本文报道大鼠水盐负荷时,其心房特殊颗粒(ASG)的数目及钙含量发生相关变化。给大鼠禁水5d或饮2%NaCl液4d造成水盐负荷,用电镜形态计量法计数ASG,以反映心房钠尿肽(ANP)的分泌水平,电镜X射线显微分析法测定ASG中钙、硫含量及肌质同终末池(TSR)中钙含量。结果显示,禁水引起ANP分泌减少时,ASG的数密度增加(6.02±2.30增至9.97±3.21个/μm3),同时伴有钙含量增加(64±16增至92±18mmol/kg);饮盐水引起ANP分泌增加时,ASG的数密度减少(6.02±2.30减至2.96±1.62个/μm3),巨伴有钙含量减少(64±16减至38±22mmol/kg),但水盐负荷对TSR中钙浓度无任何影响。据此推论ASG作为细胞内钙库,借某种机制调控其中钙的释放而参与ANP的刺激分泌耦联过程。  相似文献

通过形态计量学和免疫组织化学方法发现胰岛素诱导低血糖大鼠心房肌细胞核周区特殊颗粒(ASG)的体密度、面数密度和数密度及平均直径均高于对照组(P<0.05),但高尔基复合体各参数与对照组比较没有差别(P>0.05)。实验组的心房利钠肽(ANP)的免疫反应强度比对照组强(P<0.001)。提示胰岛素诱导低血糖对心房利钠肽的释放具有抑制作用,表明ANP作为生理和病理调节递质与代谢刺激相拮抗。  相似文献

本实验利用冠脉内给药和离体血管灌流等方法观察比较了降钙素基因相关肽(CGRP)和心房肽(ANP)对犬冠脉循环的影响及其对犬大小冠状动脉的舒张作用。结果显示,CGRP和ANP均能明显增加冠脉血流量、降低大小冠脉阻力,两者均呈剂量依赖性舒张大小脉冠脉血管。ANP的作用显著小于CGRP,其中大冠脉对两者的反应性显著小于小冠脉,CGRP和ANP对冠脉的舒张作用均无内皮依赖性。结果提示,CGRP和ANP直接  相似文献

本文采用免疫组化方法观察高血钾诱发的心律失常豚鼠(20例)和猕猴(3例)心脏心钠素(ANP)和抗心律失常肽(AM)免疫反应。结果显示:心律失常组豚鼠和猕猴心脏的ANP和AAP免疫反应比正常组的明显(P<0.05),心房肌的ANP和AAP免疫反应比传导系的明显,但心室肌未见棕色反应产物。ANP和AAP免疫反应在心脏不同部位反应强度不同可能与心脏传出神经和心肌钙通道的分布有关。  相似文献

目的和方法:血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)是新近确定的一种特异作用于血管内皮细胞的活性肽。最近发现正常心肌细胞可表达VEGF,高血压肥大心脏心肌VEGF及基因表达增强,但对运动性心肌肥大时的变化尚不清楚。本实验采用免疫组化和分子杂交方法,对游泳运动10周大鼠稳定期肥大心脏心肌VEGF及其基因表达进行研究。结果:WistarKyoto(WKY)大鼠、自发性高血压大鼠(spontaneouslyhypertensiverats,SHR)和运动大鼠心肌细胞浆内均有特异性VEGF染色颗粒,但运动大鼠心肌细胞胞浆内染色颗粒增加最明显。Northern分子杂交结果表明三组大鼠心肌均有VEGFmRNA表达,其中SHR表达最强,运动大鼠比WKY大鼠增强,但低于SHR。结论:目前对这一结果的生理意义还不清楚,推测可能与心肌肥大时细胞间质血管增生有关。  相似文献

本研究运用透射电镜及立体计量学方法结合免疫组化技术对糖尿病大鼠右心耳肌细胞心房特殊颗粒(ASG)和心房肽(ANP)免疫反应强度进行了观察和定量研究.实验动物为体重150~250克之间的SD大鼠,糖尿病的诱导采用四氧嘧啶(150mg/kg)右腹股沟区皮下注射.电镜超微结构发现糖尿病未治疗组均出现线粒体肿胀、肌原纤维缩短等病理性改变.ASG膜溶解、排空等现象多见,ASG体密度明显减少.免疫组化分析表明糖尿病未治疗组心肌细胞ANP免疫反应强度减弱.胰岛素治疗组各指标与对照组相比均无显著差别.  相似文献

收集胚胎标本共24例(经临床证实孕妇无心血管疾病),用透射电镜观察各胎龄心脏各部位心肌纤维中心特殊颗粒(ASG)分布及含量变化,同时结合免疫组织化学技术对心脏各部位心肌纤维内心钠素的表达进行研究。结果表明,胚第5周时,心肌纤维便有ASG出现,随着胎龄的增长,心房与心耳的颗粒数量逐渐增多。而心室的颗粒数量却逐渐减少,在同一胎龄,心房内的颗粒数量多于心室,心耳ASG多于心房,免疫组织化学的研究结果表明,胚第7周时,心房与心耳内可出现心钠素样免疫反应阳性的心肌细胞,其阳性反应的变化规律与ASG的变化规律一致。心室中未发现阳性反应。  相似文献

用软脑膜铺片技术,兔疫组织化学方法及全自动图像分析仪对17周龄自发性高血压大鼠脑血管床心钠素受体分布密度进行研究、结果表明,自发性高血压大鼠脑小动脉及细动脉壁上心钠素受体分布密度明显低于正常血压大鼠,P<0.01。此外,在细小动脉分叉部位心钠素受体分布密度增加,而自发性高血压大鼠中其分叉部位的受体密度明显低于正常血压大鼠。从实验结果显示,我们试用的软脑膜铺片技术显然弥补了组织切片中从血管横断面观察心钠素受体分布的局限性,清楚地展现了整个血管床心钠素受体分布特征。根据实验结果提示这些阻力性动脉心钠素受体密度减少与细小动脉舒血管反应减弱,外周阻力增加,血压升高有着明显的关系;尤其是对调节外周循环血量和外周阻力充当闸门作用的细小动脉分叉部位,心钠素受体密度降低这一特征在高血压发生机制中可能起着重要作用。  相似文献

Summary Subcellular structures of atrial myoendocrine cells in the rat heart and plasma concentrations of atrial natriuretic peptides (ANP) were examined at six evenlyspaced time points over 24 h, using morphometric techniques and radioimmunoassay.Myofibrils and mitochondria of the cells occupied 73.3% of the cytoplasm; 2% of the cytoplasm was occupied by secretory granules, rough endoplasmic reticulum and Golgi complexes, structures characteristic of endocrine cells. Plasma ANP concentration was maximal at 08.00 h, when the individual volume of secretory granules was minimal. The numerical density of secretory granules was increased at 12.00 h. The plasma ANP concentration was minimal at 20.00 h, when the numerical density was minimal and the individual volume was maximal. The fluctuation in plasma ANP concentrations over 24 h was thus parallel to that in the numerical densities of secretory granules and inverse to that in individual volumes.These results suggest that in rats the secretory activity of atrial myoendocrine cells increases at the beginning of the resting period, whereas it decreases at the beginning of the active phase.  相似文献

Summary Fine structural and cytochemical studies were performed to examine the nature of three types of specific granules found in the atrium of lamprey; specific granules of the atrial muscle cell (ASG), interstitial cell granules (ICG) and endocardial endothelial granules (ESG).Ultrastructurally, ASG and ICG appeared quite similar in size, shape and electron opacity, while ESG were much larger and less dense in opacity than the other two.None of the granules showed positive DAB reaction or acid phosphatase reaction. Only ICG revealed positive chromaffin reaction, which agreed with formaldehyde induced green fluorescence along the atrial lumen. Phosphotungstic acid at low pH stained ICG and ASG strongly positive, and ESG weakly positive. Pronase treatment in Epon sections for 24h digested ASG alone, whereas in glycol-methacrylate embedded sections, ESG were digested first, ASG were digested thoroughly after 30 min, but ICG were not digested completely after 90 min.From these results it can be concluded that the three types of specific granules have different constituents. ESG consist of protein with some polysaccharides; ASG are composed of protein carbohydrate complexes and lack catecholamines; ICG contain catecholamine as well as protein carbohydrate complexes.This work was supported by a grant from the Ministry of Education, JapanThe authors would like to express their gratitude to K. Caveolae may act as mechanosensors and function as binding sites for calcium ions. The intracaveolar localization of atrial natriuretic peptide (ANP) derived from the direct interaction of atrial granules with caveolae has been demonstrated. The aim of this study was to define the effect of caveolae on ANP secretion induced by stretch and angiotensin II. The isolated perfused beating atria from Sprague-Dawley rats were used. To disrupt caveolae, 10 mM methyl-β-cyclodextrin (MbCD) was applied for 1 h and the number of caveoli were markedly decreased. MbCD increased basal ANP secretion and atrial diastolic pressure. The molecular profile of ANP in perfusate from control atria showed mainly one major peak corresponded to synthetic ANP whereas that from MbCD-treated atria showed two major immunoreactive peaks corresponded to synthetic rat ANP and proANP. High atrial stretch induced by elevating the height of outflow catheter from 5 cm H2O to 7.5 cm H2O increased atrial contractility and ANP secretion. The response of ANP secretion to high stretch was attenuated in MbCD-pretreated atria. Pretreatment with MbCD abolished angiotensin II-induced suppression and losartan-induced stimulation of ANP secretion. However, the effect of angiotenisin (1-7) on ANP secretion was not altered by MbCD treatment. The expression of angiotensin II type 1 receptor protein was reduced by MbCD treatment. These data suggest that caveolae are essential for angiotensin II type 1 receptor-mediated ANP secretion and relate to the processing of proANP.  相似文献

The effect of short lasting hypoxia on blood pressure, plasma atrial natriuretic peptide level and number of specific atrial granules were studied in 26 male spontaneously hypertensive and 24 normotensive Wistar rats. A great difference occurred in ANP secretion between hypertensive and normotensive rats. In the hypertensive animals elevated plasma ANP concentration (130 +/- 27 pg/ml) and decreased granularity in the right atria (73 +/- 2) were found on the first day of hypoxia with a slight elevation in urinary sodium content versus normotensive controls. The blood pressure also decreased although not significantly (190 +/- 14 mm Hg). In Wistar rats increased plasma ANP (130 +/- 34 pg/ml) and decreased atrial granularity versus normotensive controls (72 +/- 10 in the left and 113 +/- 16 in the right atrium) were observed only on the third day of hypoxia without changes in blood pressure and natriuresis. In SHR the rapid but short timed ANP release might be of right atrial origin and probably the consequence of a continuous and perhaps increased secretion of the peptide in normoxic conditions too. In Wistar rats the plasma ANP elevation could be secondary due to the increased plasma level of different vasoactive hormones to hypoxia. In the altered effect of ANP in hypertensive and normotensive hypoxic animals, structural and functional changes in the vascular bed may play a role.  相似文献

Tissue levels of atrial natriuretic polypeptide (ANP) messenger RNA (ANPmRNA) and ANP in the rat heart were measured simultaneously. In Wistar rats, ANPmRNA of the same size (approximately 0.95 kbp) was detected in all four chambers of the rat heart. The ANPmRNA level was the highest in the right atrium, and the left atrial level was slightly lower than the right atrial level. Ventricular levels were more than two orders of magnitude lower than atrial levels. Tissue ANP concentrations of four chambers were roughly parallel to ANPmRNA levels. In spontaneously hypertensive rats (SHR) with the elevated plasma ANP level, the ANPmRNA level in the left atrium was substantially increased. The left/right ratio of atrial ANPmRNA level in SHR (150%) was higher than that in control Wistar Kyoto rats (WKY) (90%). In contrast, the left/right ratio of atrial ANP concentration was decreased in SHR (44%) compared with that in WKY (84%). The ratio of ANP to ANPmRNA levels in the left atrium of SHR was about three times smaller than that in the right atrium of SHR, and those in bilateral atria of WKY. These results indicate that the biosynthesis and secretion of ANP from the left atrium is preferentially increased in SHR. Thus, simultaneous determination of ANPmRNA and ANP levels is a refined strategy of investigation for the biosynthesis, storage and secretion of ANP.  相似文献

High concentration of atrial natriuretic peptide (99-126) (ANP) receptors were localized by quantitative autoradiography in superior cervical and stellate ganglia from young and adult Wistar Kyoto (WKY) rats. ANP increased cyclic GMP formation in stellate ganglia from adult rats. Both young and adult spontaneously hypertensive rats (SHR) had a much lower number of ANP receptors in the sympathetic ganglia. In spite of low receptor concentration, the cyclic GMP response to ANP in SHR was unchanged. These results suggest the existence of physiologically active ANP receptors in the rat sympathetic ganglia. These receptors may also be involved in the pathophysiology of spontaneous hypertension.  相似文献
