为获得分泌抗人β-actin蛋白单克隆抗体(McAb)的杂交瘤细胞,通过在大肠杆菌中原核表达人β-actin蛋白,以纯化的人β-actin蛋白作为抗原免疫BALB/c小鼠。经过细胞的融合及筛选获得1株能稳定分泌抗人β-actin蛋白McAb的杂交瘤细胞,命名为2B4。采用间接ELISA和Western blot方法对McAb的特异性、稳定性和适用范围进行鉴定。结果显示:蛋白的相对分子质量为43 kDa,可溶于8 mol/L尿素;杂交瘤细胞上清的抗体效价为1×10^5,腹水的抗体效价为1×10^7;间接ELISA结果表明,杂交瘤细胞在体外传20代或液氮冻存3个月后,分泌的抗体效价不变;37℃保存24 h后,抗体的效价开始下降。Western blot结果显示,单克隆抗体识别人、鼠、兔和鱼的β-actin蛋白,与其发生特异性反应。2B4分泌的单克隆抗体可以广泛的应用于细胞生物学和免疫学试验,具有良好的应用价值。  相似文献

目的:制备重组活化相关分泌蛋白1(ASP-1)的单克隆抗体,并用其鉴定保守结构域。方法:用原核表达并纯化的重组ASP-1不加佐剂免疫BALB/c小鼠,采用杂交瘤技术及有限稀释传代法筛选稳定分泌特异性抗体的杂交瘤细胞株,制备单抗腹水后用间接ELISA进行抗体特异性鉴定和效价检测,利用肽结合ELISA和Western印迹鉴定单抗识别的保守结构域。结果:获得5株能稳定分泌抗ASP-1单克隆抗体的杂交瘤细胞株,且5株单抗的识别区域均为21~28氨基酸残基的保守性结构域。结论:制备了抗ASP-1的单克隆抗体,为深入研究ASP-1佐剂的活性功能区及作用机制提供了有效工具。  相似文献

为了制备可用于胶体金快速检测试纸条的抗登革病毒2型(DEN2)E蛋白单克隆抗体(mAb),通过基因克隆获得E基因与质粒pET32a(+)的重组质粒,将重组质粒转化入大肠杆菌BL21,IPGT诱导表达重组蛋白。以DEN2重组E蛋白免疫BALB/c小鼠,采用杂交瘤技术制备抗DEN2E蛋白的mAb,以间接ELISA法和Western blot进行mAb特异性鉴定;同时采用间接ELISA法鉴定mAb的Ig亚类。结果表明获得1株可分泌特异性mAb的杂交瘤细胞(7C7),其抗体亚类为IgG1。Western blot显示该株mAb能特异识别重组pET32a-DEN2E蛋白。因此,成功制备出抗DEN2E的1株mAb,为建立快速特异橙测登革病毒感染的实验方法提供了有力的工具。  相似文献

目的 制备汉滩病毒(Hantaan virus, HTNV)PS-6株小鼠单克隆抗体(monoclonal antibody, mAb),建立HTNV抗原双抗体夹心ELISA检测方法,验证方法的检测性能并初步应用于疫苗抗原含量检测。方法 以HTNV PS-6株灭活全病毒原液作为免疫原,采用小鼠杂交瘤融合技术,筛选mAb杂交瘤细胞株,制备mAb;用间接ELISA测定mAb效价;用Western blot鉴定mAb特异性;用间接ELISA测定mAb相对亲和力;经抗体配对筛选,建立双抗体夹心ELISA病毒抗原检测方法。以I型肾综合征出血热疫苗标准品作为定量标准,验证该方法的检测限、线性范围、特异性、准确度、精密度;对6批次I型肾综合征出血热灭活疫苗原液进行检测,初步验证该方法的适用性。结果 获得4株稳定分泌抗HTNV PS-6特异性抗体的阳性杂交瘤细胞株:4B2、3H8、5D7及2A7;间接ELISA检测腹水抗体效价均在1×106~1×106~1×107;Western blot鉴定4株mAb均能特异性识别HTNV PS-6;相对亲和力为4B2>5D7>3H8>2A7;抗体ELISA配对筛选后,选用5D7作为包被抗体,4B2作为标记抗体,包被抗体工作浓度为10μg/mL,HRP标记抗体工作浓度为1∶5 000。该方法对HTNV PS-6抗原检测限为0.039 1μg/mL;检测线性范围为0.078 1~2.500 0μg/mL,R7;Western blot鉴定4株mAb均能特异性识别HTNV PS-6;相对亲和力为4B2>5D7>3H8>2A7;抗体ELISA配对筛选后,选用5D7作为包被抗体,4B2作为标记抗体,包被抗体工作浓度为10μg/mL,HRP标记抗体工作浓度为1∶5 000。该方法对HTNV PS-6抗原检测限为0.039 1μg/mL;检测线性范围为0.078 1~2.500 0μg/mL,R2> 0.97;检测与I型肾综合征出血热疫苗生产主要原辅料成分、II型肾综合征出血热疫苗、森林脑炎疫苗及甲肝疫苗均无交叉反应;准确度验证,病毒抗原回收率在95.8%~110.5%之间;试验内和试验间精密度,CV分别在6.72%~8.03%、8.24%~9.70%之间。用该方法检测6批次I型肾综合征出血热灭活疫苗原液,结果均呈剂量依赖性。结论 成功制备HTNV PS-6株mAb,建立病毒抗原双抗体夹心ELISA抗原检测方法,方法准确、可靠,可初步应用于I型肾综合征出血热灭活疫苗科研、生产过程病毒抗原检测。  相似文献

目的克隆表达HBV前S2蛋白,用纯化的重组蛋白GST-S2免疫BALB/C小鼠,通过杂交瘤技术制备抗HBV前S2蛋白的单克隆抗体(mAb),并鉴定其特性.方法利用含双拷贝乙肝病毒adw2亚型基因全序列的质粒pEcob6,经PCR方法扩增出HBV前S2片段,在GST表达系统中表达,表达产物用谷胱甘肽亲和层析纯化后,用于免疫BALB/c小鼠,采用杂交瘤技术制备抗前S2蛋白的mAb,采用间接ELISA方法和Western blot进行mAb特异性鉴定;同时采用间接ELISA方法鉴定mAb相对亲和力.结果获得一株可分泌特异性mAb的杂交瘤细胞7A6;相对亲和力均在107以上.Western blot显示该株mAb能特异识别重组前S2蛋白.结论成功地制备出抗HBV前S2蛋白的一株mAb.  相似文献

目的:采用杂交瘤技术制备抗人B7-H1单克隆抗体,并对其进行鉴定。方法:经抗原免疫的小鼠脾细胞与小鼠骨髓瘤细胞以常规方法融合;用间接ELISA法筛选分泌抗体的杂交瘤细胞株;阳性克隆用有限稀释法获得稳定分泌抗人B7-H1单克隆抗体的杂交瘤细胞株;扩增杂交瘤细胞注射进小鼠腹腔后制备腹水;纯化腹水中的单克隆抗体并对其亚型进行鉴定;用间接ELISA法测抗体效价;将肺癌组织制成石蜡切片,用抗人B7-H1抗体进行免疫组化染色。结果:获得1株稳定分泌抗人B7-H1单克隆抗体的杂交瘤细胞株,所分泌的单抗类型为IgG1;抗体效价为1×108,纯化后的抗体含量为6.76g/L;免疫组化实验中,单抗可与肺癌组织表面的B7-H1蛋白特异地结合。结论:制备了人B7-H1单克隆抗体,为B7-H1检测试剂盒的研制奠定了基础。  相似文献

以单核细胞增生李斯特菌细胞碎片免疫BALB/c小鼠,间接ELISA法成功筛选获得2株稳定分泌抗LM的单克隆杂交瘤细胞株4A7、4H11.抗体效价为1∶160 000以及1∶20 000,亚型为IgG1、IgG2a,Dot-ELISA结果表明4A7和4H11单克隆抗体具有很好的属特异性,Western blot分析表明4A7、4H11抗体分别与单核细胞增生李斯特菌62 kDa以及32 kDa外膜蛋白抗原表位结合,胶体金免疫电镜实验进一步确证以上抗体可有效识别单核细胞增生李斯特菌细胞表面抗原.  相似文献

制备Asia I口蹄疫病毒vp2单克隆抗体(mAb)并建立了单抗竞争ELISA方法。用纯化的Asia I型口蹄疫病毒vp2重组蛋白免疫BALB/c小鼠, 将免疫小鼠的脾细胞与骨髓瘤SP2/0细胞融合, 采用间接ELISA和有限稀释法筛选杂交瘤细胞。分别用ELISA、Western blotting检测mAb腹水的效价及其特异性。筛选到杂交瘤细胞2株, 腹水效价均在100×29以上; 以纯化后的Asia I型口蹄疫病毒vp2重组蛋白作为抗原, 利用Asia I型口蹄疫病毒vp2单抗酶标物建立了竞争ELISA方法用来检测Asia I型口蹄疫抗体。临床应用表明, 该方法与UBI公司的口蹄疫全病毒抗体检测试剂盒总符合率达89.0%, 和荷兰赛迪公司的口蹄疫病毒LPB-ELISA抗体检测试剂盒总符合率达86.5%。  相似文献

制备抗羊口疮病毒118(ORFV118)重组蛋白的单克隆抗体并鉴定其生物学特性。构建ORFV118原核表达重组质粒pET33b-ORFV118,将重组质粒转化大肠杆菌BL21,诱导表达出ORFV118重组蛋白;利用Ni-NTA亲和层析法纯化ORFV118重组蛋白;以纯化蛋白为抗原免疫小鼠,通过杂交瘤技术制备单克隆抗体;利用间接ELISA法、Western blot、免疫组化等方法分别对单抗特异性、效价、亚型以及ORFV118蛋白的功能进行研究。获得了3株稳定分泌单克隆抗体的细胞株,分别命名为1A2,3B5,5D10,它们诱生的小鼠腹水效价分别为110 000,16 400,18 000。其中效价最高的1A2抗体亚型为IgG1,能特异性结合其免疫原蛋白、真核表达产物以及病羊皮肤组织中的ORFV118蛋白。免疫组化结果显示单抗1A2染色局限于皮肤表皮层角化细胞及浸润至皮下组织的炎症细胞,与病毒侵染上皮组织层的特性相符。制备的单抗1A2能特异性识别ORFV118。深入研究单抗1A2将为了解ORFV118的生物学特性,为羊传染性脓疱病的诊断、预防与治疗提供可能和新思路。  相似文献

目的:制备抗心型脂肪酸结合蛋白(H-FABP)单克隆抗体(mAb),并建立侧向免疫层析方法检测血浆中H-FABP。方法:用H-FABP蛋白免疫纯系Balb/c小鼠,采用杂交瘤技术建立能稳定分泌抗人H-FABP的单克隆杂交瘤细胞株。常规制备腹水,纯化后得到特异性抗H-FABP单克隆抗体,进行效价、特异性、亲和力的鉴定分析,并在ELISA平台进行抗体配对,用所筛选到的抗体对初步建立了检测H-FABP的侧向免疫层析方法。结果:成功获得12株稳定分泌抗体的阳性细胞株,并筛选出能相互配对,并应用于侧向免疫层析平台的抗体3D1和5F4,检测临床样品与对照试剂比较总符合率为100%。结论:筛选能稳定分泌抗体的细胞株,配对抗体应用于侧向免疫层析检测方法中,能快速、特异、灵敏的检测出临床样品中H-FABP,为临床应用快速检测H-FABP指标提供了方法和关键材料。  相似文献

Identification of epitopes of trichosanthin by phage peptide library   总被引:4,自引:0,他引:4  
The phage displayed random peptide library has recently emerged as a powerful technique for analyzing Ab-Ag interactions. In this study, the method was employed to identify epitopes of trichosanthin. Two monoclonal Abs (4B5, 2E9) which recognized different epitopes of trichosanthin (TCS) were selected and a phage-peptide library with nine amino acids (9 aa) was used to screen the positive phage clones that have high affinity to the mAbs. Two groups of phage clones that carried peptide-specific binding to mAbs were identified by the screen. The identified phage clones carried peptide-specific binding to 4B5 and 2E9 mAbs were immunized in mice. To evaluate mimotope of selected phages, the specific binding activity to TCS was measured in the serum from phage-immunized mice. They all showed positive results. The conserved interaction motifs were deduced from the peptide sequences of each group of selected phage clones. When compared the motif sequence with the sequence of TCS, it was predicted that 4B5-corresponding epitope was located at 27-37 aa of TCS protein and 2E9-corresponding epitope was located at 41-48 aa of TCS. The predicted sequence of 4B5-corresponding epitope was further confirmed by site-directed mutation of TCS protein. The data showed that the expressed TCS protein mutated in 4B5-corresponding epitope was unable to bind 4B5 mAb. The results suggested that the phage display peptide library is useful to identify Ag epitopes and to raise Ab in disease diagnosis and treatment.  相似文献

目的:制备人宫颈癌基因蛋白单克隆抗体并初步用于肝癌血清检测。方法:重组表达HCCR-1蛋白胞外区(第167th-360th氨基酸)进行动物免疫,用含有HCCR蛋白抗原表位的融合蛋白筛选阳性克隆。通过ELISA、Western Blot、免疫组化鉴定抗体性质,并用于肝癌血清样本检测。结果:获得3株抗人HCCR蛋白单克隆抗体,其中B3抗体特异性、灵敏度较好。ELISA结果表明B3只识别GST-HCCRep蛋白和His-HCCR蛋白,与其他蛋白没有交叉反应;B3可检测到10 mg/L的GST-HCCRep蛋白;B3抗体亲和力常数Kaff=1.86×107(L/mol);Western Blot结果表明该抗体可识别天然HCCR蛋白;免疫组化检测表明肝癌组织中有HCCR蛋白表达而正常组织中未见表达;肝癌患者血清HCCR阳性率达到75%。结论:成功制备了HCCR单克隆抗体,为建立新的肝癌早期诊断方法奠定了基础。  相似文献

An important issue for the performance and specificity of an antibody is the nature of the binding to its protein target, including if the recognition involves linear or conformational epitopes. Here, we dissect polyclonal sera by creating epitope-specific antibody fractions using a combination of epitope mapping and an affinity capture approach involving both synthesized peptides and recombinant protein fragments. This allowed us to study the relative amounts of antibodies to linear and conformational epitopes in the polyclonal sera as well as the ability of each antibody-fraction to detect its target protein in Western blot assays. The majority of the analyzed polyclonal sera were found to have most of the target-specific antibodies directed towards linear epitopes and these were in many cases giving Western blot bands of correct molecular weight. In contrast, many of the antibodies towards conformational epitopes did not bind their target proteins in the Western blot assays. The results from this work have given us insights regarding the nature of the antibody response generated by immunization with recombinant protein fragments and has demonstrated the advantage of using antibodies recognizing linear epitopes for immunoassay involving wholly or partially denatured protein targets.  相似文献

为了制备水稻Argonaute 2(AGO2)的多克隆抗体,该研究采用RT-PCR扩增OsAGO2蛋白165~401aa片段和440~570aa片段的编码序列,并构建了2个原核表达载体。诱导表达重组蛋白后注射家兔,制备了相应的多克隆抗体,最后利用Western blot初步分析水稻AGO2蛋白的表达模式。结果表明:成功构建2个表达载体,通过诱导获得了分子量约为30kD和23kD的重组蛋白。其中,以440~570aa片段为抗原所制备的多克隆抗体免疫印记效果较好。Western blot表明在水稻花药、愈伤组织及小穗中检测到OsAGO2表达。该研究为进一步深入探讨水稻OsAGO2基因的特性与功能奠定了基础。  相似文献

【目的】原核表达空肠弯曲菌鞭毛蛋白FlaA,并制备其单克隆抗体。【方法】克隆目的基因并将其构建到pET30a(+)和pGEX-6p-1表达载体,分别以变复性纯化后的rHis-FlaA、rGST-FlaA蛋白为免疫原和检测原进行杂交瘤细胞的筛选。采用间接ELISA法测定细胞上清和单抗腹水效价,Dot-ELISA、Western blot分析单抗特异性。【结果】成功构建pET30a(+)-flaA和pGEX-6p-1-flaA重组原核表达质粒,并融合表达rHis-FlaA和rGST-FlaA蛋白,Western blot试验显示天然蛋白多抗血清能与体外表达的蛋白呈现特异性反应,表明表达蛋白具有免疫原性。筛选获得3株稳定分泌抗FlaA的单克隆杂交瘤细胞株,分别命名为2D12、5E12、6A9,其Ig亚类分别为IgG2a、IgG1、IgG1,腹水效价分别为1∶102400,1∶102400和1∶51200;Western blot试验显示,3株单抗均能与表达rHis-FlaA重组蛋白的细菌发生特异性反应;Dot-ELISA试验表明,3株单抗均能与不同来源的空肠弯曲菌分离株发生特异性反应。【结论】本研究制备的单克隆抗体有较高特异性,具有良好的应用价值。为进一步研究空肠弯曲菌鞭毛蛋白的生物学特性、致病机理,以及建立快速检测技术奠定基础。  相似文献

Fragments cDNA (nt 935-1475, 1091-1310, 935-1193) encoding N-terminal part of protein E of West Nile virus (WNV), strain LEIV-Vlg99-27889-human were obtained and cloned. Recombinant polypeptides of glycoprotein E (E1-86, E53-126, E1-180) of the WNV with corresponding amino acid sequence to the cloned fragments of cDNA and modeling the epitopes of domains I and II of surface glycoprotein E were purified by affinity chromatography. Twelve types of monoclonal antibodies (MAbs) created in our laboratory against recombinant polypeptide E1-180 interact with glycoprotein E of the WNV as results of Western blot and ELISA that is demonstrating an similarity of chemical structure of short recombinant polypeptides and corresponding amino acid sequence regions of WNV protein E. Analysis of interactions of MAbs with short recombinant polypeptides and protein E of tick-borne encephalitis virus let us reveal no less than six epitopes within domains I and II of glycoprotein E of the WNV. No less than seven types of MAbs to 86-126 aa region of the domain II were found where located peptide providing fusion of virus--cell membranes (98-110 aa). The epitope for anti-receptor MAbs 10H10 within 53-86 aa region of domain II of protein E of the WNV was mapped and it shows that the fusion peptide and co-receptor of protein E for cellular laminin-binding protein (LBP) are spatial nearness. X-ray model of protein E let us suppose that bc-loop (73-89 aa) of domain II interacts with LBP and together with cd-loop (fusion peptide) determines an initial stages of penetration virions into cell.  相似文献

The envelope fusion protein F of baculoviruses is a class I viral fusion protein which play a significant role during virus entry into insect cells. F is initially synthesized as a precursor(F_0) and then cleaved into a disulfide-linked F_1 and F_2 subunits during the process of protein maturation and secretion. To facilitate further investigation into the structure and function of F protein during virus infection, monoclonal antibodies(mAbs) against the F_2 subunit of Helicoverpa armigera nucleopolyhedrovirus(HearNPV)(Ha F) were generated. Two kinds of mAbs were obtained according to their different recognition epitopes: one kind of mAbs, as represented by 38F10,recognizes amino acid(aa) 85 to 123 of F_2 and the other kind, represented by 44D11, recognizes aa148 to 173 of F_2. Western blot and immunofluorescence assay confirmed that both of the mAbs recognized the F protein expressed in HearNPV infected cells, however, only 44D11 could neutralize HearNPV infection. The results further showed that 44D11 may not interact with a receptor binding epitope, rather it was demonstrated to inhibit syncytium formation in cells expressing the Ha F protein. The results imply that the monoclonal antibody 44D11 recognizes a region within HaF_2 that may be involved in the F-mediated membrane fusion process.  相似文献
