环介导等温扩增(loop-mediated isothermal amplification,LAMP)是一种新式核酸扩增技术,它依靠一种具有链置换活性的DNA聚合酶和2对特殊设计的引物,不需要反复的温度循环和昂贵的仪器设备,在等温条件下即可高效快速地完成扩增反应,目前已广泛应用于细菌、病毒、寄生虫等病原体的检测,及动物胚胎性别的鉴定。该文总结了LAMP技术的基本原理、相对于传统核酸检测技术的优点、产物的检测方法及其临床应用,最后指出LAMP目前存在的不足以及采取的相应措施,并对其发展前景进行了展望。  相似文献

环介导等温扩增(loop-mediated isothermal amplification,LAMP)是一种新式核酸扩增技术,它依靠一种具有链置换活性的DNA聚合酶与2对特殊设计的引物,在等温条件下即可高效快速地完成扩增反应。相较于传统扩增检测方法,LAMP技术具有特异性强、灵敏度高、操作简单快速等优点,更能在现场快速检测和基层应用中广泛推广,目前LAMP技术已广泛应用于植物病害检测、动物病害检测、食品安全检测等领域。基于此,简要介绍了LAMP技术的基本原理、反应产物的检测方法,重点阐述了LAMP技术的改进与发展,综述了近年来其在科研生产中的应用进展,并对其发展前景进行了展望,以期为LAMP技术的进一步发展提供合理的研究方向。  相似文献

The polymerase chain reaction(PCR) has been a reliable and fruitful method for many applications in ecology.Nevertheless, unavoidable technical and instrumental requirements of PCR have limited its widespread application in field situations. The recent development of isothermal DNA amplification methods provides an alternative to PCR, which circumvents key limitations of PCR for direct amplification in the field. Being able to analyze DNA in the pollen cloud of an ecosystem would provide very useful ecological information, yet would require a field-enabled, high-throughput method for this potential to be realized. Here, we demonstrate the applicability of the loop-mediated DNA amplification method(LAMP), an isothermal DNA amplification technique, to be used in pollen analysis. We demonstrate that LAMP can provide a reliable method to identify species from the pollen cloud, and that it can amplify successfully with sensitivity down to single pollen grains, thus opening the possibility of field-based, high-throughput analysis.  相似文献

环状介导等温DNA扩增(LAMP)技术是一种新的核酸扩增方法,它能够高特异性、高效、快速地进行核酸的扩增。利用LAMP法检测乙型肝炎病毒(HBV),能够在等温条件下于1h内将少量的基因拷贝数扩增至10^9,在对65份临床标本的检测中显示了较高的特异性。与现有的PCR技术相比,LAMP法更加简便快速,且在等温条件下进行,不需要复杂的仪器设备,为临床检测乙肝病毒提供了一个快速筒便的新方法。  相似文献

环介导等温扩增(Loop-mediated isothermal amplification,简称LAMP)是利用能识别靶序列上6个位点的4个特殊设计的引物和一种具有链置换活性的DNA聚合酶,在恒温条件下,特异、高效、快速地扩增核酸的新技术。该技术在1h内扩增效率可达到109-1010个数量级,扩增产物是一系列反向重复的靶序列构成的茎环结构和多环花椰菜样结构的DNA片段混合物,电泳后在凝胶上显现出由不同大小的区带组成的阶梯式图谱。近年来LAMP技术以其特异性强、等温灵敏、操作简单、产物易检测等优点已经应用于食品安全检测领域的多个方面。  相似文献

A method for nucleic acid amplification, loop-mediated isothermal amplification (LAMP) was employed to develop a rapid and simple detection system for periodontal pathogen, Porphyromonas gingivalis. A set of six primers was designed by targeting the 16S ribosomal RNA gene. By the detection system, target DNA was amplified and visualized on agarose gel within 30 min under isothermal condition at 64 degrees C with a detection limit of 20 cells of P. gingivalis. Without gel electrophoresis, the LAMP amplicon was directly visualized in the reaction tube by addition of SYBR Green I for a naked-eye inspection. The LAMP reaction was also assessed by white turbidity of magnesium pyrophosphate (a by-product of LAMP) in the tube. Detection limits of these naked-eye inspections were 20 cells and 200 cells, respectively. Although false-positive DNA amplification was observed from more than 10(7) cells of Porphyromonas endodontalis, no amplification was observed in other five related oral pathogens. Further, quantitative detection of P. gingivalis was accomplished by a real-time monitoring of the LAMP reaction using SYBR Green I with linearity over a range of 10(2)-10(6) cells. The real-time LAMP was then applied to clinical samples of dental plaque and demonstrated almost identical results to the conventional real-time PCR with an advantage of rapidity. These findings indicate the potential usefulness of LAMP for detecting and quantifying P. gingivalis, especially in its rapidity and simplicity.  相似文献

目的:建立一种基于环介导等温核酸扩增技术(Loop-mediated Isothermal Amplification,LAMP)的恶性疟原虫高灵敏可视化闭管检测方法。方法:针对恶性疟原虫核糖体DNA的序列保守区设计LAMP引物,通过优化LAMP体系中的Mg2+、甜菜碱浓度和反应温度等因素,建立环介导等温扩增法;并结合蜡封反应管对产物进行检测,检测结果可直接通过肉眼观察SYBR Green I荧光显色进行判定。结果:本方法可检测到70个拷贝/管的恶性疟原虫核酸片段,并具有高特异性,可区分检测常见的血液病毒。该法具有如下优点:1、整个反应恒温进行,无需热循环仪;2、闭管检测,极大降低了扩增产物交叉污染的风险;3、检测速度快,整个检测过程只需30 min。结论:该法的建立为恶性疟原虫的现场快速筛检提供了一种简便、高灵敏、高特异的工具。  相似文献

Loop-mediated isothermal amplification (LAMP), in which a specific DNA sequence can be directly amplified under isothermal conditions, yields DNA in large quantities of more than 500 microg/ml. We have developed a method to isolate single-stranded DNA fragments from LAMP products that are stem-loop DNAs with several inverted repeats of the target DNA. This method requires the TspRI restriction enzyme, a primer hybridized to the 3' overhanging sequence at its cleavage site, and a DNA polymerase with strand displacement activity. The LAMP products are digested with TspRI and are then extended using the primer, producing the strand-specific DNA fragments. All processes, from LAMP reaction to primer extension, can be carried out at the same temperature. The use of strand-specific DNA would be conducive for detection by hybridization technique such as DNA microarrays.  相似文献

环介导等温扩增技术(Loop-mediated isothermal amplification, LAMP)因其扩增速度快、灵敏度和特异性高、仪器要求低等优点而被广泛应用于核酸诊断领域。为充分利用LAMP技术优势、提高诊断检测的效率与可靠性、扩展其应用范围,同时节约试剂成本,近年来多重LAMP技术的研究成为一大热点。常规的LAMP扩增产物检测方法多数以聚合反应的双链DNA产物或其副产物为基础,只能判断有无扩增反应发生,而难以识别多重扩增产物的靶标来源及其特异性。为实现多重扩增产物的高特异检测,各国学者通过对该技术巧妙的改进或与其他技术相偶联,发展了一系列多重LAMP扩增检测技术。然而上述狭义的多重LAMP技术依然存在因引物间相互干扰、扩增效率存在差异而引发歧视性扩增的局限,限制了多重扩增的重数。近年研究活跃的微型扩增技术以其实现多个平行、互不干扰的小体积单重扩增的技术优势打破了这一局限,由此产生了新型的广义多重LAMP扩增技术。这些技术还具有试剂消耗少、自动化程度较高、交叉污染风险更小以及更适合对较多靶标进行现场快速检测等优势。本文分别从狭义多重LAMP的方法原理及其扩增反应体系优化、广义多重LAMP的方法原理以及多重LAMP技术在诊断检测中的应用等方面对近年来多重LAMP技术的研究进展进行了综述。  相似文献

乌梢蛇作为一种名贵中药材,市面上伪品较多,干燥熏黑处理后的样品,更是真伪难辨。本研究致力于开发一套基于环介导等温扩增(loop-mediated isothermal amplification,LAMP)为基础的快速筛查乌梢蛇的方法。本研究以乌梢蛇12srRNA基因序列为基础设计并筛选出1套LAMP引物。通过调整反应条件,建立了对乌梢蛇LAMP的检查方法。结果显示,62℃下连续反应15min左右出现典型的"S"型荧光吸收曲线,实现了对乌梢蛇12srRNA基因序列的特异扩增。根据LAMP灵敏度高的特点,本研究简化了DNA的提取方法,缩短了检测的时间。相对于常规的PCR方法,本研究建立的以快速DNA提取为基础的乌梢蛇LAMP快速筛查方法具有简单、快速、灵敏、对设备要求低等特点,适用于对中药材乌梢蛇的快速筛查。  相似文献

本文的目的在于建立用于临床检测残翅病病毒(Deformed wing virus,DWV)的等温环介导扩增技术(Loopmediated isothermal amplification,LAMP),为该疾病的预防和控制提供理论依据。在DWV基因保守序列设计4条引物,探究LAMP扩增的最优条件,并与常规的PCR(polymerase chain reaction)检测方法进行比较。建立的LAMP方法检测下限为0.89 pg,灵敏度比PCR高100倍而且特异性好。临床检测显示建立的LAMP方法可行、准确、方便、灵敏。针对DWV的LAMP建立的检测方法为养蜂生产第一线检测和预防DWV提供了技术支持,有一定的应用价值。  相似文献

Bst DNA聚合酶具有热稳定性、链置换活性及聚合酶活性,在体外DNA等温扩增反应中起重要作用. 本文利用Bst DNA聚合酶的5′→3′聚合酶、核苷酸(末端)转移酶及链置换酶活性发展了一种新的体外环式DNA扩增技术跨越式滚环等温扩增(saltatory rolling circle amplification,SRCA).在SRCA反应中,Bst DNA聚合酶以上游引物P1为模板合成其互补链RcP1,并和P1形成双链DNA|之后,Bst DNA聚合酶用其核苷酸转移酶活性在其P1的3′末端沿5′→3′方向随机掺入脱氧核糖核苷酸聚合形成寡聚核苷酸(dNMP)m序列,即DNA的合成反应跨越了RcP1 与下游引物P2之间的缺口|然后,以下游引物P2为模板形成互补序列(RcP2);接着,Bst DNA聚合酶继续将脱氧核糖核苷酸随机添加到RcP2的3′末端,形成(dNMP)n序列.继而,Bst DNA聚合酶以RcP1为模板,继续催化聚合反应合成互补新链,并通过其链置换酶活性替换P1|如此往复,形成[P1-(dNMP)m-RcP2-(dNMP)n …]序列.本文通过电泳、酶切、测序等方法对扩增产物进行分析,演绎出上述扩增过程,并就工作原理进行了讨论.该反应可能对开发等温扩增技术检测微生物有一定助益,也为解释环介导等温扩增技术中假阳性反应和滚环等温扩增反应中的背景信号提供了线索.  相似文献

Loop-mediated isothermal amplification (LAMP) yields a large amount of DNA, as well as magnesium pyrophosphate precipitate, causing a decrease in ionic strength that can be measured with a conductivity meter. There is a clear relationship between the conductivity of the LAMP mixture solution and the duration of biochemical reaction. Moreover, there is also a clear relationship between the change in conductivity and the amount of initial template DNA over the range of 0.08 to 3.2 ng. These results demonstrate the feasibility not only for detecting the LAMP product qualitatively but also for real-time monitoring the biochemical reaction progression quantitatively using conductivity measurements.  相似文献

Detection of fish nocardiosis by loop-mediated isothermal amplification   总被引:4,自引:0,他引:4  
AIMS: Loop-mediated isothermal amplification (LAMP) is a novel method that amplifies DNA with high specificity and rapidity under isothermal conditions. In this study, using the LAMP method, a protocol for detecting Nocardia seriolae which is a causative agent of fish nocardiosis, was designed. METHODS AND RESULTS: A set of four primers, two inner and two outer, were designed based on the sequence of the 16S-23S ribosomal RNA internal transcribed spacer region of N. seriolae. Time and temperature conditions for detection of N. seriolae were optimized for 60 min at 65 degrees C. Other fish pathogen was not amplified by this LAMP system. The detection of N. seriola using LAMP was found to be more sensitive than that by polymerase chain reaction. CONCLUSIONS: LAMP is a highly sensitive and rapid diagnostic procedure for detection of N. seriolae. SIGNIFICANCE AND IMPACT OF THE STUDY: LAMP is a useful diagnostic method for fish nocardiosis.  相似文献

A loop-mediated isothermal amplification (LAMP) assay for simple detection of Bordetella holmesii was developed. This assay discriminates between B. holmesii and other Bordetella species and successfully detect B. holmesii DNA in nasopharyngeal swab samples from subjects with suspected pertussis. The LAMP assay results were in complete agreement with the results of previously published real-time PCR assay, indicating that the former is a powerful tool for the accurate diagnosis and surveillance of B. holmesii.  相似文献
