因素正交试验,确定褐藻胶裂解酶产生菌WF6的最适产酶培养基为:褐藻酸钠6.0 g/L,蛋白胨5.0 g/L,酵母粉2.5 g/L,NaCl 30 g/L,K+ 5 mmol/L。进而采用最适培养基进行产酶条件的优化,优化后的发酵产酶条件为:初始pH 8.0,培养温度25℃,接种量为2%,摇瓶装液量30 ml/250 ml,培养时间39 h。优化后的褐藻胶裂解酶酶活达117.66 U/ml,是优化前的2.1倍。该酶对褐藻酸钠的酶解产物主要由聚合度为二和三的褐藻寡糖组成。  相似文献

经60 Coγ射线辐照处理获得的诱变菌株芽孢杆菌BS9418F ,其产酶活力比出发菌株提高 30 %以上。该菌株以大麦粉 7%、玉米粉 3%、豆粕 3%及适量无机盐为培养基最佳配比 ,其最适培养条件为 :培养基初始 pH 7.0 ,摇瓶装量 5 0mL/ 30 0mL三角瓶 ,种龄 16~ 2 0h ,接种量 2 %~ 3% ,培养温度 36~ 37℃ ,发酵周期 40h。在优化条件下 ,摇瓶发酵产 β 葡聚糖酶活力高达 5 5 0 0u/mL以上 ,比出发菌株初始发酵水平提高了 4倍以上  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.
