目的:构建RGD修饰的ING4和PTEN双基因共表达重组腺病毒载体(Ad.RGD-ING4-PTEN),研究其对人白血病细胞MEG01的抑制作用。方法:采用AdEasyTM腺病毒重组系统,在本科室已成功构建的pAdTrack-CMV-ING4-polyA-promoter、pAdTrack-CMV-polyA-promoter腺病毒转移质粒的基础上,在多克隆酶切位点的Not I、Xho I间插入PTEN片段,得到pAdTrack-CMVING4-polyA-promoter-PTEN重组转移载体,与RGD-4C修饰的腺病毒骨架质粒pAdEasy-1(RGD)同源重组后,经包装和扩增获得ING4和PTEN双基因共表达重组腺病毒Ad.RGD-ING4-PTEN。将Ad.RGD-ING4-PTEN体外感染人白血病细胞MEG01,检测腺病毒对MEG01细胞的感染效率,Western blot法检测外源基因ING4和PTEN在MEG01细胞中的表达,CCK8法检测外源基因的导入对MEG01细胞的生长抑制作用,流式细胞仪检测外源基因导入对MEG01细胞的凋亡和周期的影响。结果:鉴定结果显示,成功构建了Ad.RGD-ING4-PTEN双基因共表达腺病毒,其能有效地感染MEG01细胞。Western blot检测到ING4和PTEN在MEG01细胞中的表达;ING4和PTEN基因能明显抑制MEG01细胞的生长,诱导其凋亡并产生G2/M期阻滞作用,且双基因联合作用较单基因作用更明显。结论:成功构建了RGD-4C修饰的ING4和PTEN双基因共表达腺病毒载体Ad.RGD-ING4-PTEN。收获的腺病毒能有效感染人白血病MEG01细胞。Ad.RGD-ING4-PTEN具有抑制MEG01细胞的生长、诱导其凋亡及G2/M期阻滞的作用。  相似文献

旨在研究携带人IL-24基因的腺病毒表达载体(Ad-IL-24)对SGC-7901人胃癌细胞的生长抑制作用并分析其分子机制。以不同MOI(感染复数)的Ad空载体腺病毒感染SGC-7901人胃癌细胞,筛选出最佳感染剂量;Ad-IL-24以最佳感染剂量感染SGC-7901胃癌细胞,RT-PCR法和Western blotting法检测腺病毒介导的IL-24基因在SGC-7901胃癌细胞中的转录;MTT法检测Ad-IL-24对SGC-7901胃癌细胞的生长抑制作用,流式细胞仪检测其诱导SGC-7901人胃癌细胞凋亡和细胞周期改变的效应,Hoechst33258染色荧光显微镜检测其诱导胃癌细胞凋亡的核形态改变;RT-PCR半定量法进一步检测SGC-7901胃癌细胞中凋亡相关基因的转录。结果显示,100MOI为感染SGC-7901胃癌细胞的腺病毒最佳感染剂量;Ad-IL-24能成功介导IL-24基因在SGC-7901胃癌细胞中转录性表达;Ad-IL-24感染SGC-7901胃癌细胞后,能明显抑制胃癌细胞生长和诱导凋亡;Ad-IL-24能显著上调SGC-7901胃癌细胞中bax、caspase-3和p53的表达和下调bc...  相似文献

Ad-IL-24对人胶质瘤细胞生长抑制效应的体外实验   总被引:2,自引:0,他引:2  
研究携带人白介素24(IL-24)的腺病毒表达载体(Ad-IL-24)对人U251胶质瘤细胞生长的影响和抗肿瘤分子机制。将不同MOI Ad-IL-24感染U251人胶质瘤细胞后, MTT法检测Ad-IL-24对U251细胞生长的抑制作用, 流式细胞仪和Hochest 染色法检测细胞的凋亡率。RT-PCR检测bcl-2、bax、ICE、C-myc、HIF-1a和p53等基因的转录表达水平, Western blotting检测Cleaved Caspase-3的表达。结果表明100 MOI Ad-IL-24感染U251细胞后能明显抑制细胞生长, 并能明显诱导细胞凋亡, 感染72 h后细胞凋亡率可达42%, 感染4 d后细胞生长抑制率可达50%。RT-PCR检测发现Ad-IL-24能引起与细胞凋亡和血管形成相关基因bax/bcl-2、ICE、C-myc、p53的上调和HIF-1a的下调, 并促进Caspase-3的活化。本研究结果显示Ad-IL-24能明显抑制人胶质瘤细胞U251生长和诱导细胞凋亡, 其抗肿瘤机制可能与通过bax/ bcl-2、ICE、c-myc、p53的上调和HIF-1a的下调, 进而导致Caspase-3的活化而诱导肿瘤细胞发生凋亡有关。  相似文献

目的:构建抑瘤素M(OSM)重组腺病毒载体,研究其对人黑色素瘤细胞A375的抑制作用。方法:以PEGZ-OSM重组质粒为模板,通过PCR技术扩增出OSM片段,采用腺病毒载体的基因重组和体外包装技术获得表达与人OSM氨基酸序列相同的重组腺病毒子Ad-OSM,感染A375细胞,用荧光显微镜、RT-PCR、Western blot法检测OSM在A375细胞中的转录和表达;荧光显微镜观察A375细胞的形态学改变;MTT法和流式细胞术(FCM)检测Ad-OSM对A375细胞的生长抑制和细胞周期的抑制效应;半定量RT-PCR法检测OSM基因表达对A375细胞中的Bax、Bcl-2基因表达的影响。结果:基因测序和PCR分析结果显示,成功构建了Ad-OSM腺病毒表达载体;RT-PCR和Western blot法检测到OSM基因在A375细胞中的转录和表达;OSM基因的表达对A375细胞增殖有明显抑制作用,并可诱导细胞凋亡,OSM基因可通过上调细胞中Bax和下调Bcl-2基因表达诱导细胞凋亡。结论:成功构建了Ad-OSM腺病毒表达载体,感染OSM基因可明显抑制A375人黑色素瘤细胞的生长,诱导其凋亡,该现象可能是通过改变Bax、Bcl-2基因表达水平来发挥抗肿瘤作用。  相似文献

目的:应用一系列分子生物学检测技术采检测苦参碱对胶质瘤细胞中TRADD表达的影响,以进一步推测其诱导胶质瘤细胞凋亡的作用机制.方法:采用MTT法测定不同浓度苦参碱作用于C6胶质瘤细胞后的吸光值以计算其抑制率:采用Annexin/FITC染色进行流式细胞术检测来测定不同浓度苦参碱诱导C6胶质瘤细胞的凋亡率;采用ICC(免疫细胞化学)、Westem-blotting及Real-timePCR array(实时定量PCR芯片)方法来检测苦参碱对胶质瘤细胞TRADD表达的影响.结果:MTT结果显示细胞抑制率随着药物剂量的增加而增加;Annexin/FITC染色进行流式细胞术的检测结果显示胶质瘤细胞的凋亡率随着药物剂量的增加而增加;ICC、Western-blotting及Real-time PCR array结果显示苦参碱可诱导胶质瘤细胞TRADD基因及蛋白表达的上调.结论:苦参碱可以诱导胶质瘤细胞凋亡,其诱导胶质瘤细胞凋亡可能是通过上调TRADD的表达,以死亡受体途径来实现的.  相似文献

研究ING4 (肿瘤生长抑制因子4) 和IL-24 (人白细胞介素24) 双基因共表达腺病毒载体 (Ad-ING4-IL-24) 对肺腺癌的化疗增敏效应及分子机制。将Ad-ING4-IL-24感染A549肺癌细胞及联合顺铂 (DDP) 化疗药物作用,RT-PCR和Western blotting检测ING4和IL-24基因在A549肺癌细胞中的转录和表达,MTT法、流式细胞仪 (FCM) 和 Hoechst 染色法检测Ad-ING4-IL-24联合DDP (联合组) 对A549肺癌细胞的生长抑制和凋亡效应。采用A549细胞株建立人肺腺癌裸鼠模型,然后瘤体局部注射干预用药,动态测量肿瘤体积,并计算瘤重抑瘤率,免疫组化检测ING4、IL-24、bax、bcl-2、VEGF等基因的表达。结果表明,Ad-ING4-IL-24感染A549肺癌细胞后可获得成功转录和表达,体外联合组能明显抑制A549肺癌细胞生长和诱导细胞凋亡,呈现出典型细胞凋亡形态学变化。体内联合组同样能显著抑制肿瘤生长,瘤重抑瘤率达52.81%,免疫组化结果显示联合组能上调bax基因表达,同时下调bcl-2、VEGF等基因的表达。以上结果表明Ad-ING4-IL-24具有化疗增敏的作用,该作用机制可能与促进肿瘤细胞凋亡和抑制血管形成有关。  相似文献

目的:研究腺病毒载体AdING4对人MCF-7乳腺癌细胞的生长抑制及化疗增敏作用。方法:将搭载有ING-4基因的重组腺病毒载体AdING4感染人MCF-7乳腺癌细胞,用荧光显微镜观察感染后的MCF-7细胞形态学变化;RT-PCR和Western-Blot法检测ING-4基因在MCF-7细胞中的转录和表达;RT-PCR法检测凋亡相关基因在MCF-7细胞中的表达;CCK法测定Ad-ING4感染MCF-7乳腺癌细胞后所发挥的细胞增殖抑制作用。流式细胞技术检测ING-4对MCF-7乳腺癌细胞的促凋亡作用。CCK-8法分别测定病毒感染前后的MCF-7乳腺癌细胞的药物半数抑制浓度IC50,并观察Ad-ING4与化疗药物合用后对MCF-7细胞增殖抑制和化疗增敏现象。结果:MCF-7细胞在转染ING-4基因后,明显出现变圆、脱落、皱缩、聚集等现象;外源性ING-4基因在MCF-7细胞中获得成功表达;外源性ING-4基因作用下MCF-7细胞的增殖受到了明显抑制,凋亡率有所升高,凋亡相关基因Bax的表达水平明显上调,Bcl-2、Survivin的表达水平明显下调。ING-4基因感染MCF-7细胞后,使MCF-7细胞对相关化疗药物的敏感度更高;ING-4基因与化疗药物合用后对MCF-7细胞的增殖抑制作用,较之单用化疗药物更为明显。结论:MCF-7细胞在转染ING4基因后其增殖受到了明显抑制并更易凋亡,该现象可能是通过改变Bax,Bcl-2及Survivin表达水平来实现的,且对化疗药物的敏感性更高。  相似文献

本研究旨在分析腺病毒携带的IL-24基因在体内外对人骨肉瘤细胞生长抑制效应及其分子机制。将Ad-IL-24重组腺病毒感染MG-63细胞,用荧光显微镜、RT-PCR法检测IL-24在MG-63细胞中的转录和表达;MTT法、流式细胞技术和Hoechst染色法检测IL-24基因的表达对MG-63细胞的生长抑制和凋亡效应;半定量RT-PCR法检测IL-24基因的表达对MG-63细胞中的bcl-2、bax、caspase-3相关基因表达的影响。用Ad-IL-24重组腺病毒在MG-63骨肉瘤荷瘤裸鼠的瘤体内进行注射治疗,观察肿瘤生长变化,15d后处死裸鼠,摘除瘤体,称瘤重。并通过免疫组化法检测Bcl-2、Bax、Caspase-3等与细胞凋亡相关因子的表达。结果表明Ad-IL-24重组腺病毒感染MG-63细胞后,能明显抑制MG-63细胞增殖,并能通过上调细胞中bax、caspase-3和下调bcl-2基因表达,诱导细胞凋亡,呈现出典型细胞凋亡形态学变化。Ad-IL-24重组腺病毒瘤内注射MG-63裸鼠荷瘤骨肉瘤移植瘤后,能显著抑制肿瘤生长,瘤重的抑制率可达52%,免疫组化结果显示Ad-IL-24重组腺病毒能明显上调与细胞...  相似文献

利用Ad5腺病毒载体系统构建人Sema4C基因重组腺病毒表达载体并在成肌细胞系C2C12中表达,并初步探讨Sema4C基因在成肌发育过程中的可能作用。利用脂质体介导重组腺病毒载体转染HEK293细胞,包装出完整的腺病毒;将重组腺病毒载体感染C2C12成肌细胞后,利用激光共聚焦显微镜观察发现12h即有绿色荧光表达,24h后绿色荧光蛋白表达最强;流式细胞仪检测病毒的感染效率几乎达100%。WB检测结果表明感染重组腺病毒载体组C2C12细胞Sema4C蛋白的表达量明显高于空载体对照组(P<0.01)。为了进一步观察Sema4C基因对C2C12细胞增殖分化的影响,流式细胞仪检测了病毒感染48h后C2C12细胞的增殖指数,并对感染后诱导分化的C2C12细胞的分化情况进行了观察。我们的结果首次表明,过表达外源性人Sema4C基因不仅能使C2C12细胞的G0/G1期比例增加,细胞的增殖指数下降,同时在分化培养条件下还能促进C2C12细胞肌管的形成。  相似文献

目的:研究靶向抑制survivin表达对软骨多糖诱导乳腺癌MCF-7细胞凋亡的影响.方法:将survivin-siRNA转染乳腺癌MCF-7细胞.用定量PCR和Western-blotting检测转染后细胞内survivin基因表达水平,流式细胞仪和Hochest染色检测细胞凋亡的改变.结果:软骨多糖可抑制MCF-7细胞的生长,其生长抑制率与药物浓度和作用时间呈依赖关系;软骨多糖作用MCF-7细胞后,survivin表达降低;转染survivin-siRNA能促进软骨多糖诱导MCF-7细胞凋亡.结论:靶向抑制survivin表达对软骨多糖诱导乳腺癌细胞凋亡具有增敏作用.  相似文献

High risk subtype HPV16 early oncoprotein E6 contributes host cell immortalization and transformation through interacting with a number of cellular factors. ING4 is one member of the inhibitor of growth (ING) family of type II tumor suppressors and it has been shown to be involved in regulating p53 function. However, the effect and mechanism of HPV16 E6 on ING4 function remain elusive. In this study, we report HPV16 E6 combines with ING4 in vivo and in vitro. The ING4 induced p53 acetylation and its combining with p53 were attenuated by HPV16 E6 independent of p53 degradation. The enhancing function of ING4 on p53 mediated apoptosis was diminished when HPV16 E6 existed. These findings reveal that ING4 may be a common target of oncogenic viruses for driving host cell carcinogenesis.  相似文献

The known members of inhibitor of growth (ING) gene family are considered as candidate tumor suppressor genes. ING4, a novel member of ING family, is recently reported to interact with tumor suppressor p53, p300 (a major component of histone acetyl transferase complexes), and p65(RelA) subunit of NF-κB. In this study, we investigated the cellular behaviors of HepG2 cells with exogenous ING4. Interestingly, the overexpression of ING4 negatively regulated the cell growth with significant G2/M arrest of cell cycle, and moreover, enhanced the cell apoptosis triggered by serum starvation in HepG2 cells. Furthermore, the exogenous ING4 could upregulate endogenous p21 and Bax in HepG2 cells, not in p53-deficient Saos-2 cells, suggesting that G2/M arrest induced by ING4 could be mediated by the increased p21 expression in a p53-dependent manner, although there is no significant increase of p53 expression in HepG2 cells. Moreover, HepG2 cells with exogenous ING4 could significantly increase cell death, as exposed to some DNA-damage agents, such as etoposide and doxorubicin, implying that ING4 could enhance chemosensitivity to certain DNA-damage agents in HepG2 cells.  相似文献

The inhibitor of growth 4 (ING4) is known as a tumor suppressor. The expressions of ING4 were markedly reduced in human renal clear cell carcinoma (ccRCC) tissues. However, the role of ING4 in renal cell carcinoma (RCC) remains unknown. The aim of the current study was to detect the ING4 expression level and its potential role in human RCC cell lines. Our results showed that ING4 was lowly expressed in human RCC cell lines compared with that in proximal tubular cell line. Ectopic overexpression of ING4 inhibited the proliferation, migration, and invasion properties, and as well as prevented epithelial-mesenchymal transition (EMT) phenotype of RCC cells. In addition, ING4 overexpression induced cell apoptosis and autophagy in RCC cells. Furthermore, ING4 overexpression suppressed the activation of PI3K/Akt pathway in RCC cells. The activator of PI3K/Akt, insulin-like growth factor 1, abolished the effects of ING4 on RCC cells. These findings indicated that ING4 presented anticancer activity in RCC cells. The effects of ING4 on RCC cells were mediated by regulating the PI3K/Akt pathway. These findings suggested that ING4 could be used for gene therapy of RCC.  相似文献

Previous investigations have proved that microRNA (miR)-765 is significantly overexpressed in multiple tumor types. Nevertheless, the underlying molecular mechanism of miR-765 in mediating breast carcinoma cell growth and metastasis remains unclear. Quantitative real-time polymerase chain reaction was used to determine the levels of miR-765 and inhibitor of growth 4 (ING4) in breast carcinoma tissues and breast carcinoma cells. Cell proliferation, colony formation, wound healing, and Transwell invasion assays were used to analysis the role of miR-765 on breast carcinoma cell growth and aggressiveness. The expressions of ING4 were determined using Western blot analysis and immunohistochemical staining. The direct target of ING4 and miR-765 was confirmed using the luciferase reporter assay. Nude mice were subcutaneously implanted with miR-765 inhibitor transfected MDA-MB-231 cells to determine the potential role of miR-765 in tumor growth in vivo. We observed that miR-765 is overexpressed in breast carcinoma tissue and breast cancer cells. By using luciferase reporter gene bioassay, we find that ING4 is the direct target of miR-765 in breast carcinoma. The level of ING4 is inversely associated with the level of miR-765. The gain-of-function and loss-of-function experiments in vitro indicate that the downregulation of miR-765 suppresses the growth, mobility, and invasion abilities of breast cancer cells by inhibiting ING4. In addition, overexpression of ING4 suppresses the aggressiveness of the MDA-BA-231 cell that is induced by miR-761 in vitro. In this study, we prove that miR-765 regulates the growth and metastasis of breast cancer via modulating miR-765-ING4-negative feedback loop.  相似文献

The ING family of tumor suppressor proteins affects cell growth, apoptosis and response to DNA damage by modulating chromatin structure through association with different HAT and HDAC complexes. The major splicing isoforms of the ING1 locus are ING1a and INGlb. While INGlb plays a role in inducing apoptosis, the function of ING1a is currently unknown. Here we show that alternative splicing of the ING1 message alters the INGla:INGlb ratio by approximately 30-fold in senescent compared to low passage primary fibroblasts. INGla antagonizes INGlb function in apoptosis, induces the formation of structures resembling senescence-associated heterochromatic foci containing heterochromatin protein 1 gamma, the accumulation of senescence-associated beta-galactosidase activity and promotes senescent cell morphology and cell cycle arrest. Phenotypic effects may result from differential effects on gene expression since ING1a increases levels of both retinoblastoma and the p16 cyclin-dependent kinase inhibitor and ING1a and ING1b have opposite effects on the expression of proliferating nuclear cell antigen (PCNA), which is required for cell growth. Gene expression appears to be altered by targeting of HDAC complexes to gene promoters since INGla associates with several-fold higher levels of HDAC1 in senescent, compared to replication-competent cells and ING1 is found on the PCNA promoter by chromatin immunoprecipitation analysis. These data demonstrate a novel role for the ING1 proteins in differentially regulating senescence-associated chromatin remodeling vs. apoptosis and support the idea that altered ratios of the ING1 splicing isoforms may contribute to establishing the senescent phenotype through HDAC and HAT complex-mediated effects on chromatin structure.  相似文献
