True truffles (Tuber sp.) that establish ectomycorrhizal symbiosis (ECM) with trees in the Mediterranean and temporal regions have species specific abilities to assimilate soil born elements. Suitable habitats are usually inhabited by few truffle species, while distinguishing their symbiotic potentials appeared very difficult. Two species that commonly inhabit riparian forests in Serbia are the most prized one, Tuber magnatum Pico (Piedmont white truffle) and not so highly valued Tuber brumale Vitt . In order to assess potential differences between their assimilation and accumulation abilities, the differences between contents of elements that may be the subjects of the symbiotic trade between the host plant and fungi were evaluated in accumulation target (ascocarps) and their source (the soil). Essential (K, Na, Ca, Mg, Fe, P, S, and Zn) and essential trace elements (Co, Cr, Cu, Mn, and Se) in truffles and soil samples were determined by means of inductively coupled plasma with optical emission spectrometry (ICP‐OES). Their concentrations (mg/kg) in ascocarps were in the range from 1.364±0.591 (Cr) to 10760.862±16.058 (K), while in soil ranged from 23.035±0.010 (Cr) to 20809.300±122.934 (Fe). Element accumulation potential (bioaccumulation factor) was calculated in the system truffle/soil. The statistical approaches were used for establishing the differences, while the possible differentiation between symbiotic potentials of two mycelia in the defined soil conditions was discussed.  相似文献

印度块菌(Tuber indicum)菌根促生细菌的研究   总被引:1,自引:1,他引:0  
【目的】筛选对印度块菌菌根量和菌根苗长势有促进作用的菌根促生细菌(Mycorrhization helper bacteria,MHB)。【方法】选择华山松为宿主植物,自块菌菌根根际土壤中分离得到的11种细菌为供试菌株,将印度块菌菌剂与不同浓度的细菌混合于特定基质中后接种于华山松上,并通过对印度块菌与华山松形成的菌根数、华山松的株高和地径三方面的统计与分析,确认MHB。【结果】Pseudomonas sp. JCM 5481 (P143)、Streptomyces sp. EN31 (S191)、Variovorax paradoxus (V633)在浓度为2.4×109 CFU/mL时对印度块菌菌根数、株高和地径均有极显著促进作用(P<0.01);Pseudomonas chlororaphis (P11)、Pseudomonas corrugate (P127)在浓度为0.8×109 CFU/mL时对印度块菌菌根数、株高和地径均有显著促进作用(P<0.05)。4种假单胞菌浓度梯度的设置显示了不同菌株适宜的浓度不同。【结论】实验获得5种MHB,并表明细菌浓度是获得MHB的一个重要影响因素。  相似文献

The goal of the research is to determine the effect of pretreated poultry manure and irrigation on the yield and quality of potatoes (Solanum tuberosum) grown in fields located on the southern steppe of the Republic of Bashkortostan. Field experiments were repeated every three years. During vegetation, potato leaves and tubers were studied at the full blossom, leaves’ decay, and potato harvest. The results showed that applying pretreated poultry manure at the rate of 120 t/ha favored higher tuber weight both on the rainfed and irrigated plots (from 0.23 to 0.82 kg/plant and from 0.24 to 1.02 kg/plant, respectively). On the rainfed and irrigated plots where poultry mature was not applied (control fields) the tubers’ weight ranged from 0.08 to 0.31 kg/plant and from 0.16 to 0.50 kg/plant, respectively. Upon application of 40 t/ha of manure, under irrigation, the highest marketable value of tubers was 78%. On the rainfed plots the same value was 72% when applying poultry manure at a dose of 120 t/ha. Soil moisture monitoring showed that the potatoes did not get adequate water during the growing season. When the soil moisture on irrigated plots was 70% less than its minimum water capacity, potato plantings were watered.  相似文献

For the first time, a fed-batch fermentation process of Tuber melanosporum was developed for the efficient production of bioactive mycelia and Tuber polysaccharides. Each 1.67 g/L of peptone and 8.33 g/L of yeast extract were added on day 3, 6, and 9, respectively, and sucrose was fed to maintain its concentration around 35–5 g/L when its residual level decreased to 10–5 g/L. Then, the maximal biomass, the production of extracellular polysaccharides (EPS) and intracellular polysaccharides (IPS) reached 53.72 ± 2.57 g DW/L, 7.09 ± 0.62 and 4.43 ± 0.21 g/L, respectively. Compared with the batch culture conducted in the enriched medium, the biomass, the production of EPS and IPS were enhanced by 55.8%, 222.3% and 103.2%, respectively. Not only the cell density but also the production of EPS and IPS were the highest ever reported in truffle fermentation, and the biomass was also the highest as ever reported in mushroom fermentation.  相似文献

Starch from tubers potato (Solanum tuberosum), taro (Alocassia indica), new cocoyam (Xanthosoma sagitifolium), true yam (Dioscorea alata), and root cassava, (Manihot esculenta) crops was isolated and its morphology, composition and physicochemical properties were investigated before and after heat–moisture treatment (HMT) (100 °C, for 10 h at a moisture content of 30%). Native starch granules were round to oval to polygonal with smooth surfaces. The granule size (diameter) ranged from 3.0 to 110 μm.The total amylose content ranged from 22.4 to 29.3%, of which 10.1–15.5% was complexed by native lipid. The phosphorus content ranged from 0.01 to 0.1%. The X-ray pattern of potato and true yam was of the ‘B’-type. Whereas, that of new cocoyam and taro was of the ‘A’-type. Cassava exhibited a mixed ‘A+B’-type X-ray pattern. The relative crystallinity, swelling factor (SF), amylose leaching (AML), gelatinization temperature range and the enthalpy of gelatinization of the native starches ranged from 30 to 46, 22 to 54, 5 to 23%, 13 to 19 °C and 12 to 18 J/g, respectively. Susceptibility of native starches towards hydrolysis by 2.2N HCl and porcine pancreatic -amylase were 60–86% (after 12 days), and 4–62% (after 72 h), respectively. Retrogradation was most pronounced in the B-type starches. Granule morphology remained unchanged after HMT. The X-ray pattern of the B-type starches was altered (B→A+B) on HMT. However, that of the other starches remained unchanged. HMT decreased SF, AML, gelatinization enthalpy and susceptibility towards acid hydrolysis, but increased gelatinization temperatures and enzyme susceptibility. Extent of retrogradation and relative crystallinity decreased on HMT of true yam and potato starches, but remained unchanged in the other starches. The foregoing data showed that changes in physicochemical properties on HMT are influenced by the interplay of crystallite disruption, starch chain associations and disruption of double helices in the amorphous regions.  相似文献

对新近发现的块菌属一新种——攀枝花白块菌(Tuber panzhihuanense)子囊果中可培养细菌的多样性进行了研究。采用胰蛋白大豆培养基(TSA)对菌株进行分离。用毛细管电泳(HPCE)对所有获得的菌株的16S rDNA V3高变区进行筛选获得不同条带大小的菌株,对筛选出的菌株的16S rDNA进行测序,并进行细菌多样性分析和研究。结果显示,攀枝花块菌子囊果内可培养细菌在数量及种类上都表现出很高的多样性,所有细菌分属于5个门的11个属和20个种。在所分离到的变形菌门的细菌中,数量最多的菌株(4968%)属于γ Proteobacteria,其中假单胞菌属的Pseudomonas lurida为优势类群;其次为α Proteobacteria,占3742%,其中以固氮菌 Bradyrhizobium japonicum和Phyllobacterium spp.为优势类群。其余的菌株属于放线菌门(Actinobacteria) (322%)和厚壁菌门(Firmicutes) (774%),厚壁菌门中以芽孢杆菌属(Bacillus)为代表菌群。酸杆菌门中的Terriglobus roseus(194%)首次从块菌中分离获得。  相似文献

以印度块菌-云南松菌根际土壤细菌为研究对象,研究其种群组成和结构特征。(1)稀释平板法分离得到印度块菌-云南松菌根际土壤细菌的纯培养菌株,对菌株的16S rRNA序列测序分析,对测序的菌株数量和得到的OTUs数量绘制物种累积曲线,当物种累积曲线趋于平缓时,对OTUs进行系统发育分析,揭示可培养细菌的种群组成和结构特征。(2)对印度块菌-云南松菌根际土壤细菌16S rRNA基因的V3—V4区进行高通量测序,分析全部细菌类群的种群组成和结构特征。(1)分离得到菌根际可培养细菌793株,分属于3个属的61个OTUs,其中假单胞菌属(Pseudomonas)序列占总序列的86%,不动杆菌属(Acinetobacter)序列占总序列的9.8%,链霉菌属(Streptomyces)序列占总序列的6.5%。假单胞菌是印度块菌-云南松菌根际土壤可培养细菌的绝对优势类群。(2)高通量测序得到菌根际细菌序列8937条,分属于20个门、198属、2073个OTUs。隶属于变形菌门(Proteobacteria)、放线菌门(Actinobacteria)和酸杆菌门(Acidobacteria)的OTUs占总OTUs的65.9%,变形菌门、放线菌门和酸杆菌门细菌是印度块菌-云南松菌根际土壤细菌的优势细菌。隶属于黄杆菌属(Flavobacterium)、根瘤菌属(Rhizobium)和假黄色单胞菌属(Pseudoxanthomona)的OTUs占总OTUs的33%,黄杆菌属、根瘤菌属和假黄色单胞菌属细菌是印度块菌-云南松菌根际土壤细菌的优势属。印度块菌-云南松菌根际土壤可培养细菌多样性较低,假单胞菌属细菌占据绝对优势地位。印度块菌-云南松菌根际土壤细菌类群具有较高的多样性,物种种类丰富,优势菌群集中。  相似文献

Truffles are prized and nutrition‐rich edible hypogeous fungi. The aim of this study was a comprehensive investigation of chemical composition of Burgundy truffle (Tuber aestivum Vittad .). We tried to answer the question: what is the impact of the environment on the truffle quality. To know the nutritional value of Burgundy truffle we compared lipids, proteins, saccharides, polyphenolics, flavonoids, total sterols, ergosterol, volatile flavour and aroma compounds content in fruit bodies of the fungus collected in three different geographical regions, i.e., Poland, Slovakia, and Italy. A comparison of the above mentioned compounds is especially interesting due to environmental and climatic differences among the studied geographical regions. Results revealed that fruit bodies of Taestivum from Poland and Slovakia possessed nearly similar content of proteins, total sterols, and saccharides. The fruiting bodies from Italy contained significantly larger amounts of most of the investigated compounds. In turn, Polish specimens had higher content of lipids and polyphenolics than Slovak and Italian ones. We have found higher similarity of volatile compounds composition between Polish and Italian specimens than those of Polish and Slovak origin.  相似文献
