用响应面法对Burkholderiasp.SYBCLIP—Y液体发酵产低温脂肪酶的发酵条件进行了快速优化。首先利用Plackett—Burman设计对影响其产酶相关因素进行评估并筛选出具有显著效应的三个因素:牛肉膏,橄榄油,TritonX-100;用最陡爬坡路径逼近最大产酶区域后,利用响应面中心组合设计对显著因素进行优化,确定出牛肉膏,橄榄油,TritonX-100的最佳浓度分别为:牛肉膏31.8g/L、橄榄油21mL/L、TritonX-10036.55mL/L,优化后脂肪酶的酶活达到61.52U/mL,是优化前的2.62倍。  相似文献

应用响应面法对Galactomyces geotrichumY25液体发酵产脂肪酶的条件进行了优化。首先采用Plackett-Burman设计对影响产酶因素的效应进行评价,筛选出黄豆粉、玉米浆和发酵时间3个对产酶影响显著的因素。用最陡爬坡路径逼近最大产酶区域后,利用响应面设计对显著因素进行优化,得出黄豆粉、玉米浆最佳质量分数分别为2.51%、2.12%,最佳发酵时间101.95 h。优化后液体发酵液中脂肪酶活力提高到34.65 U/mL,比初始酶活力9.6 U/mL提高了3.61倍。表明响应面法可显著优化Galactomyces geotrichumY25液体发酵产脂肪酶条件。  相似文献

利用响应面法对超声提取绿茶荼多酚的工艺条件进行优化,在单因素试验的基础上,根据中心组合设计原理采用三因素三水平的响应面分析法,依据回归分析确定最优提取工艺条件。结果表明,其最佳工艺条件为:液料比为40.2mL/g,超声功率为476W,提取时间为15.1min,采用该工艺条件,茶多酚的提取得率达到10.312%,通过响应面法得到一个能较好预测试验结果的模型方程。  相似文献

本文探讨超声波作用下太子参多糖提取的最佳工艺条件。在单因素试验基础上,采用响应面法对太子参多糖提取工艺参数进行优化研究。响应面试验表明提取温度、提取时间和水料比对响应值多糖提取得率均有显著影响,优化得到太子参多糖超声提取最佳工艺条件为:提取温度为74℃;提取时间为65 min;水料比为26 mL/g;超声波功率100 W。在此条件下太子参多糖的提取得率为2.48%,与模型预测值非常接近。  相似文献

利用响应面法对小球藻(Chlorella vulgaris)在2L气升式生物反应器中对自养产生物柴油油脂的培养条件进行了优化。首先用Plackett-Burman方法对10个相关影响因素的效应进行评价并筛选出对产油有显著影响的3个因素:KNO3浓度、温度和CO2浓度;再用最陡爬坡实验逼近最大产油区域;最后由中心组合实验及响应面分析确定了影响产油主要因素的最佳条件为:KNO3浓度0.31g/L,温度26.5℃,CO2浓度6.80%,最高产油量达到0.42g/L,比优化前提高了近2倍。优化后,在10L气升式生物反应器中进行了扩大培养。  相似文献

采用响应面分析法对菊粉乙醇发酵条件进行优化,首先采用Plackett-Burman设计对影响菊粉乙醇发酵的7个因素进行筛选,获得影响最大的3个因素菊粉浓度、装液量和发酵时间.再利用响应面分析法对这3个因素进行优化,确定最佳发酵条件为菊粉浓度160.0g/L、装液量130mL、发酵时间84h,采用优化后的发酵条件进行发酵,乙醇浓度达到7.5%,取得了较好的优化效果.  相似文献

研究采用响应面法优化黑曲霉固态发酵生产单宁酶的培养条件。应用Plackett—Burman试验筛选出重要影响因子:五倍子粉含量、(NH4)2SO4浓度以及接种孢子量,最陡爬坡试验逼近最大响应区域。应用Box.Behnken响应面试验对重要影响因子进一步优化。得到最佳培养条件:每250mL三角瓶中装入1.0g五倍子粉、4.4g稻壳和0.5g麸皮、液固比(mL/g)2:1且营养盐溶液组成为(NH4)2s0421g/L、MgSO4·7H2O1g/L、NaCl1g/L,培养基pH自然,接种5.7×10^7个孢子后在30℃温度下培养4d。在此条件下,单宁酶产量从40U/g提高到114U/g,3次重复验证性试验平均值为115U/g,验证了模型的可靠性。  相似文献

利用Design Expert软件中的两水平实验设计和响应面法,对发酵生产S-酰胺酶(可用于拆分2,2-二甲基环丙甲酰胺外消旋体)的培养基进行了优化。采用Plackett-Burman(PB)设计对培养基中相关影响因素的效应进行评价并筛选出了有显著效应的葡萄糖、酵母粉及2,2-二甲基环丙甲酰胺浓度,其他因素对酰胺酶产量的影响不显著。然后用旋转中心组和实验设计及响应面分析确定了主要影响因素的最佳条件,在优化的培养基中,酰胺酶产量达到168 U/L,比优化前的80 U/L提高了110.0%。  相似文献

用响应面法对蜂房哈夫尼菌(Hafnia alvei)L-赖氨酸脱羧酶产酶诱导条件进行优化。首先通过单因素实验对产酶体系的pH、震荡培养时间、静置培养时间、诱导物添加量和Ⅷ添加量进行优化。在此基础上,用部分因子重复试验筛选出对酶活影响显著的3个因素(静置培养时间,诱导物添加量,VB6添加量),再通过Box-behnken实验对这三个因素进行优化,得出最优值。最终得到产酶最佳诱导条件为:震荡培养阶段培养基pH6.5,静置培养阶段pH5.5;摇床震荡培养11h后静置培养7.5h,诱导物L一赖氨酸加入量为5.18dL,维生素B6加入量为1.38g/L时酶活最高,达到71.2U/mL,为优化前(1.74u/mL)的41.8倍,在单因素的基础上提高了19%。  相似文献

采用单因素和效应面法对石榴皮抑菌物提取条件进行优化,并对结果进行验证。结果发现:最佳提取温度为59℃、时间为73 min、液料比为40∶1(mL/g)。实验证明石榴皮提取物对枯草杆菌具有很强的抑制作用,且所用工艺简单易行,说明效应面优化法建立的数学模型预测结果可靠。  相似文献

用响应面方法对Lactococcus lactis生产细菌素乳链菌肽的培养基进行了优化。首先用部分重复因子实验对培养基组份蔗糖,大豆蛋白胨,酵母粉,KH2PO4,NaCl,MgSO4·7H2O对乳链菌肽的影响进行评价,并找出主要影响因子为大豆蛋白胨和磷酸二氢钾,前者为负影响,后者为正效应,其它组份对乳链菌肽产量的影响不显著。第二步用最陡爬坡路径逼近最大响应区域。最后用中心组合设计及响应面分析确定主要影响因子的最佳浓度。菌株在优化培养基中的乳链菌肽产量增加1倍为2150(IU/mL)。动力学分析表明,菌株生长与细菌素的产生为部分耦联型,进入对数中期菌体比生长速率和细菌素比产率在优化培养基中均大于优化前培养基。  相似文献

研究了头孢霉(Cephalosporium sp.)菌丝体最大生产力和多不饱和脂肪酸形成积累的条件。菌丝体最适培养条件为:麦芽糖60g/L\,KNO33g/L、起始pH为60、500mL三角瓶装100mL培养基、接种25%、25℃培养10d则菌丝体达到最大干重。多不饱和脂肪酸形成积累的最适条件为:葡萄糖10~20g/L、(NH4)2SO4或NH4Cl 3g/L、培养基起始pH为40、500mL三角瓶装100mL培养基、接种10%~20%、10℃下照光培养。〖JP2〗因此,在整个生产流程中可采用不同条件分段掌握的技术原则。同时提出在多不饱和脂肪酸的形成和积累途径中油酸(18∶1)向亚油酸(18∶2)的转化是关键,为进一步探索最适培养条件和关键酶的调节提供依据。  相似文献

In order to accelerate biodegradation of feather into more amino acids, the fermentation medium of feather-biodegrading Streptomyces fradiae Var S-221 was optimized in this paper. In the first optimization step, the effects of feather powder, beet molasses, (NH4)2SO4 and KH2PO4 on amino acids formation were evaluated by using full factorial design. The results showed that feather powder and (NH4)2SO4 had significant and positive effects on feather-biodegradation into amino acids. Then, the method of the steepest ascent was used to access the optimal region of the two significant factors. In the third step, the concentration of feather powder and (NH4)2SO4 were further optimized with central composite design and response surface analysis. As a result, the composition of the optimal medium for S. fradiae Var S-221 fermentation were as follows (g/100 ml): feather powder, 19.504; beet molasses, 4.0; (NH4)2SO4, 1.467; KH2PO4, 0.3; MgSO4, 0.15; FeSO4, 0.001; ZnSO4, 0.0001; and MnSO4, 0.0001. Using this optimal fermentation medium, the amino acids concentration was increased from 4.61 to 6.13 g/100 ml.  相似文献

Rhizopus chinensis CCTCC M201021 was a versatile strain capable of producing whole-cell lipase with synthetic activity in submerged fermentation. In order to improve the production of whole-cell lipase and study the culture conditions systematically, the combination of taguchi method and response surface methodology was performed. Taguchi method was used for the initial optimization, and eight factors viz., maltose, olive oil, peptone, K2HPO4, agitation, inoculum size, fermentation volume and pH were selected for this study. The whole-cell lipase activity yield was two times higher than the control experiment under initial optimal conditions, and four significant factors (inoculum, olive oil, fermentation volume and peptone) were selected to test the effect on the lipase production using response surface methodology. The optimal fermentation parameters for enhanced whole-cell lipase yield were found to be: inoculum 4.25 x 10(8) spores/L, olive oil 2.367% (w/v), fermentation volume 18 mL/250 mL flask, peptone 4.06% (w/v). Subsequent experimental trails confirmed the validity of the model. These optimal culture conditions in the shake flask led to a lipase yield of 13875 U/L, which 120% increased compare with the non-optimized conditions.  相似文献

Enhanced growth of salmonellas in Rappaport's medium as modified by Vassiliadis et al. (1976) after pre-enrichment in buffered peptone water during the first 6 h was obtained by replacement of tryptone by soya peptone. The competing bacteria, i.e. those which grow on brilliant green agar and which may interfere with the isolation of salmonellas when Rappaport's medium (R10) incubated at 43°C is used for enrichment were inhibited or reduced in numbers when the normal amount of 5 g soya peptone/litre was used. When the amount was increased to 10 g/l, growth occurred, mainly of Enterobacter and Klebsiella species. The isolation of salmonellas was found to be largely dependent on the number and the ratio of their competitors. Every measure taken to reduce the number of competitive bacteria increases the possibility of isolating salmonellas. This explains the effect of improved selectivity of Rappaport's medium when small inocula are used. Rapid onset of growth of salmonellas by employing soya peptone introduces the possibility of using shorter incubation times than 48 h.  相似文献

采用Plackett-Burman(PB)设计和响应面分析(RSM)方法, 对一株海洋侧孢短芽孢杆菌Lh-1产抗菌肽R-1的发酵条件进行了优化。研究中数据的统计和分析均使用MINITAB 15.0。在研究中, 首先使用了Plackett-Burman (PB)设计对影响Lh-1产抗菌肽的15个因素进行了筛选, 得到了影响产量的显著因素为:葡萄糖、蛋白胨和氯化钙。在此基础上采用响应面法对该3个显著因素的最佳水平范围进行研究, 得到的最佳浓度为 15.72 g/L 葡萄糖、6.01 g/L 蛋白胨和3.29  相似文献

In this study, Prevotella ruminicola 23 (ATCC 19189), a ruminal proteolytic bacterium, was used as protease producer to examine the optimal condition for protease production. The best carbon and nitrogen sources for the maximum growth were glucose with peptone. Both sucrose and glucose could stimulate high protease production. Casein and peptone are better nitrogen sources for protease production than other choice in this study. The best enzyme production condition was 18-20 h incubation which was at late log phase in the broth of 5% glucose or sucrose as carbon source with 0.1% ammonium chloride and 0.2% peptone as nitrogen sources. Most of the protease activity was secreted into broth (65%) and on cell surface (18%). The optimal temperature and pH for protease reaction were 40 degrees C and pH 6.8, respectively. After incubation for 6h, the crude extract maintained 50% of original protease activity at 30 and 50 degrees C, and protease activity was stable between pH 6 and 8. The protease inhibitor test showed that serine, aspartic acid and metallo-protease inhibitors could cause inhibition of proteolysis. Protein feedstuff degradation experiments suggested that protease in crude extract had higher degradation ability on fish meal, whey, and feather meal (2.39, 2.60 and 1.76 micromol aminoacid/mg enzyme/h) in comparison to soybean meal and blood meal (1.11 and 1.09 micromol aminoacid/mg enzyme/h). The protease in the crude extract should have application potential in term of improving utilization of fish meal and feather meal for monogastric animals.  相似文献
