The mpd gene coding for a novel methyl parathion hydrolase (MPH) was previously reported and its putative open reading frame was also identified. To further confirm its coding region, the intact region encoding MPH was obtained by PCR and expressed in Escherichia coli as a hexa-His C-terminal fusion protein. The fusion protein was purified to homogeneity by metal-affinity chromatography. The enzyme activity and zymogram assay showed that the fusion protein was functional in degrading methyl parathion. The amino terminal sequencing of the purified recombinant MPH indicated that a signal peptide of the first 35 amino acids was cleaved from its precursor to form active MPH. A rat polyclonal antiserum was raised against the purified mature fusion protein. The results of Western blot and zymogram demonstrated that mature MPH in native Plesiomonas sp. strain M6 was also processed from its precursor by cleavage of a putative signal peptide at the amino terminus. The production of active MPH in E. coli was greatly improved after the coding region for the signal peptide was deleted. HPLC gel filtration of the purified mature recombinant MPH revealed that the MPH was a monomer.  相似文献

The sequence of a 1,693-base-pair plasmid DNA fragment from Flavobacterium sp. strain ATCC 27551 containing the parathion hydrolase gene (opd) was determined. Within this sequence, there is only one open reading frame large enough to encode the 35,000-dalton membrane-associated hydrolase protein purified from Flavobacterium extracts. Amino-terminal sequence analysis of the purified Flavobacterium hydrolase demonstrated that serine is the amino-terminal residue of the hydrolase protein. The amino-terminal serine corresponds to a TCG codon located 87 base pairs downstream of the presumptive ATG initiation codon in the nucleotide sequence. The amino acid composition of the purified protein agrees well with that predicted from the nucleotide sequence, using serine as the amino-terminal residue. These data suggest that the parathion hydrolase protein is processed at its amino terminus in Flavobacterium sp. Construction in Escherichia coli of a lacZ-opd gene fusion in which the first 33 amino-terminal residues of opd were replaced by the first 5 residues of lacZ resulted in the production of an active hydrolase identical in molecular mass to the hydrolase isolated from Flavobacterium sp. E. coli cells containing the lacZ-opd fusion showed higher levels of hydrolase activity than did cells containing the parent plasmid.  相似文献

目的:在大肠杆菌中表达猪丹毒丝菌C43065株表面保护性抗原A(SpaA)N端保护区(SpaA-N),并检测其抗原性。方法:利用PCR方法从猪丹毒丝菌C43065株基因组中扩增出spaA基因片段,构建pMD18-spaA重组质粒并对插入片段进行测序;以pMD18-spaA重组质粒为模板,PCR扩增得到spaA-N基因片段,构建重组表达质粒pGEX-spaA-N,经序列测定证实正确后转化大肠杆菌BL21(DE3),再经IPTG诱导表达GST-SpaA-N融合蛋白并纯化。结果:扩增得到的spaA基因长1881bp,编码由626个氨基酸残基构成的多肽;SDS-PAGE和Western印迹检测结果表明,诱导表达获得相对分子质量约64000的GST-SpaA-N融合蛋白,该融合蛋白能与相应抗体发生特异性反应。结论:获得了在大肠杆菌中可溶性表达的GST-SpaA-N融合蛋白,为进一步研究猪丹毒丝菌免疫保护性抗原奠定了基础。  相似文献

A heterologous phosphotriesterase (parathion hydrolase), previously cloned from a Flavobacterium species into Streptomyces lividans, was secreted at high levels and purified to homogeneity. N-terminal analysis revealed that it had been processed in the same manner as the native membrane-bound Flavobacterium hydrolase. The enzyme consisted of a single polypeptide with an apparent molecular weight of 35,000 as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Substrate specificity studies showed Kms of 68 microM for parathion, 46 microM for O-ethyl O-p-nitrophenyl phenylphosphonothioate, 599 microM for methyl parathion, and 357 microM for p-nitrophenyl ethyl(phenyl)phosphinate. Temperature and pH optima were 45 degrees C and 9.0, respectively. The purified enzyme was inhibited by 1 mM dithiothreitol and 1 mM CuSO4. After chelation and inactivation by o-phenanthroline, however, activity could be partially restored by 1 mM CuCl or 1 mM CuSO4. The results showed that the purified recombinant parathion hydrolase has the same characteristics as the native Flavobacterium hydrolase. This system provides a source of milligram quantities of parathion hydrolase for future structural and mechanism studies and has the potential to be used in toxic waste treatment strategies.  相似文献

Summary A mixed bacterial culture which was obtained in a previous enrichment grew on parathion, an organophosphate insecticide, as a sole carbon and energy source. A cell-free enzyme preparation from this culture detoxified by hydrolysis eight commercially used organophosphate insecticides.Fermentation procedures for the production of this parathion hydrolase activity were examined to determine if this enzyme activity could be produced economically. The mixed culture was grown using sterile or non-sterile procedures in 4 or 11 continuous and batch culture fermentations. A pure Pseudomonas sp isolated from the mixed culture expressed parathion hydrolase activity when grown under axenic fermentation conditions on industrially used media such as meat extract, soya bean meal, and corn extract. The optimal conditions for production of parathion hydrolase activity were determined for both pure and mixed cultures. The yield of parathion hydrolase activity/ of fermentation broth per hour was improved 22 fold by growing the pure culture on an industrial meat extract medium instead of the mixed culture on parathion.  相似文献

S-Adenosylhomocysteine hydrolase from Sulfolobus solfataricus was expressed in Escherichia coli by inserting the genomic fragment containing the gene encoding for S-adenosylhomocysteine hydrolase downstream the isopropyl-beta-d-thiogalactoside-inducible promoter of pTrc99A expression vector. An ATG positioned 25 bp upstream of the gene which is in frame with a stop codon was utilized as the initiation codon. This construct was used to transform E. coli RB791 and E. coli JM105 strains. The recombinant protein, purified by a fast and efficient two-step procedure (yield of 0.4 mg of enzyme per gram of cells), does not appear homogeneous on SDS-PAGE because of the presence of a protein contaminant corresponding to a "truncated" S-adenosylhomocysteine hydrolase subunit lacking the first 24 amino acid residues. The recombinant enzyme shows the same molecular mass, optimum temperature, and kinetic features of S-adenosylhomocysteine hydrolase isolated from S. solfataricus but it is less thermostable. To construct a vector which presents a correct distance between the ribosome-binding site and the start codon of S-adenosylhomocysteine hydrolase gene, a NcoI site was created at the translation initiation codon using site-directed mutagenesis. The expression of the homogeneous mutant S-adenosylhomocysteine hydrolase was achieved at high level (1.7 mg of mutant protein per gram of cells). The mutant S-adenosylhomocysteine hydrolase and the native one were indistinguishable in all physicochemical and kinetic properties including thermostability, indicating that the interactions involving the NH(2)-terminal sequence of the protein play a role in the thermal stability of S. solfataricus S-adenosylhomocysteine hydrolase.  相似文献

为了在原核细胞中表达青岛文昌鱼Branchiostoma belcheri tsingtaunese S-腺苷高半胱氨酸水解酶(S-adeno-sylhomocysteine hydrolase,SAHH),采取构建文昌鱼SAHH基因的原核表达重组质粒pGEX-6P-1-SAHH的方法,转化入大肠杆菌JMl09感受态细胞中,IPTG诱导蛋白表达,并进行分离纯化.结果经SDS-PAGE分析,重组质粒在JM109中表达并纯化得到的融合蛋白大约为70 kDa,成功构建了文昌鱼SAHH基因原核表达载体,且重组载体表达出融合蛋白,分离纯化得到目的蛋白.  相似文献

Whole cells of recombinant Escherichia coli were immobilized on the screen printed carbon electrode (SPCE) using glutaraldehyde. Recombinant E. coli was having high periplasmic expression of organophosphorus hydrolase enzyme, which hydrolyzes the methyl parathion into two products, p-nitrophenol and dimethyl thiophosphoric acid. Cells immobilized SPCE was studied under SEM. Cells immobilized SPCE was associated with cyclic voltammetry and cyclic voltammograms were recorded before and after hydrolysis of methyl parathion. Detection was calibrated based on the relationship between the changes in the current observed at +0.1 V potential, because of redox behavior of the hydrolyzed product p-nitrophenol. As concentration of methyl parathion was increased the oxidation current also increased. Only 20 μl volume of the sample was required for analysis. Detection range of biosensor was calibrated between 2 and 80 μM of methyl parathion from the linear range of calibration plot. A single immobilized SPCE was reused for 32 reactions with retention of 80% of its initial enzyme activity.  相似文献

摘要: 【目的】提高抗对硫磷抗体的亲和力以提高酶联免疫检测的灵敏度。【方法】本研究通过抗对硫磷单链抗体基因和核心链霉亲和素基因片段的拼接重组,获得抗对硫磷单链抗体-核心链霉亲和素融合基因(scfv-sa),并将该融合基因(scfv-sa)插入到表达载体pET28a(+)中,转化大肠杆菌BL21(DE3)进行原核表达,制备融合蛋白。SDS-PAGE和Western blot鉴定scfv-sa的表达,Ni+-NTA亲和层析柱纯化融合蛋白,并用ELISA方法测定该融合抗体的亲和力。【结果】结果表明,在大肠杆菌BL21(DE3)中该融合基因能表达出分子量约为46kD的融合蛋白,形成了四价结构域-四价聚合抗体。ELISA测定结果表明该抗体能与对硫磷特异结合,抗体效价在1:1×106以上,亲和常数为4.25×107 L /mol。 【结论】制备的抗对硫磷四价聚合抗体能与抗原特异结合,与单克隆抗体相比,抗原结合位点显著增加,ELISA检测灵敏度显著提高。  相似文献

A bacterium, Burkholderia sp. JBA3, which can mineralize the pesticide parathion, was isolated from an agricultural soil. The strain JBA3 hydrolyzed parathion to p-nitrophenol, which was further utilized as the carbon and energy sources. The parathion hydrolase was encoded by a gene on a plasmid that strain JBA3 harbored, and it was cloned into pUC19 as a 3.7-kbp Sau3AI fragment. The ORF2 (ophB) in the cloned fragment encoded the parathion hydrolase composed of 526 amino acids, which was expressed in E. coli DH10B. The ophB gene showed no significant sequence similarity to most of other reported parathion hydrolase genes.  相似文献

In this study, the mph gene encoding methyl parathion hydrolase from Pseudomonas sp. WBC-3 was expressed in Yarrowia lipolytica and the expressed methyl parathion hydrolase was displayed on cell surface of Y. lipolytica. The activity of methyl parathion hydrolase displayed on the yeast cells of the transformant Z51 was 59.5 U mg?1 of cell dry cells (450.6 U per mL of the culture) in the presence of 5.0 mM of Co2?. The displayed methyl parathion hydrolase had the optimal pH of 9.5 and the optimal temperature of 40 °C, respectively and was stable in the pH range of 4.5-11 and up to 40 °C. The displayed methyl parathion hydrolase was also stimulated by Co2?, Cu2?, Ni2? and Mn2?, and was not affected by Fe2?, Fe3?, Na?, K?, Ca2? and Zn2?, but was inhibited by other cations tested. Under the optimal conditions (OD(600 nm) = 2.6, the substrate concentration = 100 mg L?1 and 40 °C), 90.8 % of methyl parathion was hydrolyzed within 30 min. Under the similar conditions, 98.7, 97.0, 96.5 and 94.4 % of methyl parathion in tap water (pH 9.5), tap water (pH 6.8), seawater (pH 9.5) and natural seawater (pH 8.2) were hydrolyzed, respectively, suggesting that the methyl parathion hydrolase displayed on the yeast cells can effectively remove methyl parathion in water.  相似文献

死亡素是由21个氨基酸残基组成的广谱抗菌肽。为了高效表达可溶性的死亡素,本研究利用递归式PCR(recursive PCR, rPCR)扩增了死亡素基因thanatin,并将其和家蝇Musca domestica泛素基因ubiquitin构成嵌合基因,克隆到表达载体pET-32a,再与硫氧还蛋白融合后构建表达载体pET-TRX-UBI-THA。将酶切和测序鉴定正确的质粒转化表达宿主菌BL21,经0.6 mmol/L IPTG诱导,TRX-UBI-THA融合蛋白得到了高效可溶性表达。SDS-PAGE和Western blot检测结果表明融合蛋白的分子量为28.9 kD,与预期的结果一致,表达量占菌体总蛋白的46%。Western blot分析结果显示融合蛋白能与Ni-NTA鏊合物特异性的结合,表明在融合蛋白的N-端带有6×His标签。利用C-端带有6×His标签的泛素C-端水解酶对融合蛋白进行切割,切割产物经Ni2+-NTA亲和柱和HPLC纯化(纯化量为5.4 mg/L),Tricince-SDS-PAGE电泳得到单一的泛素蛋白条带。电喷雾质谱(ESI-MS)分析表明,纯化的泛素分子量为2.57 kD,与通过氨基酸预测的分子量完全一致。利用琼脂孔穴扩散法对泛素活性进行检测,结果显示纯化的泛素对大肠杆菌K12D31和金黄色葡萄球菌Staphylococcus aureus具有较强的活性抑制。本研究表明,利用泛素融合技术可以高效表达可溶性的死亡素。  相似文献

目的:表达和纯化人肿瘤坏死因子α抑制肽-抗炎酸性尾巴融合蛋白。方法:利用PCR搭接方法及基因合成方法获得目的基因,插入带有6×His标签的原核高效可溶性表达载体pET32a中,构建重组表达质粒pET32a-T9-ac-9,将重组表达质粒转化大肠杆菌BL21(DE3),经IPTG诱导目的基因表达;对融合蛋白进行Ni2+金属螯合柱纯化。结果:构建的重组表达质粒经PCR、内切酶鉴定及基因序列测定证实;目的蛋白在大肠杆菌中获得表达,SDS-PAGE显示相对分子质量为22.917×103;对表达产物进行了亲和层析纯化,从上清中获得了纯度较高的人肿瘤坏死因子α抑制肽-抗炎酸性尾巴融合蛋白。结论:获得了可溶性的人肿瘤坏死因子α抑制肽-抗炎酸性尾巴融合蛋白,为其生物学功能研究奠定了基础。  相似文献

The alpha-amino acid ester hydrolase from Acetobacter turbidans ATCC 9325 is capable of hydrolyzing and synthesizing beta-lactam antibiotics, such as cephalexin and ampicillin. N-terminal amino acid sequencing of the purified alpha-amino acid ester hydrolase allowed cloning and genetic characterization of the corresponding gene from an A. turbidans genomic library. The gene, designated aehA, encodes a polypeptide with a molecular weight of 72,000. Comparison of the determined N-terminal sequence and the deduced amino acid sequence indicated the presence of an N-terminal leader sequence of 40 amino acids. The aehA gene was subcloned in the pET9 expression plasmid and expressed in Escherichia coli. The recombinant protein was purified and found to be dimeric with subunits of 70 kDa. A sequence similarity search revealed 26% identity with a glutaryl 7-ACA acylase precursor from Bacillus laterosporus, but no homology was found with other known penicillin or cephalosporin acylases. There was some similarity to serine proteases, including the conservation of the active site motif, GXSYXG. Together with database searches, this suggested that the alpha-amino acid ester hydrolase is a beta-lactam antibiotic acylase that belongs to a class of hydrolases that is different from the Ntn hydrolase superfamily to which the well-characterized penicillin acylase from E. coli belongs. The alpha-amino acid ester hydrolase of A. turbidans represents a subclass of this new class of beta-lactam antibiotic acylases.  相似文献

目的:克隆、表达人vasorin(VASN)蛋白。方法:利用PCR方法从HepG2细胞的cDNA中扩增获得目的基因,并插入带有6xHis标签的原核高效可溶性表达载体pET28a中,构建重组表达质粒pET28a-VASN,将重组表达质粒转化大肠杆菌BL21(DE3),经IPTG诱导后目的基因获得表达,对融合目的蛋白进行Ni^2+金属螯合柱纯化。结果:内切酶鉴定及基因序列测定证实重组表达质粒构建成功;对目的蛋白进行了原核表达,SDS-PAGE显示相对分子质量为61x10^3的特异表达条带;Western印迹证实目的蛋白为VASN,且主要以包涵体形式存在;对经尿素变性的表达产物进行了亲和层析纯化,有利于以后的变性、复性过程。结论:获得了人VASN融合蛋白,为其进一步的生物学功能研究奠定了基础。  相似文献

Recombinant Escherichia coli displaying organophosphorus hydrolase (OPH) was used to overcome the diffusion barrier limitation of organophosphorus pesticides. A new anchor system derived from the N-terminal domain of ice-nucleation protein from Pseudomonas syringae InaV (InaV-N) was used to display OPH onto the surface. The designed sequence was cloned in the vector pET-28a(+) and then was expressed in E. coli. Tracing of the expression location of the recombinant protein using SDS-PAGE showed the presentation of OPH by InaV-N on the outer membrane, and the ability of recombinant E. coli to utilize diazinon as the sole source of energy, without growth inhibition, indicated its significant activity. The location of OPH was detected by comparing the activity of the outer membrane fraction with the inner membrane and cytoplasm fractions. Studies revealed that recombinant E. coli can degrade 50% of 2 mM chlorpyrifos in 2 min. It can be concluded that InaV-N can be used efficiently to display foreign functional protein, and these results highlight the high potential of an engineered bacterium to be used in bioremediation of pesticide-contaminated sources in the environment.  相似文献

Pure recombinant Flavobacterium parathion hydrolase (an organophosphorus acid anhydrase) from Streptomyces lividans was found to hydrolyze the toxic nerve agent soman at only 0.1% of the rate observed with parathion as substrate. Studies with wild-type and recombinant strains of S. lividans support the lack of significant soman breakdown by the hydrolase and also indicate the presence in S. lividans of other significant hydrolytic enzymatic activity towards soman.  相似文献

A simple, rapid and sensitive colorimetric dipstick assay for the detection of the organophosphorous insecticide methyl parathion (MPT) residue in vegetables was developed. The assay was based on the hydrolysis of MPT by a recombinant methyl parathion hydrolase (recMPH), the encoding gene of which was isolated from Burkholderia cepacia, a soil bacterium indigenous to Thailand. This reaction generates protons leading to a change in pH that correlates with the amount of MPH present. Hence, the pH indicator bromothymol blue was used to monitor the MPH hydrolysis as the associated color changes can be observed by the naked eye. The recMPH was immobilized on a PVDF membrane to establish a dipstick assay format. The assays could detect MPT residues in spiked vegetable samples at the concentration of 1 mg/L without using analytical instrumentation. The test is reusable and stable for up to 3 months in the absence of any preservatives.  相似文献
