目的:构建一种能结合钛表面的载药纳米粒及钛-载药纳米复合材料的组装和性质研究。方法:(1)多巴胺修饰的非离子表面活性剂多巴胺-泊洛沙姆188(Dop-Poloxamer188)的合成和检测;(2)Dop-Poloxamer188作为表面活性剂、PLGA作为油相基质,制备纳米粒及纳米粒载药和表征;(3)钛片的预处理及钛片与修饰后的纳米粒的结合;(4)纳米粒修饰后的钛表面的表征。结果:新合成的Dop-Poloxamer188在285 nm左右有紫外吸收峰,说明多巴胺成功的修饰在Poloxamer188的两端;Dop-Poloxamer188能和PLGA制备出很好的纳米粒,平均粒径在110 nm左右,PDI小于0.1;多巴胺修饰的纳米粒与钛片通过简单的浸渍过程结合后,通过水接触角、场发射扫面电镜(Fe-SEM)、荧光显微镜、X射线光电子能谱(XPS)等仪器检测都显示多巴胺修饰的纳米粒成功且牢固的修饰在钛片表面。结论:成功达到钛表面的载药纳米粒修饰的目的,为钛种植体的载药系统提供了新的思路和方法。  相似文献

摘要 目的:本研究旨在制备用于肿瘤可视化光治疗的多功能Mn3O4@CuS核壳型纳米粒,在磁共振成像的引导下,使用近红外光定点辐照,实现局部光热消融治疗。方法:(1)采用高温热解法制备油胺稳定的Mn3O4纳米粒,在其表面构建CuS壳层,并进行聚乙二醇修饰,得到分散于水相中的Mn3O4@CuS核壳型纳米粒。(2)通过透射电镜、紫外可见近红外吸收光谱等方法对该纳米粒进行理化性质表征,并研究其体外磁共振成像、光热升温等性能。结果:制备的水相分散的Mn3O4@CuS纳米粒,粒径均一且分散性较好,形态为近圆形,粒径为9.30±2.29 nm;紫外可见近红外吸收光谱图表明Mn3O4@CuS纳米粒在近红外区有较强吸收,最大吸收峰位于1100~1200 nm范围;磁共振成像分析结果可计算出Mn3O4@CuS纳米粒的纵向弛豫率r1为1.662 mM-1s-1,表明其具有较好的磁共振增强造影效果;光热升温曲线显示Mn3O4@CuS纳米粒可在785 nm近红外激光下升温至73.5 ℃,具备较好的光热治疗潜力。结论:本文成功制备出水相分散的Mn3O4@CuS核壳型纳米粒,具有良好的磁共振造影成像性能和光热升温效应,有望应用于磁共振成像引导下的肿瘤可视化光治疗。  相似文献

目的:建立基于聚(乳酸-羟基乙酸)纳米粒(PLGA)载DNA的基因转染体系,比较用空白聚(乳酸-羟基乙酸)纳米粒(PLG-A-E)吸附质粒DNA和用分枝PEI修饰后的PLGA纳米粒(PLGA-BPEI)吸附质粒DNA优缺点。方法:用乳化蒸发法制备纳米粒,对纳米粒进行表征研究,包括包封率、Zeta电位、粒径大小、稳定性,用荧光显微镜观察它们对NIH3T3和HEK293细胞的转染效率,用MTT检测对它们细胞的毒性。结果:制备了两种基于PLGA的纳米粒,PLGA-E和PLGA-BPEI粒径大小为200-270nm,zeta电位为0-30mV,在血清和不同的pH值时两者均较稳定,转染效率PLGA-BPEI较PLGA-E高,且释放时间早,但前者较后者对细胞毒性大。结论:这两种基于PLGA纳米粒均能有效转染质粒DNA,它们存在不同的优缺点,应根据不同需要进行选择。  相似文献

目的:通过制备RGD/FA双靶纳米金考察其与高表达整合素与叶酸受体B16细胞的协同靶向成像与热疗作用;方法:采用功能化PEG分子将靶向小分子RGD与叶酸通过强健Au-S键连接至纳米金棒表面,利用激光共聚焦与808 nm近红外激光器评价修饰纳米金的协同靶向作用;结果:RGD与叶酸分子被成功连接于纳米金表面,且双靶纳米金对小鼠黑色素瘤细胞具有较好的协同靶向作用;结论:同时靶向同一肿瘤细胞的不同表位,可克服单一靶向功能化纳米粒子难以在肿瘤位点有效积累的问题,本研究为多功能纳米金棒在临床肿瘤早期诊断与光热治疗中的应用提供研究基础。  相似文献

目的:探究纳米金壳介导的光热效应对骨肉瘤干细胞的杀伤作用。方法:采用无血清悬浮培养法富集骨肉瘤干细胞,通过实时荧光定量PCR检测所富集细胞CD133、SOX2、NANOG、OCT4 mRNA的相对表达量以进行鉴定。将纳米金壳与骨肉瘤干细胞共培养,应用波长808 nm的近红外激光器(1.5 W/cm~2)进行照射以激发光热效应,分别用CCK8法检测光热疗法对骨肉瘤干细胞的增殖抑制率,Annexin C-Fitc/PI双染法检测光热疗法对骨肉瘤干细胞凋亡的影响。结果:成功富集了高表达CD133、SOX2、NANOG、OCT4 mRNA的骨肉瘤干细胞;纳米金壳介导的光热效应显著抑制了骨肉瘤干细胞的增殖率,当纳米金壳浓度在0~250μg/mL范围内时,增殖抑制率与浓度呈正相关;细胞凋亡结果显示,纳米金壳结合近红外照射组细胞凋亡率显著高于对照组。结论:纳米金壳介导的光热效应对骨肉瘤干细胞增殖有明显抑制作用,主要诱导骨肉瘤干细胞发生中晚期凋亡。  相似文献

目的:制备粒径均一且稳定、载药率和包埋率较高的聚合物脂质纳米球。方法:将HSPC(氢化大豆卵磷脂)与PLGA(聚乳酸-羟基乙酸共聚物)两种材料结合,利用超声复乳法制备聚合物脂质纳米球,采用响应面法优化最佳制备工艺;以HSPC(氢化大豆卵磷脂)与PLGA(聚乳酸-羟基乙酸共聚物)的比例、PVA浓度、超声功率为条件优化制备参数,以粒径为响应值。结果:优化后的最佳工艺参数为:HSPC与PLGA的比例为1:10,PVA浓度为0.66%,超声功率为51.34%(205.36 W)。结论:按最优工艺制备出的聚合物脂质纳米粒的粒径为230 nm左右,多分散系数(PDI)值为0.057,与预测值偏差较小,且粒径分布均一,可作为蛋白及多肽类药物的递送载体。  相似文献

近红外(NIR)光诱导的光热治疗(PTT)因其无创、非侵入、毒副作用低、可精准靶向治疗等特性,已成为肿瘤精准治疗的新型手段。凭借其独特的表面等离激元共振(SPR)特性及其高效的光热转换效率、生物毒性与良好的光稳定性,金纳米颗粒(Au NPs)已成为理想的光热治疗剂。而高质量成像技术是实现有效光热治疗的可靠有力的工具,尤其是多模态成像技术,比起单一成像方式具有更卓越的性能,为更全面、更精准的肿瘤成像提供了可能,显著提高了非侵入性医学治疗的潜力。NIR光激发的稀土上转换纳米颗粒(UCNPs),因其丰富的4f电子结构展现出磁性、荧光、X射线衰减和放射等多功能特性,使其作为造影剂在多模态成像领域展现了重要的应用前景。因此,构建NIR光诱导的Au NPs/UCNPs复合纳米体系,可用于多模态成像引导下的光热治疗,有望成为癌症诊疗的一种新策略。本文简单介绍了Au NPs、UCNPs的光学特性,重点综述了NIR光诱导的UCNPs-Au NPs(纳米壳、纳米棒、纳米团簇)复合纳米体系在癌症光热治疗领域的最新研究进展,并对其实现诊疗一体化的未来进行了展望。  相似文献

目的:利用poly(lactide-co-glycolide)(PLGA)和poly(styrene-co-4-styrene-sulfonate)(PSS)制备带负电荷的牛血红蛋白纳米粒,并对其载氧性能和体外性能进行评价.方法:采用溶剂蒸发法制备出空白的PLGA-PSS的空白纳米粒,通过改变pH值,吸附牛血红蛋白,从而制备出牛血红蛋白PLGA纳米粒.通过TEM、粒径、Zeta电位、包裹率和载药量、体外释放及其携氧能力对该纳米粒进行了综合分析.结果:Hb-PLGA-PSS-NPsTEM电镜下呈类球形,平均粒径为226.8±23.4nm,Zeta电位为-68.62 mV,优化条件后最大包裹率约为99.3%,载药量约为28.6%,在37℃,pH7.4的PBS溶液中释放缓慢.通过对Hb结构的分析表明此工艺未对蛋白的结构造成影响,体外携氧实验测量了P50(29mmHg)和Hill系数(2.036),结果说明该Hb纳米微囊具有良好携氧功能.结论:成功制备了一种Hb-PLGA-PSS-NPs纳米粒,稳定性好,具有很好的携氧能力.  相似文献

观察以聚乳酸 (D ,L-polylacticacid,PLA)为材料制备、经吐温-80(T-80)表面改性的纳米粒对血脑屏障的穿透效果并探讨其机制 ,分别将FITC-Dextran、叶绿素铜作为PLA纳米粒的示踪标记 ,应用荧光显微镜、透射电镜及分析电镜观察经静脉注射入小鼠体内的PLA纳米粒在脑组织中的分布、穿透血脑屏障的特性。荧光显微镜观察到小鼠脑组织中散在及沿毛细血管壁分布的荧光颗粒 ,透射电镜可观察到小鼠脑毛细血管内皮细胞及周围脑组织中圆形或类圆形的外源性纳米粒 ;进一步采用分析电镜对颗粒处组织进行能谱分析证实其为叶绿素铜标记的PLA纳米粒。证实了T-80修饰的PLA纳米粒具有穿透血脑屏障的特性 ,机制可能是毛细血管内皮细胞的胞吞转运作用 ,同时 ,为研究纳米粒在组织内的定位提供了新的标记方法.  相似文献

目的:采用PLGA-PEG为聚合材料,制备RGD修饰包载超顺磁性四氧化三铁纳米粒子(RGD-NP—Fe3O4),用于脑胶质瘤细胞靶向核磁共振成像纳米探针。方法:采用沉淀法制备RGD修饰的栽超顺磁性纳米粒,考察纳米粒的粒径,电位等理化指标以及细胞毒性。通过细胞以及肿瘤球摄取实验,考察RGD.NP—Fe304的脑胶质瘤细胞靶向性。结果:制备得到的RGD-NP-Fe3O4粒径在85±7.5nm,电位为18+1.15mV。纳米粒浓度在300μg/mL范围内,对脑胶质瘤细胞均无显著毒性。经过RGD修饰后脑胶质瘤细胞U87对纳米粒的摄取效率大大提高,纳米粒穿透肿瘤球能力显著增强。结论:RGD修饰包载超顺磁性氧化铁纳米粒是一种潜在的高效的脑胶质瘤细胞靶向诊断纳米探针和靶向给药系统。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献
