野生型康氏木霉(Trichodermakoningi)854-B2经多种理化诱变因子及空间微重力辐射等因素的处理,选育到1株高活力纤维素酶变异株B-7。其固体培养物的纤维素酶,以滤纸为底物酶活力为34μ/g,以羧甲基纤维素(CMC)为底物酶力为1472μ/g。与野生菌854-B2相比,产酶活力水平分别提高5倍和7倍多。酶在滤纸上作用的最适条件为pH4.5—5.0,温度55—60°C;25°C,保温24h,pH稳定范围为pH4.0—6.5;70°C保温30分钟,剩余酶活力34.4%。  相似文献

从生霉棉布上分离出的康宁木霉CP88105经诱变处理,获得一株高产纤维素酶的突变株CP88329。在固体培养基上,28℃培养72小时,所产纤维素酶固体曲,以羧甲基纤维素钠为底物酶活力为4880u/g;以脱脂棉为底物酶活力为480u/g。产酶活力水平均为出发菌的3倍。酶在脱脂棉上作用最适条件为pH4.5-5.0,45-50℃;45℃保温4h,pH稳定范围为3.5-6.5;60℃保温1h,酶活力剩余20%。酶可用于苎麻布抛光处理和牛仔服“石磨”处理。  相似文献

野生型康氏木霉854-B_2分别经物理化学诱变因子,TDP辐射器和空间微重力辐射等因素的多级处理,得到1株形态发生明显改变的变异株B—7,其固体培养物的纤维素酶各组分酶活力,如滤纸糖酶活力(FPA)为34u/g,羧甲基纤维素酶活力(CMC—ase)为29.0u/g,β-葡萄糖苷酶活力(β-Glase)为29.0u/g,与854-β2相比,分别提高5.9倍,7.6倍和4.2倍。其固体培养的最佳条件是:pH6.0,28~30℃,96小时,最适培养基成分为:青草粉:麸皮=7:3,1.5~2.0倍水和(NH4)2SO41.5~2.0%(均以固体料计)。  相似文献

以康宁木霉( Trichoderma koningii) T215为生产菌,在 30t气升式发酵罐中进行了液体深层发酵生产纤维素酶的扩大试验。一、二级罐种子培养基由8%麦麸组成,种龄24h。产酶培养基由6%稻草粉,1%麦麸和1%蛋白胨组成,起始pH5.0。每罐装22t培养基,10%接种量,通气量0.4vvm,罐压0.08MPa,29℃±1℃培养 96h。连续试验 5批,平均发酵液酶活力: CMC-Na活力 78.3IU/mL,脱脂棉活力 1.3IU/mL,水杨苷活力1.4IU/mL,滤纸活力  相似文献

对采自全国各地的50份上样进行筛选,获得一株产酶活性较高的菌株B-9545,通过摇瓶培养确定了产酶的最适培养基Ⅱ(g/100ml):复合蛋白胨1.0;玉米淀粉1.5;玉米浆1.5;NaC10.5;K2HpO40.1;MgSO40.02;CaCO30.5;Tween800.1;pHg—10,在该条件下,30℃荡培养48h,酶活力可达596u/ml。对菌株B—9545的产酶特性研究表明:该酶对高pH环境有较强的适应性,因此,可用于碱性洗涤用酶。  相似文献

通过正交试验,对宇佐美曲霉纤维素酶固态发酵条件进行了研究,其较适培养基为:麸皮与稻草配比为2:3,氮源:尿素,含水量:200%(液固比),pH:5.5。28-30℃培养72小时,CMC酶活力为5.05(u/ml),滤纸酶活力达到1.04(u/ml)。CMC酶及滤纸酶最适pH分别为4.41、6.06;最适温度分别为65℃、55℃。酶的热稳定性与pH稳定性较高。  相似文献

根据正交试验得出无花果曲霉原生质体形成的最佳条件,用1%的混合酶液(0.5%纤维素酶+0.25%蜗牛酶+0.25%溶菌酶)作用无花果曲霉菌体细胞,原生质体产量达3.2×107个·ml-1,渗透压稳定剂为0.6mol·L-1KCl于0.2mol·L-1PO3+4(pH5.8)中,酶解时间和酶解温度分别为3.0h、30℃.比较不同酶解时间、再生稳定剂和碳源等因素对原生质体再生的影响,可确定最佳再生条件,再生率达30%以上.  相似文献

从土壤中分离到9株产生β-甘露聚糖酶的芽孢杆菌(Bacillus sp.)。Bacillus sp.M50250mL三角瓶摇瓶培养试验,以4%的魔芋粉为碳源,1.0%(NH4)2SO4为氮源,0.35%Na2CO3,30~34℃培养60h产酶达到高峰。酶活力为180~220u/mL。100L罐发酵,在30~32℃,1:0.75vvm通气量,200r/min条件下,发酵液酶活力高达330u/mL。  相似文献

在152 株脂肪酶产生菌中,链霉菌Z942 产脂肪酶活力为596u/ mL,其最适培养基(g/L) 为:糊精10 、黄豆饼粉30 、尿素10 、K2HPO4 0-5 、MgSO4 0-5 、NaCl 1 和AEO9 0 .5 ,产酶的最适条件为:初始pH9 .5 ~10-0 ,在26 ℃培养48h 。用PVA 橄榄油乳化系统测定该酶的最适pH9 .8 ,最适温度37 ℃,在pH8-6 ~10-2 于5 ℃存放24 h ,酶活力不变。0-14mol/L 的氯化钙有较大的激活作用。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Microbial resistance to antibiotics is an unresolved global concern, which needs urgent and coordinated action. One of the guidelines of the Centers for Disease Control and Preventions (CDC) to combat antibiotic resistance is the development of new antibiotics to treat drug-resistant bacteria. In our effort to find new antibiotics, we report the synthesis and antimicrobial studies of 30 new pyrazole derivatives. These novel molecules have been synthesized by using readily available starting materials and benign reaction conditions. Some of these molecules have shown activity with MIC values as low as 0.78?µg/mL against four bacterial strains; Staphylococcus aureus, methicillin-resistant S. aureus, Bacillus subtilis, and Acinetobacter baumannii. Furthermore, active molecules are non-toxic to mammalian cell line.

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

Cyclin-dependent kinases (CDKs) and Polo-like kinases (PLKs) play key role in the regulation of the cell cycle. The aim of our study was originally the further development of our recently discovered polo-like kinase 1 (PLK1) inhibitors. A series of new 2,4-disubstituted pyrimidine derivatives were synthesized around the original hit, but their PLK1 inhibitory activity was very poor. However the novel compounds showed nanomolar CDK9 inhibitory activity and very good antiproliferative effect on multiple myeloma cell lines (RPMI-8226).  相似文献
