Raffinose family oligosaccharides (RFO) have been implicated as protective agents in the cellular dehydration tolerance, especially of many plant seeds. However, their efficacy in stabilizing membranes during dehydration has never been systematically investigated. We have analyzed the effects of sucrose, raffinose, stachyose, and verbascose on liposome stability during air-drying. With increasing degree of polymerization (DP), the RFO were progressively better able to stabilize liposomes against leakage of aqueous content and against membrane fusion after rehydration. Indeed, there was a very tight linear correlation between fusion and leakage for all RFO. These data indicate that increased protection of liposomes against leakage with increasing DP is due to better protection against fusion. This is in accord with the higher glass transition temperature of the longer chain oligosaccharides. Further evidence for the influence of glass transitions on membrane stability in the dry state was provided by experiments testing the temperature dependence of membrane fusion. During incubation at temperatures up to 95 degrees C for 2 h, fusion increased less with temperature in the presence of higher DP sugars. This indicates that RFO with a higher glass transition temperature are better able to protect dry membranes at elevated temperatures. In addition, Fourier-transform infrared (FTIR) spectroscopy showed a reduction of the gel to liquid-crystalline phase transition temperature of dry liposomes in the presence of all investigated sugars. However, the RFO became slightly less effective with increasing chain length, again pointing to a decisive role for preventing fusion. A direct interaction of the RFO with the lipids was indicated by a strong effect of the sugars on the phosphate asymmetric stretch region of the infrared spectrum.  相似文献

5-Hydroxymethylfurfural (HMF) was produced from monosaccharide (fructose and glucose), polysaccharide (inulin) and the Jerusalem artichoke juice by a simple one-pot reaction including hydrolysis and dehydration using solid acid under mild condition. Hydrated niobium pentoxide (Nb(2)O(5)·nH(2)O(2)) after pretreatment showed high catalytic activities for dehydration of mono- and polysaccharide to HMF at 433 K in water-2-butanol (2:3 v/v) biphasic system, giving high HMF yield of 89% and 54% from fructose and inulin, respectively. The HMF yield was up to 74% and 65% when inulin and Jerusalem artichoke juice were hydrolyzed by exoinulinase. The solid acid made the process environment-friendly and energy-efficient to convert carbohydrates into bio-fuels and platform chemicals.  相似文献

Inulin is a polysaccharide that enhances various immune responses, mainly to T and B cells, natural killer cells, and macrophages in vivo and in vitro. Previous reports describe that inulin activates macrophages indirectly by affecting the alternative complement pathway. In this study, we examined the direct effect of inulin on PMA-treated THP-1 macrophages. Inulin treatment did not stimulate the proliferation of THP-1 macrophages at all. However, inulin treatment significantly increased phagocytosis of the polystyrene beads without the influence of serum. Doses of around 1 mg/mL had the maximal effect, and significant progression of phagocytosis occurred at times treated over 6 h. Inulin augmented phagocytosis not only with polystyrene beads but also with apoptotic cancer cells. The inulin-induced phagocytosis uptake was suppressed in Toll-like receptor (TLR) 4 mutated C3H/HeJ mice peritoneal macrophages. Moreover, inulin-induced THP-1 macrophage TNF-α secretion was inhibited using a blocking antibody specific to TLR4, suggesting that TLR4 is involved in the binding of inulin to macrophages. Furthermore, we used specific kinase inhibitors to assess the involvement of inulin-induced phagocytosis and revealed that phosphoinositide 3-kinase and mitogen-activated protein kinase, especially p38, participated in phagocytosis. These results suggest that inulin affects macrophages directly by involving the TLR4 signaling pathway and stimulating phagocytosis for enhancing immunomodulation.  相似文献

以300日龄商品代尼克T粉壳蛋鸡为试验动物,研究了菊苣提取物和菊粉对血清脂质、蛋黄总脂和胆固醇的影响.结果显示,0.1%菊苣提取物组、2.0%菊苣提取物组和0.1%菊粉组血清胆固醇(TC)、甘油三脂(TG)、低密度脂蛋白(LDL-C)均显著低于对照组(P<0.05),其中0.1%菊苣提取物组TC降低36.47%,TG降低40.71%,LDL-C降低36.09%,0.1%菊粉组LDL-C降低16.23%,差异极显著(P<0.01);2.0%菊苣提取物组高密度脂蛋白(HDL-C)比对照组高23.19%,差异极显著(P<0.01);2.0%菊苣提取物组蛋黄总脂和蛋黄胆固醇显著降低(P<0.05).表明菊苣提取物和菊粉均具有降血脂活性,但菊苣提取物比菊粉活性更强;菊苣提取物还具有降低蛋黄总脂和胆固醇的作用.  相似文献

A polysaccharide-fatty acid complex was isolated from the cell surface of Candida tropicalis growing on alkanes. This complex was solubilized by Pronase treatment of whole cells. A decrease in alkane-binding affinity was observed after Pronase treatment, resulting in 10 to 12% of the yeast dry cell weight being released as polysaccharide. The isolated polysaccharide contained 2.5% fatty acids. C. tropicalis and Saccharomyces cerevisiae grown with glucose contained only traces of fatty acids in the corresponding polysaccharide fraction. The fatty acids were not removed from the polysaccharide moiety by gel filtration. Extraction of the polysaccharide with chloroform-methanol showed that fatty acids were covalently bound to the polysaccharide. The amphipathic nature of the isolated polysaccharide and the hydrocarbon-induced formation suggest a possible role in alkane metabolism.  相似文献

A polysaccharide preparation, isolated from egg masses deposited by snails of an Ampullarius species, was purified via precipitation with Cetavlon in the presence of sodium borate, and found to contain d-galactose and a smaller proportion of d-glucose, and to have two components with sedimentation coefficients of 10S and 40S. A polysaccharide, isolated from freshly laid egg masses, was highly branched and was shown to contain nonreducing α-d-glucopyranosyl and β-d-galactopyranosyl end-groups, and 3,6-di-O-substituted β-d-galactopyranosyl residues. One or more of the polysaccharide components was a d-glucopyrano-d-galactopyranan with non-reducing α-d-glucopyranosyl end-groups (1→4)-linked to β-d-galactopyranosyl residues. The polysaccharide preparations, obtained from freshly laid egg masses and from those that were left for 10 and 15 days after being laid, were structurally different from each other. With the passage of time, progressive diminution of the 10S component and the proportion of d-glucose in the polysaccharide took place, suggesting that each constituent was consumed preferentially by the snail embryos as an energy source.  相似文献

Preservation of dried liposomes in the presence of sugar and phosphate   总被引:3,自引:0,他引:3  
It has been well established that sugars can be used to stabilize liposomes during drying by a mechanism that involves the formation of a glassy state by the sugars as well as by a direct interaction between the sugar and the phospholipid head groups. We have investigated the protective effect of phosphate on solute retention and storage stability of egg phosphatidylcholine (egg PC) liposomes that were dried (air-dried and freeze-dried) in the presence of sugars and phosphate. The protective effect of phosphate was tested using both glucose (low T(g)) and sucrose (high T(g)) by measuring leakage of carboxyfluorescein (CF), which was incorporated inside the vesicles. Liposomes that were dried with glucose or phosphate alone showed complete leakage after rehydration. However, approximately 30% CF-retention was obtained using mixtures of phosphate and glucose. Approximately 75% CF-retention was observed with liposomes that were dried with sucrose. The solute retention further increased to 85% using mixtures of phosphate and sucrose. The pH of the phosphate buffer prior to drying was found to have a strong effect on the solute retention. Fourier transform infrared spectroscopy studies showed that phosphate and sugars form a strong hydrogen bonding network, which dramatically increased the T(g). The HPO(4)(2-) form of phosphate was found to interact stronger with sugars than the H(2)PO(4)(-) form. The increased solute retention of liposomes dried in the sugar phosphate mixtures did not coincide with improved storage stability. At temperatures below 60 degrees C the rate of solute-leakage was found to be strikingly higher in the presence of phosphate, indicating that phosphate impairs storage stability of dried liposomes.  相似文献

Sugars play an important role in the desiccation tolerance of most anhydrobiotic organisms and disaccharides have been extensively investigated for their ability to stabilize model membranes in the dry state. Much less is known about the ability of oligosaccharides to protect dry membranes. However, it has been shown that different structural families of oligosaccharides have different efficacies to interact with and protect membranes during drying. Here, we have compared three families of linear oligosaccharides (fructans, malto-oligosaccharides, manno-oligosaccharides) for their chain-length dependent lyoprotective effect on egg phosphatidylcholine liposomes. We found increased protection with chain length for the fructans, a moderate decrease in protection with chain length for malto-oligosaccharides, and a strong decrease for manno-oligosaccharides. Using Fourier-transform infrared spectroscopy and differential scanning calorimetry, we show that the degree of lyoprotection of the different sugars is closely related to their influence on the gel to liquid-crystalline phase behavior of the dry membranes and to the extent of H-bonding to different groups (C=O, P=O, choline) in the lipids. Possible structural characteristics of the different oligosaccharides that may determine the extent to which they are able to interact with and protect membranes are discussed.  相似文献

A partially purified preparation of callose plug was obtainedfrom Camellia japonica pollen tube by limited enzymolysis anddensity centrifugation. The plug preparation consisted of proteinand polysaccharide at about 24% and 58% of dry weight, respectively.The plug polysaccharide was demonstrated to be a rß-1,3-glucanby staining reaction, acid hydrolysis, enzymatic digestion,infrared spectroscopy and methylation analysis. The averagedegree of polymerization of this glucan was calculated to beat least 90. This indicates that the plug callose has a longerchain than the tube wall callose. When the extracellular polysaccharide from the black yeast NRRL Y-6272, composed of two parts N-acetyl-D-glucosamine and one part N-acetyl-D-glucosaminuronic acid, is isolated at maximum culture viscosity, adhering black pigment gives the polysaccharide preparations a gray-to-black appearance. Precipitation of the polysaccharide from cell-free culture supernatants with either ethanol of hexadecyltrimethylammonium bromide failed to remove the pigment. Various other methods were therefore tried for obtaining a high-viscosity polysaccharide product free of pigment. By systematically varying ingredients of defined and semidefined media, an improved medium was found that not only gave polysaccharide preparations of increased viscosity, but also increased yield. A key ingredient in this medium is L-asparagine. Also, adding autoclaved bovine serum albumin or egg albumin to this medium at the time of inoculation allowed a pigment-free polysaccharide to be isolated by standard procedures. None of several other proteins of synthetic polyamides tested were as effective as bovine serum albumin or egg albumin. In an alternate approach, pink mutants obtained by irradiation of the parent black strain with ultraviolet light, apparently produce the same extracellular polysaccharide free of any pigment but in lower yields or inferior in quality.  相似文献

A novel conjugate of a polysaccharide and a Gd(III) chelate with potential as contrast agent for magnetic resonance imaging (MRI) was synthesized. The structure of the chelate was derived from H5DTPA by replacing the central pendant arm by a phosphinic acid functional group, which was covalently bound to the polysaccharide inulin. On the average, each monosaccharide unit of the inulin was attached to approximately one (0.9) chelate moiety. The average molecular weight is 23110 and the average number of Gd3+ ions per molecule is 24. The ligand binds the Gd3+ ion in an octadentate fashion via three nitrogen atoms, four carboxylate oxygen atoms, and one P-O oxygen atom, and its first coordination sphere is completed by a water molecule. This compound shows promising properties for application as a contrast agent for MRI thanks to a favorable residence lifetime of this water molecule (170 ns at 298 K), a relatively long rotational correlation time (866 ps at 298 K), and the presence of two water molecules in the second coordination sphere of the Gd3+ ion. Furthermore, its stability toward transmetalation with Zn(II) is as high as that of the clinically used [Gd(DTPA)(H2O)]2-.  相似文献

分析了来自不同地区的菊芋成分,干物质含量在23%~26%,菊粉多糖含量为17%~18%(鲜重)。制备菊芋汁中的主要固形物成分是菊粉多糖,游离的还原糖和可溶性蛋白质含量很低,菊芋用于果糖或果糖糖浆生产具有较高的经济价值;通过摇瓶发酵试验确定了利用菊芋汁生产果糖糖浆的工艺,用自动模拟发酵罐进行了生产模拟实验。产品总糖含量为61%,其中果糖95%,葡萄糖5%,通过发酵法生产果糖糖浆总糖得率为90%。  相似文献

采用电镜和细胞化学技术对地钱(Marchantia polymorpha)卵发生过程进行了研究,根据卵发生过程中细胞化学和超微结构特征可将卵发育过程分为幼卵、中期卵和成熟卵3个阶段.幼卵阶段,卵细胞、腹沟细胞及颈沟细胞间有发达的胞间连丝,但卵与腹沟细胞间的胞间连丝很快退化,幼卵细胞内具大量透明的囊泡,均匀分布于细胞质中;卵发育中期,突出特征是卵细胞质内产生嗜锇性的脂滴,位于囊泡中,与此同时,腹沟细胞退化,其细胞质内产生大型囊泡,囊泡内分泌物与卵细胞外的物质类似,呈PAS反应阳性,表明该物质应为多糖类;卵成熟时,腹沟细胞和颈沟细胞完全退化,卵细胞外包被大量粘性多糖类物质,卵细胞核表面不规则,产生明显的核外突,众多的小泡围绕着细胞核,脂滴聚集成簇,卵细胞内其他细胞器不易区分.卵发育过程中,质体不含淀粉粒,线粒体退化,高尔基体相对发达.地钱卵发育的这些特征显著区分于蕨类植物.  相似文献

Inulin is a polysaccharide composed mainly of D-fructose units and is the most reliable indicator of the glomerular filtration rate. We have proposed an inulin detection method that involves the hydrolysis of inulin to D-fructose using inulinase and the selective binding of D-fructose from inulin using 5-quinolineboronic acid. In this method, the fluorescence of 5-quinolineboronic acid increases, depending on inulin concentration. For inulin in plasma, the detection and quantitation limits were calculated to be 3.7 and 11 μg/ml, respectively.  相似文献

The main objective of this study was to investigate if egg size (mass) at spawning is invariant for Scotia-Fundy summer and autumn (SFSH) and Icelandic summer (ISSH) spawning herring Clupea harengus. Oocyte dry mass measurements for SFSH females collected in 2001 and ISSH females collected in 1999 and 2000 showed a large variation. Difference in egg dry mass among fish was found to vary by as much as twofold in each stock. For ISSH, variation in egg mass was also apparent from oocyte volume measurements made jointly with a histological examination of the ovaries. Approximately 20% of the variation in egg mass could be explained by maternal whole-body mass or total length, indicating that length or age composition in the stocks can potentially influence the recruitment success. This implies that fisheries management strategies should aim to maintain a broad range in age composition.  相似文献

用超声波辅助提取法从柴胡中提取、分离得到多糖SAP3。高效液相色谱法测定其相对分子质量,红外光谱、气相色谱和原子力显微镜对其结构进行初步分析。结果表明:柴胡多糖SAP3是组分均一的多糖,由鼠李糖、木糖、甘露糖、葡萄糖、半乳糖组成,平均相对分子质量为3.3×105。红外光谱分析表明:柴胡多糖具有一般多糖类物质的特征吸收峰,单糖残基以吡喃环的形式存在。原子力显微镜显示柴胡多糖糖链具有分支结构,并缠绕形成环状结构。在0.2~1.2 mg/mL的实验浓度范围内,随着多糖质量浓度的升高,柴胡多糖SAP3对.OH的清除率逐渐增大,最大清除率可达45.2%。  相似文献

Inulin, a polydisperse reserve polysaccharide from chicory, was chemically modified via alkoxylation using ethylene oxide, in a water free medium. The reaction resulted in a range of products with very distinct properties, such as a highly increased water solubility, moderate surface-active properties and high cloud points in electrolyte media. Because of the unique characteristics of inulin, such as its molecular weight range, and because of the high water solubility of the ethoxylates, the products were evaluated as additive in water-blown polyurethane foams. The addition of inulin ethoxylates resulted in an increased foam hardness and density, the latter in fact being unwanted. The foam properties were evaluated based on the indentation test, the foam density, the SAG factor, and the hysteresis curves of standard cubes. Based on these parameters inulin ethoxylates were shown to have a beneficial effect on the foam properties. The inulin ethoxylate with a theoretical degree of substitution of 0.5 proved to be the best derivative, since the increase in hardness was the highest, while the increase in density was negligible.  相似文献
