猪源双歧杆菌的分离与鉴定   总被引:1,自引:0,他引:1  
运用双歧杆菌选择性培养基,从1~4周龄乳猪的粪便中共分离纯化到52个菌株。通过染色镜检、生化反应、代谢产物分析、抗生素敏感性试验研究表明这些菌株均与双歧杆菌属特征相符。根据糖发酵试验初步鉴定结果,其中45个菌株为小猪双歧杆菌,5株为猪双歧杆菌,2株为嗜热双歧杆菌。部分菌株的急性毒性试验表明受试菌株对小白鼠无任何毒性副反应。  相似文献

目的从肉仔鸡肠道中筛选出耐酸、耐胆盐和耐消化酶的优良双歧杆菌,研究其生长特性,并优化其发酵参数,为转化生产力提供理论依据。方法通过无菌采样并分离得到多株双歧杆菌,对分离获得的双歧杆菌进行形态学、生化特性研究,然后采用牛津杯法,测定90株双歧杆菌对大肠埃希菌和沙门菌的抑制作用,采用改良MRS培养基,模拟鸡胃肠道逆环境,对其耐消化道特性进行研究,筛选出优良双歧杆菌,再进行生长特性研究及发酵参数优化。结果从肉仔鸡肠道分离出90株双歧杆菌,初步挑选出23株作为候选菌株,抑菌试验测得双歧杆菌B1、B2和B3具有良好的抑菌效果,然后经过耐受消化道逆环境试验,发现B2菌株的耐受能力最好,初步鉴定双歧杆菌B2为小鸡双歧杆菌,并将其定名为Bifidobacterium pullorum B2,对其生长特性的研究发现经18 h发酵细菌总数可以从8.3×105CFU/mL升高到1.3×109CFU/mL,运用优化的发酵培养基进行中试试验,发酵后的活菌数可达1.41×1010CFU/mL。结论本实验从肉仔鸡肠道中分离筛选并初步鉴定了Bifidobacterium pullorum B2,优化了制备Bifidobacterium pullorum B2发酵液的发酵条件,降低了生产成本。  相似文献

目的分离筛选出一株对甲胺磷敏感的双歧杆菌菌株。方法双歧杆菌菌株传代筛选,取一系列浓度(4.0、2.0、1.0、0.5和0.25μg/mL)的甲胺磷农药标准溶液30μL加入含TPY液体培养基的检测管溶液内,双歧杆菌菌株接种到检测管内,37℃厌氧培养12h,测定检测管液体吸光度(A)值。结果筛选一株甲胺磷敏感的短双歧杆菌菌株LJM-006,微生物抑制法的最佳接种浓度为107 CFU/mL,最低检测限为0.5mg/L。结论筛选出一株甲胺磷敏感的双歧杆菌LJM-006菌株,并可作为微生物法检测甲胺磷残留的菌株。  相似文献

一株携带质粒的人两歧双歧杆菌的分离与鉴定   总被引:3,自引:1,他引:2  
目的:分离携带天然质粒的人双歧杆菌.方法:用自制的改良型Blb双歧杆菌选择培养基,从人新鲜粪便分离双歧杆菌,对初步质粒检测阳性的单菌落通过糖发酵试验、(G C)mol%测定和16S rDNA序列分析,进行菌株鉴定.结果:筛选到一株携带天然质粒的人双歧杆菌,编号B200304,在1.0%琼脂糖凝胶上,测得质粒的相对分子质量约为22 kb.通过对该菌株的形态学观察和糖发酵试验等生理生化特征研究,证明该菌株为两歧双歧杆菌(Bifidobacterium bifidum);HPLC法测得其(G C)mol%为55.6,16SrDNA序列分析进一步证实该菌株为两歧双歧杆菌.结论:分离得到一株携带天然质粒的人两歧双歧杆菌新菌株.  相似文献

利用改良的乳酸细菌(MRS)培养基从健康人群的粪便中进行双歧杆菌的分离筛选。结果表明:在添加有5-溴-4-氯-3-吲哚-β-D-半乳糖苷(X-Gal)的MRS培养基上双歧杆菌的菌落呈典型的乳白色并带有蓝色,其他菌株的菌落呈白色或深蓝色。实验共分离出10株疑似菌株,经生理生化试验和Biolog自动微生物分析系统鉴定,结果发现其中4株为双歧杆菌,这4株菌中有3株为两歧双歧杆菌,1株为长双歧杆菌。和传统MBS培养基相比较,改良MRS培养基能显著提高筛选效率。  相似文献

为研制食品双歧杆菌制剂,从健康长寿老人肠道正常菌群分离并鉴定具有较高生物活性及保健作用的双歧杆菌菌株。对30份未明菌株标本,进行了分离,纯化,鉴定,经生物学特性,生化试验,同时做代谢产物分析,各菌均产生乙酸和乳酸为主。还做了DNA(G+Cmol%)含量测定,测定值在56~63范围。而确定3株为婴儿双歧、5株为青春双歧,共选出8株双歧杆菌,为选育优良的双歧杆菌菌株提供了依据。  相似文献

目的:观察大豆低聚糖和所含糖对肠道菌群中双歧杆菌、肠杆菌体外促生长作用。方法:按1%大豆低聚糖、0.36%水苏糖、0.52%蔗糖、0.11%棉子糖含量配制培养液,分别接种各试验菌株,在0、12、24小时测定各培养液吸光度A值(分光光度计.550nm)和pH值。结果:经24小时培养,加大豆低聚糖的双歧杆菌标准株和临床分离株培养液A值分别为>1.5和1.307,大于肠杆菌标准株和临床分离株的1.1和1.173,而其pH值小于肠杆菌;加水苏糖的双歧杆菌培养液A值为1.47,大于蔗糖的1.4和棉子糖的0.53。结论:大豆低聚糖促双歧杆菌生长作用大于促肠杆菌生长,水苏糖是促双歧杆菌增殖生长的主要成分。  相似文献

蒙古族儿童源益生特性双歧杆菌的筛选及鉴定   总被引:10,自引:0,他引:10  
【目的】双歧杆菌在人和动物胃肠道中发挥着重要的生理作用,然而双歧杆菌能否耐受胃酸、肠液及胆汁酸是影响活菌制剂益生效果的关键因素,本研究旨在从蒙古族儿童粪便中分离筛选出具有良好益生特性的双岐杆菌。【方法】本文采用双岐杆菌选择性培养基对样品进行分离纯化,并对菌株进行生理生化鉴定,以耐受人工胃肠液、耐受牛胆盐为手段对各菌株的益生特性进行评价,并且对B. animalisV9进行了16S rDNA分子生物学鉴定。【结果】本文从12位健康蒙古族儿童粪便中分离得到11株双歧杆菌,经传统生理生化试验鉴定为5株B. adolescentis:A1、H3、G4、A8、V10;3株B. longum:C6、C7、D11;1株B. pseudocatenlatum:B2;1株B. bifidum:G5;1株B. animalis:V9。B. animalis V9具有较强的耐酸性,在pH2.0的人工胃液中厌氧培养3h后存活率为92.4%,而其它10株双歧杆菌在此条件下的存活率均小于31.25%;B. animalis V9在pH2.0的人工胃液中厌氧培养3h后接入pH8.0的人工肠液中消化8h,存活率为99.7%,并且可以耐受0.3%的牛胆盐。进一步对V9菌株进行16S rDNA分子生物学鉴定,发现与Bifidobacterium animalis subsp. lactic BB12的同源性为99%。【结论】本研究结果显示B. animalis V9来源安全,并且具有良好耐酸、耐胆盐益生特性,有望在乳制品及保健类产品中得到广泛的应用。  相似文献

Bifidobacteria are important members of the intestinal microbiota and are considered to contribute to maintaining health. However, the level of bifidobacteria colonising the intestine of elderly subjects tends to be lower than in younger adults. Therefore, two Bifidobacterium longum strains, isolated from healthy elderly, were chosen for supplementation of the endogenous Bifidobacterium microbiota in the elderly. Bifidobacteria are generally regarded safe for human consumption. However, since the strains are intended for consumption by the elderly, whom may be more prone to disease, it is important to ascertain their safety. For this purpose, the strains were given to healthy adult volunteers. No side effects were reported and no undesirable changes observed in the immune parameters measured. Based on this study it appears that the two strains are well tolerated by human subjects and there are no reservations about their food use.  相似文献

The Lactobacillus and Bifidobacterium population in the feces of 26 animals (16 species) were studied by culture-dependent and culture-independent techniques. Lactobacilli were detected from a few herbivores, all carnivores and some omnivores. Lactobacillus johnsonii, Lactobacillus reuteri, Lactobacillus salivarius, Lactobacillus vaginalis and Lactobacillus ingluviei were the most dominant lactobacilli in carnivores. These species were, however, not predominant in herbivores and omnivores. Lactobacillus brevis, Lactobacillus casei, Lactobacillus parabuchneri, Lactobacillus plantarum, Lactobacillus sakei, Leuconostoc mesenteroides and Leuconostoc pseudomesenteroides, usually present in raw plant material, were present in omnivores but not in carnivores. Bifidobacteria were detected in only four herbivores and two omnivores. Bifidobacterium pseudolongum was the only Bifidobacterium species detected in herbivores. Bifidobacteria detected in the two omnivores are phylogenetically not closely related to known species and are possible novel species in the genus.  相似文献

Bifidobacteria are gaining commercial significance due to their probiotic properties. However, little is still known about the production of these bacteria and their behavior in bioreactors. Two Bifidobacterium longum strains were sensitive to light when grown in a transparent (glass) bioreactor under microaerophilic growth conditions (i.e. no gases added and slow mixing). The sensitivity was less clear the more anaerobic the initial conditions were. In a darkened bioreactor in microaerophilic conditions, the two strains grew with maximum specific growth rates of 0.36 h(-1) and 0.48 h(-1). In an illuminated bioreactor neither strain grew. In comparison, Lactobacillus reuteri was not sensitive to light under the same conditions.  相似文献

目前,由于抗生素的广泛、大量使用,耐药菌株急剧增多,加剧了耐药菌对人类健康和生态环境的威胁。有研究表明,益生菌自身含有的耐药基因或耐药质粒等可通过基因水平转移传递给人体肠道中的致病菌,导致耐药菌感染。随着双歧杆菌相关微生态制剂的广泛应用,通常以活菌形式进入人体的双歧杆菌,与肠道内原籍菌群混合生长,致使其携带的耐药性基因片段在肠道菌群中水平转移,从而导致某些致病菌具有耐药性。因此,研究双歧杆菌等益生菌的耐药性基因转移有着十分迫切的意义。本文总结了双歧杆菌的耐药性,分析了双歧杆菌的耐药机理,为进一步筛选安全的双歧杆菌菌株提供理论依据。  相似文献

应用TGGE技术分析人肠道中双歧杆菌的组成   总被引:5,自引:0,他引:5  
用温度梯度凝胶电泳(TGGE)技术结合16SrDNA克隆、测序,对健康人肠道中双歧杆菌的组成进行了分析。10例健康人肠道双歧杆菌的TGGE分析显示:人肠道内双歧杆菌的组成具有宿主特异性,不同人肠道双歧杆菌种的多样性和种类不同。通过对一健康儿童肠道双歧杆菌属特异性PCR扩增产物的测序及TGGE电泳行为的分析发现,该个体双歧杆菌TGGE图谱中的条带分别代表Bifidobacteriumbifidum、B.infantis、B.longum、B.adolescentis、B.pseudocatenulatum等种和一新种,其中B.pseudocatenulatum(假小链双歧杆菌)是大多数个体共有且较优势的种类。用传统培养方法只检出B.pseudocatenulatum和B.longum两种。基于双歧杆菌属特异性PCR基础上的TGGE方法结合16SrDNA克隆文库分析可较灵敏、直观地反映人肠道中双歧杆菌的组成。  相似文献

One hundred and twenty-two strains of Bifidobacterium and Lactobacillus species have been tested against 12 antibiotics and two antibiotic mixtures by a commercial system (Sensititre Anaero3; Treck Diagnostic Systems). The upper limits of some minimum inhibitory concentrations (MICs) were completed on MRS agar plates by the NCCLS procedure. All strains were sensitive to chloramphenicol and imipenem and most of the strains were resistant to metronidazole. Bifidobacteria isolates were susceptible to cefoxitin, whereas about half of the lactobacilli were resistant. Approximately 30% of the Bifidobacterium isolates were resistant to tetracycline, as well as five Lactobacillus strains belonging to four different species. None of the tested Bifidobacterium isolates was resistant to vancomycin, whereas a species-dependent resistance was found among the lactobacilli. Single strains of Bifidobacterium longum, Bifidobacterium pseudocatenulatum, Lactobacillus acidophilus, Lactobacillus rhamnosus, and Lactobacillus brevis were resistant to erythromycin and/or clindamycin. Most of the observed resistances seemed to be intrinsic, but some others could be compatible with transmissible determinants.  相似文献

Bifidobacteria from breast-fed infants, formula-fed infants, or premature babies fed by parenteral methods were isolated and identified. The persistence of these microorganisms in the gastrointestinal tract of mice, after oral administration, was studied to determine the optimal dose and frequency of translocation to the liver and spleen. The rate of isolation among infants varied between 19 and 82% depending on the origin of the samples, with the highest values seen in breast-fed babies. The predominant species found in all cases was Bifidobacterium adolescentis. The optimal dose for oral administration of bifidobacteria to mice was 10(7) cells per day per animal for up to 2, 5, or 7 days. These bacteria remained up to 5 days postfeeding, even if feeding was interrupted. The results of bacterial translocation assays showed differences for the different strains and doses tested.  相似文献
