 为阐明Ⅱ型糖尿病( N I D D M )患者红细胞膜的化学组成在非酶糖基化( N E G)影响下发生的变化,采用毛细管气相色谱法和分光光度法测定了 20 名正常人和 19 例Ⅱ型糖尿病患者红细胞膜的 4 种结合中性糖与 2 种氨基糖和唾液酸含量以及 4 种寡糖链上的末端中性糖和唾液酸含量.结果表明, N I D D M 患者红细胞膜几种结合单糖( Glc, Fuc, Glc A, Gal A)与游离单糖( Glc, Gal, Man, Fuc)含量以及唾液酸含量均较正常对照组明显降低( P< 001 或 005).据此推测,由于患者红细胞膜蛋白的重度糖基化导致某些膜结构蛋白的氧化损伤,细胞膜糖类含量的减少,可能是重度糖基化的继发后果.  相似文献

目的探讨大鼠实验性肝癌发病中刺五加对肌体免疫功能和抗氧化酶活性的影响。方法46只SD雄性大鼠被随机分成对照组(喂普通饲料)、3-甲基4-双甲氨基偶氮苯(3-Me-DAB)组(喂含0.06%3Me-DAB饲料 10周)和刺五加组(饲喂同 3-Me-DAB外、另加入刺五加 4.5g/kg饲料,用常规方法检测全血谷光甘肽过氧化物酶(GSH-PX)、血清超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,用微量化学发光造检测吞噬细胞活性(PMN-CL)。结果1.PMN-CL检测峰值、积分值和吞噬细胞指数,3-MeDAB组较正常组和刺五加组均有显著升高(P<0.05和P<0.01)2.全血GSH-PX活性、SOD活性,刺五加组较3-MeDAB组均有显著升高(P<0.05)。MDA含量刺五加组和3-MeDAB组均较正常组升高(均P<0.05)。结论刺五加在大鼠实验性肝癌诱发过程中有提高抗氧化酶活性和对抗致癌剂引起的机体中性粒细胞吞噬功能代偿性增高的作用。  相似文献

本文采用Ca~2+指示剂的分光光谱法测定巨噬细胞(Mφ)内Ca~2+浓度([Ca~2+]i)、APAAP桥联酶标法检测Mφ膜上Ⅰa抗原的表达,研究肌醇磷脂代谢中第二信使分子甘油二酯(DG)在去甲肾上腺素(NE)促进MφⅠa表达效应中的作用,以进一步探讨NE效应的跨膜信息传递机制。结果表明:蛋白激酶C(PKC)抑制剂4αPDD(25μg/ml)虽不影响NE(10 ̄-8mol/L)升高Mφ[Ca ̄2+]i的效应,却显著减弱了NE促进MφⅠa抗原表达的效应;而PKC激动剂PMA(10nmol/L)本身促进MφⅠa抗原表达的作用不明显,也不能进一步增强NE促进MφⅠa抗原表达的效应。结果提示:DG激活的PKC系统也参与了NE促进MφⅠa抗原表达的信息传递过程,并与另一第二信使分子肌醇-1,4,5-三磷酸(IP_3)介导的Ca ̄2+途径协同发挥作用。  相似文献

通过培养的人主动脉平滑肌细胞(hASMC)及脐静脉内皮细胞(hUVEC),应用3H-TdR参入、Northernblot分析、逆转录多聚酶链反应(RT-PCR)、放射免疫分析(RIA)、和紫外比色法等技术观察了人主动脉中硫酸乙酰肝素蛋白聚糖(HSPG)对hASMC和hUVECDNA合成的作用及对血小板源生长因子(PDGF)、PDGF受体、转化生长因子β(TGF-β)、内皮素-1(ET-1)或碱性成纤维细胞生长因子(bFGF)基因表达和肾素-血管紧张系统(RAS)的影响,结果显示,HSPG明显抑制培养的hASMC基础的DNA合成(cpm值为:10385±3263vs,25541±6421,P<0.01)及外源性PDGF诱导的DNA合成(cpm值为:9878±1947vs.13481±44l0,P<0.05);抑制PDGFA链、TGF-Bp和ET-1mRNA表达,提高PDGFa和β受体mRNA的表达;显著降低hASMC培养液中血管紧张素Ⅱ(AngⅡ)的浓度和血管紧张素转换酶(ACE)的活性,推测HSPG抑制PDGFA链、TGF-β及ET-1mRNA表达,降低ACE活性及AngⅡ浓度是其抑制hASMC增殖的重要机  相似文献

DDPH[1-(2.6-二甲基苯乙氧基)-2-(3.4二甲氧基苯乙胺基)丙烷盐酸盐]是南京药科大学合成的降压新化合物,也具有降低肺动脉高压和抑制肺动脉平滑肌细胞增殖作用。本实验用细胞培养、免疫细胞化学、图像分析、3H-TdR、细胞周期测定等方法,进一步探讨DDPH对缺氧性肺动脉平滑肌细胞(PASMCS)增殖的抑制机制。结果:缺氧促进肺动脉内皮细胞(PAECs)的PDGF·BB和bFGF两种生长因子的表达(积分光密度OD值)增高。缺氧内皮细胞条件培养液(HECCM)能促进PASMCS的PDGF·BB的OD值增高,bFGF的OD值无明显改变。加药组(HEC-CM+DDPH)的PDGF·BB和bFGF的OD值均显著降低,尤以PDGF·BB的OD值减少最多.提示:DDPH能抑制HECCM引起PASMCS的PDGF·BB和bFGF表达增多和细胞增殖。结果与大鼠实验观察相符。  相似文献

在海拔4300m地区,对18名移居汉族、24名世居藏族和21名高原红细胞增多症(HAPC)患者测定了2,3—二磷酸甘油酸(2,3—DPG)和肺通气功能,并进行了血气分析。结果显示:HAPC患者的全血和红细胞内2,3—DP6浓度均显著高于健康组,但世居、移居健康组之间无明显差异。HAPC组的红细胞2,3—DPG和Pdo_2呈显著负相关(r=—0.771,P<0.01),而在健康组无显著相关(r=—0.26,P>0.05)。HAPC组与健康组相比,pH、Pao_2和Sao_2降低,Paco_2和肺泡动脉氧分压差增高。HAPC病人P_(50)为3.75±0.66kPa,健康组为3.40±0.12kPa(P<0.05),P_(50)与2,3-DPG呈正相关(r=0.592,P<0.05)。HAPC组最大呼气中段流量和50%肺活量最大呼气量明显低于健康组(P<0.01)。本研究提示:①HAPC患者的低氧血症可能与血中2,3-DPG浓度过高有关;②轻度肺功能异常亦可促使红细胞进一步增多。  相似文献

Comparison of human and primate erythrocyte membrane sialoglycoproteins showed that common chimpanzee, dwarf chimpanzee, gorilla, orangutan, and gibbon have major periodic acid Schiff-positive proteins resembling human glycophorin A (GPA) monomer and dimer in electrophoretic mobility on sodium dodecyl sulfate-polyacrylamide gels. Immunoperoxidase staining of Western blots with monoclonal antibodies to human GPA showed that these primate bands express some GPA antigenic determinants. A new sialoglycoprotein analogous to human glycophorin B (GPB) was detected in common chimpanzee. Although human MN blood group phenotype results from an amino acid polymorphism of GPA, Western blots showed that in chimpanzee sialoglycoprotein (GPAch) always expresses the M blood group, whereas chimpanzee sialoglycoprotein (GPBch) expresses either the N blood group or a null phenotype. This result explains the detection of M and MN, but not of N, blood group phenotypes in chimpanzee. GPBch has higher apparent m.w. than human GPB, is present in the erythrocyte membrane in greater quantity than human GPB, and contains trypsin cleavage site(s) and the 10F7 determinant (both found on human GPA but not GPB). Expression of human GPA antigenic determinants was consistent with the phylogeny of the hominoid primates; common and dwarf chimpanzee expressed most of the determinants tested, gorilla and orangutan an intermediate number, and gibbon and siamang the least. Of the GPA antigenic determinants examined, the MN blood group determinants were most consistently expressed during evolution of the hominoid primates. The results suggested that variability in expression of GPA antigenic determinants between species was due to both differences in amino acid sequence and glycosylation.  相似文献

Rat reticulocytes contain an isoproterenol-sensitive adenylate cyclase activity which is lost with maturation to erythrocytes despite no change in the density of β-adrenergic receptors. To explore this observation, a cytosol factor, previously shown to be important in the expression of catecholamine-sensitive adenylate cyclase in the reticulocyte, was compared to a cytosol factor obtained in a similar manner from mature erythrocytes. The cytosol factor from reticulocytes augmented isoproterenol-responsive adenylate cyclase activity in reticulocyte and erythrocyte membranes half-maximally at 0.7 ± 0.1 (SEM) and 1.1 ± 0.3 μg/ml, respectively. These concentrations of reticulocyte-derived cytosol factor were significantly lower (P < 0.01) than those concentrations of the factor from erythrocytes necessary to augment isoproterenol-responsive adenylate cyclase activity in reticulocyte (9.7 ± 2.3) and erythrocyte (7.5 ± 1.0) membranes. Cytosol factor from reticulocytes also caused greater total isoproterenol responsiveness than that from erythrocytes both in reticulocyte (784 ± 107 vs 525 ± 65 pmol/mg protein) and in erythrocyte membranes (54 ± 6 vs 36 ± 3); P < 0.05. Neither reticulocyte nor erythrocyte cytosol factor affected the concentration at which isoproterenol half-maximally stimulated adenylate cyclase in either set of membranes. However, the cytosol factor from reticulocytes markedly decreased the binding affinity of isoproterenol for β receptors in reticulocytes from 0.8 ± 0.2 to 6.9 ± 1.4 μm; P < 0.001. This reticulocyte factor had no significant effect on the binding affinity of isoproterenol for erythrocyte membranes. Erythrocyte factor did not change the binding affinity for isoproterenol in either reticulocyte or erythrocyte membranes.  相似文献

Zinc status was assessed in 53 diabetic patients: 18 insulin-dependent diabetic patients (IDDM), 22 noninsulin-dependent diabetic patients (NIDDM) treated with oral antidiabetic agents, and 13 insulin-treated, noninsulin-dependent diabetic patients (IRDM). Plasma zinc concentrations were in the usual range for healthy subjects in these three groups (15.3±0.9 μmol/L). Urinary zinc excretions were elevated in the IDDM group (18.3±4.1 μmol/24 h;p<0.01 vs normal) and in the NIDDM group (17.5±3.5 μmol/24 h;p<0.01 vs normal), but normal in the IRDM group (11.3±2.4 μmol/24 h). In 14 NIDDM patients treated with transient continuous sc insulin injections, urinary zinc decreased from 16.5±2.2 μmol/24 h before insulin treatment to 11.5±0.3 μmol/24 h after insulin treatment without any modification in plasma zinc concentrations.  相似文献

Piwi-interacting RNAs (piRNAs) play a role in gene silencing of retrotransposons, maintenance of spermatogenesis and maturation in germlines. The piRNA and PIWI protein are essential for fertility. To reveal piRNA function associated with testosterone, we investigated the expression of piRNA and piwi protein in normal male rats and testosterone-treated rats. Normal Sprague–Dawley (SD) rats were randomly selected and sacrificed at neonatal to late adolescence stage stages (2, 9, 16, 20, 24, 28, 35, and 42 days, n = 6 each). Additional SD rats were divided into four groups: group 1 received weekly injections of testosterone enanthate (8 mg/100 g) during 1–3 weeks; group 2 during 3–5 weeks; group 3 during 1–5 weeks; and group 4 was the control (n = 20 each). These animals were sacrificed at an age of 60 days. We investigated piRNA, PIWI, and Ago3 protein levels using real-time PCR, Western blot, and immunohistochemistry in each group. In normal rats, PIWI protein and piRNA were expressed at P24. The expression of PIWI and piRNA gradually increased from adolescence to adulthood on Western blot, real-time PCR and immunohistochemistry. In testosterone-treated rats, the expression of PIWI protein was analyzed by Western blot and shown to be significantly increased in group 1 (neonatal to juvenile injection). In real-time PCR, the expression of piRNA after testosterone treatment was increased in all groups (G1 166.8 ± 2.7; G2 113.3 ± 4.6; G3 70.2 ± 1.5 vs. control, 32.87 ± 2.0, all p < 0.001). The expression of testosterone in adolescence induces the development of male genitourinary organs and spermatogenesis. At the same time, the sexual hormones may activate the piRNA and PIWI protein. Our data demonstrate that patterns of piRNA and PIWI expression are similar to the secretion pattern of testosterone, and that piRNA expression was increased after testosterone treatment. Therefore, testosterone may affect testis function through the regulation of piRNA expression in rats.  相似文献

应用竞争性逆转录聚合酶链式反应(RT-PCR)测定了10例急性粒细胞白血病M_2型(AML)患者外周血网织红细胞中α/β珠蛋白mRNA的相对含量,其中8例表现不同程度增高,2例正常,均值为1.513±0.182(±s),与正常对照组(1.24±0.083)进行t检验比较,有非常显著差异(P<0.01).此外,PCR-SSCP分析显示AML患者β珠蛋白基因启动子区序列(5'端-135至+122位核苷酸)无明显异常。说明AML患者珠蛋白基因表达失衡系由于转录异常,很可能系AML的发生对α/β珠蛋白基因的平衡表达产生了某种影响,从而表现为获得性β地中海贫血特征。实验结果为进一步探讨白血病的发病机理及其α/β珠蛋白基因表达失衡的机制奠定了基础。  相似文献

Background

Neuroacanthocytosis (NA) denotes a heterogeneous group of diseases that are characterized by nervous system abnormalities in association with acanthocytosis in the patients' blood. The 4.1R protein of the erythrocyte membrane is critical for the membrane-associated cytoskeleton structure and in central neurons it regulates the stabilization of AMPA receptors on the neuronal surface at the postsynaptic density. We report clinical, biochemical, and genetic features in four patients from four unrelated families with NA in order to explain the cause of morphological abnormalities and the relationship with neurodegenerative processes.

Case presentation

All patients were characterised by atypical NA with a novel alteration of the erythrocyte membrane: a 4.1R protein deficiency. The 4.1R protein content was significantly lower in patients (3.40 ± 0.42) than in controls (4.41 ± 0.40, P < 0.0001), reflecting weakened interactions of the cytoskeleton with the membrane. In patients IV:1 (RM23), IV:3 (RM15), and IV:6 (RM16) the 4.1 deficiency seemed to affect the horizontal interactions of spectrin and an impairment of the dimer self-association into tetramers was detected. In patient IV:1 (RM16) the 4.1 deficiency seemed to affect the skeletal attachment to membrane and the protein band 3 was partially reduced.

Conclusion

A decreased expression pattern of the 4.1R protein was observed in the erythrocytes from patients with atypical NA, which might reflect the expression pattern in the central nervous system, especially basal ganglia, and might lead to dysfunction of AMPA-mediated glutamate transmission.  相似文献

Unhealthy dietary habits are key determinants of obesity in adolescents. Assuming that dietary fat profile influences membrane lipid composition, the aim of this study was to analyze structural changes in the erythrocyte membrane of obese compared to normal-weight adolescents. The study was conducted in a group of 11 obese and 11 normal-weight adolescent subjects. The lipid profile, lipid peroxidation and acetylcholinesterase enzyme (AChE) activity were analyzed by conventional methods. The structural properties of reconstituted erythrocyte membrane were characterized by X-ray diffraction. Erythrocyte membrane from obese adolescents had a lipid profile characterized by a higher cholesterol/phospholipid ratio, an increase in saturated fatty acid and a decrease in monounsaturated and n-6 polyunsaturated fatty acid concentrations. Differences in lipid content were associated with changes in the structural properties of reconstituted membranes and the oxidative damage of erythrocyte membrane. The lower oxidative level shown in the obese group (0.15 ± 0.04 vs. 0.20 ± 0.06 nmol/mg for conjugated diene concentrations and 2.43 ± 0.25 vs. 2.83 ± 0.31 nmol/mg protein for malondialdehyde levels) was related to a lower unsaturation index. These changes in membrane structural properties were accompanied by a lower AChE activity (1.64 ± 0.13 vs. 1.91 ± 0.24 nmol AChE/[min mg protein]) in the obese group. The consequences of unhealthy dietary habits in adolescents are reflected in the membrane structural properties and may influence membrane-associated protein activities and functions.  相似文献

Human skin allografts are one of the best temporary biological coverings for severely burned patients. Cryopreserved (CPA) and glycerol-preserved (GPA) allografts are the most widely used types. This study compared the allograft efficiency of both preservation methods under the same conditions. To simulate actual clinical conditions, we used a porcine wound model. In addition, we evaluated the macroscopic and microscopic scoring of graft performance for each method. Porcine cadaver skin 1 mm thick was obtained from one pig. Cryopreserved skin cell viability was 20.8 %, glycerol-preserved skin was 9.08 %, and fresh skin was 58.6 %. We made ten partial-thickness wounds each in two pigs. The take rates on day 2 were 96.23 and 82.65 % in the GPA and CPA group (both n = 9), respectively. After 1 week, the take rates of both groups were nearly equal. The removal rate at week 5 was 98.87 and 94.41 % in the GPA and CPA group, respectively. On microscopic findings at week 2, inflammation was greater in the CPA group. Other findings such as fibroblast hyperplasia and neovascularization were not significantly different between both groups. At week 5, the score of collagen fiber synthesis was 2.67 ± 0.47 and 2.33 ± 0.47 in the GPA and CPA group, respectively. The epidermal-dermal junction was 2.22 ± 0.79 and 2.00 ± 0.47 in the GPA and CPA group, respectively. These findings suggest that wound healing takes longer in the CPA group. The preservation method of allografts is not a absolute factor in the wound healing process in this wound model.  相似文献
