Tropic 1808基因在大鼠损伤神经组织中的表达   总被引:2,自引:1,他引:1  
目的观察Tropic 1808基因在大鼠正常和损伤坐骨神经组织中的表达,探讨Tropic 1808基因在周围神经损伤与再生过程中的作用.方法采用地高辛标记的Tropic 1808 cDNA探针、抗大鼠S-100蛋白抗体,以原位杂交和免疫组织化学双重染色法,观察Tropic 1808基因在正常和损伤大鼠坐骨神经组织中的表达.结果免疫组化结果显示,大鼠正常坐骨神经可表达S-100蛋白,但表达量较低;神经损伤后,其远侧端S-100蛋白的表达量明显增加.原位杂交结果显示,大鼠正常坐骨神经组织未见Tropic 1808 mRNA杂交信号;损伤神经的远侧端呈现较强的阳性信号,而且在部分S-100强阳性反应区可见Tropic 1808 mRNA杂交信号.结论 Tropic 1808基因在正常坐骨神经组织中未见表达;坐骨神经损伤后,其远侧端增殖的雪旺氏细胞可表达Tropic 1808 mRNA.提示,Tropic 1808是一种周围神经损伤后特异表达的基因.  相似文献

Tropic1808基因重组蛋白单克隆抗体的制备及鉴定   总被引:1,自引:1,他引:0  
目的:制备Tropic1808基因重组蛋白的单克隆抗体,并对其生物学特性进行鉴定。方法:用Tropic1808基因重组蛋白作为抗原免疫BALB/C小鼠,取小鼠脾细胞与小鼠骨髓瘤(SP2/0)细胞融合,经ELISA筛选和有限稀释法获得分泌单克隆抗体的细胞株,Western Blot等方法对其生物学特性进行鉴定。结果:获得2株识别Tropic1808基因重组蛋白的单克隆抗体的细胞株Ⅱ4B、Ⅲ4C。WesternBlot法显示该抗体特异性地识别Tropic1808基因重组蛋白;ELISA法测定杂交瘤细胞培养上清及体内成瘤后产生的腹水的抗体效价分别为1:80、1:600;杂交瘤细胞染色体数平均为90-100;亚类鉴定单抗的重链为小鼠IgG1,轻链为κ型。结论:成功地制备了Tropic1808基因重组蛋白的单克隆抗体,为进一步研究Tropic1808基因重组蛋白的功能提供了良好的基础。  相似文献

目的:比较原代培养许旺细胞(Schwann cells,SCs)和冷冻保存的SCs移植对损伤后坐骨神经再生的作用。方法:原代培养和液氮保存的SCs分别移植到桥接缺损坐骨神经的硅胶管内。在移植后不同时间(第6和8周末),硅胶管远端神经干内注射HRP,逆行追踪背根神经节和脊髓前角的标记神经元数量;测量再生神经纤维的复合动作电位传导速度;电镜观察再生神经纤维的髓鞘形成。结果:原代培养和冷冻保存SCs在移植后不同时间其背根神经节和脊髓前角神经元HRP标记细胞数量、再生神经纤维的复合动作电位传导速度基本一致,再生神经纤维髓鞘的形成未见明显差别。结论:冷冻保存的SCs仍具有促进损伤后周围神经再生的能力。  相似文献

采用大鼠坐骨神经切断损伤模型,行神经外膜端端对线缝合,术中依不同组别,动物于神经缝合处远端0.5cm处分别注射人的正义和反义bcl-2重组腺病毒(Ad/s-bcl-2、Ad/as-bcl-2),报道基因重组腺病毒(Ad/lacZ)和生理盐水。术后48h,7d,15d和30d常规灌注固定大鼠,取L4-L6脊髓节段,应用X-gal染色、bel-2原位杂交和免疫组化染色、TUNEL染色以及乙酰胆碱酯酶(AChE)组织化学染色方法,观察到外源基因能在脊髓中表达,同时外源性Ad/s-bcl-2能显著减少L4到L6节段脊髓前角运动神经元凋亡的数目,减少脊髓前角运动神经元中因坐骨神经切断导致的AChE活性的降低幅度,并加快其恢复。而Ad/as-bcl-2可显著增加坐骨神经切断诱导的脊髓前角运动神经元凋亡数目以及AChE活性降低幅度,并延缓其恢复。这些观察结果表明,外源性bcl-2能保护周围神经切断后引起的脊髓运动神经元损伤。  相似文献

目的:研究周围神经损伤后,局应用碱性成纤维细胞生长因子(basic fibroblast growth factor ,bFGF)对脊髓前角运动神经元的保护作用。方法:切为SD大鼠右侧坐骨神经,近端套接单盲硅胶管后,管内立好注入20μlbFGF(浓度为4000u/ml)或等量生理盐水,术后2周,观察L3-L5脊央前角外侧群大、中小型神经元乙酰胆碱酯酶(acetyl cholinesterase,AChE),酸性磷酸酶(acid phosphatas,Acp)和一氧化氮合酶(nitric oxide synthase,NOS)的阳性细胞数、反应强度和平均灰度。结果:坐骨神经损伤后,脊髓腰节段运动神经元因受损伤的影响发明明显改变。表现为AChE降低,Acp,NOS活性升高,局部应用bFGF后,大型运动神经元(α运动神经元)阳性细胞数,反应强度和平均灰度接近正常侧,与对照组比较具有显性意义(P<0.01)。结论:bFGF可以有效地防止周围神经损伤引起的脊髓前角α运动神经元退变,促进其恢复。  相似文献

目的探讨外源性碱性成纤维细胞生长因子(bFGF)对晚期周围神经再生的作用.方法50只SD大鼠随机分治疗组、对照组各25只,切断右侧坐骨神经,12周后予以修复,修复术后每日分别给予bFGF和生理盐水,行神经电生理和组织学检查.结果治疗组和对照组修复处远段神经均有不同程度再生,4周时已可见到再生轴突,且治疗组多见.计量分析治疗组运动神经传导速度、神经肌肉动作电位幅值、髓鞘厚度、再生轴突直径和截面积明显优于对照组.治疗组与对照组相比,差异有显著性.结论bFGF能促进晚期周围神经再生.  相似文献

为了研究体内表达的脑源神经营养因子(Brain-derivedNeurotrphicfactor,BDNF)对脊髓运动神经元存活及切断神经再生的作用,我们将人BDNFcDNA克隆到真核表达载体pCB6中,使BDNF基因在CMV启动子控制下表达。在雏鸡出生后3小时内及第二天直接将pCB-BDNFcDNA·lipofectin混合物注射到坐骨神经预切断位点附近肌肉内。第二天切断坐骨神经,神经切断10天后进行实验检测,观察到,BDNFcDNA转染阻止了切断神经一侧的腰脊髓内(L4-L6)运动神经元的大量死亡。并显著的促进了切断坐骨神经的再生。这些结果表明直接注射含BDNFcDNA的质粒对损伤的神经进行基因治疗,具有良好的前景;lipofectin介导重组质粒进行基因转染是导入外源基因到体内的一个可行方法。  相似文献

嗅鞘细胞复合PLGA导管修复周围神经缺损的研究   总被引:1,自引:1,他引:0  
探讨嗅鞘细胞(OECs)复合聚乳酸-聚羟基乙酸共聚物(PLGA)导管对大鼠坐骨神经缺损的修复作用。方法:SD大鼠80只,随机分成4组,切除右侧部分神经干造成10mm的神经缺损。OECs PLGA组用充满细胞外基质凝胶和OECs悬液(CM-DiI预标记)的PLGA导管桥接坐骨神经缺损;OECs 硅胶管组用含相同内容物的硅胶管桥接;PLGA组和硅胶管组则分别用充满细胞外基质凝胶和DMEM/F12培养基的PLGA导管和硅胶管桥接。术后每周进行感觉运动功能检测,8周时行腓肠肌湿重恢复率、乙酰胆碱脂酶(AChE)染色、电生理和组织形态学分析等检测,同时移植细胞的两组每周进行细胞示踪观察。结果:移植细胞沿神经纵轴分布;除坐骨神经功能指数(SFI)指标外,OECs PLGA组的各项再生功能指标均优于其它三组。结论:OECs复合PLGA导管能够促进再生神经的成熟和靶组织功能的恢复,二者联合移植是一种有效的周围神经缺损修复方法。  相似文献

采用硅管套接大鼠切断的坐骨神经模型 ,局部给予胶质细胞源性神经营养因子 (GDNF) ,应用尼氏染色、酶组织化学染色方法 ,观察到外源性GDNF能减少脊髓修复侧前角运动神经元死亡的数目 ,降低脊髓前角运动神经元及脊神经节感觉神经元中胆碱酯酶 (CHE)及酸性磷酸酶 (ACP)变化的幅度。这表明外源性GDNF能保护周围神经切断后引起的神经元损伤。  相似文献

观察谷氨酸转运体抑制剂苏-羟天冬氨酸(Threo-hydroxyaspartate,THA)对器官型培养的脊髓片的影响,探讨谷氨酸在运动神经元损伤中的作用。取出生后8天乳鼠的腰段脊髓组织切片做脊髓器官型培养,在培养液中加入不同浓度THA(50μmol/L、100μmol/L、500μmol/L),用神经元的特异性免疫组化染色剂SMI-32,非磷酸化神经丝标记物,对脊髓腹角α运动神经元进行鉴定,用单克隆抗钙网膜蛋白(calretinin)抗体对背角中间神经元进行记数,测定培养液中乳酸脱氨酶(LDH)的含量,并与对照组比较。结果显示对照组α运动神经元数目恒定,THA可以引起剂量依赖性的培养液中LDH含量增高和α运动神经元数目减少,而脊髓背角的中间神经元损伤相对较轻,其中THA100μmol/L组在体外培养4周后出现类似于肌萎缩侧索硬化(ALS)的病理改变:α运动神经元数目较对照组明显减少,而脊髓背角的中间神经元数目无显著变化。细胞外谷氨酸增高主要对运动神经元造成损伤,脊髓运动神经元较感觉神经元对谷氨酸的兴奋毒作用更加敏感。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.
