为探究裸果木再生体系建立的影响因素,确定其不定芽发生的起源,该研究以裸果木健壮植株的茎段为外植体,采用6 BA和IBA不同浓度组合,筛选愈伤增殖及不定芽再生的最佳浓度组合,确定生根诱导的关键影响因素,建立再生体系,并对其不定芽分化进程进行解剖结构分析,以确认其起源。结果表明:(1)裸果木茎段的最佳愈伤增殖培养基为MS+1 mg·L-1 IBA+1 mg·L-1 6 BA+30 g·L-1蔗糖+7 g·L-1琼脂,主体间效应分析表明IBA为关键影响因素;愈伤大小随IBA浓度增加呈现先升高后下降的趋势。(2)最佳不定芽诱导培养基为MS+0.5 mg·L-1 6 BA+30 g·L-1蔗糖+7 g·L-1琼脂,诱导不定芽数量为4.9个/块,生芽率达92.3%。(3)生根诱导中,SH基本培养基和蔗糖浓度为关键因素,最佳生根培养基为SH+0~10 g·L-1蔗糖+7 g·L-1琼脂,生根率达91.3%。(4)解剖结构观察发现,不定芽起源于愈伤表层的分生细胞,为外起源。该研究通过器官发生途径建立了裸果木的再生体系,确定了不定芽为外起源,为裸果木这一珍稀濒危的林木种质资源保护及可持续利用奠定了研究基础,并为其未来的发展利用提供了有效途径。  相似文献

以中国传统菊花品种‘小林静’叶片为外植体,建立了‘小林静’较好的再生体系及遗传转化体系.结果表明,‘小林静’叶盘最适不定芽分化培养基为MS+2.0 mg/L 6-BA+1.5 mg/L NAA,不定芽分化率为92.76%,平均再生不定芽数为2.3767个;试管苗最佳生根培养基为1/2MS,生根率达100%.移栽采用灭菌的蛭石,再生苗移栽成活率达90%以上.采用根癌农杆菌C58C1介导的叶盘转化法进行‘小林静’的遗传转化试验,农杆菌OD600=0.5-0.6,侵染10 min后,将叶片外植体接种到MS+2.0 mg/L 6-BA+1.5mg/L NAA的培养基中黑暗共培养2d,之后转接到附加10 mg/L硫酸卡那霉素和400 mg/L羧苄青霉素的分化筛选培养基中进行转化细胞的筛选,待长出抗性芽后转接至生根培养基中进行培养,最终建立了菊花品种‘小林静’的遗传转化体系.  相似文献

为探讨水稻(Oryza sativa L.)中CPR5 基因的功能,以其cDNA 文库为基础进行序列比对,并将1 个同源性较高的序列命名为OsCPR5.生物信息学分析表明,OsCPR5 的开放阅读框长度为1551 bp,编码516 个氨基酸.软件预测该编码蛋白可能是1 个具有5 个跨膜区的膜蛋白和核定位蛋白.组织表达和亚细胞定位分析表明,OsCPR5 在根中的表达水平较高,且广泛分布在细胞膜、细胞质和细胞核上.逆境胁迫分析实验表明,植物激素脱落酸(Abscisic acid, ABA)、过氧化氢(Hydrogen peroxide,H2O2)、氯化钠(NaCl)、甲基紫精(Methyl viologen, MV)等环境胁迫可诱导OsCPR5 的上调表达,其中氧化胁迫相关的MV 和H2O2 处理效果最为明显.拟南芥(Arabidopsis thaliana)转基因株系atcpr5/OsCPR5 在浓度为0.5 μmol L-1、 1.0 μmol L-1 的MV以及6 mmol L-1、 8 mmol L-1 的H2O2 处理下,种子萌发率明显高于atcpr5 突变体.这揭示了OsCPR5 基因在植物抗氧化胁迫响应中具有一定的作用.  相似文献

选用东北地区主栽品种‘黑农41’的子叶节和胚尖作为外植体,建立了‘黑农41’的子叶节和胚尖再生体系,并对2种再生体系进行了比较.结果表明子叶节最适芽诱导培养基为MSB5+1.0 mg/L6-BA+0.2 mg/LIBA,最适生根培养基为MSB5+1.0mg/LIBA;胚尖最适芽诱导培养基为MSB5 +0.2 mg/L6-BA +0.2 mg/LIBA,最适生根培养基为MSB5+2.0 mg/LIBA,子叶节更适合作为遗传转化的受体材料.  相似文献

该研究以大花蕙兰‘红酒''(Cymbidium hybridum ‘hongjiu'')×莲瓣兰‘边草素花''(C. tortisepalum ‘biancaosuhua'')F1代杂交种原球茎和根状茎为材料,比较了不同激素配比增殖分化、生根的培养基,建立了适用杂交兰组培快繁体系。结果表明:1/2MS+6-BA 1.0 mg·L-1+NAA 1.0 mg·L-1+AC 0.05%+香蕉80 g·L-1对原球茎增殖效果最佳,增殖率达到307%; 1/2MS+6-BA 1.5 mg·L-1+NAA 1.0 mg·L-1 +AC 0.05%+香蕉80 g·L-1有利于原球茎分化,分化率为82%; 1/2MS+TDZ 2.0 mg·L-1+NAA 0.1 mg·L-1+AC 0.05%+香蕉80 g·L-1对根状茎增殖分化效果最佳,增殖率为293%,分化率为79%; 1/2MS+IBA 0.5 mg·L-1+NAA 0.3 mg·L-1+AC 0.05%+香蕉80 g·L-1为最佳生根培养基,生根率达到84.7%,且根粗苗壮,叶色浓绿。此体系为杂交兰种苗的规模化生产提供了技术支持。  相似文献

以河南温县铁棍山药带腋芽的茎段为外植体进行铁棍山药种苗快繁及珠芽离体再生体系的研究。结果表明:(1)75%乙醇浸泡30 s和5% NaClO消毒15 min配合使用灭菌效果最好;腋芽诱导最适培养基为MS+0.5 mg·L-1 6-BA+0.1 mg·L-1 NAA,培养20 d后诱导的多芽体倍数最高,为2.22,高度最高为3.3 cm;继代增殖最适培养基为MS+1.5 mg·L-1 6-BA,增殖倍数可达4.1;生根培养最适培养基为1/2MS+0.2 mg·L-1 6-BA+1.0 mg·L-1 NAA+0.02%活性炭,平均生根天数为12 d,生根率达100%,根系最长为1.04 cm。(2)用单芽带外植体的接种方式,其珠芽诱导率及诱导的珠芽数显著高于只接单芽的接种方式,珠芽诱导率达88.9%,平均珠芽数为1.50,大小为0.38 cm×0.54 cm;蔗糖浓度为1%~3%有利于珠芽诱导,珠芽整齐度好,形状规则,试管苗叶色浓绿;离体珠芽芽诱导的最适培养基为MS+1.5 mg·L-1 6-BA+0.2 mg·L-1 NAA,珠芽在18~22 d发芽,30 d后诱导率最高为83.3%。该实验结果为铁棍山药试管苗的工厂化生产奠定了技术基础。  相似文献

以吉粳88为材料,通过对不同培养基和激素的筛选,确定最佳的再生体系。结果表明:NMB培养基为吉粳88最适的愈伤组织诱导培养基。通过L9(33)正交实验研究最适激素浓度配比,筛选出吉粳88愈伤组织诱导的最佳激素配比为2.0 mg/L 2,4-D+0.2 mg/L 6-BA+0.10 mg/L NAA。在添加2.0 mg/L 6-BA+0.5 mg/L NAA的NMB基本培养基条件下,吉粳88的分化率最高,成培苗率也最高。  相似文献

为揭示外源蔗糖参与干旱胁迫下高表达转玉米C4 型磷酸烯醇式丙酮酸羧化酶(phosphoenolpyruvate carboxylase, PEPC)基因(C4 pepc)水稻(简称:PC)种子萌发的生理机制,该研究以 PC及其未转基因野生型受体‘Kitaake’(简称:WT)的种子为材料,研究外施不同浓度蔗糖联合模拟干旱(10% PEG 6000)处理下,其种子发芽参数、总可溶性糖及可溶性蛋白含量、蔗糖非发酵1 (sucrose nonfermenting 1, SNF1)相关蛋白激酶(SNF1 related protein kinase 1s, SnRK1s)基因以及PEPC基因表达等参数的变化。结果表明:(1)PEG 6000模拟干旱处理均显著抑制两材料发芽,但明显促进胚根的生长;外施蔗糖则呈现浓度效应,高浓度蔗糖(>150 mmol·L-1)进一步加剧了干旱对发芽的抑制效应,而低浓度(<30 mmol·L-1)则可缓解干旱的抑制,但与WT(<30 mmol·L-1)相比,促进PC水稻萌发的外施蔗糖浓度(<6 mmol·L-1)更低,且各处理的发芽表现与其α 淀粉酶活性的动态表现一致。(2)与WT相比,外施3 mmol·L-1蔗糖联合干旱处理下,显著提高了PC种子的发芽率,且伴随PC内源蔗糖含量、总可溶糖和可溶性蛋白含量显著增加;且外施3 mmol·L-1蔗糖使PC中内源C3 pepc基因表达下调,而外源导入C4 pepc基因表达显著增加。(3)与WT相比,干旱处理下外施3 mmol·L-1蔗糖,PC的糖信号相关基因SnRKs亚家族基因(包括SnRK1s:OsK1a OsK24 OsK35和SnRK2s:SAPK6)的表达也显著增加。研究发现,外施低浓度蔗糖通过上调PC水稻种子中可溶性糖和可溶性蛋白含量,增强SnRK1s亚家族基因和外源C4 pepc基因的表达,提高了α 淀粉酶活性,从而缓解了干旱胁迫对PC种子萌发的抑制。  相似文献

以珙桐冬芽为材料进行组织培养和植株再生研究,结果表明:珙桐冬芽直接诱导丛生芽的最适培养基为WPM+NAA 0.2 mg·L-1+6-BA 3.0 mg·L-1+AC 2.0 g·L-1;珙桐带芽茎段增殖的适宜培养基为WPM+NAA 0.05 mg·L-1+6-BA 1.0 mg·L-1+GA3 2.0 mg·L-1+AC 2.0 g·L-1;生根最佳培养基为White+IBA3.0 mg·L-1+6-BA 1.0 mg·L-1+AC 2.0 g·L-1,在此条件下,根发育良好,植株健壮;组培苗炼苗后移栽,成活率可达80%。  相似文献

以江西铅山红芽芋脱毒苗为试材,研究不同因素对红芽芋脱毒苗球茎愈伤组织诱导及其再生体系的影响,以期对红芽芋脱毒苗的再生体系进行优化。结果表明,红芽芋脱毒苗球茎愈伤组织诱导的最佳培养基是MS+TDZ 2 mg·L-1+2,4-D 1 mg·L-1。红芽芋脱毒苗球茎愈伤组织分化的最佳培养基是MS+TDZ 2 mg·L-1+NAA 1 mg·L-1。红芽芋脱毒苗不定芽生根的最佳培养基是1/2MS+NAA 0.5 mg·L-1+PP333 0.5 mg·L-1。红芽芋再生苗最好的移栽基质为发酵后的腐锯木屑。红芽芋脱毒苗球茎愈伤组织再生苗移栽时最佳的PP333浓度为20~50 mg·L-1。本试验成功建立了红芽芋脱毒苗球茎愈伤组织的再生体系,为红芽芋脱毒苗转基因的研究和种质创新奠定了基础。  相似文献

Neutral glycolipids from the brain of a patient with Fucosidosis were analyzed and two complex glycolipids containing five and eight sugars were isolated from the cortical grey matter. These two glycolipids reacted with antibodies recognizing the SSEA-1 [Lex(X)] carbohydrate determinant. SSEA-1 glycolipids are normally expressed in human embryonic brain but are found in only small amounts in postnatal human brain. The accumulation of the two SSEA-1 glycolipids in Fucosidosis brain thus represents a defect which affects the normal developmentally regulated decrease in postnatal, expression of these glycolipids, and may be a contributing factor in the abnormal brain development associated with the disease. Komárek has recently reviewed the various species assigned to the green algal genusNeochloris Starr (Chlorococcales, Chlorococcaceae) and removed those with uninucleate vegetative cells to a new genus,Ettlia. Watanabe & Floyd, unaware ofKomárek's work, also reviewed the species ofNeochloris and distributed them among three genera—Neochloris, Chlorococcopsis gen. nov., andParietochloris gen. nov.—on the basis of details of the covering of the zoospore and the arrangement of the basal bodies of the flagellar apparatus. This paper reconciles these two treatments and makes additional recommendations at the ranks of genus, family, order, and class.  相似文献

Two burgeoning research trends are helping to reconstruct the evolution of the Hox cluster with greater detail and clarity. First, Hox genes are being studied in a broader phylogenetic sampling of taxa: the past year has witnessed important new data from teleost fishes, onychophorans, myriapods, polychaetes, glossiphoniid leeches, ribbon worms, and sea anemones. Second, commonly accepted notions of animal relationships are being challenged by alternative phylogenetic hypotheses that are causing us to rethink the evolutionary relationships of important metazoan lineages, especially arthropods, annelids, nematodes, and platyhelminthes.  相似文献

The cardinalfishes (Apogonidae) are a diverse clade of small, mostly reef-dwelling fishes, for which a variety of morphological data have not yielded a consistent phylogeny. We use DNA sequence to hypothesize phylogenetic relationships within Apogonidae and among apogonids and other acanthomorph families, to examine patterns of evolution including the distribution of a visceral bioluminescence system. In conformance with previous studies, Apogonidae is placed in a clade with Pempheridae, Kurtidae, Leiognathidae, and Gobioidei. The apogonid genus Pseudamia is recovered outside the remainder of the family, not as sister to the superficially similar genus Gymnapogon. Species sampled from the Caribbean and Western Atlantic (Phaeoptyx, Astrapogon, and some Apogon species) form a clade, as do the larger-bodied Glossamia and Cheilodipterus. Incidence of visceral bioluminescence is found scattered throughout the phylogeny, independently for each group in which it is present. Examination of the fine structure of the visceral bioluminescence system through histology shows that light organs exhibit a range of morphologies, with some composed of complex masses of tubules (Siphamia, Pempheris, Parapriacanthus) and others lacking tubules but containing chambers formed by folds of the visceral epithelium (Acropoma, Archamia, Jaydia, and Rhabdamia). Light organs in Siphamia, Acropoma, Pempheris and Parapriacanthus are distinct from but connected to the gut; those in Archamia, Jaydia, and Rhabdamia are simply portions of the intestinal tract, and are little differentiated from the surrounding tissues. The presence or absence of symbiotic luminescent bacteria does not correlate with light organ structure; the tubular light organs of Siphamia and chambered tubes of Acropoma house bacteria, those in Pempheridae and the other Apogonidae do not.  相似文献

Functionally active preparations of Na+,K+-ATPase isozymes from calf brain that contain catalytic subunits of three types (1, 2, and 3) were obtained using two approaches: a selective removal of contaminating proteins by the Jorgensen method and a selective solubilization of the enzyme with subsequent reconstitution of their membrane structure by the Esmann method. The ouabain inhibition constants were determined for the isozymes. The real isozyme composition of the Na+ pump from the grey matter containing glial cells and the brain stem containing neurons was determined. The plasma membranes of glial cells were shown to contain mainly Na+,K+-ATPase of the 11 type and minor amounts of isozymes of the 22(1) and the 31(2) type. The axolemma contains 21 and 31 isozymes. A carbohydrate analysis indicated that 11 enzyme preparations from the brain grey matter substantially differ from the renal enzymes of the same composition in the glycosylation of the 1 isoform. An enhanced sensitivity of the 3 catalytic subunit of Na+,K+-ATPase from neurons to endogenous proteolysis was found. A point of specific proteolysis in the amino acid sequence PNDNR492 Y493 was localized (residue numbering is that of the human 3 subunit). This sequence corresponds to one of the regions of the greatest variability in 1-, 2-, 3-, and 4-subunits, but at the same time, it is characteristic of the 3 isoforms of various species. The presence of the 3 isoform of tubulin (cytoskeletal protein) was found for the first time in the high-molecular-mass Na+,K+-ATPase 31 isozyme complex isolated from the axolemma of brain stem neurons, and its binding to the 3 catalytic subunit was shown.  相似文献

The mature embryo of seven species belonging to five genera of Indian bamboos is described. In all these the basic pattern of embryo organisation is same: the scutellar and coleoptilar bundles are not separated by an internode, the epiblast is absent, the lower portion of the scutellum and the coleorhiza are separated by a cleft and the margins of embryonic leaves overlap. The features unique to fleshy fruited bamboos are: presence of a massive scutellum, the juxtaposition of plumule and radicle and the occurrence of a bud in the axil of the coleoptile. The fleshy fruit bearing bamboos should be classified into one group, the tribeMelocanneae. Evidence is provided to recognise additional groups in the subfamilyBambusoideae.  相似文献

A molecular phylogeny of Hebeloma species from Europe   总被引:2,自引:1,他引:1  
In order to widen the scope of existing phylogenies of the ectomycorrhizal agaric genus Hebeloma a total of 53 new rDNA ITS sequences from that genus was generated, augmented by sequences retrieved from GenBank, and analysed using Bayesian, strict consensus and neighbour joining methods. The lignicolous Hebelomina neerlandica, Gymnopilus penetrans, and two species of Galerina served as outgroup taxa. Anamika indica, as well as representatives of the genera Hymenogaster and Naucoria, were included to test the monophyly of Hebeloma, which is confirmed by the results. Hebeloma, Naucoria, Hymenogaster and Anamika indica cluster in a strongly supported monophyletic hebelomatoid clade. All trees largely reflect the current infrageneric classification within Hebeloma, and divide the genus into mostly well-supported monophyletic groups surrounding H. crustuliniforme, H. velutipes, H. sacchariolens, H. sinapizans, and H. radicosum, with H. sarcophyllum being shown at an independent position; however this is not well supported. The section Indusiata divides with strong support into three groups, the position of the pleurocystidiate Hebeloma cistophilum suggests the possible existence of a third subsection within sect. Indusiata. Subsection Sacchariolentia is raised to the rank of section.  相似文献

The Drosophila Pax6 homolog twin of eyeless (toy) is so far the first zygotically expressed gene involved in eye morphogenesis in Drosophila. The study of its expression during embryogenesis is therefore informative of the initial events of eye development in Drosophila. We have analyzed how the initial expression domain of toy at cellular blastoderm is regulated. We show that the three maternal patterning systems active in the cephalic region (the anterior, terminal and dorsal-ventral systems) cooperate with zygotically activated gap genes to shape the initial expression domain of toy. Whereas Bicoid, Dorsal and Torso signaling synergistically act as activators, Hunchback, Knirps and Decapentaplegic act as repressors.  相似文献

Scirtothrips perseae Nakahara was discovered attacking avocados in California, USA, in 1996. Host plant surveys in California indicated that S. perseae has a highly restricted host range with larvae being found only on avocados, while adults were collected from 11 different plant species. As part of a management program for this pest, a “classical” biological control program was initiated and foreign exploration was conducted to delineate the home range of S. perseae, to survey for associated natural enemies and inventory other species of phytophagous thrips on avocados grown in Mexico, Guatemala, Costa Rica, the Dominican Republic, Trinidad, and Brazil. Foreign exploration efforts indicate that S. perseae occurs on avocados grown at high altitudes (>1500 m) from Uruapan in Mexico south to areas around Guatemala City in Guatemala. In Costa Rica, S. perseae is replaced by an undescribed congener as the dominant phytophagous thrips on avocados grown at high altitudes (>1300 m). No species of Scirtothrips were found on avocados in the Dominican Republic, Trinidad, or Brazil. In total, 2136 phytophagous thrips were collected and identified, representing over 47 identified species from at least 19 genera. The significance of these species records is discussed. Of collected material 4% were potential thrips biological control agents. Natural enemies were dominated by six genera of predatory thrips (Aeolothrips, Aleurodothrips, Franklinothrips, Leptothrips, Scolothrips, and Karnyothrips). One genus each of parasitoid (Ceranisus) and predatory mite (Balaustium) were found. Based on the results of our sampling techniques, prospects for the importation of thrips natural enemies for use in a “classical” biological control program in California against S. perseae are not promising.  相似文献

Intracellular cysteine aspartate-specific proteases (caspases) play both signaling and effector roles in realizing the program of cell death. Caspases function as proteolytic cascades unique for each cell type and signal triggering apoptosis. All parts of the proteolytic cascades are duplicated and controlled by feedback signals. Amplification cycles between pairs of caspases (the third and the sixth, the ninth and the third, the twelfth and the sixth, and others) help multiply the initial apoptotic signal. The presence of physiological inhibitors of apoptosis that directly interact with caspases creates a multilevel regulatory network of apoptosis in cell. The caspase proteolytic cascades are also regulated by sphingolipid secondary messengers, among them ceramide, sphingosine, and their phosphates. Moreover, an association of the caspase signaling with ubiquitin-dependent proteolysis is shown in cells. In particular, the use of extracellular activators and inhibitors of caspases allows irreversible activation of apoptosis in tumor cells or the prevention of apoptosis in cortical neurons under neurodegenerative diseases.  相似文献
