采集全国各地红曲霉制品中的红曲霉资源,经分离获278株红曲霉菌株。高效液相色谱(HPLC)法测定各菌株发酵液中莫纳可林K(Monacolin K)和桔霉素含量,筛选获1株Monacolin K产量较高、桔霉素含量较低的红曲霉菌株编号为M-22。依据形态特征和ITS基因序列,参照红曲霉属分类检索表,鉴定M-22菌株为紫色红曲霉(Monascus purpureus)。通过摇瓶发酵对温度、初始pH、碳源、氮源、碳氮比(C/N)等因素进行优化,确定M-22菌株摇瓶发酵产Monacolin K适宜条件为发酵温度26℃、初始pH 5.0、转速160 r/min,甘油为碳源、蛋白胨为氮源、碳氮比(C/N)为5:1,Monacolin K产量显著提高,最高为107.16 mg/L。以优化的发酵条件对M-22菌株进行5 L发酵罐发酵,发酵液中Monacolin K产量最高为189.83 mg/L,桔霉素含量32.53μg/L,红曲色素色价为16.38 U/m L。  相似文献

【背景】洛伐他汀(lovastatin)是红曲霉的次生代谢产物,是重要的临床用降血脂药物。在液态发酵条件下,红曲霉的洛伐他汀产量较低,难以满足工业化生产的要求。【目的】筛选获得一株高产洛伐他汀的红曲霉株,并通过优化液态发酵条件提高洛伐他汀的产量。【方法】从红曲米中筛选获得一株高产洛伐他汀的红曲霉株,依据形态学特征、生理生化特性及18S rRNA基因序列分析对分离菌株进行鉴定;通过响应面法对其产洛伐他汀的液态发酵条件进行优化。【结果】获得一株产洛伐他汀的紫红曲霉(Monascus purpureus M4),该菌在甘油57.80g/L、酵母浸粉5.52 g/L、接种量为6.90%条件下,洛伐他汀产量(173.60 mg/L)较优化前提高了4.8倍。【结论】菌株M4产洛伐他汀最优液态发酵条件的建立,为洛伐他汀的大规模生产及该菌株的工业化应用提供了技术支撑。  相似文献

从180余份海水、海泥样品中筛选得到60株产海藻糖较高的菌株,编号为2-14的菌株海藻糖产量最高,为127.9mg/g cell。对2-14菌株进行形态特征、培养特征及生理生化试验,鉴定该菌株为红酵母属(Rhodotorula sp.)。研究摇瓶发酵条件对红酵母海藻糖产量的影响,结果为:初始pH5.5,发酵温度28℃,装液量75mL(250mL三角瓶中)。采用优化后发酵条件红酵母海藻糖产量为193.3mg/g cell,优化前对照值为132.1mg/g cell,优化后的结果是优化前的1.46倍。在5L发酵罐中培养得到最佳发酵时间为54h,发酵罐培养发酵液中海藻糖含量最高达2.5g/L,为摇瓶培养的1.6倍。  相似文献

对不同样品中的真菌进行了L-精氨酸高产菌株的筛选,从红曲米中分离出1株高产L-精氨酸菌株红曲霉H13,其初始发酵液中L-精氨酸产量为1.67 g/L.通过单因素实验对其液体发酵进行研究.结果表明,最佳碳源是可溶性淀粉,最佳浓度为10%,最佳氮源是蛋白胨,最佳浓度为3%,最佳pH值4.5,温度30℃,种子培养时间为18 h,摇瓶装液量50 mL/250 mL.转速为120 r/min,发酵时间7d.在此条件下进行发酵培养,所得发酵液产L-精氨酸4.08 g/L,菌丝体干重为10.32 g/L,菌丝体中L-精氨酸含量为24.17 mg/g.  相似文献

从不同生境收集的自然发酵样品中分离纯化获86株曲霉纯菌株;用薄层层析法(TLC)和高效液相色谱法(HPLC)测定曲霉各菌株发酵液中洛伐他汀(Lovastatin)含量进行筛选;获1株较高产Lovastatin的编号为A-8曲霉菌株,产量为58 g/L。根据形态特征和ITS基因序列,鉴定A-8菌株为杂色曲霉。单因素实验初步优化了A-8曲霉菌株发酵条件和培养基配方,结果表明A-8菌株产生Lovastatin较适宜的发酵条件为:温度28℃、初始pH 5.2、摇床转速180 r/min、接种量为10%;培养基配方为:乳糖为碳源、含量为100 g/L,蛋白胨为氮源、含量为12 g/L,碳源/氮源为15∶1.5;在该条件下A-8曲霉菌株发酵产生Lovastatin的水平最高,可达130.04μg/mL,比优化前提高了约2.24倍。A-8曲霉菌株是较有潜力的工业菌株。  相似文献

以红曲米中筛选到的红色红曲霉菌株G为原始菌株,通过农杆菌介导T-DNA插入突变技术,成功构建了含有1 483株红曲霉突变子的T-DNA插入突变库。用HPLC等方法从突变库中筛选出10株γ-氨基丁酸(GABA)产量稳定高于原始菌株G的突变子,薄层层析结合HPLC技术分析了10株突变子发酵液中桔霉素的含量;其中突变子1047的GABA产量较高,为1.169g/L,是原始菌株G(GABA产量0.472g/L)的247.67%,桔霉素含量稳定低于原始菌株G。以红曲霉菌株G和突变子1047为实验材料,通过5 L发酵罐发酵并定时取样,HPLC等方法精确分析发酵液各种活性物质的含量;结果显示,突变子1047生长速度稍快于原始菌株G;GABA、莫纳可林K(Monacolin K)、红曲色素色价分别为2.201 g/L、83.892 mg/L、21.984 U/mL,是原始菌株G的279.67%、108.01%和182.35%;而桔霉素产量为1.976 mg/L,是原始菌株G的41.71%。因此,利用TDNA插入的方法对红曲霉进行育种,能产生稳定的遗传变化,在红曲霉资源的保护和利用上有一定潜力。  相似文献

为获得高产MonacolinK的红曲菌菌株,将经农杆菌介导转化获得的携带潮霉素抗性基因且以甘油为原料液态发酵高产MonacolinK的发白红曲菌H2和以大米为原料固态发酵产Monaco-1inK的烟色红曲菌9908作为亲本,对其原生质体分别进行热灭活及紫外灭活,然后对灭活双亲用PEG作融合剂进行原生质体融合。从融合子中选出有潮霉素抗性的突变株,通过发酵与亲本对比,筛选得到一株以大米为原料固态发酵高产MonacolinK的融合株F12.11,其MonacolinK产量达到8.73mg/g;较发白红曲菌H2与烟色红曲菌9908分别提高了100.23%和48.98%;一株以甘油为原料液态发酵高产MonacolinK的融合株F13-2,其MonacolinK的产量达到1752.46mg/L,较发白红曲菌H2与烟色红曲菌9908分别提高了32.98%和1979.33%。  相似文献

高产洛伐他汀棒曲霉菌株的筛选、鉴定及发酵条件优化   总被引:1,自引:0,他引:1  
从不同生境(食品、土壤、空气、有机质等)收集到的自然发酵样品中分离得到150株曲霉属菌株.用高效液相色谱(HPLC)法检测发酵液中洛伐他汀(Lovastatin)含量,筛选获得1株稳定高产Lovastatin的曲霉菌株(编号:Ac-32).根据菌落形态特征并结合18 S rDNA测序,鉴定其为棒曲霉(Aspergillus clavatus).通过摇瓶发酵单因素实验优化了碳氮源种类、碳氮源含量、碳氮比(C/N)、发酵温度、初始pH、转速、种龄和接种量,确定了棒曲霉菌株Ac-32摇瓶发酵产Lovastatin的适宜条件为:乳糖为碳源、蛋白胨为氮源、碳源含量为100 g/L、氮源含量为12 g/L、碳氮比(C/N)为15:1.8、温度28℃、转速180 r/min、初始pH 5.2、种龄4 d、接种量6%.采用Minitab 17软件的P-B实验设计法,筛选对Lovastatin产量有显著影响的因素为:温度、pH、碳源含量和氮源含量.根据P-B实验结果,运用响应面法分析,确定棒曲霉菌株Ac-32产Lovastatin的最优条件为:碳源含量100 g/L,氮源含量11.8 g/L,温度28℃,pH 5.2.在此条件下,Lovastatin最高产量为236.221μg/mL.  相似文献

谷氨酸棒杆菌可发酵葡萄糖产L-鸟氨酸。通过连续筛选与驯化筛选到了产量较高的生产菌株CG1006。以GC/MS对发酵液成分的定性分析及鸟氨酸的产量为依据,优化了培养条件,产量从最初的14.346g/L提高到40.416g/L。初步考察了酵母菌CG1006-1对发酵液成分及产量的影响。结果表明混合菌种发酵可纯化发酵液成分。  相似文献

从数株红酵母中选出了1株产类胡萝卜素能力较强的红酵母RY-98(生物量,类胡萝卜素含量和产量分别为19.9g/L,334.8ug/g和6.7mg/L);研究了该菌株产类胡萝卜素的最适营养与环境条件,获得了最佳的发酵生理学条件;葡萄糖40g/L,(NH4)2SO4 10g/L,酵母膏3g/L,蕃茄汁2mL/L,花生油0.5mL/L,接种量30mLL初始pH6.0和通气量(培养基装置040ml/250mL:,在此初步优化的培养条件下,红酵母RY-98经72小时摇瓶发酵其生物量,类胡萝卜素含量和产量分别可达26.8g/L,386.9ug/g和10.4mg/L,依次比初筛中提高了34.7%,15.6%,和55.2%。  相似文献

将单因素实验结果与响应面法相结合,对高产Monacolin K的紫色红曲霉Mp-24菌株进行发酵工艺条件优化。通过摇瓶发酵对碳源、氮源、碳源含量、氮源含量、培养时间等进行单因素优化,确定Mp-24菌株摇瓶发酵适宜条件:乳糖为碳源、酵母膏为氮源、碳源含量7%、氮源含量2%、培养时间12 d,Monacolin K产量为167 mg/L。应用Box-Behnken中心组合试验设计建立数学模型,进行响应面分析优化发酵条件,结果显示最佳发酵工艺条件为:碳源(乳糖)8%,氮源(酵母膏)3%,培养时间11 d,在此条件下Monacolin K的含量达到247.8 mg/L,比优化前提高1.5倍。  相似文献

利用紫外线和氯化锂对一株紫红曲Monascus purpureus菌株进行复合诱变试验。在最佳紫外照射时间为45s和最佳氯化锂浓度为1.0‰的情况下,获得了一株MonacolinK高产突变菌株Monascus purpureus ZT32且连续传接5代产量稳定,用紫外分光光度法检测其MonacolinK的含量为219.9μg/mL,较原始菌株高出2倍多,然后将其用于固态发酵,结果表明:红曲米中的MonacolinK的含量达到8.33mg/g,是原始菌株的3.3倍。  相似文献

In China, Monascus spp., a traditional fungus used in fermentation, is used as a natural food additive. Monascus spp. can produce a secondary metabolite, monacolin K namely, which is proven to be a cholesterol-lowering and hypotensive agent. Hence, recently, many researchers have begun focusing on how to increase the production of monacolin K by Monascus purpureus. In the present study, we investigated the effect of the fungal elicitor and the mutagenesis of UV & LiCl on the amount of monacolin K produced by Monascus purpureus. The fugal elicitor, Sporobolomyces huaxiensis, was isolated from tea leaves and its filtrate was added into the culture filtrate of Monascus purpureus during growth to induct the production of monacolin K. The results showed that the highest amount of monacolin K produced by the liquid fermentation was 446.92 mg/mL, which was produced after the fungal elicitor was added to the culture filtrate of Monascus purpureus on the day 4; this amount was approximately 6 times greater than that of the control culture filtrate, whereas the highest amount of monacolin K produced by the mutated strain was 3 times greater than the control culture after the irradiation of UV light in the presence of 1.0 % LiCl in the medium.  相似文献

对三株在YES培养基中不产桔霉素的红曲霉菌种,在摇瓶中研究了它们液体发酵生产莫纳可林K的情况。在大米粉培养基中,红色红曲霉不产莫纳可林K,但是紫色红曲霉和烟灰色红曲霉均能产莫纳可林K,前者产量高于后者。在葡萄糖.甘油培养基中,后两者的产量均很低,但是如果在该培养基中添加酵母膏,紫色红曲霉能产生较为可观的莫纳可林K。在2L的发酵罐中,利用添加了酵母膏的葡萄糖-甘油培养基,紫色红曲霉在第13d的莫纳可林K产量可达104mg/L,培养过程中无桔霉素产生。  相似文献

Monacolin K is a secondary metabolite of Monascus and is known to decrease cholesterol levels in humans. There are 9 genes (mokA-mokI) controlling its biosynthesis, of which mokH is thought to act as a pathway-specific regulator. In this study, the Monascus purpureus M1 strain was compared with mokH gene deletion strains (△H1) and overexpression strains (H7). The monacolin K yields in the △H1 strain were reduced by 52.05 %, and increased in the H7 strain by 82 %. The mycelium samples of the M1, △H1, and H7 strains were found to vary with scanning electron microscopy. Compared to the M1 strain, some mycelium of the △H1 strain showed obvious folding and expansion, while the mycelium of the H7 strain was fuller. Besides, these results indicate that the mokH gene can increase the yield of monacolin K by regulating the expression level of mokA-mokI genes, and influence the production of Monascus pigment. The study is the first to combine deletion and overexpression techniques to further verify the mokH gene and get the desired results in M. purpureus.  相似文献

Monacolin K (MK) is a naturally occurring hypocholesterolemic agent that specifically inhibits HMG‐CoA reductase. As a natural source of MK, Monascus‐fermented products are of special interest; however, some Monascus strains could produce citrinin, which is a nephrotoxin, as a contaminant in Monascus‐derived products. A Monascus pilosus strain (MS‐1) that produces high amounts of MK, but no citrinin, was screened in previous investigations. Herein, liquid‐state fermentation parameters of the MS‐1 strain were optimized using statistical methods to maximize the MK yield with potato juice as a basic medium. The maximum MK yield (326.74 μg/mL) was predicted with 50 mL of medium in a 250‐mL conical flask containing 30 g/L sucrose, 38.75 g/L soybean flour, 0.00105 mol/L Mg2+ at pH 5.48, and 8% v/v seed inoculum precultured for 42 h at 30°C, incubated at 30°C for 3 days, followed by further incubation for 11 days at 24.7°C. The verified MK yield was 390.68 μg/mL and the MK yield increased to 565.64 μg/mL after 21 days of fermentation. No citrinin was detected in MS‐1‐fermented products. The results suggest that citrinin‐free MK can be obtained from natural medium through liquid‐state fermentation in an economical way. This method will be of practical value to the industrial production of MK.  相似文献

红曲霉(Monascus purpureus)是我国重要的传统食用和药用微生物,能分泌红曲色素、莫纳卡琳(Mo-nacolin K)、γ-氨基丁酸、麦角固醇等物质,被广泛用于生产食品发酵剂、食品着色剂、保健食品和药品等,在我国有上千年的应用历史。本实验以根癌农杆菌(Agrobacterriumtum efaciens)介导的红曲霉突变体库中754株突变子菌株为材料,通过乙醇提取法筛选出了9株产红曲色素发生显著变化的色素突变子,其中S50在505 nm波长处的色价降为原始菌株S的0.12倍,几乎变为白化株。300~600 nm波长下UV-VIS扫描图谱特征显示,色素突变子不仅色价发生了变化,而且吸收波长、吸收峰值也发生了不同程度的变化。最后,对色素突变子菌落形态、显微结构、潮霉素抗性、遗传稳定性以及产桔霉素能力等进行分析,为研究红曲霉产色素相关基因奠定了基础。  相似文献

红曲霉出发菌株经紫外线 (UV)、硫酸二乙酯 (DES)、氯化锂 (LiCl)等诱变剂反复诱变处理 ,获得供试红曲霉菌株 10株。用薄层层析法初筛得到可能产MonacolinK的菌株 4株 ,再经高效液相色谱法对初筛菌株进一步进行检测 ,证实其中 3株具有较强的产MonacolinK能力 ,特别是诱变株M7的含量与出发菌株相比提高了 4 1倍  相似文献
