以马铃薯品种"夏波蒂"为材料,设保水剂穴施、涂层、拌种处理和对照4个处理,分析了保水剂不同施用方式对马铃薯生长及产量的影响。结果表明:施用保水剂有利于提高马铃薯的出苗率和成苗率,以穴施处理的出苗率和成苗率最高,较对照分别提高22%和4%;株高和株幅随生育时期的推进逐渐增大,进入淀粉积累期时,穴施处理的株高显著高于其他处理,比对照提高44%;穴施处理各时期的叶绿素含量最高,维持了植株高水平的光合速率;马铃薯叶片中脯氨酸含量随生育时期的推进逐渐增加,进入淀粉积累期时,对照的脯氨酸含量明显低于拌种处理,而显著高于穴施和涂层处理;各处理叶片中可溶性糖含量先增加后降低,并逐渐向薯块中转移;植株鲜质量、干质量均随生育时期的推进逐渐增加,块茎形成期后增加较快,进入淀粉积累期时,施用保水剂处理的生物量均显著高于对照;穴施处理产量最高,较对照提高57%,其次是涂层处理;施用保水剂处理均利于旱作马铃薯增产。  相似文献

以‘云薯505’马铃薯(Solanum tuberosum ‘Yunshu 505’)为材料,测定马铃薯块茎发育初期四个阶段茉莉酸含量,并以叶面喷施方式,研究茉莉酸甲酯对马铃薯生长和块茎产量的影响。结果表明,马铃薯块茎膨大过程中,茉莉酸的积累水平不断升高。在微型薯生产中,使用100 μmol·L-1茉莉酸甲酯在结薯期以不同频率喷施叶面,测量并统计植株、块茎性状及产量变化。结果表明,与对照(CK)相比,1次·d-1处理茎粗增加36.1%,2次·d-1处理的叶绿素含量降低20.1%。此外,植株的叶色、茎色、花色、株型等生长性状及块茎大小整齐度、薯形、皮色、肉色、薯皮类型、芽眼深浅、裂薯率、大薯空心率等块茎性状在各组间没有显著差异。2次·d-1、1次·d-1、1次·2d-1、CK四种处理的植株存活率分别为45.57%、100.00%、100.00%、87.29%;前三种喷施频率处理折合产量较CK分别增加-15.61%、8.77%、12.11%。综合分析,马铃薯在块茎形成初期茉莉酸积累水平不断升高,以1次·2d-1频率叶面喷施100 μmol·L-1茉莉酸甲酯,马铃薯微型薯的产量增加最大且不影响生长。  相似文献

高温对马铃薯块茎形成期光合及抗氧化特性的影响   总被引:1,自引:0,他引:1  
块茎形成期高温是影响宁南山区马铃薯产量和品质的重要因素。本研究通过连续2年试验,选用当地主栽品种"青薯9号",设置自然温度(对照)、低温、高温3个处理,研究马铃薯块茎形成初期、中期、后期、块茎膨大期及主要淀粉积累期的光合特性和抗氧化酶活性。结果表明:高温降低了马铃薯功能叶片的净光合速率(P_n)、荧光综合指标(P_I)、光能供应化学反应的最大效率(F_v/F_m)及潜在光化学活性(F_v/F_o); P_n在马铃薯块茎形成后期高温较自然温度降低34.55%,较低温处理降低53.15%,PI在块茎形成后期高温较低温降低了42.22%;高温处理的根系活力、超氧化物歧化酶(SOD)及过氧化物酶(POD)活性也有所降低,丙二醛(MDA)、脯氨酸(Pro)和过氧化氢酶(CAT)均有不同程度的升高;相关性分析表明,除抗氧化系统中的CAT活性、MDA和Pro含量与产量呈负相关外,其余指标与产量呈正相关,与根系活力的相关性最为显著;马铃薯块茎形成中期高温天气不利于马铃薯功能叶片的光合作用,降低了P_n、P_I等光合指标,使功能叶片的抗氧化系统遭到一定的破坏,导致产量下降。  相似文献

遮荫对掌叶半夏生长、产量及光合生理特性的影响研究   总被引:2,自引:0,他引:2  
目的:研究遮荫对掌叶半夏生长、药材产量和光合生理指标的影响,分析适宜掌叶半夏药材高产的遮荫度,探寻其产量差异形成的光合生理机制。方法:采用遮荫网遮荫,共设置了5种光强处理,透光率分别为全光对照CK(100%)、T1(75%)、T2(65%)、T3(50%)、T4(25%),测定各遮荫处理下掌叶半夏的株高、单株叶片数、单株叶面积、单片叶面积、药材产量、叶绿素含量、光合日进程和叶绿素荧光参数。结果:(1)遮荫可显著(P<0.01)提高掌叶半夏的株高、单株叶面积和叶绿素含量;(2)光合日进程曲线测定结果表明,除T4处理外其他处理均发生光合"午休"现象,日平均光合速率(Pn)以T1处理最高,绝对光能利用率(Eu)以T3处理最高;(3)叶绿素荧光参数测定结果表明,CK的Fv/Fm最低,发生的原因可能与PSⅡ反应中心的光化学伤害有关;(4)药材产量以T2处理最高,较CK增产56.34%。结论:掌叶半夏更适宜在遮荫条件下栽培,透光率在65%时有利于掌叶半夏药材高产,其产量差异的形成可能与日均光合速率的不均衡有关。  相似文献

以‘早大白’马铃薯脱毒苗为试验材料,通过人工温室苗床栽培探讨外源激素萘乙酸(NAA)、吲哚丁酸(IBA)和硼酸生根处理对扦插脱毒苗生长和生理特征的影响,为马铃薯微型薯的实际生产提供理论依据。结果显示:(1)外源激素生根处理较对照扦插苗根系长势好,根活力、根系可溶性蛋白含量增加。(2)外源激素生根处理较对照脱毒扦插苗光合速率提高、叶绿素含量增加,光合物质的形成与积累增多。(3)外源激素生根处理较对照叶片SOD、POD、CAT活性和游离氨基酸含量均表现出不同程度的上升,MDA含量下降,衰老减缓。(4)外源激素生根处理有利于小区产量和单株结薯数的增加。研究表明,不同外源激素生根处理可改善脱毒马铃薯扦插苗农艺性状和生理指标,并以NAA 100mg/L+IBA 50mg/L+硼酸17.5mg/L配方处理植株的长势最好,叶绿素含量、保护酶活性及游离氨基酸含量最高,净光合速率大、小薯膨大速度快且单株结薯数量及产量增加显著,更利于发挥脱毒薯的增产优势。  相似文献

甘肃省中部沿黄灌区是全国重要的加工型马铃薯生产基地,然而因集约化生产带来的连作障碍问题已经严重影响到当地马铃薯种植业的健康发展。结合田间试验和相关的室内分析,从马铃薯块茎产量和品质、植株生理特征和土壤真菌群落结构等角度,初步评估土壤灭菌和生物有机肥联用(Ammonia Disinfection plus Bio-organic Fertilizer Regulation,ABR)对马铃薯连作障碍的防控效果。同对照相比,ABR处理的块茎产量和商品薯率分别显著增加约71.1%—152.1%和39.2%—53.3%,但块茎化学品质变化不大。ABR处理叶绿素含量和根系活力较CK均显著增加,而叶片和根系丙二醛含量显著下降。PCR-DGGE分析发现,ABR处理显著影响了马铃薯连作土壤的真菌群落结构,表现为真菌群落的多样性指数较CK相比显著下降。ABR处理还有效抑制了土传病害的滋生,植株发病率和收获后的病薯率较CK分别显著下降约67.2%—82.2%和69.1%—70.5%。采用Real-time PCR评估连作土壤中3种优势致病真菌的数量变化,显示ABR处理下立枯丝核菌、茄病镰刀菌和接骨木镰刀菌的数量在生育期内较CK均有不同程度的下降。综合来看,土壤灭菌和生物有机肥联用技术在防控甘肃省中部沿黄灌区马铃薯连作障碍上具有较大的应用潜力,而对土传病害的抑制和微生物群落结构的改善是其主要的作用机理。  相似文献

为筛选出抗旱效果明显的保水剂,提高甜菜成苗率和产量,采用温室结合田间试验的方法,研究了不同保水剂对甜菜植株生长和产量的影响。结果表明:4种保水剂中以TM-3005KM保水剂处理效果最佳;TM-3005KM保水剂较CK定植后幼苗成活率高7.22%;苗期末期,TM-3005KM保水剂处理的株高、根粗、冠幅、叶长和叶宽值均最大,且显著高于CK,根鲜重、根干重、茎叶鲜重和茎叶干重较CK分别高27.83%、38.46%、65.21%和59.15%;各处理甜菜叶片叶绿素含量均呈单峰曲线,并于叶丛快速增长期达到峰值,各生育期施用保水剂各处理的叶绿素含量均显著高于CK,且以TM-3005KM保水剂处理含量值最高;各处理甜菜叶片光合速率变化与叶绿素含量基本一致;各处理甜菜叶片脯氨酸含量随生育时期的推进,表现出升高→降低→升高→降低的趋势,各生育期均以TM-3005KM保水剂处理含量最低,CK最高;TM-3005KM保水剂处理可溶性糖从地上部叶片向块根中转移的相对较早,且其含量变化相对稳定;收获时,TM-3005KM保水剂处理较CK生物产量、经济产量和折光糖度分别提高50.08%、47.53%和2.10%。研究表明,TM-3005KM保水剂处理有利于提高甜菜幼苗成活率,促进幼苗快速生长,增强植株抵御干旱的能力,为叶绿素含量积累、光合速率增大和产量的提高奠定良好的基础。  相似文献

马铃薯叶片光合作用是影响其块茎产量和品质的重要因素之一。为了选择适宜湖南栽培的马铃薯品种和提高马铃薯的栽培效益,本文时中薯3号、大西洋、费乌瑞它、东农303、PB04、PB08和滇薯6号7个马铃薯品种(系)在匍匐茎伸长期、块茎形成期、块茎膨大期和块茎完熟期4个时期的光合特征参数(叶片净光合速率、气孔导度、蒸腾速率、胞间CO_2浓度)、SPAD值和相对产量进行了比较研究。结果表明,中薯3号的叶片净光合速率、气孔导度、胞间CO_2浓度、蒸腾速率和平均每株产量都最高,适宜于在湖南栽培。  相似文献

以降香黄檀(Dalbergia odorifera T. Chen)为材料,分析不同移植方式和外施钾肥等培育措施对其光合参数和叶绿素(Chl)含量的影响。结果显示,断根处理后植物的最大净光合速率(Pnmax)比去冠、全冠移、对照(CK)和去冠移4种处理方式分别提高了19.25%、34.79%、40.88%和219.86%。光合参数分析结果显示,光饱和点(LSP)最高的为去冠处理,光补偿点(LCP)最高的为断根处理。断根处理后植物的净光合速率(Pn)和蒸腾速率(Tr)最大。断根、去冠、全冠移3种处理的Chla、Chlb、Chl(a+b)含量及胡萝卜素均高于CK,且断根处理后上述各指标达到最高值。外施钾肥处理中,降香黄檀最大净光合速率随钾肥用量的增加而增加,K2处理下的LSP值最大。随着钾肥施用量的增加,植物叶片的Pn、气孔导度呈上升的趋势,且植株的Chla、Chlb和Chl(a+b)含量均随钾肥用量的增加而增加,CK处理下Chla/Chlb比值显著高于K1、K2处理。研究结果表明断根、去冠、施用钾肥等处理均可显著提升降香黄檀叶片的叶绿素含量,有利于植株进行光合作用,促进生长发育。  相似文献

通过大田试验,研究了沟垄集雨种植结合不同补灌量处理对冬小麦光合器官、光合速率、产量和水分利用效率的影响.结果表明:沟垄宽度各为60 cm时,集雨种植不灌溉(T1)、返青期种植沟补灌375 m3·hm-2(T2)和种植沟补灌750 m3·hm-2(T3)3个处理较平作灌水750 m3·hm-2(畦灌,T4)处理的小麦籽粒产量分别提高2.8%、9.6%和18.9%,收获系数提高2.0%~ 8.5%,旗叶叶绿素含量提高41.9% ~64.4%,整个生育期内0~ 40 cm土壤含水量增加了0.1%~4.6%;开花期和灌浆期的叶片光合速率分别较T4处理提高了22.3%~ 54.2%和-4.3%~67.2%,农田总水分利用效率较T4处理分别提高17.9%、10.4%和15.4%,比平作不灌水处理(CK)提高69.3%、58.6%和65.7%;降水利用效率较CK提高94.3% ~ 124.5%;T2、T3处理各生育阶段叶面积均显著高于T4处理,灌溉水利用效率分别比T4处理提高119.1%和18.8%.在灌溉量减少50%的条件下,集雨种植比畦灌处理能维持较高的籽粒产量,显著提高灌溉水利用效率,尤其是在降雨量偏少的年份,可以显著提高小麦水分利用效率.  相似文献

目的:克隆壳聚糖酶基因于大肠杆菌中实现高表达,制备壳寡糖。方法:以枯草芽孢杆菌总DNA为模板扩增壳聚糖酶基因(CSN),克隆至载体pET23a(+)上,转化菌株BL21(DE3)。重组子经0.5 mmol/L IPTG诱导后,SDS-PAGE和质谱检测与鉴定重组酶。酶纯化后水解壳聚糖,薄层色谱分析其水解产物。结果:质谱证明壳聚糖酶(31.5kDa)成功表达,表达量占菌体总蛋白的45%左右。纯化后重组酶浓度为900 mg/L,纯度95%、回收率85%,酶活力为10 000 U/mg。壳聚糖降解产物为壳二糖至壳四糖。结论:原核表达载体pET23a(+)-CSN构建正确,壳聚糖酶表达量与活性高,适用于水解壳聚糖制备壳寡糖。  相似文献

研究放牧对草地植物生理活动的影响,对于揭示草地放牧演替的生理机制有重要意义.大量研究表明,家畜放牧对牧草光合作用、呼吸作用以及C和N吸收与转运的影响,可以分为生理伤害和生理恢复2个阶段.放牧通过改变草地冠层结构影响牧草光合作用,净光合作用速率短期内迅速下降,随着叶面积指数增加又逐渐上升,呼吸作用有相似的变化趋势.牧草放牧后再生长所需的C和N最初主要来自根系和留茬中的贮藏物质,此后随着牧草生长恢复逐渐由同化作用供给,C代谢与土壤N水平负相关.放牧后牧草生理活动变化与牧草遗传特性、种间竞争、家畜放牧特征、非生物环境等因素密切相关.  相似文献

N -substituted phenothiazines (PTs) and phenoxazines (POs) catalyzed by fungal Coprinus cinereus peroxidase and Polyporus pinsitus laccase were investigated at pH 4–10. In the case of peroxidase, an apparent bimolecular rate constant (expressed as k cat/K m) varied from 1 ×107 M−1 s−1to 2.6×108 M−1 s−1 at pH 7.0. The constants for PO oxidation were higher in comparison to PT. pH dependence revealed two or three ionizable groups with pK a values of 4.9–5.7 and 7.7–9.7 that significantly affected the activity of peroxidase. Single-turnover experiments showed that the limiting step of PT oxidation was reduction of compound II and second-order rate constants were obtained which were consistent with the constants at steady-state conditions. Laccase-catalyzed PT and PO oxidation rates were lower; apparent bimolecular rate constants varied from 1.8×105 M−1 s−1 to 2.0×107 M−1 s−1 at pH 5.3. PO constants were higher in comparison to PT, as was the case with peroxidase. The dependence of the apparent bimolecular constants of compound II or copper type 1 reduction, in the case of peroxidase or laccase, respectively, was analyzed in the framework of the Marcus outer-sphere electron-transfer theory. Peroxidase-catalyzed reactions with PT, as well as PO, fitted the same hyperbolic dependence with a maximal oxidation rate of 1.6×108 M−1 s−1 and a reorganization energy of 0.30 eV. The nanomechanical properties of the coiled-coils of myosin are fundamentally important in understanding muscle assembly and contraction. Force spectra of single molecules of double-headed myosin, single-headed myosin, and coiled-coil tail fragments were acquired with an atomic force microscope and displayed characteristic triphasic force-distance responses to stretch: a rise phase (R) and a plateau phase (P) and an exponential phase (E). The R and P phases arise mainly from the stretching of the coiled-coils, with the hinge region being the main contributor to the rise phase at low force. Only the E phase was analyzable by the worm-like chain model of polymer elasticity. Restrained molecular mechanics simulations on an existing x-ray structure of scallop S2 yielded force spectra with either two or three phases, depending on the mode of stretch. It revealed that coiled-coil chains separate completely near the end of the P phase and the stretching of the unfolded chains gives rise to the E phase. Extensive conformational searching yielded a P phase force near 40 pN that agreed well with the experimental value. We suggest that the flexible and elastic S2 region, particularly the hinge region, may undergo force-induced unfolding and extend reversibly during actomyosin powerstroke.  相似文献

以白术(Atractylodes macrooephala Koidz.)二倍体组培苗为材料,对其四倍体诱导方法进行研究,共获得45个白术同源四倍体株系,为优良株系的选育提供了材料。此外,还分析比较了其中8个白术四倍体株系与二倍体的过氧化物酶同工酶(POD)的酶谱差异,发现四倍体各株系过氧化物酶同工酶谱比二倍体的均多了Rf0.310的谱带,且总过氧化物酶比活力也发生了很大改变,对探讨白术四倍体优良株系的生理生化机理具有一定的参考价值。  相似文献

3-O-Carboxymethylcoumestrol was prepared as the hapten for immunoassay by a partial alkylation of coumestrol with ethyl chloroacetate in acetone alkalized with potassium carbonate. 3-O-Ethoxycarbonylmethylcoumestrol was separated by column chromatography and finally was hydrolyzed with formic acid. 1H and 13C NMR data (APT, COSY, HMQC, and HMBC) revealed that the reaction was regioselective, as 3-O-ethoxycarboxymethylcoumestrol was the only monosubstituted derivative. The hapten was then conjugated to bovine serum albumin and used for immunization of rabbits. A radioimmunoassay (RIA) system was established based on the polyclonal antiserum and a 125I-labeled hapten-tyrosine methyl ester conjugate as the radioligand. Parameters of the RIA: sensitivity: 12 pg per tube, 50% intercept: 140 pg per tube, working range: 20-4000 pg per tube. The cross-reactivity of a panel isoflavonoid and lignan phytoestrogens was either negligible (e.g. formononetin 0.07%; biochanin A 0.06%) or not detectable at all. The major immunoreactive peak in HPLC fractions from an alfalfa extract had the same retention time as coumestrol standard and represented 94.8% of the signal. The remaining 5.2% of immunoreactivity was distributed between five minor peaks. We conclude that after the validation for particular matrices, the method will be a useful tool for analysis of coumestrol, especially in low volume and low concentration samples.  相似文献

Among tetrapods, evidence for postrenal modification of the urine by the distal digestive tract (including the colon and cloaca) is highly variable. Birds and bladderless reptiles are of interest because the colon and cloaca represent the only sites from which water and ions can be reclaimed from the urine secreted by the kidney. For animals occupying desiccating environments (e.g., deserts and marine environments), postrenal modification of the urine may directly contribute to the maintenance of hypo‐osmotic body fluids. We compared the morphology and distribution of key proteins in the colon, cloaca, and urogenital ducts of watersnakes from marine (Nerodia clarkii clarkii) and freshwater (Nerodia fasciata) habitats. Specifically, we examined the epithelia of each tissue for evidence of mucus production by examining the distribution of mucopolysaccharides, and for evidence of water/ion regulation by examining the distribution of Na+/K+‐ATPase (NKA), Na+/K+/Cl? cotransporter (NKCC), and aquaporin 3 (AQP3). NKCC localized to the basolateral epithelium of the colon, urodeal sphincter, and proctodeum, consistent with a role in secretion of Na+, Cl?, and K+ from the tissue, but NKA was not detected in the colon or any compartment of the cloaca. Interestingly, NKA was detected in the basolateral epithelium of the ureters, suggesting the urothelium may play a role in active ion transport. AQP3 was detected in the ureters and coprodeal complex, consistent with a role in urinary and fecal dehydration or, potentially, in the production of the watery component of the mucus secreted by the coprodeal complex. Since no differences in general cloacal morphology, production of mucus, or the distribution of ion transporters/water channels were detected between the two species, cloacal osmoregulation may either be regulated by proteins not examined in this study or may not be responsible for the differential success of N. c. clarkii and N. fasciata in marine habitats.
