Pokemon基因即POK红系髓性致癌因子,也被称为FBI-1,LRF,OCZF,TIP,它是POK转录抑制物家族成员之一,其编码的产物具有BTB/POZ域和锌指结构,它是第一个被发现的可变阅读框基因ARF(Altemative reading frame,ARF)的特异性转录抑制物,Pokemon通过特异抑制ARF转录来调控细胞周期、诱导肿瘤发生、促进肿瘤的发生发展,在一些人类肿瘤如食管鳞癌、肺癌、结肠癌、人贲门癌、膀胱癌,前列腺癌和乳腺癌等肿瘤组织中具有较高的表达水平。Pokemon的特殊性在于还能与其它癌基因如Bcl-2,MDM2等结合之后发挥作用来促进其他癌基因的活性。因此可以认为Pokemon是肿瘤的总开关,对Pokemon基因的进一步研究有助于为肿瘤的诊断和治疗提供新的途径和方法。  相似文献

垂雄瘤转化基因1(PTTG1),也被称为分离酶抑制蛋白基因,是近几年从大鼠垂体肿瘤中发现的癌基因。它不但可以与分离酶结合,使分离酶失活,从而抑制姐妹染色单体的分离,还具有转录激活活性。已有的染色质免疫共沉淀结合芯片数据显示,PTTG1不仅可以直接调控基因的转录,也可以与其他蛋白,如PTTG1结合因子(PBF)、p53、Spl、上游刺激因子1(USF1)等相互作用来调控下游基因的转录。在NIH3T3细胞中,PTTG1激活c-Mvc的转录,增强NIH3T3细胞在裸鼠体内的成瘤能力。PTTG1也能激活肿瘤细胞中成纤维细胞生长因子2(FGF2)的转录,从而促进肿瘤血管生成。PTTG1结合p53、抑制p21表达、激活周期蛋白D3的能力,提示它在凋亡、细胞周期和衰老方面廿.发挥作用。另外,PTTG1在肿瘤转移和肝癌的发生发展中也发挥着重要作用。我们简要综述了PTTG1的靶基因,及其在肝癌及肿瘤转祷中的研究进展。  相似文献

生长素响应因子(Auxin response factor, ARF)作为一类调控生长素响应基因表达的转录因子, 是生长素研究的重要内容。它可与生长素响应基因启动子区域内的生长素响应元件结合, 促进或抑制基因的表达。文章介绍了植物体内ARF家族的分子生物学近年来的研究进展, 同时也讨论了ARF转录因子的结构、ARF基因的表达调控、ARF在植物生长发育及信号转导中的作用以及ARF对靶基因的调控机制等内容。植物ARF成员都有一定的同源性, 大多含有4个结构域, 在多种组织和器官中都有表达, 其表达受到转录及转录后调控, 并且在介导生长素与其它激素之间相互作用方面扮演重要角色。  相似文献

C/EBPα基因是抑癌基因,有抑制增殖、促进分化和调节DNA损伤反应等作用。各种机制如对抗性的蛋白质-蛋白质相互作用、基因突变和翻译后修饰等导致C/EBPα转录抑制或活性丧失,可能诱发肿瘤。本文结合国内外研究现状,介绍C/EBPα基因的结构、功能及与肿瘤的诊断、治疗和预后的关系。  相似文献

NPR1蛋白是水杨酸信号和系统获得性抗性的转录调节因子,它的功能受蛋白质降解酶体CUL3.E3的控制。植物的发育主要受生长素信号通路的控制,生长素反应因(ARF)参与生长素信号转导转录调控。植物转录因于NPR1和ARF8分别在蛋白质降解酶体CUL3-E3与CUL1-E3控制下,调控抗病防卫与生长发育。烟草TTG2促进ARF8从细胞质向细胞核转运及其转录调控作用,因此促进生长发育;相反,TTG2把NPR1扣留在细胞质,阻止它对防卫反应基因的转录调控作用,从而抑制抗病性。TTG2与NPR1或ARF8并不直接互作,说明存在协助因子。  相似文献

泛素蛋白连接酶MDM2(Murine double minute 2)具有癌基因活性, MDM2高表达会导致抑癌基因p53失活而诱发肿瘤, 但在至少7%的肿瘤中p53基因正常而mdm2异常扩增, 表明MDM2还具有其他底物分子, 以p53不依赖的方式促进肿瘤的发生。鉴于MDM2的重要作用, 文章在基因水平、转录水平、翻译后修饰水平、相互作用分子的调节等方面系统总结了目前对MDM2调控的主要研究机制及其进展。  相似文献

转录因子BACH1在哺乳动物组织中广泛表达,参与调控血红素稳态和氧化应激等生理过程。BACH1不仅在转录水平调控HMGA2、CXCR4、MMPs、RKIP、HOXB8、HO-1、CXCR3-B和TKT等基因,还通过高甲基化修饰等表观遗传调控机制使特定抑癌基因转录沉默,从而促进肿瘤发生发展。近年研究发现,BACH1参与肿瘤代谢重编程,促进肿瘤血管和淋巴管的新生和重构,从而增强肿瘤细胞的侵袭转移。该文综述了近年BACH1在肿瘤中的研究进展。  相似文献

目的:探讨食管鳞癌(esophageal squamous cell cacinoma,ESCC)组织中Pokemon和P14ARF表达的关系及其对ESCC临床病理特征和预后的判定价值。方法:应用SP免疫组织化学方法检测Pokemon和P14ARF二种蛋白在96例ESCC组织及20例癌旁正常组织中的表达,分析它们与ESCC病理学特征的关系,以及Pokemon和P14ARF的相关性,并结合随访资料观察上述蛋白表达对ESCC长期预后的影响。结果:癌旁正常组织中未见Pokemon蛋白表达,P14ARF蛋白阳性表达率为90.0%;在ESCC组织中Pokemon和P14ARF阳性表达率分别为75.0%和30.2%。Pokemon表达与P14ARF表达呈负相关(r=-0.458,P=0.000)。Pokemon和P14ARF的表达与TNM分期和淋巴结转移相关(P<0.05)。Pokemon表达阳性组的5年生存率分别为20.8%,显著低于阴性组(P=0.004);P14ARF表达阳性组的5年生存率为41.4%,显著高于阴性组(P=0.011)。结论:Pokemon蛋白的高表达和P14ARF蛋白低表达可能与ESCC的发生、发展关系密切,它们对于ESCC预后评估有一定的临床意义。  相似文献

胶质瘤相关癌基因蛋白(glioma-associated oncogene1,Gli)是Hedgehog(Hh)信号通路的转录因子,定位于细胞核和细胞浆,将信号传送至核内。脊椎动物中已鉴定出3个成员,分别为Gli 1、Gli2和Gli3,该蛋白家族成员只有在维持全长时才具有转录激活子的功能,羧基端被蛋白酶体水解后,就形成了转录抑制子。近年来,Gli与肿瘤的关系日益受到人们的重视,以前普遍认为的Gli目的基因的调控和Gli蛋白的转录后修饰是通过Hh通路实现受到挑战,越来越多的研究证明有许多非经典机制可以不通过Hh通路来调节Gli目的基因的表达。Gli研究将有助于我们对肿瘤的认知和治疗。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献
