莱克多巴胺核酸适配体电化学生物传感器的研制   总被引:1,自引:0,他引:1  
莱克多巴胺(RAC)被大量非法用于畜牧生产,易在动物组织残留,对人体造成危害。因此,研发灵敏、快捷的检测RAC的新方法是有效控制RAC滥用的关键环节之一。通过等温滴定量热法筛选到了一条对莱克多巴胺有高亲和力(Kd=1.66×10-6mol/L)的核酸适配体,利用该适配体作为识别分子成功的构建了莱克多巴胺适配体电化学生物传感器。差分脉冲伏安法分析,在0.5~1.0×102ng/ml浓度范围内,峰电流值的差值ΔIp与莱克多巴胺浓度的对数呈现良好的线性关系,相关系数R2=0.977 0,检测限达到0.1 ng/ml,反应时间为15 min。对同一浓度的莱克多巴胺重复检测7次,其峰电流值的RSD值为3.8%;说明该传感电极具有良好的检测重现性。不仅如此,该适配体传感器还具有良好的选择性。  相似文献

从传统中药川牛膝(Cyathula offcinalis kuan)中分离提取到了一种具有生物活性的多糖RCP.核磁共振、甲基化分析、还原裂解和GC-MS分析揭示了RCP是一高度分支的果聚糖,它以(2→1)连接为骨架,其上有大量的(2→6)连接的分支,且属于新蔗果三糖系列.在93.17%果糖残基中,24.15%是末端果糖,26.24%是1-连接果糖,20.46%是6-连接果糖.在6.83%的葡萄糖残基中,2.14%是末端葡萄糖,4.69%是6-连接葡萄糖.RCP的平均聚合度是15.  相似文献

Knowing that Alfred Donné was the discoverer of an important human parasite, and finding that he was also a pioneer of photomicrography, it occurred to me that his parasite might well have become a subject of his photography. It was a simple matter to confirm that this was indeed the case. The parasite he discovered was Trichomonas vaginalis; and, in collaboration with Foucault, Donné made a photomicrograph showing several protozoan parasites lying among vaginal epithelial cells. His publication of an engraved image of the photomicrograph in 1845, was a landmark in the history of photography and microbiology.  相似文献

应用AdMax腺病毒载体系统构建携带siSPK1基因的重组腺病毒,进一步研究SPK1基因对N2a细胞凋亡的影响。设计可形成小发夹结构的SPK1-siRNA模板cDNA序列,克隆至质粒pDC316-siRNA,构建SPK1-siRNA穿梭质粒pDC316-SPK1,经鉴定正确后,将pDC316-SPK1与辅助包装质粒(pBHG loxΔE1,3 Cre)共转染至HEK293细胞,包装纯化并扩增重组腺病毒颗粒,终点稀释法测定病毒滴度;病毒感染N2a细胞,Western blot法检测SPK1蛋白的表达;Hoechst33258染色检测SPK1基因对N2a细胞凋亡的影响。酶切后PCR分析、测序鉴定表明,pDC316-SPK1构建成功,病毒纯化后滴度为2.50E+10 PFU/mL;重组病毒可在蛋白水平抑制SPK1的表达;SPK1基因抑制后,N2a细胞凋亡增加(P〈0.001)。上述结果表明,含有SPK1-siRNA的重组腺病毒构建成功,且具有抑制SPK1蛋白表达的功能,该基因沉默后,N2a细胞凋亡增加。  相似文献

In this paper the development and feasibility of a novel detection system for a low molecular weight chemical, in which a peptide was utilized as a binding molecule, are described. Surface plasmon resonance (SPR) apparatus was used as a transducer. The porphyrin binding peptide, PSP2, was used as a model peptide ligand, while a porphyrin derivative, H2TMpyP, was used as a model low-molecular-weight chemical. PSP2 was covalently immobilized onto the SPR sensor chip and SPR measurement using the PSP2-immobilized chip for various concentrations of porphyrin was carried out. H2TMpyP was detectable in the range from 100 ng ml−1 to 10 μg ml−1 with a linear correlation and good precision and the PSP2-immobilized chip could be regenerated within 1 min after measurement in this system. From comparison of the detection manners of three porphyrin derivatives, the ability of a short peptide to discriminate between differences in molecular structure was demonstrated. Moreover, the self-assembled monolayer (SAM) of PSP2 was successfully prepared on the gold substrate and H2TMpyP could be detected using the PSP2-SAM chip.  相似文献

大豆(Glycine m ax L.) 下胚轴可溶性蛋白提取液进行自磷酸化,以SDS-PAGE电泳分析其标记产物时发现,当有较高浓度的Ca2+ 存在于反应液中时,有一条18 kD蛋白带被高强度标记,同时也可观察到另一条标记强度不高的67 kD蛋白带. 当反应时间延长到15 或30m in 时,它们的标记强度都逐渐减弱,最终从放射自显影底片上消失;在反应液中加入钙螯合剂EGTA 时,则只有67 kD 被高强度标记;在磷酸化反应过程中加入非标记ATP,蛋白中的32P逐渐被非标记磷取代,表明反应体系处于磷酸化-脱磷酸化的平衡过程中,并有结果显示这一过程是钙依赖性的. 组蛋白H1 可以使反应进程加快,表明提取液中的蛋白激酶可以利用它作为底物. 综合结果表明,18 kD和67 kD蛋白可能是具有自磷酸化能力且对Ca2+ 敏感的蛋白激酶,它们对Ca2+ 的不同反应,使得钙信号的传递更具可控性  相似文献

在光镜和电镜下对嫁(虫戚)(Cellana toreuma)和斗嫁(虫戚)(C.grata)的齿舌形态进行观察比较。2种嫁(虫戚)的齿式都为1.1.0.1.1,即具有1枚侧齿和1枚缘齿,缺乏中央齿。齿舌前端都有1小段弯曲,齿片排列松散且存在明显的磨损现象。嫁(虫戚)和斗嫁(虫戚)的侧齿形状很相似,侧齿呈镰刀型且具1个齿尖,基部近似三角形且具突起,尖齿部分细长。两种嫁(虫戚)的缘齿存在一定的差异,嫁(虫戚)缘齿具3个齿尖,第2尖齿靠近第3尖齿。斗嫁(虫戚)缘齿具2个齿尖且比较细长,第2尖齿靠近缘齿基部。本文用17个参数对这两种嫁(虫戚)的齿舌带及其前中后3段上的齿片进行了测量比较,发现斗嫁(虫戚)齿舌带的长宽比明显大于嫁(虫戚)齿舌带的长宽比,即斗嫁(虫戚)的齿舌带显得更加细长。齿舌带前、中、后3段各比例参数的值存在一定的关系,即中段大于前段、中段大于后段。据此认为用齿舌作为2种嫁(虫戚)的分类依据是可行的。  相似文献

Batch and continuous hydrolysis of olive oil in an organic-aqueous two-phase system using the live whole cell of Pseudomonas putida 3SK as a source of a lipase is investigated. The strain was not only fully viable and grown well, but also produced extracellular lipase simultaneously. The degree of hydrolysis, depending on olive oil concentration in the solvents, was maximal at 13.5% (w/v) and decreased with the increase of the substrate concentration. At the optimal condition, a degree of hydrolysis higher than 95% was achieved with 24 h at 30 degrees C when the reaction was carried out in a two-phase batch stirred reactor. For long-term operation a continuous stirred reactor was designed. When the reaction was carried out in a continuous stirred reactor, the degree was hydrolysis reached 86% at a dilution rate of 0.2 h(-1). Satisfactory performance of a two-phase bioreactor was obtained in a long-term continous operation, which lasted for at least 30 days by feeding organic solvent containing olive oil and aqueous media separately. 中国西南季风常绿阔叶林不同恢复阶段凋落物动态分析   总被引:1,自引:0,他引:1  
为探索季风常绿阔叶林不同恢复阶段群落凋落物产量及其动态变化规律,于云南普洱地区通过设置凋落物承接网并定期收集网内的凋落物,对中国西南季风常绿阔叶林区不同恢复阶段群落凋落物产量及其动态变化进行研究。结果表明:(1)不同恢复阶段季风常绿阔叶林年凋落物总量在8 133.1~8 798.3kg/hm2之间,年凋落物总量大小关系为恢复30年群落老龄林群落恢复40年群落。其中叶凋落量最高,其次为枝凋落量,两者贡献量超过总凋落量的3/4。(2)季风常绿阔叶林不同恢复阶段群落中凋落物随时间的动态变化趋势大致相同,2月份达到高峰值,随后逐渐减少,在9月份降至最低,随后又有所升高,为单峰或多峰曲线;在不同凋落物组分凋落量时间动态上,不同恢复阶段群落叶凋落量随月份变化均为单峰曲线;枝凋落量在恢复群落中为单峰曲线,而在老龄林中则为多峰曲线;皮凋落量随月份的变化在恢复30年及老龄林群落间均为单峰曲线,但在恢复40年群落中为平缓曲线,月份间变化不大;繁殖体凋落物在恢复30年及老龄林群落间均为多峰曲线,但在恢复40年群落中为单峰曲线;半分解物凋落量在恢复30年及老龄林群落中随月份呈单峰曲线,在恢复40年群落中则为多峰曲线。(3)在短刺栲、刺栲和红木荷3种优势物种中,短刺栲叶片年凋落量在所有群落中均最大(分别占恢复30年群落的53.93%、恢复40年群落的47.83%、老龄林的28.32%),红木荷次之(分别占恢复30年群落的8.45%、恢复40年群落的10.71%、老龄林的31.69%),刺栲最少(分别占恢复30年群落的6.1%、恢复40年群落的7.53%、老龄林的6.36%)。短刺栲叶凋落量随月份的变化在恢复群落中呈单峰曲线,而在老龄林中则呈现多峰曲线;红木荷在3种群落中则均为单峰曲线;刺栲则是在恢复30年及老龄林中呈单峰曲线,而在恢复40年群落中呈多峰曲线。  相似文献

A carbamoylase enzyme was purified from a cell-free extract of Agrobacterium sp. with an overall yield of 81%. It was judged to be homogenous on sodium dodecyl sulfate-polyacrylamide gel electrophoresis, with a subunit molecular weight of 38,000 daltons. Further studies on the native enzyme suggested that the active enzyme was present as a dimer, with a pI of 5.5. It was able to cleave a variety of N-carbamoyl substrates, but was strictly D(−) specific. It was found to have a Km of 0.82 m and a Vmax of 31 U mg−1 for D(−) N-carbamoyl hydroxyphenylglycine in the presence of 10 m dithiothreitol. It showed no metal ion requirements but was inhibited by iodoacetic acid and iodoacetamide, both thiol reagents. The N-terminal amino acid sequence of the enzyme was elucidated.  相似文献

A single-step chromatographic method for purification of a synthetic 20-mer oligonucleotide is described. Method optimisation was conducted at laboratory scale where 30 mg crude sample was purified per run with a yield of 17 mg pure oligonucleotide. The protocol was scaled-up in steps to achieve 5-, 58- and a final 230-fold scale-up. At the final scale, 7.0 g of crude material was purified with a yield of 4.1 g product. The purity of the oligonucleotide was in all scales higher than 97%. The cycle time was 110 min, which corresponds to a purification capacity of about 90 g crude oligonucleotide material per 24 h.  相似文献

In 5 experiments thirsty rats received an injection of lithium chloride or of saline, and their consumption of fluid was monitored at 5-min intervals for 30min. The novelty of the fluid and the novelty of the test context was varied. In Experiment 1 a novel fluid (a sucrose solution) was offered in a novel context; in Experiment 2 the fluid was novel and the context was familiar (the home cage); in Experiment 3 the fluid was familiar and the context was novel; and in Experiment 4 both fluid and context were familiar. Lithium influenced fluid consumption in those designs that included at least one novel feature (Experiments 1, 2, and 3, but not in Experiment 4). Consumption was initially enhanced (with respect to the controls) when the context was novel, but was suppressed when the fluid was novel. In Experiment 5, the flavor was over-ingested after lithium treatment when it was presented in a short (5min) test conducted in a novel place, but was rejected in a subsequent consumption in the home cages. It is argued that the effect of lithium depends on two factors: enhanced attention to salient cues that modifies the exploratory responses evoked by a novel context; rapid conditioning of an aversion when the fluid consumed is novel. Implications for the use of fluid consumption as an index of lithium-induced nausea are considered.  相似文献

插入序列IS2插入某转录子后产生极性效应,研究Ⅰ发现左插和右插的极性效应不同。本研究(Ⅱ)表明,不同IS2左插后的极性效应也有很大差异。对IS2的DNA序列分析表明,当IS2中ORF的方向和被插入转录子的转录方向相同时为左插,极性效应较小,当IS2中ORF的方向和被插入转录子的转录方向相反时为右插,极性效应较强。当IS2发生碱基突变出现终止密码子时,将增加左插的极性效应。  相似文献

The mating system of the coreid bug, Colpula lativentris, was studied, and compared with those in other hemipteran bugs. Copulating pairs formed a compact aggregation on a shoot of the Japanese knotweed, Polygonum cuspidatum, and there continued copulation and feeding for more than one day. This aggregation was defined as “a shoot aggregation”. Other than this aggregation composed of copulating pairs, there were loose aggregations on the ground composed of males and females not in copula. This loose aggregation was defined as “a ground aggregation”. Males searched for receptive females mostly in a ground aggregation. Sex ratio was constant and variance of sex ratio was also very small in any shoot aggregations and in large ground aggregations. However, in small ground aggregations, sex ratio was strongly male biased and variance of sex ratio was fairly large. This was because receptive females tended to leave a small ground aggregation and join a large one. These results suggest that a male can attain higher mating success in a larger ground aggregation.  相似文献

An experiment was carried out to assess the significance of inter-plot competition in a yield trial of potato cultivars. Seventeen cultivars were deliberately chosen and assessed for yield in single-drill and four-drill plots. Inter-plot competition for fresh-weight yield was a significant factor in the single-drill plots. It was modelled using a common competition coefficient with a covariate based on neighbour fresh-weight yields. In contrast, there was no statistically significant inter-plot competition for specific gravity. After adjustment for inter-plot competition, varietal ranking in estimated monoculture yield differed little from that based on unadjusted means. However, there was a reduction in the range of yield estimates, and a closer agreement with the observed pure-stand yields from the inner two drills of the four-drill plots. The adjustment for monoculture performance was most pronounced for the higher and lower yielding varieties, as expected from the assumption that the performance of high yielding varieties was enhanced in a competitive environment at the expense of low yielding ones. A general and flexible method of estimating competition coefficients in variety trials, together with a suitable algorithm, was developed and is explained in an appendix. It was used to check for inter-plot competition in a number of potato trials with single-drill plots and a large number of entries. Competition was found in some trials but not in others. Thus, where potato tubers are grown in single-drill plots for assessment of fresh-weight yield, adjustment should be made for inter-plot competition when evidence of inter-drill competition is found.  相似文献

In this study, a novel three-compartmented electrochemical bioreactor (3-CEB) was designed in an effort to overcome the disadvantages of the two-compartmented electrochemical bioreactor (2-CEB) separated with a cation-selective membrane for enrichment of strict anaerobes. The 3-CEB was comprised of an anode, outlet, and a cathode compartment. The outlet compartment was positioned between the anode and cathode compartment, and it was separated with the anode side by a rubber plate and with the cathode side by a porous glass membrane. A platinum wire bridging the anode and outlet compartment operated as a redox passage, however, through which no material could permeate. Butyrate fermentation bacteria were enriched on the basis of the metabolite production. Butyrate generated by strict anaerobes was significantly more abundant in the 3-CEB than in the 2-CEB. Acetic acid and lactic acid generated by facultative anaerobes was relatively higher in the 2-CEB than in the 3-CEB. Meanwhile, butyrate was not generated in the bioreactor utilized for the control test, to which the electrochemical potential was not charged. In a continuous culture using the 3-CEB, the majority of the glucose was fermented to butyrate, and the acetate additionally supplied to the bacterial culture was metabolically reduced to butyrate. More lactate than butyrate was generated from glucose in the 2-CEB.  相似文献

A carboxypeptidase B-like enzyme was purified 116-fold with a recovery of activity of 29% from a crude extract of camel pancreas by a four-step procedure consisting of two anion exchange chromatographies in succession, gel filtration and hydrophobic interaction chromatography. The enzyme was homogeneous on SDS and non-denaturing gel electrophoresis and on gel isoelectric focusing. Its molecular mass was found to be 31.5 kDa and its isoelectric point was estimated as 6.1. It was active towards a number of substrates that are cleaved by carboxypeptidases B from other species and was also susceptible to inhibition by inhibitors of such enzymes. The camel enzyme showed a pH optimum of 8.0 and it was seen to be a relatively potent kinase in vitro. The enzyme purified in this study was very similar to carboxypeptidases B isolated from other species in size, charge, substrate specificity and susceptibility to inhibition and thus it can be identified as camel carboxypeptidase B.  相似文献

The luminol test for blood was carried out on a set of interior fittings and surfaces inside three different makes of modern motor car. The surfaces and fittings provided little interference with the test for blood, although there was some detectable chemiluminescence when the test was applied to blood-free material from a seatbelt, a boot-lining and a gear-knob. The case with which haemoglobin samples could be washed off interior car surfaces was also examined for seat fabrics, carpets, roof-linings and various other plastic interior surfaces. A standard wash with water alone was not very effective and removed only ca. 50% of the haemoglobin. A standard wash with soapy water or with a proprietary multipurpose car cleaner removed ca. 90% of the haemoglobin from the tested surface. The effect of high car interior temperatures on haemoglobin samples that were subsequently used in the luminol test was also examined. It was shown that the sensitivity of the luminol test was not decreased but was increased by the prior heating of a haemoglobin sample. This effect was attributed to the thermal conversion of haemoglobin to the more brighter catalyst for chemiluminescence, methaemoglobin. The enthalpy of this conversion in the solid state was found to be 14.1 kJ/mol.  相似文献
