嗜卷书虱抗气调品系的选育及其适合度研究   总被引:2,自引:0,他引:2  
于恒温条件下以人工饲料饲养的嗜卷书虱Liposcelis bostrychophila,在35% CO2和1% O2组配的气调环境中以50%左右的选择压力处理,以选育其抗气调性。至30代获得抗性品系(CA-R),抗性倍数达5.6倍,且有继续增强的基因潜能。该抗性品系对气调的抗性不太稳定,在无选择压力的情况下经室内5代饲养,抗性衰退了63.2%。CA-R品系与敏感品系在无气调选择压力的条件下相比具有一定程度的繁殖不利性;但没有发现CA-R品系在生长发育特性上的不利性。以净增殖率来衡量,CA-R品系相对于敏感品系具有0.39的适合度。  相似文献

杀虫剂胁迫下嗜卷书虱和嗜虫书虱能源物质的代谢比较   总被引:4,自引:0,他引:4  
采用比色法测定嗜卷书虱(Liposcelis bostrychophila)和嗜虫书虱(L.entomophila)体内甘油三酯、多糖和可溶性蛋白的贮存量。结果表明,嗜虫书虱体内3种物质的贮存量均显著高于嗜卷书虱。在毒死蜱和丁硫克百威的胁迫下,嗜虫书虱体内3种物质的变化幅度均大于嗜卷书虱,其中甘油三酯的含量先增高后降低,整体呈现下口抛物线趋势;多糖和可溶性蛋白的消耗率增加。这些说明在毒死蜱和丁硫克百威的作用下,书虱更多地启动了多糖和可溶性蛋白的代谢,降低了对甘油三酯的利用,以应对杀虫剂的胁迫。  相似文献

嗜卷书虱成虫于28°C、70%—80%RH条件下,在35%CO2。和1%O2。组配的气调环境中连续暴露30代,选择压力保持在70%左右,以选育其抗气调性。至第30代,以LT50。为指标衡量其抗性倍数达5.6倍。生化分析表明,抗性选育过程中离体羧酸酯酶(CarE)和超氧化物歧化酶(SOD)活力均随选育代数的增多而显著增强,且酶活力与抗气调水平显著相关。第30代,抗性品系(CA-R)的CarE和SOD活力分别是敏感品系(CA-S)的4.06和5.22倍。两品系暴露于气调环境中,其CarE活力均受抑制,但对CA-S品系的抑制率显著强于CA-R品系。短时间的气调暴露对SOD具有诱导作用,但诱导作用的时间及强度CA-R品系明显长(强)于CA-S品系。离体过氧化氢酶(CAT)活力也随着抗气调性的增强而增强,但两着之间并无显著的相关性。抗性选育过程中离休酸性磷酸酯酶(ACP)和碱性磷酸酯酶(ALP)活力均未发生明显的变化。过氧化物酶(POD)在两品系中均未检测出。由此看出,CarE和SOD活力增强是嗜卷书虱抗气调性形成的主要机制,CAT可能对低氧气调抗性的形成具有辅助作用。  相似文献

室内恒温条件下以人工饲料饲养的嗜卷书虱 Liposcelis bostrychophila Badonnel成虫,分别在35%和55%CO_2。(21%O_2,N_2作为平衡气体使用)组配的气调环境中以70%的选择压力处理,以选育其抗高CO_2气调性。选育至30代获得抗性品系HCC_1和HCC_2,以LT_50为标准,其抗性倍数分别达4.6和5.3倍。整个选育过程中,气调暴露时间对数值与死亡率机率值间的回归直线基本平行,且斜率值均低于敏感基线的斜率值,这表明嗜卷书虱对高CO_2抗性发展的基因潜能至第30代仍未耗尽。两个抗性品系的抗性均不太稳定,去除选择压力后抗性显著下降,且两抗性品系均具有繁殖不利性即适合度缺陷。以净增值率为指标衡量,HCC_1和HCC_2相对于敏感品系分别具有0.52和0.45的适合度。  相似文献

为了分析气调处理对实验昆虫解毒酶性质的影响, 探讨酶活性变化与气调抗性形成的潜在关系, 本研究用高浓度二氧化碳胁迫处理咖啡豆象Araecerus fasciculatus (De Geer), 研究其羧酸酯酶(carboxylesterase, CarE)、 酸性磷酸酯酶(acid phosphatase, ACP)和谷胱甘肽转移酶(glutathione S-transferases, GSTs)的生物化学与毒理学性质。结果表明: 高浓度二氧化碳气调胁迫处理咖啡豆象3 h, 6 h和9 h, 其CarE酶活力分别升高 35.41%, 55.02%和88.98%。 CarE酶促动力学参数Vmax分别升高26.13%, 31.77%和57.12%, Km没有显著改变; 相应处理下, ACP活力分别升高34.53%, 72.45%和126.37%; GSTs 活力分别升高5.40%, 8.40%和17.59%。可见, 二氧化碳气调胁迫下, 实验昆虫可以通过调节部分解毒酶的酶活力或酶促动力学参数来应对不利环境; 酶与底物间的亲和力增强可能是昆虫在气调胁迫下保护机体免受伤害的一种代谢反馈信息。本研究可为昆虫气调杀虫及其抗(耐)气性形成机制提供一些基础信息。  相似文献

1 引言 嗜卷书虱(Liposcelis bostrychophila)属啮虫目虱啮科。其体形微小,食性复杂,栖息场地多在粮食仓库及食品加工厂、图书馆、标本馆等地。大量发生时可造成严重损失,传播微生物,并以其尸体、排泄物等污染储藏的粮食和其它储藏物品。王进军等调查发现,在我国“双低”和“三低”贮粮的粮仓中,书虱已成为优势种群,对贮粮安全构成了威胁。使用病原真菌和细菌来防治嗜卷书虱,很难达到理想效果,即使采用菊酯类杀虫剂也难以奏效,贮粮熏蒸中普遍使用的溴甲烷(CH_3Br)和磷化氢(PH_3)对嗜卷书虱的防治效果也不很理想。利用昆虫生长调节剂防治嗜卷书虱的研究已有一些报道,但在具体应用上还存在着一些问题。气调措施可应用于控制书虱种群的增长,但书虱的抗气性发展很快,  相似文献

在10%CO_2,5%O_2(其余部分为N_2,下同);35%CO_2,11%CO_2;35%CO_2,21%O_2;75%CO_2,11%O_2,及75%CO_2,21%O_2,五种气调环境下对腐食酪螨Tyrophagus putrescentiae(Schrank)进行连续25~26代的抗气性诱导筛选。结果表明,抗性都有不同程度的增加,在75%CO_2,11%O_2下抗性系数最大为5.0,在10%CO_2,5%O_2下抗性系数最小为3.7,在其它气调环境下,抗性系数都在4以上。  相似文献

选用有机磷类杀虫剂(敌敌畏、毒死蜱、对氧磷)和氨基甲酸酯类杀虫剂(丁硫克百威),通过生物测定(药膜法)和生化测定(比色法)比较了嗜卷书虱和嗜虫书虱对所选药剂的敏感差异性。根据LC50可知嗜虫书虱对所选药剂比嗜卷书虱敏感。离体酶活性分析结果显示嗜卷书虱和嗜虫书虱的羧酸酯酶只对敌敌畏敏感,且嗜卷书虱比嗜虫书虱更敏感;4种药剂对乙酰胆碱酯酶均有强烈的抑制作用,同样是嗜卷书虱比嗜虫书虱敏感。乙酰胆碱酯酶的动力学研究结果和离体酶活性测定相一致。聚丙烯酰胺凝胶电泳分析显示,4种杀虫剂离体条件下对2种书虱的酯酶同工酶的抑制能力有明显差异,其中敌敌畏的抑制力最强;但对不同同功酶的抑制趋势(对大分子的抑制似乎较强)是一致的。酶的敏感性分析结果与生测结果比较表明,2种书虱的耐药力差异与其乙酰胆碱酯酶和酯酶对药剂的敏感性无关。如要弄清耐药力机制,需做进一步研究。  相似文献

两种书虱微卫星富集文库的构建及比较   总被引:1,自引:0,他引:1  
利用链霉亲和素与生物素之间的强亲和性原理,将链霉亲和素偶联的磁珠与微卫星探针(AC)12、(TC)12、(ATC)8、(ATG)8、(AAC)8、(ATAC)6及(GATA)6退火结合后,再亲和捕捉含接头和微卫星序列的单链书虱基因组DNA限制性酶切目的片段,经PCR扩增形成双链后进行克隆、建库。结果表明本研究成功构建了嗜卷书虱和嗜虫书虱共13个微卫星富集文库,包括6个嗜卷书虱文库,7个嗜虫书虱文库,其平均阳性克隆率为71.17%。经检测发现共得到两种书虱260个微卫星位点。这两种书虱微卫星富集文库的建立和高多态性微卫星位点的筛选将为嗜卷书虱和嗜虫书虱的种群遗传与进化、基因连锁图谱构建、分子系统发育研究等提供大量分子遗传标记,对其在实仓中的持续控制提供遗传学信息。  相似文献

Abstract  One population of Liposcelis bostrychophila Badonnel (CA selected) was exposed to a controlled atmosphere (CA) (35% CO2+ 1% O2, balance N2) for 30 generations. Another population (control) was reared under natural atmospheric conditions. Reserves of triacylglycerol, polysaccharides and free amino acids were evaluated in adults of the CA selected and the control populations in generations F15 and F30. The utilization rate of triacylglycerol and polysaccharides in the CA exposure were also determined in generation F30. The results indicate that the reserves of triacylglycerol and polysaccharides increased significantly during selection for CA tolerance; the higher the tolerance level, the greater the reserves. A total of 15 free amino acids constituents were detected in both populations. The total amino acid content in the CA selected population was obviously higher than that in the control population. Exposure of this population to a controlled atmosphere was associated with a steady utilization of reserves. In contrast, the unselected population responded to the controlled atmosphere by accelerated utilization of triacylglycerol and polysachharides. Comparison of utilization rates during CA exposure showed that triacylglycerol is the main energy source, and polysaccharides contribute only a small extent to the metabolic energy supply.  相似文献

Abstract: Two populations of the psocid, Liposcelis bostrychophila Badonnel, were exposed to two CO2-enriched atmospheres (35% CO2 + 21% O2, and 55% CO2 + 21% O2, balance N2) for 30 generations. Controls were reared in normal atmospheres. The reserves of triacylglycerol and polysaccharides were evaluated in adults of the two experimental and the control populations in generations F15 and F30. The utilization rate of triacylglycerol and polysaccharides in the CO2-enriched atmospheres were also determined in generation F30. The results indicated that the reserves of triacylglycerol and polysaccharides increased significantly during selection for CO2 resistance; the higher the resistance level, the greater the reserves. Exposure of these populations to controlled atmosphere was associated with a steady utilization of the reserves. By contrast, the unselected population responded to controlled atmospheres by accelerated utilization of triacylglycerol and polysaccharides. How insects evolve resistance or counter-resistance against antagonists is a basic issue in the study of host-parasitoid coevolution. One of the factors that affect their coevolution is fitness costs of resistance and counter-resistance. Here, we assess fitness costs of resistance against the parasitoid Leptopilina victoriae in Drosophila bipectinata on the basis of selection experiments. We made a base population by mixing three geographic fly populations that differed in resistance. The established base population was divided into four populations, two for selection of resistance against a L. victoriae population and two for control. Resistance increased rapidly in response to selection and reached a very high level within four generations in the selected populations, while resistance of the control populations remained low during 20 generations. High resistance of the selected populations was maintained at least for 10 generations after selection was stopped. Both selected populations had lower female longevity than the control populations, and at least one of the selected populations had shorter thorax length, lower female desiccation tolerance and adult heat tolerance than both or either of the control populations. On the other hand, the selected populations had higher male starvation tolerance and longevity than the control populations. There were no significant differences in resistance against another population of L. victoriae and two other parasitoid species between the selected and control populations. These results suggest that the resistance against the L. victoriae population in D. bipectinata may incur some but not so high costs and act parasitoid-species- and/or parasitoid-population-specifically.  相似文献

Maximal aerobic metabolic rate (MMR) is an important physiological and ecological variable that sets an upper limit to sustained, vigorous activity. How the oxygen cascade from the external environment to the mitochondria may affect MMR has been the subject of much interest, but little is known about the metabolic profiles that underpin variation in MMR. We tested how seven generations of artificial selection for high mass-independent MMR affected metabolite profiles of two skeletal muscles (gastrocnemius and plantaris) and the liver. MMR was 12.3% higher in mass selected for high MMR than in controls. Basal metabolic rate was 3.5% higher in selected mice than in controls. Artificial selection did not lead to detectable changes in the metabolic profiles from plantaris muscle, but in the liver amino acids and tricarboxylic acid cycle (TCA cycle) metabolites were lower in high-MMR mice than in controls. In gastrocnemius, amino acids and TCA cycle metabolites were higher in high-MMR mice than in controls, indicating elevated amino acid and energy metabolism. Moreover, in gastrocnemius free fatty acids and triacylglycerol fatty acids were lower in high-MMR mice than in controls. Because selection for high MMR was associated with changes in the resting metabolic profile of both liver and gastrocnemius, the result suggests a possible mechanistic link between resting metabolism and MMR. In addition, it is well established that diet and exercise affect the composition of fatty acids in muscle. The differences that we found between control lines and lines selected for high MMR demonstrate that the composition of fatty acids in muscle is also affected by genetic factors.  相似文献

Responses of a salt tolerant and a normal population of an oilseed crop,Eruca sativa Mill. were assessed after four weeks growth in sand culture salinized with 0 (control), 100, 200, or 300 mol m?3 NaCl. The salt tolerant plants produced significantly greater dry biomass than the normal population. The populations did not differ significantly in leaf osmotic potential, relative water content and leaf soluble proteins. However, the tolerant population accumulated significantly greater amounts of soluble sugars, proline and free amino acids in the leaves compared with the non-tolerant population. It is established that leaf soluble sugars, proline, and free amino acids are important components of salt tolerance inEruca sativa.  相似文献

We have been exploring the behavioral response ot insect pests to heterogeneous distribution of toxins (low dose with refugia), and its genetic correlation with physiological tolerance to these toxins. A field-collected population of diamondback moth, Plutella xylostellu (L.) (Lepidoptera Plutellidae), from Celeryville, OH, was selected with permethrin to determine whether low heterogeneous doses could lead to increased susceptibility to permethrin by selecting indirectly on behavior. Two replicates of each of three selection regimes: uniform high concentration hypothesized to result in increased physiological tolerance, heterogeneous low concentration hypothesized to result in increased susceptibility through indirect selection on behavior, and a control with no exposure to permethrin, were maintained in 1-in3 cages in a greenhouse, for 33 generations. All life stages of the diamondback moth were exposed to the selection regimes, and new generations were started with a random selection of pupae from the previous generation. Lines selected with uniform high concentrations developed 76-fold levels of resistance to permethrin by the 17th generation, with little changes thereafter. For generations 1-20, lines selected with heterogeneous low concentrations remained slightly lower in LC50 but not significantly different from the unselected control lines. Based on confidence intervals from probit analyses, the LC50 of the lines selected with heterogeneous low concentration, however, were significantly lower than those of the control lines in generations 21-33. Our results demonstrate that selection on behavioral responses can result in greater susceptibility than no selection at all, despite exposure to the toxin and ample genetic variation and potential for increased physiological tolerance. The implications of our findings, which are based on selection scenarios that could take place in field situations, are that behavioral responses can prevent and even decrease the levels of resistance in insect populations, an important result with respect to resistance and resistance management.  相似文献

Abstract In order to understand the differentiation of amino acid requirments in host‐related populations of the brown planthopper (BPH), Nilaparvata lugens (Stål), we established three BPH populations by separately maintaining them on three rice varieties, i.e. TN1 (common susceptible control), Mudgo (containing resistant gene Bph 1) and ASD7 (containing resistant gene bph2) for over 30 generations, and then reared them on 20 holidic diets, each with distinct overall amino acids and ratio of essential amino acid (EAA) to nonessential amino acid (NEAA). The emergence rate, brachypter rate, nymphal duration and weight of newly molted adults were evaluated and compared. The results showed significant difference among the three populations in the effect of amino acid variation on BPH performance, ascending in an order of Mudgo population ASD7 population > TN1 pupulation. The results also indicated that the required optimum concentrations of overall amino acids by BPH populations on Mudgo and ASD7 were 4.0% ‐4.8% and 4.0%, respectively, higher than that of TN1 population (2.4% ‐ 3.2%). In addition, it was found that Mudgo and ASD7 populations were more sensitive to the concentration of EAAs than TN1 population. We propose that there is substantial differentiation in responses to dietary amino acid conditions among different host‐associated BPH populations and that such differentiation may be closely related to the induced virulence shift on resistant rice varieties.  相似文献

Abstract. The metabolic rates, as expressed by oxygen (O2) consumption, carbon dioxide (CO2) production, and losses in wet and dry weights, were examined for adults of three strains of the red flour beetle Tribolium castaneum (Herbst), during exposure to two modified atmospheres (MAs). Exposure of a strain selected for resistance over twenty-one generations to an atmosphere of 65% CO2, 20% O2 and the balance nitrogen (N2), termed a high carbon dioxide concentration atmosphere (HCC) and exposure of an unselected strain to HCC, showed considerable levels of aerobic metabolism during exposure. For the unselected strain water loss and mobilization of energy reserves were rapid and mortality was followed by rapid desiccation. For the HCC-resistant strain water balance was maintained and energy reserves were utilized more slowly over a prolonged period. Exposure of a strain selected for resistance over twenty-one generations to a low oxygen concentration atmosphere (LOC) of 0.5% O2 in N2, and an unselected strain to LOC, revealed that even at 0.5% O2, metabolism was largely aerobic in both strains. Maintenance of water balance was not a major factor causing mortality of either strain during exposure to LOC. In air, metabolic rates of both the resistant strains were lower than that of the unselected strain.  相似文献
