本研究用昆明种小鼠十二指肠上段提取的α-葡萄糖苷酶,以4-硝基苯-α-D-吡喃葡萄糖苷为反应底物,建立α-葡萄糖苷酶抑制剂高通量筛选模型。用该模型对1276株放线菌和132株细菌等共计2224个发酵液粗提物样品进行筛选。初筛得到抑制率超过50%的阳性样品23个,初筛阳性率为1.03%。将阳性菌株再次进行发酵提取,得到新发酵液的阳性样品数为17,复筛阳性率为0.74%。其中细菌3株,放线菌13株。3个阳性样品经α-葡萄糖苷酶验证后的抑制率均在75%以上。本研究建立的α-糖苷酶抑制剂高通量筛选模型具有很强的实用价值,可用于新型糖尿病药物的开发。  相似文献

黄曲霉毒素B1降解菌的筛选及在玉米贮藏中的应用   总被引:2,自引:0,他引:2  
【目的】本研究分离筛选出一株对黄曲霉既有抑制作用又能降解其毒素的拮抗细菌菌株,并将其应用于玉米中的黄曲霉污染防治研究。【方法】试验通过平板筛选法结合玉米活体筛选法对黄曲霉毒素B1 (AFB1)的拮抗细菌进行初筛,以AFB1的降解率和抑制率为指标进行复筛。【结果】分离到的菌株对黄曲霉菌的抑制率为79.20%,对1μg/mL的黄曲霉毒素B1的降解率为68.39%。贮藏期玉米含水量在15%–30%时,该菌株对黄曲霉污染的抑制率与玉米含水量成反比,即玉米含水量在15%时其抑制率达92.46%,玉米含水量在30%时其抑制率为19.41%。玉米含水量在28%时,菌株对黄曲霉污染玉米的防治效果达到36.39%。【结论】筛选出的拮抗菌株为枯草芽孢杆菌,该菌株不仅对黄曲霉菌有抑制作用,而且能减少AFB1对玉米的污染。  相似文献

一株具有抑制单胺氧化酶作用的干酪乳杆菌筛选   总被引:3,自引:0,他引:3  
【目的】通过体外模型从健康人体粪便内分离筛选出具有抑制单胺氧化酶(MAO)活性的乳酸菌,为今后乳酸菌体内抗衰老的研究提供参考。【方法】采用单胺氧化酶体外抑制模型对乳酸菌的发酵上清及无细胞提取物进行了筛选,并对筛选出的样品进行了两种指标的测定,即样品的剂量效应,以及样品与酶的预保温时间对酶活抑制率的影响;同时利用膜分离技术对不同分子量范围的样品进行了MAO的抑制测定。以筛选出的菌株JH-23为目的菌,通过16S rDNA序列分析及API细菌鉴定系统对菌株进行鉴定。【结果】筛选出的菌株JH-23无细胞提取物对MAO的抑制率达到33.7%。样品经冻干后,在反应浓度为16 mg/mL时抑制率达到53.2%,且MAO抑制率随预保温时间的增加而上升,在30 min之后抑制效果趋于平稳;粗样品经48 h透析后,透析液中的MAO抑制率较透析前明显升高。菌株JH-23的鉴定结果显示其属于干酪乳杆菌。【结论】开发了一种以单胺氧化酶作为靶位酶的新式体外筛选模型,该模型方便快捷且灵敏性高,对之后的抗衰老体内研究有所帮助。筛选出的干酪乳杆菌JH-23细胞裂解物对MAO有抑制作用,其中起到MAO抑制作用的主要是细胞内的小分子类物质。  相似文献

两株生防芽孢细菌筛选、鉴定及拮抗研究   总被引:1,自引:0,他引:1  
【目的】筛选出广谱、高效的生防芽孢细菌,并对其拮抗作用进行研究。【方法】以8种植物病原真菌为靶标菌,通过皿内拮抗和发酵液拮抗能力的测定筛选出2株广谱性和高效性的芽孢细菌B06和B07。【结果】B06对8种植物病原真菌的R2/R1为0.4-1.8,无菌滤液对8种植物病原真菌的抑制率为66.7%-87.5%。B07对8种植物病原真菌的R2/R1为0.23-1.21,无菌滤液对8种植物病原真菌的抑制率为55.56%-81.25%。经16S rRNA序列鉴定,菌株B06和B07都被鉴定为解淀粉芽孢杆菌。【结论】芽孢细菌能够抑制多种植物病原真菌,具有较好的抑病作用。广谱和高效芽孢细菌的筛选在农业生物防治方面具有很大的开发和应用价值。  相似文献

本研究采用胰脂肪酶抑制剂筛选模型结合淀粉酶、蛋白酶抑制活性的测定,对来源于土壤的微生物进行高通量筛选,得到三株胰脂肪酶抑制剂产生菌FIM\|1013、FIL\|19和FIMe\|3,其发酵产物对胰脂肪酶活性的抑制率分别为61%、53%和42%,对淀粉酶活性的抑制率分别为0、7%和3%,对蛋白酶活性的抑制率分别为6%、4%和0。结果显示,筛选所得的三个菌株均能产生具有选择性的胰脂肪酶抑制剂,值得进一步的研究。  相似文献

Sec途径(分泌途径,Secretion pathway)是蛋白质转运的主要途径。其中,SecA ATPase是蛋白质转运途径中的"动力泵",它通过ATP的水解循环驱使蛋白质前体穿过细菌内膜。SecA蛋白在细菌中是独有且不可缺少的。克隆和高效表达绿脓杆菌PasecAN75蛋白(绿脓杆菌SecA蛋白N端645个氨基酸残基组成的片段,大小约75 kD)并优化其ATPase酶活测定体系,在此基础上建立了更为灵敏的SecA蛋白ATPase活性抑制剂的筛选模型。运用该模型从化合物库的3220个样品中筛选得到可抑制绿脓杆菌SecA ATP酶的活性阳性化合物4个,从7196个微生物发酵液中得到66个阳性样品,筛选阳性率为0.67%(以抑制率大于30%为筛选阳性标准)。而后通过已建立的细胞水平筛选模型对其抗菌活性进行验证。研究结果表明3个化合物样品和6个发酵液样品在酶水平和细胞水平对绿脓杆菌SecA ATPase均有较好的抑制作用,值得进一步研究。  相似文献

目的:研究内生细菌B10对水稻稻瘟病菌的抑制作用,为菌株的应用提供理论依据。方法:采用菌丝生长速率法、孢子萌发法和田间试验测定内生细菌发酵上清液和菌液对稻瘟病菌的抑制作用。结果:内生细菌B10发酵上清液对稻瘟病菌菌丝生长和孢子萌发有较强的抑制作用,100倍稀释液对菌丝生长的抑制率为79.37%,对孢子萌发的抑制率为63.42%,对稻瘟病的田间防治效果达70.2%以上。结论:内生细菌B10对稻瘟病有较强的抑菌作用。  相似文献

谷子(Setaria italica(L.)P.Beauv.)是我国重要的杂粮作物,具有抗旱耐贫瘠、水分利用率高、适应性广、营养丰富等特点,但谷子田间除草一直制约着谷子的集约化栽培和有机旱作产业规模化的推广与发展。为快速选育抗除草剂谷子品种,本试验以晋谷21突变体M2群体为试验材料,通过喷施0.33%拿捕净除草剂对晋谷21突变群体进行初次筛选和再次筛选,进而快速获得抗拿捕净除草剂突变体植株并对其进行分子鉴定。结果显示,在大田苗期喷施0.33%拿捕净除草剂的初次筛选中,筛选出抗拿捕净除草剂的晋谷21突变体共39个株系;对这39个突变体株系再次喷施0.33%拿捕净除草剂,筛选获得9个有抗拿捕净除草剂特性的株系。对筛选获得的抗拿捕净突变体株系的其中3个株系共15个植株进行分子鉴定,发现有4个突变体植株的ACCase基因编码的第1780个氨基酸-异亮氨酸突变为亮氨酸。该结果可为今后利用分子生物学手段改良名优谷子品种晋谷21、加速抗除草剂谷子品种的选育提供理论和材料基础。  相似文献

采用微量筛选模型考察48种中药提取物的乙酰胆碱酯酶抑制活性。采用不同极性溶剂对中药材进行提取,采用微量筛选模型对不同浓度提取物的乙酰胆碱酯酶抑制活性进行考察,并采用Origin8.0计算其IC50值。结果表明,几种中药材的不同极性溶剂(水、80%乙醇、乙酸乙酯、石油醚)提取物有不同程度的乙酰胆碱酯酶抑制活性。其中黄柏、元胡的80%乙醇提取物,丹参的乙酸乙酯提取物的抑制率较高。当上述提取物在实验体系中的终浓度为76.92 mg/L时,抑制率分别为66.72±0.02%、52.67±0.04%、46.84±0.03%,其IC50值分别为295.12、229.09、1202.26 mg/L。本研究结果为从中草药中快速筛选乙酰胆碱酯酶抑制剂提供了方法和数据支持。  相似文献

【目的】分离并鉴定具有较好除草活性的植物病原真菌菌株,进一步分离活性化合物,为开发新型生物源除草剂奠定一定的基础。【方法】采用固体培养基接种法分离纯化植物病原真菌,通过形态学特征观察和5.8S rDNA测序鉴定目标菌株;在活性筛选追踪下,对活性组分进行追踪分离及纯化,经波谱分析确定活性单体化合物结构;采用培养皿生物分析法测定活性单体化合物的除草活性及对常见作物的安全性。【结果】茶叶致病菌CY-H具有较好的除草活性,其发酵液对稗草和反枝苋根的抑制率分别为94.6%和77.3%。CY-H被鉴定为间座壳属菌(Diaporthe sp.)。从CY-H菌中分离得到单体化合物CY1被鉴定为cytosporaphenones C。在供试浓度为100μg/mL时,CY1具有较好的抑制反枝苋根的活性,抑制率为57.1%,且其对小麦和油菜的安全性较好,抑制率均在20%左右。【结论】茶叶致病菌CY-H具有开发成微生物源除草剂的潜力。  相似文献

采用离体和活体试验方法分别测定了黄花草木樨不同溶剂提取物对12种植物病原真菌的抑菌活性.结果表明:各溶剂提取物对12种植物病原真菌均具有不同程度的抑菌活性,其中以乙酸乙酯提取物的抑菌活性最高,对油菜菌核病菌、玉米大斑病菌和白菜黑斑病菌抑制菌丝生长的EC50分别为0.62、0.83、0.64g/L,对稻瘟病菌和玉米大斑病菌抑制孢子萌发的EC50分别为0.67、0.97g/L.离体组织法测定表明其乙酸乙酯提取物对番茄灰霉病菌具有较高的保护和治疗作用,在浓度为5.0g/L时,防治效果分别为75.41%和59.18%(6d).活体试验表明乙酸乙酯提取物对小麦白粉病和小麦条锈病也有一定的保护作用,在浓度为10.0g/L时,防治效果分别为73.39%和63.27%.  相似文献

Aedes aegypti control is crucial to reducing dengue fever. Aedes aegypti larvae have developed resistance to organophosporous insecticides and the use of natural larvicides may help manage larval resistance by increasing elements in insecticide rotation programs. Here, we report on larvicidal activity of Moringa oleifera flower extract against A. aegypti L(1), L(2), L(3), and L(4) as well as the effect of flower extract on gut trypsin and whole-larval acetylcholinesterase from L(4.) In addition, the heated flower extract was investigated for larvicidal activity against L(4) and effect on larval gut trypsin. Moringa oleifera flower extract contains a proteinaceous trypsin inhibitor (M. oleifera flower trypsin inhibitor, MoFTI), triterpene (β-amyrin), sterol (β-sitosterol) as well as flavonoids (kaempferol and quercetin). Larvicidal activity was detected against L(2), L(3), and L(4) (LC(50) of 1.72%, 1.67%, and 0.92%, respectively). Flower extract inhibited L(4) gut trypsin (MoFTI K(i) = 0.6 nM) and did not affect acetylcholinesterase activity. In vivo assay showed that gut trypsin activity from L(4) treated with M. oleifera flower extract decreased over time (0-1,440 min) and was strongly inhibited (98.6%) after 310 min incubation; acetylcholinesterase activity was not affected. Thermal treatment resulted in a loss of trypsin inhibitor and larvicidal activities, supporting the hypothesis that flower extract contains a proteinaceous trypsin inhibitor that may be responsible for the deleterious effects on larval mortality.  相似文献

Jirds (Meriones unguiculatus) were immunized with irradiated (35 krad) stage-3 larvae (L3) of Acanthocheilonema viteae. The induced resistance against homologous challenge infection and the antibody response of the animals were studied. Immunization with 3, 2, or 1 dose of 50 irradiated L3 induced approximately 90% resistance. Immunization with a single dose of only 5 irradiated L3 resulted in 60.8% protection while immunization with a single dose of 25 L3 induced 94.1% protection. The protection induced with 3 doses of 50 irradiated L3 did not decrease significantly during a period of 6 months. Sera of a proportion, but not all resistant jirds, contained antibodies against the surface of vector derived L3 as defined by IFAT. No surface antigens of microfilariae or adult worms were recognized by the sera. Vaccinated animals had antibody responses against antigens in the inner organs of L3 and in the cuticle and reproductive organs of adult worms as shown by IFAT. Immunoblotting with SDS-PAGE-separated L3 antigens and L3-CSN revealed that all sera contained antibodies against two exported antigens of 205 and 68 kDa, and against a nonexported antigen of 18 kDa. The 205-kDa antigen easily degraded into fragments of 165, 140, 125, and 105 kDa which were recognized by resistant jird sera. Various antigens of adult worms, but relatively few antigens of microfilariae, were also recognized. To test the relevance of exported antigens of L3 to resistance, jirds were immunized with L3-CSN together with a mild adjuvant. This immunization induced 67.7% resistance against challenge infection and sera of the immunized animals recognized the 205- and 68-kDa antigens of L3.  相似文献

Samples (98 plaque and 72 saliva) from 93 patients with dental caries were investigated for Lactobacillus species which comprised 65 (62.5%) of 104 isolates. Yeasts (20.1%), Streptococcus spp. (8.7%), Staphylococcus spp. (2.9%) and a few unidentified species (5.8%), were also found. The Lactobacillus isolates were L. brevis (24.6%) L. fermentum (18.5%) L. casei (16.9%), L. delbrueckii (15.4%), L. plantarum (9.23%), L. acidophilus (7.69%), L. jensenii (4.62%), L. salivarius (1.54%) and L. gasseri (1.54%). The most common species was L. brevis (24.6%). The strains tested for beta-lactamase production showed 75.4% positive. All the Lactobacillus strains were tested for bacteriocin production against Escherichia coli, Salmonella spp., Shigella dysenteriae, S. sonnei, Klebsiella spp. and Campylobacter sp. All the lactobacilli except L. jensenii produced bacteriocin against at least one of the indicator organisms. The involvement of Lactobacillus in dental caries was established, although its role and mechanism is not well understood. The ability of Lactobacillus spp. to protect their host against certain diseases by inhibiting the growth of potential pathogens was evident.  相似文献

To understand the factors that influence selection for anthelmintic resistance, it is necessary to examine the impact of drug treatment, particularly persistent drugs, on all phases of the worm life cycle. The efficacy of various avermectin/milbemycin anthelmintics was determined against resident worms, incoming larvae (L3) and development of eggs in faecal culture. Homozygote-resistant and maternal and paternal F1-heterozygote genotypes of Haemonchus contortus were used to infect sheep before or after treatment with ivermectin (IVM) oral, IVM capsule, moxidectin (MOX) oral or MOX injectable. Total worm count and quantitative larval culture were used to determine efficacy against parasitic and free-living stages, respectively. Selection for resistance by IVM capsules occurred at the adult and L3 stages because of poor efficacy against these stages for all resistant genotypes. However, the selective advantage of these surviving worms was reduced due to the low development of their eggs to L3 in faecal culture. For MOX, selection for resistance predominantly occurred after treatment because of high efficacy against resident adult worms of all resistant genotypes but poor efficacy against resistant L3 ingested after drug administration. The results indicated no evidence of sex-linked inheritance for IVM resistance. Mean IVM efficacies against homozygous and heterozygous resistant adult worms were not different, and IVM capsule efficacy against incoming L3 was approximately 70% for all resistant genotypes, consistent with a dominant trait. MOX was highly effective against adults of all resistant genotypes and approximately 76% effective against incoming L3 regardless of resistance genotype, also consistent with a dominant trait. These results will enable the impact of persistent drugs on worm control and anthelmintic resistance to be estimated. The results indicate that IVM capsules should not be used in populations where avermectin/milbemycin resistance is present.  相似文献

从华凤仙(Impatiens chinensis L.)、问荆(Equisetum arvense L.)和轮叶狐尾藻(Myriophyllum verticillatum L.) 3种植物中共分离到内生真菌155株。经鉴定, 它们分属于26个不同的分类单元。以杨桃炭疽菌(Colletotrichum gloeosporioides)、香蕉疫霉(Phytophthora nicotianae)和腐皮镰刀菌(Fusarium solani)等6种植物病原真菌为指示菌, 对3种水生植物内生真菌代谢产物进行抗菌活性研究。结果表明, 有37株内生真菌(占分离菌株总数的23.9%)显示出对一种或多种病原菌的抑菌活性。来自华凤仙、问荆和轮叶狐尾藻的抗菌活性菌株比例分别为29.2%、13.9%及37.1%, 都远低于已报道的陆生植物内生真菌中活性菌株的比例。抗菌活性菌株主要分布于枝孢属(Cladosporium)、木霉属(Trichoderma)和地霉属(Geotrichum)等5个属中。  相似文献

Litsea salicifolia is one of the many plants used as phytopesticide, traditionally by various tribes of Assam. Of the five extracts of L. salicifolia tested for the bioactivity against Aedes aegypti and Culex quinquefasciatus, the aqueous extract was more effective compared to other extracts exhibiting bioactivity at 72 ppm against A. aegypti. The hexane extract (2000 ppm) exhibited 70% repellent activity for 3 hr against A. aegypti and 46% activity for 3 hr against C. quinquefasciatus. This is the first report on the insecticidal properties of L. salicifolia which can be further developed as an eco-friendly biopesticides of the future.  相似文献

Lactococcus garvieae and Aeromonas hydrophila are bacterial pathogens affecting salmonids and other fish species and cause of heavy losses in aquaculture. Diseases caused by these bacteria can be controlled satisfactory by immunization using monovalent vaccines. In this study, the protective efficacy of two bivalent vaccines against L. garvieae and A. hydrophila was evaluated in rainbow trout (Oncorhynchus mykiss). Bivalent formulations, containing formalin-inactivated bacteria, were prepared as an aqueous bacterin and as an adjuvanted vaccine using montanide ISA-763. Protection against L. garvieae and A. hydrophila was tested at day 30 and 90 post-vaccination. High levels of protection were achieved for the aqueous and adjuvanted bivalent vaccines against L. garvieae (RPS of 100% and 95.3%) and A. hydrophila (RPS of 100% and 95.3%) at day 30 post-vaccination. Significant differences (p < 0.05) were found between the RPS at days 30 and 90 post-immunization with a decrease in the protection levels for the aqueous bivalent vaccine against L. garvieae (RPS 76.2%) and A. hydrophila (RPS 85%), but not for the adjuvanted vaccine (RPS of 90% against L. garvieae and 95% against A. hydrophila). In addition, high antibody levels were observed in the vaccinated fish at day 15 post-immunization using both vaccines. Our results demonstrate that these bivalent vaccines can effectively protect rainbow trout against L. garvieae and A. hydrophila and could offer an appropriate strategy to prevent these infections in rainbow trout farms.  相似文献

从土壤中分离筛选出1株编号为Z40的链霉菌菌株,其代谢产物具有明显的除草活性。室内生物测定结果表明,Z40菌株发酵液干物质的除草活性主要表现为抑制生长作用。对反枝苋的主根和主茎生长抑制中浓度(EC50)分别为276.76 mg/L和1 609.37 mg/L,对狗尾草的主根和主茎生长抑制中浓度分别为501.81mg/L和629.01 mg/L。浓度为10 000 mg/L对油菜的萌发抑制率和主茎生长抑制率均为100.00%,而对小麦的生长发育没有明显影响。田间试验结果表明,Z40菌株代谢产物对稗、狗尾草、铁苋菜的防治效果均在80%以上,抑制率分别为80.30%,81.59%和88.71%。  相似文献

美洲斑潜蝇是严重危害瓜、豆类蔬菜的世界性害虫之一,主要以幼虫潜伏在叶表皮下的叶肉组织中为害,给防治带来很大困难。本研究测定了葫芦素B对美洲斑潜蝇成虫产卵和取食的抑制作用。在非选择性试验中,当葫芦素B使用浓度为500~2000 mg/L时,对美洲斑潜蝇成虫的产卵抑制率达31.2~69.9%,对美洲斑潜蝇成虫的取食抑制率达61.7~64.9%;在选择性试验中,葫芦素B使用浓度达1000~2000 mg/L时,对美洲斑潜蝇成虫的产卵抑制率和取食抑制率分别为54.8~71.1%和57.2~65.2%;研究结果表明:利用葫芦素B可有效地抑制美洲斑潜蝇成虫的产卵和取食,减轻其幼虫的为害,为安全、有效地控制美洲斑潜蝇提供了一条新途径。  相似文献
