神经生长因子在6—OHDA化学性去交感神经中的保护作用   总被引:1,自引:0,他引:1  
本实验用6-OHDA造成成年小白鼠颌下腺化学性去交感神经,观察了神经生长因子对该神经的保护作用。6-OHDA处理后24小时腺体内去甲肾上腺素(NE)含量降至正常水平的2%以下。若在6-OHDA处理同时开始多次给予神经生长因子(NGF),则NE残留量明显提高。减小6-OHDA剂量到10mg/kg,NE残留量增加,同时NGF的作用亦较用6-OHDA15mg/kg时列为显著。若提前24小时给予NGF,尽  相似文献

本文旨在研究针对EGFR mRNA的反义寡核苷酸片段对BEL-7404细胞EGFR基因表达及其对细胞生长的影响。合成了互补于EGFR基因5’起始编码区的21聚脱氧寡核苷酸(ODNs)。实验结果显示,3.2umol/L ODNs明显抑制BEL-7404细胞生长,「3H」Tdr掺入抑制试验显示其对细胞DNA合成的抑制作用表现剂量依赖性;密度扫描分析显示ODNs处理6、24h后,肝癌细胞EGFR mRN  相似文献

低分子量硫酸葡聚糖对小鼠造血干细胞动员作用的研究   总被引:3,自引:0,他引:3  
给小鼠静脉注射低分子量(<10 ̄4u)硫酸葡聚糖(DS)15mg/kg后外周血中白细胞、单个核细胞(mononuclearcek,MNC)、CFU-GM、BFU-E和CFU-Mix产率等指标出现时相性变化。给药后1h开始升高,2h达到高峰、分别为药前值的2.2、2.6、3.8、4.4和3.0倍,7h时趋向正常。给药后2h上述各类细胞在外周血中的含量随着DS的剂量增加而增加。白细胞、MNC计数在DS180mg/kg时达到峰值,均为对照组的4倍。240mg/k8时未见明显增加。不同剂量DS对各系祖细胞均有不同程度的动员作用,DS剂量15-30mg/kg效果最好,每升血中CFU-GM、BFU-E、CFU-Mix的数量分别相当于对照组的5.0、11.9和8.8倍。其峰值出现时间与白细胞、MNC不同,表明DS对不同类型细胞的作用机制也不尽一致。经口给小鼠投以DS240和48omg/kg后,未见外周血中白细胞、MNC计数有显著性升高,提示对造血干细胞没有动员作用。  相似文献

用酶组织化学和免疫组织化学双标技术,观察了正常SD大鼠基底前脑内侧隔核(MS)、斜角带垂直支(VDB)和水平支(HDB)中NOS阳性神经元的形态和分布及NOS与胆碱能神经元标志物ChAT、NGF受体(NGF-R)和AChE之间的共存关系。结果发现,MS、VDB和HDB的头端NOS阳性神经元较多、胞体较大、突起多,尾端NOS阳性神经元数目较少、胞体较小、突起少而短。NOS+ChAT双标神经元占NOS阳性神经元总数的90%,占ChAT阳性神经元总数的39%;NOS+NGF-R双标神经元占NOS阳性神经元总数的83%,占NGF-R阳性神经元总数的40%;NOS+AChE双标神经元占NOS阳性神经元总数的96%,占AChE阳性神经元总数的39%。这些结果为研究Alzheimer'sdisease病理过程中基底前脑隔区胆碱能神经元退变与NO的关系提供了形态学依据。  相似文献

根据基因库中的顺序,设计了胶质细胞源神经营养因子(GDNF)基因的PCR引物,以此从人基因组DNA中扩增并克隆了GDNF的编码序列,经DNA测序确认后,该片段克隆到表达质粒pET-3a中,转化大肠杆菌BL21(DE3).培养的重组菌经IPTG诱导,在T7启动子调控下表达出hGDNF蛋白.经电泳分析表明GDNF主要存在于细菌包涵体中.从培养菌中制备包涵体,经充分洗涤,溶解于含8mol/L尿素的变性缓冲液中.经SP-Sepharose柱层析分离,梯度洗脱,以15%SDS-PAGE检查含GDNF的部分.将含单体GDNF部分进行复性,再次用SP-Sepharose离子柱分离同源二体GDNF.最后经SDS-PAGE制备电泳纯化,纯度大于95%.经N端测序表明序列正确.经测定,每升培养菌可得约10mg纯化的GDNF.  相似文献

本研究采用原代培养大鼠海马神经元,观察睫状神经营养因子(ciliary neurotrophic factor,CNTF)对NO引起细胞毒性反应的影响。NO供体硝普钠与S-亚硝基-乙酰青霉胺,NOS底物L-Arg及钙载体ionomycin,均可引起海马神经元存活率下降,LDH漏出增加;提前24h给予不同浓度CNTF,均能提高神经元的存活率,减少LDH漏出,其作用呈剂量依赖性。  相似文献

神经节苷脂对6-OHDA损毁交感神经末梢的对抗作用   总被引:1,自引:0,他引:1  
单次6-OHDA (15mg/kg.i.p.)注射后24h,可使雌性成年小鼠颌下腺内儿茶酚胺荧光神经末梢几乎完全消失;同时用 HPLC 测得腺体内去甲肾上腺素(NA)和多巴胺(DA)的含量下降至正常值的3—4%以下。随着受损交感神经末梢再生过程,NA 和 DA 水平有缓慢的恢复。在损毁2周时 NA 和 DA 含量分别达到正常水平的50%和28%,且在4周时完全恢复。在注射6-OHDA 的同时,和在损伤后12h 内给动物注射4次神经节苷脂(每次50mg/kg.i.p.)并在其后的一周內每天注射一次,可使颌下腺内 NA 含量维持在正常水平;在损毁后4h 及损毁前4d 开始施用神经节苷脂,也可不同程度地对抗交感神经末梢损伤,但作用强度不如前者。实验结果提示:(1)神经节苷脂通过减弱6-OHDA 及其代谢产物的损伤效应能够保护交感神经末梢膜,它可能还有促损伤末梢再生性长芽的作用;(2)损伤后神经节苷脂处理得越早,其效果越好。  相似文献

The effects of i.v. 6-hydroxydopamine (6-OHDA), 100 mg/kg, have been studied on parotid glands of rats at 12, 24, 48, 72 hr and 3 weeks after avulsion of the right superior cervical sympathetic ganglion. The salivary flow from normal left control glands and from right glands 12 hr after ganglionectomy were similar, but at longer times after ganglionectomy the secretory response from the test glands was greatly reduced. Morphological assessment showed that 6-OHDA induced a massive depletion of secretory granules from all control glands and also at 12 hr after ganglionectomy but at 48 and 72 hr there was considerably less depletion of granules on the ganglionectomized side. It is thought that at the longer times after ganglionectomy the secretion from the test glands is caused by circulating catecholamines released by the action of 6-OHDS on adrenergic nerves elsewhere, plus a possible small direct secretogogue effect oomy are thought to be attributable to the release of catecholamines from adrenergic nerves within the gland.  相似文献

6-Hydroxydopamine (6-OHDA) is often used in models of Parkinson's disease since it can selectively target and kill dopaminergic cells of the substantia nigra. In this study, pre-treatment of PC12 cells with nerve growth factor (NGF) inhibited apoptosis and necrosis by 6-OHDA, including caspase activity and lactate dehydrogenase release. Notably, cells exposed to 6-OHDA in the presence of NGF were subsequently capable of proliferation (when replated without NGF), or neurite outgrowth (with continued presence of NGF). Following 7 days growth in the presence of NGF, expression of betaIII tubulin and tyrosine hydroxylase and increased intracellular catecholamines was detectable in PC12 cells, features characteristic of functional dopaminergic neurons. NGF-pre-treated PC12 cells retained expression of betaIII-tubulin and tyrosine hydroxylase, but not catecholamine content following 6-OHDA exposure. These data indicate that NGF-protected cells maintained some aspects of functionality and were subsequently capable of proliferation or differentiation.  相似文献

Administration of nerve growth factor (NGF) twice daily for 2 days to rats 11 days at the time of the initial injection resulted in a 6.6-fold increase in [3H]thymidine levels of pancreas, when comparison was made to levels of untreated controls. Isoproterenol (ISO), a beta-adrenergic agonist known to produce marked increase in [3H]thymidine incorporation into DNA of salivary glands, caused increases in levels of [3H]thymidine in pancreas that were similar in magnitude to those induced by NGF. The combined administration of ISO and NGF did not cause any increase above those observed with either agent alone. Administration of propranolol (3 mg/kg body wt) prior to administration of ISO prevented the usual ISO-induced increase in DNA synthesis, but propranolol in either a 3- or 9-mg/kg body wt dose, caused only a 50% inhibition of NGF-induced thymidine incorporation. In the absence of the submandibular-sublingual glands, the ISO failed to induce the usual high levels of thymidine incorporation, whereas NGF induced the same high levels observed in rats with submandibular glands intact. NGF did not alter the distribution of beta 1- and beta 2-adrenoceptors in the pancreas but did increase norepinephrine when the initial administration was at age 5 days, but not when it was given at age 10 days. Since NGF increased DNA synthesis in the absence of submandibular-sublingual glands, whereas ISO did not, this suggests that ISO requires NGF to induce beta 1-activation and subsequent synthesis and that NGF is a direct activator.  相似文献

Abstract: Removal of nerve growth factor (NGF) from sympathetic neurons initiates a neuronal death program and apoptosis. We show that pituitary adenylyl cyclase-activating polypeptide (PACAP) prevents apoptosis in NGF-deprived sympathetic neurons. PACAP (100 nM) added to culture medium at the time of plating failed to support neuronal survival. However, in neurons grown for 2 days with NGF and then deprived of NGF, PACAP prevented cell death for the next 24–48 h. Uptake of [3H]norepinephrine ([3H]NE) was used as an index of survival and decreased >50% in NGF-deprived cultures within 24 h. PACAP (1–100 nM) restored [3H]NE uptake to 92 ± 8% of that of NGF-supported controls. Depolarization-induced [3H]NE release in neurons rescued by PACAP was the same as that in NGF-supported neurons. PACAP rescue was not mimicked by forskolin or 8-bromo-cyclic AMP and was not blocked by the protein kinase A inhibitor Rp-adenosine 3′,5′-cyclic monophosphothioate. Mobilization of phosphatidylinositol by muscarine failed to support NGF-deprived neurons. Thus, PACAP may use novel signaling to promote survival of sympathetic neurons. The apoptosis-associated caspase CPP32 activity increased approximately fourfold during 6 h of NGF withdrawal (145 ± 40 versus 38 ± 17 nmol of substrate cleaved/min/mg of protein) and returned to even below the control level in NGF-deprived, PACAP-rescued cultures (14 ± 7 nmol/min/mg of protein). Readdition of NGF or PACAP to NGF-deprived cultures reversed CPP32 activation, and this was blocked by lactacystin, a potent and specific inhibitor of the 20S proteasome, suggesting that NGF and PACAP target CPP32 for destruction by the proteasome. As PACAP is a preganglionic neurotransmitter in autonomic ganglia, we propose a novel function for this transmitter as an apoptotic rescuer of sympathetic neurons when the supply of NGF is compromised.  相似文献

Thyroid hormone, nerve growth factor (NGF) and norepinephrine (NE) and weight-bearing affect bone metabolism, yet interactions between these factors and osseous tissue have not been investigated. Therefore, the aims of the study were to measure NGF and NE concentrations in weight-bearing and non-weight-bearing bones from euthyroid (control) and hyperthyroid (HT) rats. Hyperthyroidism was induced by oral intake of triiodothyronine (90 mg/kg/day) for 21 days. Histomorphometry on distal femurs verified significant trabecular bone loss in HT rats compared to euthyroid animals. NGF concentrations were assayed via ELISA, whilst NE concentrations were measured via HPLC and ECD. In euthyroid rats: (i) the concentration of NGF in ribs (914 ng/g) was almost 3-fold greater than in femurs (326 ng/g wet weight of tissue) (ii) the concentrations of NE in ribs (74.7 ng/g) and calvaria (87.4 ng/g) were 2.5-3.5-fold greater than either femurs (24.0 ng/g) or tibiae (30.5 ng/g) and (iii) NE concentrations were comparable between ribs (74.7 ng/g) and calvaria (87.4 ng/g) and similar between tibiae (30.5 ng/g) and femurs (24.0 ng/g). In HT rats: (i) the concentration of NGF in ribs (1802 ng/g) was 4-fold greater than in femurs (402 ng/g) (ii) NE concentrations in ribs (23.3 ng/g) and calvaria (13.6 ng/g) were 4.5-fold and 2.6-fold greater respectively than in tibiae (5.2 ng/g), while ribs had almost a 2-fold higher concentration of NE than calvaria. In HT rats compared to euthyroid animals: (i) NGF concentrations almost doubled in ribs but there was little change in the NGF concentration in femurs (ii) there was a reduction in NE concentrations in calvaria by 84%, in ribs by 69% in tibiae by 83% and 55% in femur (NS). Conclusions: (i) Non-weight-bearing is associated with higher concentrations of NGF and NE than weight-bearing in bones in euthyroid and HT rats; (ii) Hyperthyroidism exerts opposite effects on NGF and NE in bone and (iii) Hyperthyroidism interacts with weight-bearing to determine NGF and NE concentrations in bone. Therefore, the influence of thyroid hormone on NGF and NE in bone may need to be taken into account when considering the action of thyroid hormone on bone in either euthyroid or hyperthyroid states.  相似文献

The effects of 6-hydroxydopamine (6-OHDA) on the bioluminescent response of Porichthys photophores were investigated as part of a pharmacological study of the neural control of luminescence in this fish. Subcutaneous injections of 6-OHDA induce a luminescent response similar to that of norepinephrine (NE), suggesting a sympathomimetic action. The luminescent response to electrical stimulation is almost completely and irreversibly abolished within 24 hours following low-dose treatment of the photophores with 6-OHDA, while the sensitivity of these organs to exogenous NE is increased significantly over the few days post-treatment. During this period the photophores continuously emitted a steady low-level glow. Electronmicroscopic studies of such photophores revealed progressive destruction of the nerve endings. Photophore luminescent sensitivity to NE subsequently became sub-normal, and at this stage electron microscopy revealed an increasingly larger number of damaged photocytes, supportive cells and, in one case, lens cells. From these results it is suggested that 6-OHDA initially impairs neuro-photocyte transmission by destroying catecholaminergic nerve endings. In turn, the transmitter reuptake mechanism is also impaired, thus accounting for development of supersensitive responses to exogenous NE. Subnormal luminescent responses to NE appear as a result of loss of photocyte competence due to structural deterioration. The latter are interpreted as the consequence of removal of trophic factors supplied by the photophore adrenergic innervation.Suppression of luminescent response to both electrical stimulation and exogenous NE in photophores treated with higher doses of 6-OHDA, may be due to a direct effect of this drug on the receptor sites of the photocytes.  相似文献

Drugs of abuse, such as phencyclidine (PCP), methamphetamine (METH), and cocaine (COC) are known to affect several behaviors in rats, such as motor activity, stereotypy, and circling. In this study, we evaluated whether these drugs produce circling preferences in the presence or absence of unilateral 6-hydroxydopamine (6-OHDA)-induced lesions of the caudate nucleus. Adult male CD rats were lesioned with 10 μg 6-OHDA/site. Animals were dosed with PCP (15 mg/kg, ip), its congener, (+) MK-801 (0.15 mg/kg, ip), METH (2 mg/kg, ip), COC (60 mg/kg, ip), or apomorphine (0.2 mg/kg, ip). circling preference was recorded in control and lesioned rats for 2 h before animals were sacrificed to determine monoamine levels by HPLC/EC. In control animals, administration of these drugs produced 60–70% left circling. In, lesioned animals, these drugs produced 78–90% ipsilateral (toward the lesion) circling, except apomorphine, which produced 60–80% contralateral (away from the lesion) circling. Dopamine (DA) and its metabolites 3,4-dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA) concentrations significantly decreased ipsilaterally in lesioned caudate nucleus (CN) and substantia nigra (SN). However, no significant changes were observed in nucleus accumbens (NA) and olfactory tubercles (OT). These data demonstrate that drugs of abuse like PCP, its congener (+) MK-801, METH, and COC produce a greater preference to turn toward the left than the right, a finding similar to that found in human psychosis. Since 6-OHDA lesions enhanced the circling bias and depleted DA and its metabolites DOPAC and HVA, it also suggests that the dopaminergic system may be involved in the circling behavior.  相似文献

用放射免疫法测得兔动脉的亮氨酸脑啡肽(L-ENK)和甲硫氨酸脑啡肽(M-ENK)样免疫活性物的含量(pg/mg组织湿重)分别为耳动脉38.99±17.29,134.67±8.11;肾动脉31.10±7.76,93.60±18.22,肠系膜动脉25.70 13.60,88.43±18.16。动物经注射交感神经末梢化学切割剂6-OHDA后,这三种动脉中L-ENK样免疫活性物的含量均明显下降(P<0.001);切除颈上神经节使支配耳动脉的交感神经溃变后,或离体耳动脉条受电场刺激后,脑啡肽(ENK)样免疫活性物的含量也都显著下降(P<0.001);但注射利血平则无明显影响,用荧光法测定培育动脉条的浴槽液中NE的含量,观察到ENK可抑制电场刺激所引起的NE的释放。结果提示,动脉壁的L-ENK和M-ENK存在于交感神经末梢中,它可因受电场刺激而释放,可能通过激活突触前膜阿片受体,减少交感神经末梢释放NE,从而抑制动脉的收缩活动。  相似文献

Intracerebroventricular injection of lipopolysaccharide (LPS) induces a marked increase in circulating interleukin (IL)-6 levels and in IL-6 mRNA expression in brain and peripheral organs. Recently, it was reported that intraperitoneal administration of alpha-adrenoceptor antagonists inhibits centrally injected LPS-induced increases in plasma IL-6 levels, suggesting the involvement of the norepinephrine (NE) system in the central LPS-induced IL-6 response. However, the localization (either central or peripheral) of NE involvement in the central LPS-induced IL-6 response has not been characterized. In the present study, mice were pretreated with 6-hydroxydopamine (6-OHDA) administered intracerebroventricularly or intraperitoneally to deplete central or peripheral stores of NE, respectively. Intracerebroventricular LPS (50 ng/mouse) markedly increased plasma IL-6 levels and IL-6 mRNA expression in choroid plexus, hypothalamus, pituitary, adrenals, heart, liver, spleen, and lymph nodes, but with minimal effect in lung, kidney, and testis, as revealed by RT-PCR. Pretreatment with intracerebroventricular 6-OHDA (50 microg/mouse) decreased the LPS-induced plasma IL-6 levels by 39% and the LPS-induced IL-6 mRNA expression in liver, spleen, and lymph nodes, but not in choroid plexus, hypothalamus, pituitary, adrenals, and heart. Pretreatment with intraperitoneal 6-OHDA (100 mg/kg) decreased the LPS-induced plasma IL-6 levels by 36% and the LPS-induced IL-6 mRNA expression in all the peripheral organs displaying increased IL-6 mRNA. Central LPS-induced increase in plasma corticosterone levels was decreased slightly by central but not by peripheral NE depletion. These results suggest that central NE and peripheral NE are differentially involved in the central LPS-induced IL-6 mRNA expression in peripheral organs.  相似文献
