本研究从安徽淮南采集储藏根茎类中药材样本共30种,采用直接镜检法和光照驱虫法分离其中孳生的书虱,光镜下鉴定书虱种类并计数。结果表明,30份样本中书虱的孳生率为933%(28/30),检获的书虱为嗜卷书虱(Liposcelis bostrychophila Motschulsky,1852);该书虱年平均孳生密度为57头/克,孳生指数为142。本研究表明安徽淮南部分储藏根茎类中药材有书虱孳生,在中药材储藏和加工过程中应注意书虱的防制。  相似文献

选用有机磷类杀虫剂(敌敌畏、毒死蜱、对氧磷)和氨基甲酸酯类杀虫剂(丁硫克百威),通过生物测定(药膜法)和生化测定(比色法)比较了嗜卷书虱和嗜虫书虱对所选药剂的敏感差异性。根据LC50可知嗜虫书虱对所选药剂比嗜卷书虱敏感。离体酶活性分析结果显示嗜卷书虱和嗜虫书虱的羧酸酯酶只对敌敌畏敏感,且嗜卷书虱比嗜虫书虱更敏感;4种药剂对乙酰胆碱酯酶均有强烈的抑制作用,同样是嗜卷书虱比嗜虫书虱敏感。乙酰胆碱酯酶的动力学研究结果和离体酶活性测定相一致。聚丙烯酰胺凝胶电泳分析显示,4种杀虫剂离体条件下对2种书虱的酯酶同工酶的抑制能力有明显差异,其中敌敌畏的抑制力最强;但对不同同功酶的抑制趋势(对大分子的抑制似乎较强)是一致的。酶的敏感性分析结果与生测结果比较表明,2种书虱的耐药力差异与其乙酰胆碱酯酶和酯酶对药剂的敏感性无关。如要弄清耐药力机制,需做进一步研究。  相似文献

两种书虱微卫星富集文库的构建及比较   总被引:1,自引:0,他引:1  
利用链霉亲和素与生物素之间的强亲和性原理,将链霉亲和素偶联的磁珠与微卫星探针(AC)12、(TC)12、(ATC)8、(ATG)8、(AAC)8、(ATAC)6及(GATA)6退火结合后,再亲和捕捉含接头和微卫星序列的单链书虱基因组DNA限制性酶切目的片段,经PCR扩增形成双链后进行克隆、建库。结果表明本研究成功构建了嗜卷书虱和嗜虫书虱共13个微卫星富集文库,包括6个嗜卷书虱文库,7个嗜虫书虱文库,其平均阳性克隆率为71.17%。经检测发现共得到两种书虱260个微卫星位点。这两种书虱微卫星富集文库的建立和高多态性微卫星位点的筛选将为嗜卷书虱和嗜虫书虱的种群遗传与进化、基因连锁图谱构建、分子系统发育研究等提供大量分子遗传标记,对其在实仓中的持续控制提供遗传学信息。  相似文献

杀虫剂胁迫下嗜卷书虱和嗜虫书虱能源物质的代谢比较   总被引:4,自引:0,他引:4  
采用比色法测定嗜卷书虱(Liposcelis bostrychophila)和嗜虫书虱(L.entomophila)体内甘油三酯、多糖和可溶性蛋白的贮存量。结果表明,嗜虫书虱体内3种物质的贮存量均显著高于嗜卷书虱。在毒死蜱和丁硫克百威的胁迫下,嗜虫书虱体内3种物质的变化幅度均大于嗜卷书虱,其中甘油三酯的含量先增高后降低,整体呈现下口抛物线趋势;多糖和可溶性蛋白的消耗率增加。这些说明在毒死蜱和丁硫克百威的作用下,书虱更多地启动了多糖和可溶性蛋白的代谢,降低了对甘油三酯的利用,以应对杀虫剂的胁迫。  相似文献

采用拌粮法 ,用硅藻土单剂、配方 1 (硅藻土 +95 %马拉硫磷EC)、配方 2 (硅藻土 +2 . 5 %溴氰菊酯EC)、配方 3 (硅藻土 +80 %敌敌畏EC)、配方 4(硅藻土 +0 . 4%天惠虫清EC)、配方 5 (硅藻土 +5 %双氧威WP)、配方 6(硅藻土 +5 %抑太保EC)对嗜卷书虱Liposcelisbostrychophila进行防治研究 ,结果表明 :配方 4为优选配方 ,处理 2 4h后 ,3种浓度的致死率均达到 1 0 0 % ;配方 1、配方 2、配方 3、配方 5、配方 6对嗜卷书虱的致死率处理 48h后达到 1 0 0 % ;单用硅藻土处理 96h后达到 1 0 0 % ,各配方处理间差异极显著。  相似文献

应用PCR-RFLP技术,对我国4种常见仓储书虱即嗜虫书虱Liposcelis entomophila(Enderlein)、嗜卷书虱L.bostrychophila(Badonnel)、无色书虱L.decolor(Pearman)和小眼书虱L.paeta(Pearman)开展快速识别方法的研究。采用CTAB法从上述4种书虱体内提取基因组DNA,选用1对引物扩增16S rDNA基因区域,分别用5种限制性内切酶对PCR产物进行酶切。结果表明,PCR扩增片段大小约为500bp,用限制性内切酶DraI酶切得到的片段可将供试书虱区分开。该方法不受4种供试书虱地理种群、虫态(成虫、若虫)和性别的影响,可用于4种书虱的快速识别。  相似文献

报道了嗜卷书虱对高二氧化碳,低氧气调抗性形成过程中能源物质的积累以及抗性形成后在气调胁迫下能源物质的利用情况。结果表明随着抗性水平的提高。嗜卷书虱体内甘油三酯,多糖以及游离氨基酸含量均显升高。在气调胁迫环境下,嗜卷书虱抗性品系能源物质的消耗率明显低于敏感品系,甘油三酯的积累以及在气调摁迫下缓慢消耗可能是其抗气调性的主要内在机理之一。  相似文献

记述并描绘了中国虱啮属昆虫两新种:离首虱啮Liposcelis capitisecta sp.nov.和褐虱啮Liposcelis badia sp.nov..离首虱啮采集于四川成都一农户家的粮柜内,隶属于D组,其前胸颜色较浅,肉眼或在低倍镜下观察头部与身躯似乎分离.褐虱啮采自湖北省钟祥市野外,根据其特征划分为A组,其个体较大,躯体颜色较深,体表毛较密集.其与暗褐虱啮L.brunnea Motschulsky的区别是:PNS毛有2~3根;Se毛圆筒形,不弯曲.  相似文献

储粮书虱Liposcelis spp.和锈赤扁谷盗Cryptolestes ferrugineus(Stephens)由于对磷化氢具有较高的抗性而成为目前粮食储藏过程最难防治的两类害虫,研究储粮防护剂和磷化氢联用对储粮书虱和锈赤扁谷盗的防治效果,对于缓解其对磷化氢的抗性以及其在实仓中综合防治具有重要的经济意义。本文用55%甲基嘧啶磷乳油和2%溴氰·甲嘧磷粉剂分别和磷化氢联用对广东、广西、安徽和河南4个不同气候条件的粮库中的书虱和锈赤扁谷盗进行防治效果的研究。结果表明,在磷化氢熏蒸粮食之前用55%甲基嘧啶磷乳油或2%溴氰·甲嘧磷粉剂处理粮面表层的粮食,在4个粮库的试验粮仓内书虱和锈赤扁谷盗的防治效果有显著差异:在北方干燥少雨的河南汤阴库防治效果最好,可达100%,其次是安徽舒城库、广西柳州库,防治效果分别为71.8%和71.9%。效果最差的是高温高湿的是广东湛江库,防治效果为50.4%。用55%甲基嘧啶磷乳油或2%溴氰·甲嘧磷粉剂和磷化氢联用对书虱和锈赤扁谷盗具有很好的防治效果,说明储粮防护剂和磷化氢联用对于储粮书虱和锈赤扁谷盗具有更好的防治效果。  相似文献

Field experiments were conducted in steel bins containing 13,600 kg of hard red winter wheat, Triiticum aestivum L. One bin was treated with ozone and the second bin served as a control. Stored grain insects were placed in bins for 1-, 2-, 3-, and 4-d exposure periods in sampling tubes to test ozone concentrations of 0, 25, 50, and 70 parts per million by volume (ppmv). Ozone treatments on eggs and larvae of Plodia interpunctella (Hübner) were not effective, but pupae were more susceptible. Sitophilus oryzae (L.) adults were the most susceptible species with 100% mortality reached after 2 d in all ozone treatments. However, some progeny were produced at all concentrations and exposure periods. Tribolium castaneum (Herbst) adults had 100% mortality only after 4 d at 50 or 70 ppmv. No T. castaneum progeny were produced after 2-4 d at 70 ppmv. For Rhyzopertha dominica (F.), Cryptolestes ferrugineus (Stephens), and Oryzaephilus surinamensis (L.), 100% mortality was never achieved and progeny were produced at all ozone concentrations. Laboratory experiments, testing the effectiveness of ozone in controlling psocids, were conducted in two polyvinyl chloride cylinders each containing 55 kg of hard red winter wheat. Ozone treatment at a concentration of 70 ppmv was highly effective against adult female Liposcelis bostrychophila Badonnel and Liposcelis paeta Pearman after only 1 d of exposure. However, it was not effective against eggs of both species at all exposure periods. Ozonation has potential for the control of some stored grain insect pests on wheat.  相似文献

Four organophosphate insecticides, azamethiphos, fenitrothion, chlorpyrifos-methyl, and pirimiphos-methyl, were tested as surface treatments on concrete (porous surface) and galvanized steel (nonporous surface) panels (0.3 by 0.3 m) against adults of three Liposcelid psocid spp.--Liposcelis bostrychophila Badonnel, Liposcelis entomophila (Enderlein), and Liposcelis paeta Pearman. Residual efficacy of these chemicals was assessed at 30 +/- 1 degrees C, 70 +/- 2% RH, and a photoperiod of 12:12 (L:D) h from 1 d after treatment (0 wk) and thereafter at weeks 1, 2, 4, 6, and 8, and then every 4 wk up to week 40. Mortality was recorded at exposure periods of 6 h and then every 24 h until end-point was achieved. L. bostrychophila was the most susceptible species to the organophosphates tested, followed by L. paeta and L. entomophila. We conclude that for long-term protection, azamethiphos is the preferred organophosphate against L. bostrychophila (up to 36 wk on steel and 24 wk on concrete storage surfaces) and L. paeta infestations only on steel surface (up to 28 wk). None of the four organophosphates tested, however, would provide long-term protection against L. paeta on concrete surface and against L. entomophila infestations on either concrete or steel storage surfaces.  相似文献

The insect growth regulator pyriproxyfen was evaluated as a surface treatment for control of three stored-product psocid pests Liposcelis bostrychophila Badonnel, Liposcelis decolor (Pearman), and Liposcelis paeta Pearman (Psocoptera: Liposcelididae). Nymphs were exposed for 35 d on a concrete surface treated with 2.3 mg of active ingredient/m2 pyriproxyfen. Exposure to pyriproxyfen significantly reduced the numbers of both adults and nymphs in comparison with untreated controls. In adults, the greatest reduction (> 90%) was for L. decolor and L. bostrychophila, whereas for L. paeta it was 49%. Few adults of any species were found in the pyriproxyfen treatments. The greatest numbers of nymphs were recorded for L. bostrychophila for both pyriproxyfen treatments and controls. Few adults of any species were found in the pyriproxyfen treatments. The results indicate that pyriproxyfen is effective for control of L. bostrychophila, L. decolor, and L. paeta on concrete, and although complete control was not achieved, the results warrant further long-term study to determine whether pyriproxyfen can completely eliminate psocid populations over time.  相似文献

Abstract

Red flour beetle, Tribolium castaneum Herbst (Coleoptera: Tenebrionidae) is a prominent pest of stored products particularly cereal flour. Since resistance of this pest to common chemical insecticides is well documented, we were examined the synergistic/antagonistic interaction between Satureja hortensis L., Trachyspermum ammi L., Ziziphora tenuior L., Cuminum cyminum L. and Foeniculum vulgare Miller essential oils, ethanolic extracts and powders with Diatomaceous earth (DE) against T. castaneum adults under laboratory conditions at 27 ± 1 °C, 65 ± 5% RH and continuous darkness. We assayed repellency of ethanolic extracts and essential oils of mentioned plants on the pest. Results showed that DE had high toxicity to the pest. Plant essential oils and ethanolic extracts (except ziziphora) synergized the performance of DE. Nevertheless, plant powders elicited antagonistic effects (except ziziphora that exhibited synergistic effect). The most repellent EO and extract was cumin which exhibited mean repellency value on adult insect equivalent to 92.58 and 51.47%, respectively.  相似文献

Endosymbiotic bacteria that potentially influence reproduction and other fitness-related traits of their hosts are widespread in arthropods, and their appeal to researchers' interest is still increasing. In this study, the effects of removal of Cardinium infection on development, survival, and reproduction of Liposcelis bostrychophila Badonnel (Psocoptera: Liposcelididae) were investigated in the laboratory. The Cardinium-free strain was obtained by the removal of Cardinium infection by using 1% rifampicin treatment on the Cardinium-infected strain (control) for 4 wk, and no Cardinium gene product was detected in this strain throughout the experiment. The results showed that the removal of Cardinium infection had negative effects on fitness of L. bostrychophila. Compared with the control strain, the Cardinium-free strain (both in first [F1] and second [F2] generation) had a similar developmental time, reduced survivorship of immature stages, as well as reduced fecundity, which resulted in much smaller r(m) values. Using r(m) values, the fitness for Cardinium-free F1 and F2 relative to the control was calculated as 0.81 and 0.74, respectively. We concluded that the use of antibiotics combined with heat treatment might be a good control measure for L. bostrychophila.  相似文献

Six plant essential oils alone as repellent and fumigant, and in combination with the controlled atmosphere against Liposcelis bostrychophila Badonnel were assessed in the laboratory. These essential oils were extracted from the leaves of six source plants: Citrus tangerina Tanaka, Citrus aurantium L., Citrus bergamia Risso et Poiteau, Pinus sylvestris L., Cupressus funebris End]., and Eucalyptus citriodora Hook. The repellency test indicated that L. bostrychophila adults were repelled by filter paper strips treated with six essential oils. Of these essential oils, the C. funebris oil was most effective followed by that of F. sylvestris, C. tangerina, C. bergamia, and E. citriodora. The average repellency of the C. aurantium oil against L. bostrychophila adults was significantly lower than other five test oils by day 14. These essential oils had a high level of toxicity in the fumigation assay against L. bostrychophila adults at both 10 and 20 ppm. When combined with two controlled atmosphere treatments (12% CO2 + 9% O2, and 10% CO2 + 5% O2, balanced N2), the toxicity of plant oils was enhanced significantly.  相似文献

Insects face several (environmental) abiotic stressors, including low temperature, which cause the failure of neuromuscular function. Such exposure leads insects toa reversible comatose state termed chill-coma, but the consequences of this state for the organism biology were little explored. Here, the consequences of the chill-coma phase were investigated in two of the main stored product pest species – the red flour beetle Tribolium castaneum (larvae and adults) and the rice weevil Sitophilus oryzae (adults). For this purpose, a series of low-temperature shocks were used to estimate the chill-coma recovery time (CCRT), survival, nutrition and weight gain/growth of T. castaneum (larvae and adults) and S. oryzae, as well as the development of T. castaneum life stages. The relatively long CCRT was characteristic of beetle larvae, at different low-temperature shocks, and CCRT increased with decreasing temperatures and increasing exposure intervals for both pest species. The survival was little affected by the low-temperature shocks applied, but such shocks affected insect feeding and growth. Tribolium castaneum larvae was more sensitive than adults of both insect species. Moreover, the relative consumption and weight gain of S. oryzae adults were lower than those of T. castaneum adults and mainly larvae, while feeding deterrence was not affected by low temperature shocks, unlike food conversion efficiency. Low-temperature shocks, even under short duration at some temperatures, significantly delayed development. The lower the temperature and the higher the exposure period, the more delayed the development. Thus, the physiological costs of chill-coma are translated into life-history consequences, with potential implications for the management of this insect pest species in stored products and even more so on red flour beetles and rice weevils.  相似文献

Anagasta kuehniella is a polyphagous pest that feeds on a wide variety of stored products. The possible roles suggested for seed proteinase inhibitors include the function as a part of the plant defensive system against pest via inhibition of their proteolytic enzymes. In this study, a trypsin inhibitor (ApTI) was purified from Adenanthera pavonina seed and was tested for insect growth regulatory effect. The chronic ingestion of ApTI did result in a significant reduction in larval survival and weight. Larval and pupal developmental time of larvae fed on ApTI diet at 1% was significantly longer; the larval period was extended by 5 days and pupal period was 10 days longer, therefore delaying by up to 20 days and resulting in a prolonged period of development from larva to adult. As a result, the ApTI diet emergence rate was only 28% while the emergence rate of control larvae was 80%. The percentage of surviving adults (%S) decreased to 62%. The fourth instar larvae reared on a diet containing 1% ApTI showed a decrease in tryptic activity of gut and that no novel proteolytic form resistant to ApTI was induced. In addition, the tryptic activity in ApTI ‐fed larvae was sensitive to ApTI. These results suggest that ApTI have a potential antimetabolic effect when ingested by A. kuehniella. © 2010 Wiley Periodicals, Inc.  相似文献

为了评估桉树精油和α-蒎烯对白腹皮蠹Dermestes maculatus Degeer的杀虫作用,采用生测方法测定了不同剂量和不同处理时间下,其幼虫和成虫的死亡率。结果表明:用桉树精油及其活性成分α-蒎烯处理白腹皮蠹成虫和幼虫后,可显著影响其死亡率(P<0.001)。白腹皮蠹的反应因化学物种类、剂量和暴露时间不同而异。当剂量为32 μL/cm3的桉树精油处理白腹皮蠹幼虫72 h后,死亡率可超过90%。同样的剂量对其成虫熏蒸72 h后也表现高毒。在相同的剂量和相同的暴露时间下,α-蒎烯对白腹皮蠹的成虫和幼虫也表现高毒,且成虫的抗性比幼虫强。Probit检验结果表明桉树精油比α-蒎烯更有效。以上结果说明这些天然产物对白腹皮蠹种群的控制是很有用的。  相似文献
